Abstract

Infeksi cacing menjadi salah satu masalah kesehatan yang paling umum tersebar diseluruh dunia. Salah satu penyebab infeksi kecacingan adalah penularan Soil Tranmitted Helminth (STH) melalui tanah ke sayuran. Jenis cacing yang ditularkan melalui tanah yaitu Ascaris lumbricoides (cacing gelang), Trichuris trichiura (cacing cambuk), Ancylostoma duodenale dan Necator americanus (cacing tambang). Sayuran selada merupakan jenis sayuran yang kontak langsung dengan tanah sehingga kemungkinan besar lebih beresiko terkontaminasi oleh parasit. Tujuan penelitian ini adalah untuk mengidentifikasi telur STH sebelum dan sesudah pencucian pada sayuran selada yang dijual di pasar Anyar Kota Tangerang. Penelitian ini bersifat deskriptif. Sampel penelitian ini diambil dari pedagang sayuran dengan menggunakan teknik purposive sampling. Sampel penelitian berjumlah 10 sampel. Pengumpulan data dengan melakukan pemeriksaan sayuran menggunakan metode sedimentasi. Hasil penelitian dapat disimpulkan bahwa terdapat telur STH Ascaris lumbricoides (20%) pada sayuran selada sebelum pencucian dan tidak terdapat telur STH pada sayuran selada sesudah pencucian. Nilai signifikan dari Uji Wilcoxon terhadap sayur selada sebelum, dan sesudah pencucian adalah < 0,05 yaitu 0,000 maka maka Ha diterima atau Terdapat perbedaan Penelitian ini menunjukkan bahwa adanya perbandingan sayur selada sebelum pencucian Penelitian ini menunjukkan bahwa masih terdapat sayuran selada yang terkontaminasi telur STH. Dengan ini perlu dilakukan upaya-upaya dari berbagai pihak yang terkait untuk mencegah terjadinya kontaminasi telur STH.

Abstract

Tanaman Pare (Momordica charantia L) adalah salah satu tanaman yang paling banyak mendapat perhatian karena khasiat anti diabetiknya. Diperkirakan bahwa sepertiga dari pasien diabetes menggunakan beberapa bentuk pengobatan komplementer dan alternatif. Dilakukan pengujian toksisitas bertujuan untuk menentukan nilai LD50 pada pemberian ekstrak etanol buah dan biji pare menggunakan metode Thompson-Weil pada mencit putih jantan (Mus musculus) sebanyak 23 ekor dengan bb 25 gram. Pemberian ekstrak buah dan biji pare peroral dengan dosis awal 0,5 mg/kgBB. Adapun variasi dosis toksisitas yang yaitu 100 mg/kgbb, 50 mg/kgbb, 25 mg/kgbb dan 12,5 mg/kgbb serta aquadest sebagai kontrol. Mencit diamati secara individu selama 1 jam, 2 jam, 4 jam dan 24 jam setelah pemberian ekstrak dengan melihat jumlah hewan yang mati dan gejala toksik. Hasil penelitian pada ekstrak buah pare mencit mati pada dosis 100 mg/kgbb : 1 ekor (20%). Pada ekstrak biji pare mencit mati pada 100 mg /kgbb : 1 ekor (20%), dosis 50 mg/kgbb : 2 ekor (40%), dan pada dosis 12,5 mg/kgbb : 1 ekor (20%). Hal ini menunjukkan ekstrak buah dan biji pare termasuk kategori tidak toksik. Pemberian bahan uji ektrak tidak menimbulkan gejala toksik di mana semua tikus beraktifitas normal, sehingga buah dan biji pare aman dikonsumsi.

