Abstract

Guru memiliki peran penting dalam mengembangkan pembelajaran yang kreatif, kontekstual, dan berwawasan lingkungan. Namun, pemanfaatan vegetasi lokal sebagai media pembelajaran dan produk kreatif masih belum optimal. Kegiatan pengabdian ini bertujuan meningkatkan keberdayaan guru MAN 1 Kepulauan Meranti melalui pelatihan ecoprint berbasis vegetasi lokal. Metode yang digunakan meliputi pemaparan materi menggunakan presentasi, demonstrasi, praktik langsung (hands-on training), diskusi interaktif, pemberian reward kepada peserta aktif, serta evaluasi menggunakan kuesioner pre-test dan post-test dengan skala Likert empat tingkat. Hasil kegiatan menunjukkan peningkatan pengetahuan dan keterampilan peserta dalam memahami konsep ecoprint, memilih bahan, melakukan perlakuan kain, menyusun motif, dan melakukan proses pengukusan. Seluruh peserta berhasil menghasilkan produk ecoprint berupa syal dengan memanfaatkan vegetasi lokal. Evaluasi juga menunjukkan meningkatnya kepercayaan diri peserta untuk menerapkan ecoprint dalam pembelajaran serta munculnya minat mengembangkan produk ecoprint sebagai peluang ekonomi kreatif. Tingkat kepuasan peserta terhadap pelaksanaan pelatihan tergolong sangat tinggi, ditunjukkan oleh 87% peserta menyatakan sangat puas. Kegiatan ini berhasil meningkatkan kompetensi, kreativitas, dan motivasi guru dalam memanfaatkan sumber daya lokal sebagai media pembelajaran inovatif sekaligus mendukung pengembangan kewirausahaan berbasis lingkungan.

Abstract

Hutan mangrove merupakan ekosistem pesisir yang kaya akan bahan organik dan mendukung pertumbuhan mikroorganisme penghasil enzim selulase. Salah satu kelompok mikroorganisme penting di lingkungan ini adalah bakteri selulolitik yang mampu mendegradasi selulosa. Penelitian ini bertujuan untuk isolasi dan karakterisasi bakteri selulolitik dari sedimen dan rizosfer tanaman alami mangrove di Desa Kelapapati, Kabupaten Bengkalis, Riau. Penelitian dilakukan melalui pengambilan sampel sedimen dan rizosfer, isolasi bakteri menggunakan medium Cellulose Congo Red Agar (CCRA), uji aktivitas selulolitik berdasarkan zona bening yang terbentuk di sekeliling koloni, dan karakterisasi makroskopis dan mikroskopis terhadap isolat yang menunjukkan aktivitas selulolitik. Sebanyak 36 isolat berhasil diisolasi, terdiri atas 21 isolat asal rizosfer dan 15 isolat asal sedimen. Hasil uji menunjukkan bahwa sebanyak 20 isolat konfirmasi membentuk zona bening dengan indeks selulolitik pada kategori tinggi sebanyak 13 isolat, kategori sedang sebanyak empat isolat, dan kategori rendah sebanyak tiga isolat. Isolat MAT 1.1 menunjukkan indeks selulolitik tertinggi mencapai 10,8±0,6 dan potensial sebagai agen biofertilizer. Sebagian besar isolat tergolong bakteri Gram negatif berbentuk bacil dan streptobacil. Lingkungan mangrove dengan pH 6, suhu rata-rata 30°C, dan salinitas 12,67 dS/m mendukung pertumbuhan bakteri selulolitik. Penelitian ini menunjukkan bahwa hutan mangrove Desa Kelapapati, Bengkalis, Riau merupakan sumber potensial bakteri selulolitik yang dapat dikembangkan sebagai agen biofertilizer.

Abstract

The purpose of this study was to determine the genetic diversity and heritability values of the morphological characters of several genotypes of soybean plants. The study used a completely randomized design (CRD) consisting of fourteen genotypes and three replications. The observed parameters were flowering age, harvesting age, plant height, number of nodes, crown width, stem diameter, number of fruitful pods per plant, total number of pods per plant, total number of seeds per plant, number of fruitful seeds per plant, number of seeds per pods, 100 seed weight, and seed weight per plant. Data were analyzed by analysis of variance using SAS version 9.0, followed by Duncan's multiple range test (DNMRT) at the 5% level. The results showed that the characters were flowering age, harvesting age, plant height, number of nodes, stem diameter, total number of pods per plant, number of seed pods per plant, total number of seeds per plant, number of seeds per plant, number of seeds per pod, weight seed per plant, has a wide genetic diversity. Crown width characteristics have narrow genetic diversity. All characters showed wide phenotypic diversity. The heritability results showed that the characteristics were flowering age, harvesting age, plant height, number of nodes, crown width, stem diameter, total number of pods per plant, number of seed pods per plant, total number of seeds per plant, number of seeds per plant, and number of seeds per plant. Pods and seed weight per plant were classified as having a high heritability. The characteristics of flowering age, harvesting age, plant height, number of nodes, stem diameter, total number of pods per plant, number of fruitful pods per plant, total number of seeds per plant, number of fruitful seeds per plant, number of seeds per pod, and seed weight per plant can be used as selection criteria because they have a wide genetic diversity and high heritability value.

