Abstract

The aims of this study was to isolate and select Bacillus from the gut of catfish as probiotic candidates in the fish feed production. Isolation was conducted by heating samples at 80 °C for 10-15 minutes using Triptone Soy Agar (TSA) media which have been added with 1% skim milk for proteolytic activity and 1% starch for amylolytic activity. Selection was conducted based on pathogenicity test, antibiotic susceptibility test and total suspended solids. Isolate that have ability to degrade feed would be made the growth curves, analysis of protease and amilase activites and also combination of bacteria isolate with feed. Selected isolates as candidate probiotic were identified furthermore using 6S-rRNA gene. Among 16 isolates, there were 7 isolates that have gamma hemolytic activity (PTB 1.1, PTB 1.2, PTB 1.4, PTB 1.7, STB 1.6, STB 1.1 and STB 2.1). Antibiotic susceptibility test showed that 3 isolates were sensitive to the tested antibiotics (PTB 1.4, PTB 1.7 and STB 1.6). These three selected isolates were tested for their ability to degrade fish feed. PTB 1.4 isolate was able to degrade the feed with the smallest residue on the filter paper (0.0068 g). PTB 1.4 isolate also has proteolytic and amylolytic index of 0.61 and 0.60, respectively. Amylase activity of PTB 1.4 isolate added with 1.2% feed reached the highest peak in 120-hour of observation time (0.399 µ/mL) and the highest protease activity was in 72-hour of observation time (6.595  µ/mL). PTB 1.4 isolate has the ability to degrade the feed with the amount of 106 CFU/mL inoculum. Based on 16S-rRNA gene sequences isolate PTB 1.4 was 99% homolog with Bacillus megaterium. Isolation and selection of probiotic candidate from Clarias sp. get PTB 1.4 was a best isolate that there were not pathogenic, sensitive to antibiotic test, had protease and amilase activities. Abstract

Based on the raw materials, inasua consists of two types namely inasua with sap and inasua without sap. Research of inasua with sap has never been done and considered as the novelty of this research. The sensory characteristics and shelf life of two types of inasua were different. The research aims to analyze the microbiological and physicochemical characteristics of two types of inasua during fermentation. The microbiological analyzes include the total number of bacteria and lactic acid bacteria, while physicochemical analyzes include temperature, pH, water activity, proximate analysis, salt, alcohol, histamine, amino acids and fatty acids contents. The total number of bacteria and lactic acid bacteria has decreased during fermentation. At the end of the fermentation the total number of bacteria and lactic acid bacteria inasua with sap were 3.2x107 CFU/g and 3.0x107 CFU/g, while inasua without sap were 5.4x105CFU/g and 3.5x105 CFU/g, respectively. The moisture, protein, alcohol contents and water activity decreased, otherwise the salt, fat, ash, amino acids, and fatty acids contents increased during fermentation. Generally, microbiological and physicochemical characteristics of inasua with sap was better than inasua without sap. The results of this research to improve the quality of this fermentation product in the future.

Abstract

Bawang hutan Eleutherine bulbosa (Mill.) Urb.merupakan tanaman obat yang memiliki kandungan oligosakarida, berpotensi sebagai prebiotik yang mampu meningkatkan kesehatan usus udang vaname (Litopenaeus vannamei). Tujuan dari penelitian ini adalah menguji efek dari ekstrak bawang hutan dengan konsentrasi berbeda terhadap perubahan pH media pada stimulasi pertumbuhan probiotik secara in vitro. Konsentrasi ekstrak bawang hutan terdiri dari : 10, 5, 2.5, 1.25, 0.625, 0.313, 0.156 mg/mL. Bakteri probiotik yang digunakan adalah Pseudoalteromonas piscicida 1Ub dan Bacillus sp. NP5,sedangkan bakteri enterik menggunakan Vibrio parahaemolyticus. Waktu inkubasi selama 24 jam dengan pengamatan warna dan pH media dilakukan pada jam ke-0, 14, 18, dan 24. Hasil penelitian menunjukkan bahwa setelah inkubasi selama 24 jam terjadi perubahan warna dan pH media. Nilai pH pada perlakuan probiotik lebih rendah dibandingkan dengan bakteri enterik. Nilai pH pada P. piscicida 1Ub lebih rendah dibandingkan dengan Bacillus sp. NP5. Perubahan pH yang lebih rendah menandakan probiotik mampu memanfaatkan oligosakarida yang ada pada ekstrak bawang hutan.Kata kunci : Eleutherine bulbosa, oligosakarida, pH, prebiotik, probiotik.

Abstract

Application of bioflocs technology and probiotics has improved water quality and production of Pacific white shrimp (Litopenaeus vannamei) culture. This experiment was to verify the effect of probiotic bacteria addition and different carbon:nitrogen (C:N) ratio on water quality and performance of Pacific white shrimp nursery culture. Nursery culture was carried out for 25 days in an aquarium under laboratory condition with stock density of one Post-Larvae (PL) (poslarval) per liter (24 PL/aquarium) of PL16 shrimp. Different C:N ratio resulted a significant difference on shrimp production performance. Treatment of 10 C:N ratio demonstrated the best shrimp growth (20.37 + 0.48% per day in weight and 6.05 + 0.41% per day in length), harvesting yield (1180 + 62 g/m3) and feed efficiency (121 + 6%). There was however no significant difference observed between treatments in water quality.