Abstract

Indonesia memiliki jenis tanaman obat yang banyak ragamnya, kelompok tanaman obat mencapai lebih dari 1000 jenis. Tanaman herbal adalah tumbuhan yang dapat dimanfaatkan untuk pengobatan tradisional, dan biasanya dikenal sebagai tumbuhan obat. Daun papaya, (Carica papaya) daun Sambiloto (Andrographis paniculata), daun Miana (Coleus scutellarioides L) mengandung beberapa zat aktif yang diduga dapat mempengaruhi/menghambat pertumbuhan koloni bakteri. Konsentrasi terendah dari ekstrak daun pepaya dan daun Sambiloto dan daun Miana yang memberikan antiadhesi (50, 25, 15%), dijadikan sebagai konsentrasi ekstrak yang digunakan sebagai antiadhesi (indikator daya tahan tubuh). Daya hambat adhesi ekstrak daun papaya dan daun Sambiloto dan daun Miana terhadap bakteri TBC-Resisten Rifampisin yang memiliki adhesin yang dihasilkan bakteri akan tergantung kepada lamanya variasi waktu kontak antara daun papaya, daun Sambiloto dan daun Miana dengan bakteri uji. Hasil konsentrasi antiadhesi dari masing masing ekstrak daun papaya, daun Sambiloto dan daun Miana digunakan untuk konsentrasi antibakteri antara ekstrak daun papaya, daun Sambiloto dan daun Miana terhadap bakteri uji, dengan pengamatan ada tidaknya pertumbuhan koloni bakteri uji. Berdasarkan hasil dari artikel yang telah direview terlihat bahwa ekstrak daun pepaya, daun sambiloti dan daun miana dapat berpotensi dalam menghasilkan senyawa antibakteri. Hal ini dapat disebabkan komponen senyawa metabolik yang di miliki oleh tanaman tersebut. Kandungan senyawa metabolit sekunder telah terbukti bekerja sebagai derivat antikanker, antibakteri dan antioksidan, antara lain adalah golongan alkaloid, tanin, golongan polifenol dan turunanya.

Abstract

The use of traditional medicine as an option for treatment is an alternative that is in great demand because traditional medicine has been proven to be relatively safe if used correctly and with the right indications and rarely causes side effects. This is one of the factors for people to use natural ingredients or traditional medicine as an alternative treatment. One of these efforts is to explore and find antioxidant compounds such as beta carotene, astasantine, alkaloids and phenols in plants. To take bioactive compounds directly from the plant, a lot of biomass or parts of the plant are needed, so to make the method of obtaining these bioactive compounds more efficient, specific endophytic microbes obtained from the inside of the plant are used which are expected to be able to produce a number of bioactive compounds from the plant. Endophytic microbes consist of bacteria, fungi and actinomycetes which have the potential to produce bioactive compounds such as those produced by plants without causing damage to the plant. The aim of this research was to examine the antagonism ability of the Namnam leaf endophytic fungus (Cynometra cauliflora L.) against pathogenic fungi. The stages of the research method are identification of endophytic fungi macroscopically and microscopically and inoculation of endophytic fungi into PDA media, followed by preparation of test pathogenic fungi, the pathogenic fungi tested are Candida albicans and Malassezia furfur, antagonism test by inoculating pathogenic fungi into PDA media and After that, place the endophytic fungi on media that has been inoculated with pathogenic fungi, incubate at room temperature for 3-5 days and measure using a ruler. The results of macroscopic and microscopic identification of endophytic fungi, isolate code P1-1, were Penicillium sp, while isolate codes P4-1 and P5-1 were identified as Mucor sp. Based on the antagonist test that was carried out, it was seen that endophytic fungi grew in the test media but did not provide significant inhibition to the growth of the test fungi, namely Candida albicans and Malassezia furfur.