Isolation and characterization of endophytic bacteria from sterile leaf of Acrostichum aureum from Bengkalis Island (Riau, Indonesia) and its potency for antidiabetic. Biodiversitas 24: 1580-1588. Endophytic bacteria are found in symbiotic plant tissues. These bacteria pose potential sources for the natural product due to their health benefit, e.g., for antidiabetic activity. The present study aimed to isolate and characterize endophytic bacteria isolates from the sterile leaf of sea fern (Acrostichum aureum Linn.) from Bengkalis Island (Riau Province), Indonesia. Furthermore, to investigate their potency as an antidiabetic agent. The characteristics of isolates bacteria were described based on colony morphology and biochemical tests. A total of six endophytic bacteria were isolated from the sterile leaf of Acrostichum aureum. Their characteristics vary in shape, color, and size. The phytochemical screening indicated the presence of alkaloids and saponin in all the isolates. The results exhibited that the extract of the sterile leaf of A. aureum also gave a similar phytochemical compound of isolate endophytic bacteria. The dendrogram constructed from morphological and biochemical characteristics showed the separation of D.SB 4.2 from other six bacteria strains due to the positive antibacterial activity for Escherichia coli ATCC35218, Staphylococcus aureus ATCC29213, Salmonella typhi ATCC14028, and Bacillus subtilis ATCC11774. Out of six isolates, only isolate D.SB 5.2 was selected for the antidiabetic test. The isolates showed positive results on the activity as a-amylase inhibitor (18.12%). This study provides important information on endophytic bacteria isolates on A. aureum and their potency for antidiabetic agents.

Nusantara Bioscience 10: 146-150. Despite the abundant amount of phosphorus (P) in the soil, P uptake by the plants is very limited. In acidic soil, phosphorus (P) is bound to aluminum (Al) and iron (Fe), whereas in the alkaline soil, phosphorus (P) is bound to calcium (Ca). The improvement of efficiency and availability of P to plants can be made by utilizing a group of solubilizing phosphate microorganisms. Potency test to investigate the P solubility by Phosphate Solubilizing Bacteria (PSB) has been conducted by isolating the bacteria from peat soil at Biosphere Reserves of Giam Siak Kecil Bukit Batu, Riau, Indonesia. The semi-quantitative test revealed that the PSB were able to dissolve Ca3 (PO4)2, FePo4 and phosphate rock. However, the adaptation ability and potency of PSB from indigenous Riau peat soil inoculated into soybean (Glycine max L. Merr) plants in the mineral soil have not yet been investigated. The present study was carried out from March to June 2015 on the alluvial soil in Babussalam Village, Rambah Sub-district, Rokan Hulu District, Riau. The aim of this study was to determine the effect of PSB inoculation on bacterial population and phosphatase activity. The study also aimed to determine the available P and P uptake and their impact on soybean growth and production. The study employed a factorial experiment laid out in a completely randomized design (CRD) consisted of two factors, i.e., soil treatment and PSB. The first factor comprised of two levels, i.e., T0: non-sterilized soil, T1: sterilized soil. The second factor consisted of 4 levels, i.e., B0: without PSB inoculation, B1: inoculation using 2 isolates of PSB (BB_UB6 and BB_K9), B2: inoculation using 3 isolates of PSB (BB_UB6, BB_K9 and BB_K2), and B3: inoculation using 4 isolates of PSB (BB_UB6, BB_K9, BB_K2, and BB_HS13). The results showed that inoculation of starter 3 had the highest phosphatase activity rate of 12.43 ?g p NP g-1 hour-1. The highest available P was produced by starter 2, while the P uptake on non-sterilized soil was higher than that on the sterilized soil at 2.63 mg plant-1. PSB inoculation and soil sterilization did not significantly affect the population of phosphate solubilizing bacteria.

