Abstract

Research on the antioxidant activity of the local onion roots of Palu (Allium cepa Var Aggregatum L.) with various polarity solvents has been carried out. This study aims to determine the potential of Palu local shallot root extract as an antioxidant and determine the IC50 value of the Palu local shallot root antioxidant activity. The method applied in this research is maceration using n-hexane, ethyl acetate, and ethanol. The extracts from each solvent were tested for their antioxidant activity by the DPPH method. The results of the study showed that the IC50 value in n-hexane extract, ethyl acetate extract, and ethanol extract were 579.98 ppm, 518.28 ppm 315.83 ppm and as a comparison used ascorbic acid obtained IC50 of 53.69 ppm. Based on the results obtained, the best antioxidant activity is in polar solvents (ethanol extract) compared to other solvents. Abstract

Liquid smoke contains acid, carbonyl, and phenolic compounds, so it is very appropriate to be used as a preservative. Traditional smoking can be replaced with liquid smoke because it is safer and more practical. The study of the use of coconut shell liquid smoke has been used in skipjack fish to determine the effect of liquid smoke concentration and storage time on dissolved protein content and total bacteria in smoked fish. The quality of smoked fish was determined by two treatments, namely the variation in the concentration of liquid smoke (2, 3, 4, and 5%) and storage time (2, 4, 6, 8, and 10 days). The results showed that the best concentration of liquid smoke to produce smoked skipjack fish is 3% with a water content of 62.41% and dissolved protein content of 17.58%. The maximum storage time of smoked skipjack tuna is 6 days with a total plate count (TPC) value or total bacteria 1.8 x 105.

Abstract

Black sea cucumber is one type of sea cucumber whose protein content is quite high at 74-82%, not yet widely used by the community as a source of protein. One way to use it is to process the protein into a protein hydrolyzate using the bromelain enzyme which was isolated from pineapple stems. This study aims to determine the concentration of the bromelain enzyme extract and the optimum hydrolysis time to the value of the hydrolysis degree of the hydrolyzate of black sea cucumber protein. Protein hydrolysates are produced by variations of the crude bromelain concentration of 9%, 10%, 11%, and 12% (v/v) and hydrolysis time of 120, 180, 240, and 300 minutes. The results showed that the optimum degree of hydrolysis was obtained with the use of bromelain extract with a concentration of 12% with a value of 11.43% and a hydrolysis time of 240 minutes with a value of 19.17%.

Abstract

This research was conducted by making VCO-based liquid soap with the addition of ethanol extract from Moringa leaves (Moringa oleifera). The purpose of this research was to determine the mass ratio of ethanol extracts of Moringa leaves and Virgin Coconut Oil (VCO) which produces soap with the highest antibacterial activity against tested bacteria. The other purpose is to determine the effect of the mass ratio against the viscosity of liquid soap. The research design used was a Completely Randomized Design (CRD) with the independent variables being the mass ratio of Moringa leaf ethanol extract and VCO which consists of five levels of 0:75 (F1), 1.5:73.5 (F2), 3:72(F3), and 4.5:70.5(F4) (w/w). The soap produced was tested for antibacterial activity againts Staphylococcus aureus, Escherichia coli, and Staphylococcus epidermidis using agar diffusion method as well as viscosity tests. The results showed that the antibacterial activity of liquid soap tends to be higher with increasing levels of ethanol extract of Moringa leaves. The liquid soap formula with the ratio of Moringa leaf ethanol extract: VCO of 3:72 (w/w) (Formula F3) had relatively higher antibacterial activity against the three tested bacteria. The inhibition zones against Staphylococcus aureus, Escherichia coli, and Staphylococcus epidermidis were 22.302 mm, 33.230 mm, and 36.490 mm, respectively, while the viscosity value of liquid soap was 47.433 cP.