Abstract

Pemantapan Mutu Laboratorium bertujuan menjamin ketelitian dan ketepatan hasil pemeriksaan laboratorium. Di bidang hematologi, pemeriksaan hemoglobin merupakan beban kerja utama, maka diperlukan suatu bahan kontrol yang selalu tersedia. Salah satu aspek pemantapan kualitas laboratorium adalah penggunaaan bahan kontrol sebagai pemantauan kinerja pemeriksaan. Di negara berkembang, ketidaktersediaan dan harga bahan kontrol komersial menjadi masalah tersendiri. Bahan kontrol buatan sendiri dapat menjadi alternatif untuk memangkas biaya kendali mutu pemerikasaan laboratorium. Bahan kontrol buatan sendiri berasal dari 2 kantong darah yang dibuat menjadi hemolisat dengan kadar hemoglobin rendah, normal, dan tinggi, kemudian masing-masing ditentukan kadarnya menggunakan rerata ± standar deviasi. Selanjutnya, dilakukan uji homogenitas dan uji stabilitas untuk mengetahui apakah hemolisat (kontrol Hb) yang telah dibuat stabil dan homogen sehingga layak dijadikan bahan kontrol Hemoglobin. Pada bahan kontrol buatan sendiri kadar Hb rendah yaitu 5,4-5,6 g/dL, Hb normal 12,48-12,60, dan Hb tinggi 16,27-16,40 sedangkan pada kontrol Hb yang berasal dari pabrikan, kadar Hb rendah 5,2-5,8 g/dL, Hb normal 12,2-13,2 g/dL, dan Hb tinggi 15,9-17,3 g/dL. Uji homogenitas dan stabilitas menunjukan bahwa baik hemolisat maupun pabrikan memberikan hasil uji homogen dan stabil. Perbedaan bermakna terdapat pada nilai homogenitas dan stabilitas Hb rendah antara Hemolisat dan pabrik (Sig.000) sedangkan nilai homogenitas dan stabilitas Hb normal dan tinggi pada hemolisat dan pabrik tidak terdapat perbedaan yang bermakna. Bahan kontrol buatan sendiri dengan kadar Hb rendah, normal, dan tinggi didapatkan homogen dan stabil. Uji t independen menunjukan adanya perbedaan bermakna kadar Hb rendah antara hemolisat dan pabrikan.

Abstract

Indonesia is known as a mega biodiversity country rich in biodiversity, animals and their ecosystems. The parts of the binahong plant (Andredera cordifolia) that can be utilized come from flowers, leaves, stems, roots, and tubers that have metabolite compounds such as flavonoids, tannins, alkaloids, terpenoids and saponins. This can be used as a basis for utilizing binahong leaves from which endophytic fungi are taken to determine the potential for secondary metabolites from endophytic fungi in binahong leaves by means of isolation, identification and antibacterial potential against Staphylococcus aureus bacteria. The purpose of the study was to determine the presence or absence of endophytic fungi in binahong plants and to determine their potential as antibacterials in endophytic fungi isolated from binahong plants against Staphylococcus aureus. The methods in this study were isolation, purification of endophytic fungi, preparation of Staphylococcus aureus test bacteria, antibacterial test using the agar diffusion method. The results showed that there were 5 isolates of endophytic fungi, with isolate codes AK. H, AK. M, BT 1.1, BT 2.1, and DA 1, microscopically the fungi obtained were Penicillium sp, and Mucor sp. And have the ability to inhibit the growth of Staphylococcus aureus bacteria included in the strong category, with an average inhibition zone of 20 mm, 20.5 mm, 15.5 mm, 21 mm, 22.5 mm respectively. The conclusion of this study is that endophytic fungal isolates in binahong plants have the potential as antibacterials against Staphylococcus aureus.

Abstract

The use of traditional medicine is in great demand because it is relatively safe and rarely causes side effects. Bioactive compounds in plants generally require large biomass to be extracted, so endophytic microbes, especially actinomycetes, are used as an alternative source because they are able to produce secondary metabolites without damaging the host plant. This study aims to isolate and test the antibacterial activity of endophytic actinomycetes from turmeric and galangal plants against Staphylococcus aureus and Escherichia coli. Actinomycete isolation was carried out using Humic Vitamin Agar (HVA) media, and purification using ISP 4 media and continued with antibacterial activity tests. The results of the research showed that of the 26 isolates of endophytic actinomycetes from turmeric and galangal plants that had been isolated, there were 15 isolates that had the ability to inhibit the growth of Staphylococcus aureus and Escherichia coli bacteria. The diameter of the inhibition zone against Staphylococcus aureus ranged from 7.14-14.16 mm, while against Escherichia coli it ranged from 8.11-18.17 mm, with isolate DK4 showing the highest antibacterial activity. These results indicate that endophytic actinomycetes from turmeric and galangal plants have the potential to produce secondary metabolites that are antibacterial.