Abstract

The development of biofertilizers based on indigenous microorganisms is an important strategy to sustainably enhance crop productivity. Mangrove ecosystems represent potential sources of bacteria with diverse metabolic capabilities, including phosphate-solubilizing bacteria (PSB). However, information regarding the multifunctional characteristics of PSB isolated from the Bengkalis mangrove ecosystem, Riau, Indonesia remains limited. This study aimed to evaluate the potential of 25 PSB from the Bengkalis mangrove ecosystem as biofertilizer candidates by assessing their cellulolytic activity, potassium-solubilizing ability, indole-3-acetic acid (IAA) production, nitrogen-fixing ability, plant safety test, and plant growth-promoting effects on maize (Zea mays L.). The results showed that 12 isolates exhibited cellulolytic activity, 10 isolates were capable of solubilizing potassium, 14 isolates showed nitrogen-fixing ability, and all isolates produced IAA at varying concentrations. Based on a scoring approach integrating multiple plant growth promoting traits, isolate SMK 2.1 showed the highest potential, followed by MA 2.3, SMB 2.1, and RA 1.2 as promising isolates. In the maize growth assay at 7 days after planting, isolate SMK 2.1 was identified as the most promising isolate, producing the highest fresh weight (0.94 ± 0.21 g), root length (7.72 ± 3.21 cm), and shoot length (19.50 ± 5.27 cm) among all treatments. In addition, all isolates showed negative results in the plant safety test. Although the evaluation was limited to the early growth stage, these findings demonstrate that PSB isolated from the Bengkalis mangrove ecosystem have considerable potential as biofertilizer candidates and represent valuable indigenous microbial resources for sustainable agriculture. Ekosistem mangrove berpotensi menjadi sumber bakteri dengan kemampuan beragam, termasuk bakteri pelarut fosfat (BPF). Namun, informasi mengenai karakter multifungsi BPF asal mangrove di Bengkalis, Riau masih terbatas. Penelitian ini bertujuan untuk mengevaluasi potensi 25 isolat BPF asal ekosistem mangrove Bengkalis sebagai kandidat agen biofertilizer melalui pengujian aktivitas selulolitik, pelarutan kalium, produksi hormon indole-3-acetic acid (IAA), kemampuan penambatan nitrogen, uji keamanan tanaman, serta potensinya dalam memacu pertumbuhan tanaman jagung (Zea mays L.). Hasil penelitian menunjukkan bahwa 12 isolat memiliki aktivitas selulolitik, 10 isolat mampu melarutkan kalium, 14 isolat mampu menambat nitrogen, dan seluruh isolat mampu menghasilkan hormon IAA dengan konsentrasi yang bervariasi. Berdasarkan sistem skoring yang mengintegrasikan berbagai karakter pemacu pertumbuhan tanaman, isolat SMK 2.1 memperoleh nilai tertinggi, diikuti isolat MA 2.3, SMB 2.1, dan RA 1.2 sebagai isolat potensial. Pada uji pemacuan pertumbuhan tanaman jagung umur 7 hari setelah tanam, isolat SMK 2.1 merupakan isolat paling potensial yang ditunjukkan oleh berat segar (0,94±0,21 g), panjang akar (7,72±3,21 cm), dan panjang tajuk (19,5±5,27 cm) yang lebih tinggi dibandingkan perlakuan lainnya. Seluruh isolat juga menunjukkan hasil negatif pada uji keamanan tanaman. Meskipun pengujian masih terbatas pada fase awal pertumbuhan, hasil ini menunjukkan bahwa BPF asal mangrove Bengkalis berpotensi sebagai kandidat biofertilizer dan sumber agen hayati lokal untuk pertanian berkelanjutan. Abstract

Microalgae inhabiting urban waters and peat-influenced aquatic systems experience variable environmental pressures, potentially leading to adaptive responses to conditions with fluctuating nutrient availability. This study aimed to identify microalgae cultured from several urban water bodies and peat waters in Pekanbaru and to evaluate their growth responses in culture media with different nutrient compositions using an image-based approach. Water samples were collected from multiple locations and cultured in modified BG-11 and BBM media. The growing microalgae were identified based on their morphological characteristics, while culture growth was qualitatively assessed using the mean pixel intensity (MPI) values of digital images. The study recorded 16 species from 10 microalgal genera belonging to the classes Cyanophyceae, Chlorophyceae, and Zygnematophyceae, exhibiting unicellular, colonial, and filamentous morphologies. Culture observations revealed changes in MPI values over the observation intervals, indicating increased microalgal growth rates in the growth media. These findings suggest that local microalgae from urban and peat waters possess a high tolerance to limited nutrient conditions and provide baseline information on the diversity and growth responses of indigenous microalgae in tropical urban and peat aquatic systems.