Abstract

This study evaluated the effect of biofloc technology (BFT) application on water quality and production performance of red tilapia Oreochromis sp. at different stocking densities. Three different fish densities were applied, i.e. 25, 50, and 100 fish/m3, and for each density there were Control (without external carbon input) and BFT treatments. Mixed sex red tilapia with an initial average body weight 77.89 + 3.71 g was cultured in 3 m3 concrete tanks for 14 weeks. Molasses was added on BFT treatments as the organic carbon source at a C/N ratio of 15. Control treatments of each density tested showed more fluctuated water quality parameters throughout the experimental period. The highest TAN and nitrite-nitrogen were observed in control treatment at a stocking density of 100 fish/m3 (3.97 mg TAN/L and 9.29 mg NO2-N/L, respectively). The highest total yield was observed in control treatment at the highest density treatment (43.50 kg), whereas the highest survival was obtained by BFT treatment at a density of 25 fish/m3 (97.78 + 0.77%). Total feed used in BFT was lower than that of control treatments in particular at 50 fish/m3 density (P < 0.05) suggesting that biofloc could be continuously harvested by the fish as other source of food.

Abstract

This study evaluated the effects on viral immune responses and bacterial co-infection, of different feeding frequencies of a synbiotic supplemented diet given to Pacific white shrimp (L. vannamei). A synbiotic-supplemented diet was formulated from probiotic Vibrio alginolitycus SKT-bR and prebiotics from sweet potato (Ipomoea batatas L.) oligosaccharide. Pacific white shrimp were fed the synbiotic diet at different frequencies, i.e. daily (P1), twice a week (P2), and once a week (P3) for a 30 day pre-challenge test. After the 30 day feeding period, the shrimps were challenged by intramuscular injection of Infectious Myonecrosis Virus (IMNV) and Vibrio harveyi. The results showed that shrimp treated with a synbiotic-supplemented diet showed  significantly higher growth performance than control groups (P<0.05). Shrimp treated under regime P1 showed the highest values for phenoloxidase (PO) and respiratory burst (RB) parameters compared to shrimp given the other treatments. Following the challenge test, higher survival rate were seen in the P1 treatment group, in comparison to the positive control, and the P1 treatment group showed the highest values in total haemocyte count (THC), PO, and RB.

Abstract

The aim of this study was to evaluate the effect of encapsulated synbiotic (Bacillus sp. NP5 and oligosaccharide) dietary at different dosages on growth performance, survival rate, feed conversion ratio, and immune responses of Litopenaeus vannamei against Vibrio infection. The shrimps of the main treatments were fed by the diet that contained three different dosages of encapsulated synbiotic [0.5% (A), 1% (B), and 2% (C) (w/w)] with feeding rate of 5% of shrimp biomass (4 times a day). The shrimps of two control treatments (negative control and positive control) were fed only by commercial feed without supplementation of encapsulated synbiotic. The growth, feed conversion ratio, and survival rate were observed after 30 days of encapsulated synbiotic dietary. The shrimps were then challenged by injection of Vibrio harveyi (6 log colony forming units/mL) 0.1 mL/shrimp, excluded the negative control treatment. Afterward, the survival and immune responses were observed for 9 days after experimental infection. The shrimps treated with 2% encapsulated synbiotic (treatment C) in the diet showed the highest growth performance (2.98 ± 0.42%), feed conversion ratio (1.26 ± 0.19), and better immune responses i.e. total hemocyte counts, differential hemocyte count, phenoloxidase, and intestine bacteria observation compared to those of positive control treatment.

Abstract

This study aimed to analyze the effect of Bacillus megaterium PTB 1.4 on the population of intestinal microflora, digestive enzyme activity, and the growth of catfish. Gnotobiotic and normal fish were used. Treatment using gnotobiotic was divided into gnoto (with feed and 100 μg/mL rifampicin) and gnotoplus (with feed, 100 μg/mL rifampicin, and 1% probiotic); whereas treatment using normal fish was divided into normalplus (with feed and 1% probiotic) and normal (only feed). The amount of bacteria on gastrointestinal tract was measured 30 days after treatments using the total plate count method. The results indicated no significant difference in bacterial growth between gnotobiotic and normal fish. The total amount of probiotic bacteria with normalplus treatment was significantly different with gnotoplus. The activity of protease and amylase enzymes, and specific growth rate in normalplus treatment were significantly higher (p < 0.05) than other treatments. Bacillus megaterium PTB 1.4 increased the activity of digestive enzymes and the growth of catfish.

Abstract

Bacterial disease attack occurs at the hatchery stage, which is considered to be the most serious threat, and often results in mass mortality of shrimp larvae by vibrosis which is that caused by a luminous bacterium identified as Vibrio harveyi. This research was carried out to obtain local isolates of probiotic bacteria that were able to inhibit the growth of V. harveyi and effectively apply it as a biocontrol of vibriosis in shrimp cultures. The research was carried out as follows: (1) In vitro and in vivo selection of probiotic bacteria candidates, (2) Study of the action mechanism and characterization of the selected pro biotic bacteria, (3) Study on application of the selected probiotic bacteria as a biocontrol agent in shrimp cultures. Results of in vitro and in vivo selection provided the best three isolates, which were 1Ub, SKT-b and Ua. The survival rate of shrimp larvae which were not only inoculated by V. harveyi but also with 1Ub, SKT-b and Ua probiotic bacteria were 88.33, 83.33, and 81.67% respectively; where as the positive control treatment (merely inoculated with V. harveyi) gave a 41.67% survival rate and the negative control (without bacterial addition) was 68.33%. Studies using a rifampicin resistant marker (RfR) demonstrated that the number of V. harveyi MR5339 RfR cells in treatments without probiotic addition were higher than the treatment with the probiotic bacteria, in dead larvae, living larvae, as well as in the culture media. Partial sequencing of the I6S-rRNA gene showed that the I Ub isolate was similar to Pseudoalteromonas piscicida, whereas the SKT -b and Ua isolates were similar to Vibrio alginolyticus. Selected probiotic bacteria could be applied directly to shrimp larva culture media, or orally through enrichment of both natural and artificial food.
