Abstract

Insulin is an important factor in glucose transport. Glucose transporter (GLUT-2) in pancreatic β cell membranes is responsible for glucose transport that will stimulate insulin secretion. The purpose of this research was to investigate the antihyperglycemic activities of extract of Trigonella foenum-graecum L. (fenugreek) seed and the effects on GLUT-2 protein expression in pancreatic β cells on Streptozotocin-Nicotinamide-induced (STZ-NA) rats. This research utilized five groups of male Wistar rats. Group I was normal control group, while Groups II-V were induced with STZ-NA. Group II was negative control group (CMC Na 1%); group III was positive control group (Glibenclamide); group IV and V was examined group (fenugreek seed ethanol extract of 100mg/kg bw and 200 mg/kg bw)). The test compounds were orally administered for 14 days. Measurement of blood glucose levels and observation of the expression of GLUT-2 protein in pancreatic β cells in rats were conducted by immunohistochemistrical method. The research results revealed that the examined group dose of 200 mg/kg bw of fenugreek seed extract appeared to show blood glucose level decline that was proportionate to that of the positive control group. The increased density of GLUT-2 protein in pancreatic β cells significantly occurred in the group injected with 200 mg/kg bw compared to the group injected with 100 mg/kg bw. Hence, 200 mg/kg bw of fenugreek seed ethanol extract was able to decrease blood glucose levels and increase the GLUT-2 protein expression in pancreatic β cells in STZ-NA-induced rats with the effectiveness that was comparable to the positive control group

Abstract

Diabetes Mellitus (DM) type II occurs in individual with insulin resistance or β pancreaticcells damage which is not able to respond glucose and produce insulin. Insulin is an importanthormon in glucose transport. Glucose transporter (GLUT-2) in β pancreatic cell membranes isresponsible for glucose transport that will stimulate insulin secretion. Abstract

Diabetes melitus merupakan suatu penyakit dengan adanya gangguan sekresi insulin baik karena penurunan sensitifitas maupun kerusakan pada sel beta. Tujuan penelitian ini adalah membuktikan ekstrak daun petai cina (Leucaena leucocephala (Lam.) de Wit) memiliki aktivitas menurunkan kadar gula darah mencit hiperglikemik pada dosis yang efektif. Penelitian antihiperglikemik ini menggunakan mencit putih sebanyak 30 ekor. Pengujian aktivitas antihiperglikemik dilakukan pada 6 kelompok perlakuan yaitu kelompok I (kontrol normal), kelompok II (kontrol negatif, induksi aloksan 200 mg/kg BB dan akuades), kelompok III (induksi aloksan dan glibenklamida 10 mg/kg BB), kelompok IV (induksi aloksan dan ekstrak daun petai cina 200 mg/kg BB), kelompok V (induksi aloksan dan ekstrak daun petai cina 400 mg/kg BB), kelompok VI (induksi aloksan dan ekstrak daun petai cina 300 mg/kg BB). Semua kelompok perlakuan diberikan perlakuan tersebut selama 14 hari dan dilakukan pengukuran kadar gula darah pada hari ke-0, 3, 10, dan 17. Aktivitas antihiperglikemik ekstrak daun petai cina ditunjukkan dengan cara menghitung daya hipoglikemik masing-masing perlakuan. Hasil penelitian menunjukkan bahwa ekstrak etanol daun petai cina mempunyai aktivitas antihiperglikemik pada mencit yang diinduksi aloksan. Pada dosis uji ekstrak etanol daun petai cina 600 mg/kg BB mempunyai aktivitas antihiperglikemik yang efektif dibanding dengan dosis 400 mg/kg BB dan 200 mg/kg BB yang sebanding dengan kelompok kontrol positif.

