Abstract

Analgesic and anti-inflammatory tests of eel (Anguilla bicolor bicolor) oil on animal models have been performed. Previous studies have proven that oral administration of EPA and DHA exhibits analgesic and anti-inflammatory effects. Gas Chromatography analysis shows that eel contains EPA and DHA. In this research, the analgesic activity was evaluated with the acetic acid-induced writhing test and hot plate test, while the anti-inflammatory properties were identified using carrageenan-induced inflammation. In the writhing test, 25 male Swiss Webster mice were divided into five groups. Group I was given 0.5% CMC-Na as a negative control, group II was given 65 mg/kg b.w. of Acetosal as a positive control, group III-V was given eel oil at different doses, namely 2400, 4800, and 9600 mg/kg b.w. of the mouse. For the hot plate test, 6.5 mg/kg b.w. of tramadol acted as the positive control. Similar to the analgesic effect analysis, the anti-inflammatory test also divided 25 male Wistar rats into five groups. Group I as a negative control was given 0.5% CMC-Na,  group II was given 9 mg/kg b.w. of diclofenac potassium as a positive control, and group III-V were given eel oil at different doses, namely 1500, 3000, and 6000 mg/kg b.w. of the rat. The results of the acetic acid-induced writhing test and hot plate test showed that when compared with the positive and negative controls, eel oil had a potential analgesic activity with a significance value of p< 0.05. The analgesic effects were noticeable at doses of 2400, 4800 and 9600 mg/kg b.w. in the writhing test and at 4800 and 9600 mg/kg b.w. in the hot plate test. The anti-inflammatory test showed that eel oil was efficacious when administered at the doses of 1500, 3000, and 6000 mg/kg b.w. with percentage inhibition of 34.35%, 35.132%, and 40.28%, respectively.

Abstract

The fatty acids, like EPA and DHA, were known as anti-inflammation. It works on inflammatory tissue, brought by edema plasma, and changed into resolvins, protectins, and maresins by the enzyme reaction. Anguilla bicolor and Sea cucumber fish are known to contain EPA and DHA. This study analyzes the inhibition Cox-2 effect of the combination of Anguilla bicolor oil with Sea cucumber extract as an active ingredient in a cream preparation. This experiment used 7 groups of male BALB/C strain mice: normal control; negative control; positive control; A. bicolor oil cream; H. leucospilota extract cream; a combination of A. bicolor : H. leucospilota (2:1) cream; a combination of A. bicolor : H. leucospilota (1:2) cream. The anti-inflammatory activity was evaluated by the amount of inflammatory cell, the thickness epidermis, and the amount of cell expression COX-2  on mice's back skin tissue induced by croton oil (0,1 %). After 3 days, histopathological skin tissues were made. The data were analyzed statistically by One Way ANOVA followed by LSD to know each group's differences at a significance level of 0.05. The experiment results showed that the formula with the best anti-inflammatory activities is the combination of A. bicolor and H. leucospilota (2:1) cream.  The decrease of the amount of inflammatory cell (75.97%), the thickness of the epidermis (43.88%), and the amount of COX-2 cell expressions (60.52%) from the formula did not differ significantly with the positive control (p>0,05). It can be concluded that A. bicolor and H. leucospilota have anti-inflammation activity based on the experiment results.

Abstract

Indonesia is a high sun exposure country. Exposure to ultraviolet (UV) causes various kinds of skin disorders such as erythema, sunburn, aging, and cancer. Mountain papaya fruit (Vasconcellea pubescens A.DC.) and mangosteen peel (Garcinia mangostana L.) contains metabolite compounds that can protect the skin from sunlight because of its antioxidants activity. The purpose of this study to determine whether the combination of the mountain papaya fruit and mangosteen peel extracts in skin lotion can be used as sun protectors through the in-vitro and in-vivo study. The experiment was done by extracting the mountain papaya fruit and mangosteen peel through the maceration method. The extracts were formulated into skin lotion in three different formulas with the ratio of mountain papaya fruit extract: mangosteen peel extract as follows F1(1:1), F2(1:3), and F3(3:1). In vitro test was done by using UV-VIS spectrophotometry to determine the SPF value and in vivo test was used erythema-induced rats by exotera beam light. The result of in vitro test gained a high enough SPF value for all three formulas F1=23,23; F2=21,70 and F3=28,64 and the result of in vivo test showed that all three formulas did not indicate the existence of erythema value.         It can be concluded that three skin lotion formulas containing mountain papaya fruit and mangosteen peel ethanol extract have the effect of sun protection.

Stress is generally a normal condition and does not cause health problems. Aromatherapy balms with the main ingredients of essential oils can be used as an alternative to relax the body and play a role in emotional regulation. This community service is the transfer of knowledge and assistance in training on making aromatherapy balm. The activities included providing information on making balm, followed by a training demonstration on aromatherapy balm. The service activity, carried out on October 25, 2022, resulted in an aromatherapy balm product that was handmade by the present residents, accompanied by a team from UNS Pharmacy D3. This activity has achieved the desired target, namely the ability of the residents of Mojosongo Village to make a small-medium-scale product that has a high value both in terms of use and finances. This mentoring activity is the first step in establishing the independence of the local community to produce aromatherapy balm independently or in groups within the scope of Micro Small and Medium Enterprises (MSME).
