Abstract

DNA (deoxyribonucleic acid) extraction method is the process of separating DNA from the sample. In this process, the DNA obtained must be protected from contamination by RNA, carbohydrates, lipids, and proteins. Contamination of RNA, carbohydrates, lipids, and proteins can increase DNA purity. DNA purity was measured using a NanoDrop 2000 spectrophotometer measured by the absorbance ratio at 260 nm and 280 nm wavelengths. Good quality DNA will have an A260/A280 ratio of 1.7-2.0 and a concentration > 0.03 pg. This study aimed to obtain the appropriate DNA extraction method for fresh meat samples (a mixture of rat and chicken meat). This research consisted of two stages: the DNA extraction stage using the Progenus EasyFast™ Extraction Kit for Meat Products and the amplification stage using the EASYFAST™ Rat Detection Kit. This study used 16 samples of a mixture of rat meat and chicken with concentrations of rat meat: 5, 10, 15, and 20%. At the extraction stage, the incubation time was optimized for 15, 30, 45 minutes, and 1 hour. The results showed that the one hour incubation had a lowest CT value in the results of PCR amplification.

Abstract

Perkembangan produk halal dewasa ini sangat meningkat seiring dengan semakin tingginya kepedulian Masyarakat, khususnya muslim terhadap pola hidup sehari-hari. Undang-undang tentang Jaminan Produk Halal (JPH) yang mendorong munculnya lembaga-lembaga independent atau yang ditunjuk pemerintah dalam memfasilitasi pengakuan kehalalan produk tersebut. Produk kesehatan halal adalah produk yang dalam pembuatan serta pengemasannya mengikuti proses halal sesuai prosedur yang ditetapkan pemerintah melalui Sistem Jaminan Produk Halal (SJPH). Pengenalan produk obat dan kosmetika halal bertujuan untuk mengedukasi masyarakat dalam menggunakan produk yang digunakan sehari-hari dan menjadi kebutuha utama setelah makanan yang berdampak pada kesehatan tubuh. Penting bagi masyarakat mengetahui kehalalan setiap produk yang mereka gunakan dan menghindari rasa khawatir dari terpaparnya barang syubhat dan najis/haram. Edukasi diberikan dengan cara tatap muka langsung dan menyampaikan informasi lewat brosur serta media sosial, serta dilakukan penilaian sebelum dan setelah peserta mendapatakan edukasi. Berdasarkan hasil yang diamati selama kegiatan berlangsung, terdapat peningkatan pemahaman peserta atau warga terkait informasi dan cara memilih obat dan kosmetika halal, sehingga meningkatkan keamanan dan rasa nyaman bagi warga.

Abstract

Pletekan leaves are found in many regions in Indonesia, traditionally used for the treatment of hypertension, diabetes and antioxidants. Pletekan plants contain tannin compounds with phenol groups which are known to have biological effects such as antioxidants through mechanisms as free radical traps and electron donors. The aim of the research was to determine antioxidant activity using the ABTS (2,2-azinobis 3- ethyl benzothiazoline-6-sulfonic acid) method and testing total tannin content using the spectrophotometric method and Folin-Ciocalteu reagent and tannic acid as standards. Based on the research results, the 70% ethanol extract of Pletekan leaves has antioxidant activity as a free radical reducer which is in the medium category with an IC50 value of 215.07 ± 11.65µg/ml while the total tannin content is 114.60 ± 4.1 mg TAE/g.