Abstract

Inflamasi merupakan respon protektif normal terhadap luka jaringan yang disebabkan oleh trauma fisik, zat kimia yang merusak, atau zat-zat mikrobiologi. Salah satu bahan alam yang memiliki khasiat antiinflamasi adalah minyak atsiri daun cengkeh (Syzygium aromaticum (L.) Merr. & L.M.Perry). Penelitian ini bertujuan untuk mengetahui kualitas minyak atsiri daun cengkeh, formulasi gel minyak atsiri, serta uji sifat fisik dan uji iritasinya. Minyak atsiri diperoleh dengan destilasi uap dan air. Minyak atsiri yang didapat digunakan dalam sediaan gel dengan konsentrasi hidroksipropil metilselulose (HPMC) 3% (FI), HPMC 3% dan  minyak atsiri 6% (FII), HPMC 6% (FIII), HPMC 6% dan minyak atsiri 6% (FIV), HPMC 10% (FV), serta HPMC 10% dan minyak atsiri 6% (FVI). Pengamatan terhadap gel meliputi uji organoleptik, homogenitas, pH, uji daya lekat, dan uji daya sebar. Gel  yang telah diuji sifat fisiknya, dievaluasi iritasinya terhadap kulit dengan metode Draize test. Data hasil pengujian dianalisis statistik dengan ANAVA satu arah dengan taraf kepercayaan 95%. Dari penelitian yang telah dilakukan diperoleh hasil minyak atsiri daun cengkeh yaitu berwarna kuning kecoklatan dan berbau khas daun cengkeh dengan nilai rendemen sebesar 1,6%, indeks bias sebesar 1,525, dan bobot jenis sebesar 1,02. Gel daun cengkeh memenuhi standar kualitas gel  yaitu homogenitas, daya sebar,  pH, dan daya lekat. Analisis statistik  terhadap masing-masing percobaan tidak menunjukkan perbedaan yang signifikan. Hasil percobaan terhadap uji iritasi menunjukkan bahwa sediaan gel tidak menimbulkan iritasi pada kulit marmut.

Abstract

Depresi merupakan suatu gangguan emosional dalam waktu jangka yang panjang yang disebabkan turunnya kadar neurotransmiter didalam otak. Daun pepaya (Carica papaya L.) memiliki kandungan senyawa alkaloid, flavonoid, saponin, dan tanin yang digunakan sebagai antidepresan. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antidepresan dan dosis yang efektif pada ekstrak etanol daun pepaya (Carica papaya L.). Pada penelitian ini menggunakan ekstrak etanol daun pepaya dengan uji antidepresan dan metode forced swimming test dengan mengamati immobility time pada mencit. Sebanyak 25 ekor mencit jantan dibagi kedalam 5 kelompok uji. Kelompok pertama diberi CMC Na 0,5% sebagai kontrol negatif. Kelompok kedua diberi amitriptilin dengan dosis 0,065 mg/20 g BB mencit sebagai kontrol positif. Kelompok ekstrak daun pepaya (Carica papaya L.) dibuat dengan menggunakan metode maserasi dan pelarut etanol 96% dengan 3 variasi dosis (150 mg/kgBB; 250 mg/kgBB; dan 350 mg/kgBB). Masing masing kelompok uji sebelumnya telah diinduksi depresi selama 7 hari dengan cara direnangkan. Setelah itu mencit diberi perlakuan sampai hari ke-14. Pengamatan Immobility time pada mencit diamati pada hari ke 1, 7, dan 14. Hasil dari penelitian ini menunjukkan bahwa ekstrak daun pepaya mempunyai aktivitas sebagai antidepresan yang dilihat dari immobility time dengan metode forced swimming test. Dosis yang paling efektif untuk menurunkan waktu imobilitas secara signifikan pada mencit adalah dosis 350 mg/kgBB. Abstract

Methicillin-resistant Staphylococcus aureus (MRSA) is a Staphylococcus aureus bacterium that is resistant to methicillin-type antibiotics. This study aims to determine the antibacterial activity of the active fraction of sweet corn hair extract on the growth of Methicillin-Resistant Staphylococcus aureus (MRSA) bacteria. Extraction was carried out by the remuneration method in 70% ethanol. In fractionation, liquid vacuum column chromatography with n-hexane, ethyl acetate, and methanol solvents was used. The fraction with the largest clear zone of methanol (100%) is with a clear zone diameter of 0.752 cm. Testing the antibacterial activity of ethanol extract at concentrations of 10%, 15%, and 20% obtained by clear zone with an average of 1,013; 1,073; 1,159 cm and in the active fraction with an average of 0,703; 0.903; 1,004 cm. Ciprofloxacin 0.005% was used as a positive control and DMSO as a negative control. The results showed that ethanol extract and an active fraction of sweet corn hair (Zea mays f saccharata Kornicke & Werner) had antibacterial activity against Methicillin-Resistant Staphylococcus aureus (MRSA). One-way ANAVA test results showed that there were significant differences in providing antibacterial activity (p> 0.05) between ethanol extract and active fractions at concentrations of 10%, 15%, and 20%. Flavonoid compounds, alkaloids, saponins, tannins, and triterpenoids in the active fraction of corn hair have antibacterial activity against MRSA bacteria.