Abstract

Infectious diseases caused by Staphylococcus aureus are one of the health problems occurring in Indonesia. Infectious diseases can be treated with antibiotics. However, antibiotics used without proper prescription can trigger antibiotic resistance. To overcome antibiotic resistance, a combination of natural ingredients such as kirinyuh leaves (Chromolaena odorata L.) and mango leaves (Mangifera indica L.) can be used. This study aims to determine the inhibitory activity and effective concentration of the combination of mango leaves extract (Mangifera indica L.) and kirinyuh leaves  extract (Chromolaena odorata L.) as antibacterial agents against Staphylococcus aureus. The methods used in this study included maceration to prepare mango leaves and kirinyuh leaves extract, phytochemical screening to identify the presence of secondary metabolites in the kirinyuh leaves and mango leaves extract, and the disk diffusion method for antibacterial testing. The results showed inhibitory activity at all concentrations due to the presence of secondary metabolites in the kirinyuh leaves and mango leaves extracts. At a 1:3 concentration, an inhibition zone of 18 mm was obtained; at a 1:1 concentration, an inhibition zone of 19 mm was obtained; at a 3:1 concentration, an inhibition zone of 20 mm was obtained. The effective concentration of the combination as an antibacterial agent is the 3:1 concentration. The results of this study fall into the strong category, indicating that the combination of mango leaves and kirinyuh leaves extracts has antibacterial efficacy against Staphylococcus aureus.

Abstract

Antibiotik merupakan obat yang paling banyak digunakan pada infeksi yang disebabkan oleh bakteri. Penggunaan antibiotik tidak luput dari penggunaan yang tidak rasional. ESBL adalah enzim yang dapat menghidrolisis dan menonaktifkan antibiotik β-laktam spektrum luas, seperti penisilin (ampisilin dan amoksisilin), sefalosporin generasi ketiga (cefotaxime, ceftazidime, dan ceftriaxone), dan monobactam (aztreonam). Enterobacteriaceae, seperti Escherichia coli, Klebsiella, Salmonella merupakan bakteri yang mendominasi ESBL. Di Indonesia, prevalensi infeksi bakteri penghasil ESBL mencapai 65%. Peneliti bermaksud memberikan informasi kepada rumah sakit tentang keberadaan ESBL agar pihak rumah sakit dapat mengambil tindakan untuk mengurangi penggunaan antibiotik yang tidak rasional dengan memberikan informasi tentang ESBL yang dihasilkan oleh bakteri E.coli yang terdapat di RSUD Tangerang. Rancangan penelitian menggunakan deskriptif dengan pendekatan uji laboratorium, untuk uji resistensi dan molekuler untuk skrining ESBL. Hasil penelitian ini menemukan 5 isolat ESBL yang dikonfirmasi secara fenotipe dengan metode double-disk diffusion test (DDST), dan secara genetik ditemukan 2 isolat yang memiliki gen TEM dan SHV dari 5 isolat E. coli yang berasal dari pasien dengan ESBL, berjenis kelamin laki-laki dan orang) dan perempuan (1 orang), dari bangsal ICU (3 orang) dan ruang Dahlia (2 orang). Lima isolat tersebut berasal dari sampel jaringan (3 buah), feses (1 buah), dan pus (1 buah).

Abstract

Background: Typhoid fever is a systemic infectious disease caused by Salmonella typhi and remains a public health problem due to its high incidence rate and the increasing antibiotic resistance. Objectives: This study aimed to determine the antibacterial activity of endophytic fungi isolated from turmeric (Curcuma longa Linn.) against Salmonella typhi. Methods: This study employed a descriptive exploratory research design, involving the isolation of endophytic fungi from the leaves, stems, and rhizomes of turmeric plants using Potato Dextrose Agar (PDA) supplemented with chloramphenicol, followed by the identification of the isolates based on their macroscopic and microscopic characteristics. Furthermore, antibacterial activity was evaluated using the diffusion method on Mueller Hinton Agar (MHA). Results: Four endophytic fungal isolates were obtained, consisting of three isolates from the stem (B1, B2, and B3) and one isolate from the rhizome (R). Isolates B1 and B2 were identified as Acremonium sp., B3 as Mucor sp., and R as Aspergillus sp. Antibacterial activity testing against Salmonella typhi showed that all isolates exhibited very strong activity. The mean inhibition zone diameters were 29 mm for isolates B1 and B2, 27 mm for isolate B3, and 22 mm for isolate R. Meanwhile, the positive control using chloramphenicol discs showed an inhibition zone diameter of 30.5 mm. Conclusions: This study provides data on the diversity of endophytic fungi successfully cultured from various parts of the turmeric plant and demonstrates strong antibacterial activity against Salmonella typhi, thereby contributing to the enrichment of scientific knowledge regarding the potential of endophytic fungi as alternative sources of antimicrobial agents.