Abstract

Dangke is one of traditional food from Enrekang Province Sulawesi Selatan which is made from buffalo milk and enzimatically processed using papain from papaya’s gland secretion. Research on this local product as source of lactic acid bacteria (LAB) has been done. Counted 30 LAB was successfully isolated and 3 of them were potentially to be able to yield antimicrobial compound. Enterococcus faecium DU55 is one of LAB isolate available to be applied for producing bacteriocin compound through fermentation. Optimum condition of fermentation was specified by determining the highest antimicrobial activity generated by filtrate of fermentation result. Antimicrobial activity examination was carried out through diffusion agar method by measuring inhibition zone growth of pathogen bacterium Salmonella typhimurium FNCC 0050. Research results indicate that maximum bacteriocin compound production by BAL isolate E. faecium DU55 was obtained at condition of optimum fermentation at 30 °C during 42 hour using M1 medium with the same composition to medium MRS (Man Rogosa and Sharpe).Keyword: dangke, LAB isolate, bacteriocin, antimicrobial activity

Abstract

Desa Bale merupakan salah satu daerah yang terletak di Kabupaten Donggala, Sulawesi Tengah. Penduduk di Desa Bale umumnya sebagai petani kelapa. Pembuatan produk olahan kelapa yang inovatif masih sangat kurang di Desa Bale. Minuman isotonik dan susu coklat santan kelapa dapat menjadi produk olahan baru bagi masyarakat setempat. Tujuan kegiatan ini adalah untuk menambah keterampilan masyarakat di desa Bale dalam mengolah air kelapa tua dan santan kelapa menjadi produk yang menarik dan memiliki nilai jual yang menjanjikan. Metode yang digunakan pada kegiatan ini adalah penyuluhan yang diikuti dengan demonstrasi. Hasil dari kegiatan yang dilakukan adalah keterampilan masyarakat mitra di desa Bale bertambah dalam membuat minuman isotonik dan susu coklat santan kelapa dengan menggunakan metode yang sederhana dan murah. Hasil pengukuran tingkat kepuasan menunjukkan bahwa 80% peserta atau mitra sangat puas terhadap pelaksanaan kegiatan pengabdian. Masyarakat mitra siap untuk mengembangkan produk olahan hasil pelatihan dalam bentuk produk yang siap dipasarkan.

Abstract

The study of the potential of Gempol wood stem extract (Nauclea orientalis L.) with three solvent polarities has been exercised to determine the extract with a certain polarity and the smallest concentration that can inhibit the growth of bacteria Staphylococcus aureus and Escherichia coil. Extraction is performed using the maceration method with n-hexane, ethyl acetate, and ethanol as solvents. Furthermore, the potency in inhibiting the growth of bacteria was observed by the diffusion method with a range of extract concentrations from the largest to the smallest. The extract's ability to inhibit bacterial growth in the time range of up to 24 hours was measured using the turbidimetric method. The results showed that ethyl acetate and ethanol extracts were able to inhibit the growth of both bacteria in a wide concentration range (100 %– 5%), although n-hexane extracts showed the highest percentage in inhibiting both bacteria (15.27 mm and 16.89 mm, respectively). Turbidimetric assays showed the extract was stronger in inhibiting S. aureus compared to E. coli in the logarithmic phase range of 12 hours. This study showed that semipolar and polar extracts can inhibit the growth of bacteria with a wider concentration range compared to non-polar extracts despite having a higher inhibition capability.

Abstract

Coconut pulp wasted from processed coconut oil has a protein content of up to 18.20%. The potential coconut pulp protein can be processed in protein concentrate which has many benefits. This research was conducted to determine the effect of NaOH and (NH4)2SO4 concentrations in isolating protein and producing protein concentrates that have the highest yield and protein content. Isolation of coconut pulp protein was carried out using NaOH at several concentrations, namely 0, 0.2, 0.4, 0.6, 0.8, and 1 M, while the protein concentrate was prepared using the salting out method with a saturation level of the ammonium sulfate salt of 55%, 60%, and 65%. Analysis of protein content was carried out using the Kjeldahl method. The use of NaOH 0.2 M and (NH4)2SO4 with a saturation level of 60% was the chosen treatment with crude protein content and yield of 75.63% and 33.83%, respectively.