Abstract

Telang flower (Clitoria ternatea L.) has antioxidant effects because it contains compounds such as phenols, anthocyanins, flavonol glycosides, kaempferol glycosides, terpenoids, steroids, flavonoids, and tannins. This study aims to determine whether telang flower ethanol extract has antioxidant activity, whether telang flower ethanol extract ointment can heal cuts in rabbits and determine the effective concentration of telang flower ethanol extract ointment for healing cuts in rabbits. This study used telang flower extract macerated with 70% ethanol. The concentration of the extract was made as much as 3, namely 0.1%, 0.2%, 0.4%. The extract was tested for antioxidant activity using the DPPH method. The tested extract was then made into an ointment and applied to the rabbit's back, and wound healing was observed. Based on the results of statistical tests using SPSS®, variations in the concentration of the ethanol extract of the butterfly pea flower affected the antioxidant activity. The 0.2% concentration gave a significant difference compared to the 0.1% and 0.4% concentrations.  Telang flower ethanol extract ointment can heal the incision wound on the rabbit's back. Based on the results of statistical tests using SPSS® on the healing of cuts in rabbits showed a significant difference. The 0.2% concentration differed significantly from the 0.4% concentration, where the time required for the 0.2% concentration was faster than the 0.4% concentration.

Abstract

The subchronic toxicity test of Noni (Morinda citrifolia L.) ethanol leaf extract have been studied. The purpose of this study was to determine the effect of subchronic toxicity of noni ethanol leaf extract (Morinda citrifolia L.) on increased levels of creatinine and BUN (Blood Urea Nitrogen) of white rats. Noni leaves were extracted by using the maceration method with 96% ethanol dissolution for 3 times 24 hours while occasionally stirring until a thick extract was obtained. Then the blood creatinine and BUN levels were determined before administering the test preparation and on day 28. The research was carried out for 28 days then continued for 14 days for the satellite group. The results of the subchronic toxicity testing that was carried out showed a double increase in creatinine and BUN levels compared to normal values. Noni leaf extract treatment caused an increase in creatinine and BUN levels in the subchronic toxicity test at doses of 250, 500 and 1000 mg/kg BW, and the highest increase in levels occurred in the 1000 mg/kg BW group

Abstract

Acrylamide is a food contaminant that has the potential to cause cancer. Acrylamide is a substance that is formed due to high temperature processing of food such as frying and baking. Chicken nuggets are a popular fast food, easy to get and can be served quickly. This study aims to determine the levels of acrylamide in chicken nugget with variations in processing using frying and oven with High Performance Liquid Chromatography (HPLC). Acrylamide in chicken nuggets is extracted with dichlormethane and ethanol. Measurement of acrylamide levels using High Performance Liquid Chromatography with a Supercolsil C18 column, UV detector at a wavelength of 210 nm and a flow rate of 0.5 mL/min with 85% phosphoric acid as mobile phase in acetonitrile and aquabidest (5:95). Method validation was carried out to determine linearity, accuracy, precision, LOD and LOQ. The results of the method validation were declared valid for the assay as indicated by the linearity value (r) of 0.99947, percent recovery of 97.29% and the results of the precision test obtained an RSD value of 1.36%. The LOD value was 0.3933 ppm and the LOQ was 1.1919 ppm. The results showed that acrylamide can be formed in the processing of chicken nuggets in 3 replications using the oven method with sequential levels of 0.8459 ppm; 0.7936 ppm and 0.7807 ppm. Whereas with the frying method the acrylamide content formed was 1.4591 ppm; 1.4721 ppm and 1.0554 ppm. The results of statistical analysis using the Independent Simple t test showed that there were significant differences with a significance value of 0.028 in the processing with the oven and frying methods.
