Abstract

AbstrakPenelitian ini dirancang untuk mengetahui pengaruh fraksi air G. lucidum dalam menurunkan kadar glukosa darah dan ekspresi p53 pada jaringan pankreas tikus diabetes yang diinduksi aloksan. Tiga puluh enam tikus dibagi menjadi enam kelompok yaitu normal, diabetes, glibenklamid dan tiga dosis fraksi air pengobatan kelompok G. lucidum (225, 450, dan 675 mg / KgBB). Tikus diabetes diinduksi aloksan intraperitoneal pada dosis 150 mg / KgBB. Keadaan diabetes terjadi pada hari ke-3 setelah pemberian aloksan dan menunjukkan peningkatan kadar darah sampai 274,00 ± 15,11 mg / dL. Pemberian oral fraksi air G. lucidum dan glibenklamid selama 9 hari setelah kondisi diabetes secara signifikan mengurangi kadar glukosa darah (P <0,05). Pada hari ke-10 setelah pengobatan, tikus yang dimatikan dan jaringan pankreas diambil untuk pengujian Haematoxyllin eosin. Hasil penelitian menunjukkan bahwa fraksi air G. lucidum menghambat apoptosis sel β. Perubahan morfologi jaringan pankreas dalam keadaan diabetes secara signifikan menurun dan berubah menjadi keadaan normal setelah pemberian berulang fraksi air G. lucidum.  Kata kunci: fraksi air  G. lucidum, Aloksan monohidrat, Diabetes melitus  AbstractThe present study was designed to investigate the influence of water fraction of G. lucidum toward lowering blood glucose level and p53 expression in pancreatic tissues in alloxan-induced diabetic rat. Thirty six rats were divided into six groups i.e. normal, diabetic, glibenclamide and three dosage of treatment water fraction of G. lucidum groups (225, 450, and 675 mg/KgBW).  Diabetic rats were induced by alloxan monohidrate intra peritoneally at the dose of 150 mg/KgBW. Diabetic state occurred on the 3rd day after alloxan administration and it was showed increasing blood level until 274.00 ± 15.11 mg/dL. Orraly administrations of water fraction of G. Lucidum and glibenclamide for 9 days after diabetic condition  were significantly reduce of blood glucose level (P<0.05). At the day 10th after treatment, rats were sacrified and the pancreatic tissues were harvested for Haematoxyllin eosin. The results showed that water fraction of G. lucidum inhibit the β cell apoptotic. The morphological change of pancreas tissue in a diabetic state was significantly decreased and turned into normal state after repeated administration of water fraction of G. Abstract

Diabetes mellitus is one of the degenerative diseases of concern to government and society, because the incidence of sufferers is increasing from year to year. Lingzhi (Ganoderma lucidum) mushroom is a traditional medicinal plant which one of the benefits to treat diabetes mellitus. This study aimed to identify and seek the most effective dose of lingzhi mushroom extract to decrease blood glucose levels in white male rats of wistar strain. Lingzhi mushroom extracted by maceration with ethanol 96% and infuse it with aquadest. The ethanol extract was tested on each group of test animals with a dose of 1.18 mg/200 g BW; 2:36 mg/200 g BW and 4.71 mg/200 g BW. Negative control group given CMC 1% and the positive control group was given glibenclamide (90 mg/200gBB). Aqueous extract obtained was tested after the optimal dose of ethanol extract. Antidiabetic effect is obtained by measuring blood glucose levels for 2-week with intervals measure every week, then performed statistical tests with two-way analysis of variance with a level of 95% followed Dunnett test. The results showed that the lingzhi mushroom has antidiabetic activity in male rats. Ethanol 96% extract of lingzhi mushroom (1.18 mg/200 g BW) most effectively to decrease blood glucose. Water extract at a dose equivalent to 1.18 mg/200gBB have equally strong effects with ethanol extracts.

Abstract

Onion (Allium cepa L.) is one of medical plants that widely used, and onion tuber is supposed to have beneficial to decrease cholesterol level. The aim of this study was to find out the effect of water fraction of onion ethanol extract that given orally on decreasing total cholesterol level in blood serum of white rats wistar and to see the dose that gave the effect equal with simvastatin in decreasing total cholesterol level. The study to determine cholesterol level of the animal testing was conducted by CHOD-PAP method. Test animals were divided into 5 groups and each group consists of 5 rats. Group I as negative control was contain CMC 1%. Group II was contain CMC 1% and treated water fraction of onion etanol extracts with three variations of dose, namely 25, 50, 100 mg/200 g body wight obtained from the ethanol extract fraksinasi using water and ethyl acetate. Group III as positive control was contain simvastatin and CMC 1%. Test animals were given pig oil and cholesterol powder for one week until hypercholesterolemia condition, and then were given test preparation until the 14th day. The cholesterol level was measured on the 1st, 7st and 14st day. The results of this study were that water fraction of onion ethanol extracts that given orally could decrease total cholesterol level in blood serum of white male rat. According to the results of one way anova statistical test, it was indicated that water fraction of onion ethanol extracts of 100 mg/ 200 g body wight dose had the effect to decrease total cholesterol level equal with simvastatin.

Abstract

Kamboja (Plumeria acuminata Ait.) plant is traditional medicine used as medication for abscess and yaws. Flavonoid is a component of chemical compound in kamboja leaves that supposed to have antibacterial activity. The aim of the experiment was to find out the ability of kamboja leaves fractions creams in healing the infection of Staphylococcus aureus on rabbit skin and to know the most effective concentration. Kamboja leaves were extracted by soxhletation method using 96% ethanol solvent, and then the obtained extract was fractionated using n-hexane, ethyl acetate and 70% ethanol solvents. The antibacterial activities of the three fractions were tested in the form of creams by in vivo method. It was found that ethyl acetate fraction was the most effective, and then various doses were conducted with concentrations of 15%, 20%, and 25% using test animals infected by Staphylococcus aureus.The result of the experiment showed that ethyl acetate fraction had the best ability compared with n-hexane and ethanol 70% fractions. Ethyl acetate fraction cream concentration of 15%, 20%, and 25% could heal the infection for 11.40, 10.60 and 9.40 days, respectivelly.

Abstract

The aim of this experiment was to find out the effect of Bandotan (Ageratum conyzoides, L) herbs ethanol extract that given orally on decreasing bleeding and blood coagulation time in white male rats (Rattus norvegicus) wistar strain. Bandotan herbs were extracted by soxhlet using 70% ethanol as solvent. The animals test were divided in five groups, each group consist of 5 mice. Group I was given dose 69,58 mg, group II was given dose 104,37 mg, and group III was given dose 139,16 mg/200 g BW. Group IV as positive control was given tranexamic acid 4,5 mg/200 g BW. Group V as negative control was given CMC 1%. The animals test were given sub cutaneous heparin injection for 5 days to induce bleeding and inhibit blood coagulation, and then were given test preparation for 6 days and the time of bleeding ceasing and blood coagulation were recorded once every 2 days during test preparation was given. \The statistic results of the study showed that ethanol extract of goat weed herbs given orally could decrease bleeding time and blood coagulation in white male rats. According to the result of two ways anova statistical test, ethanol extract of goat weed herbs with dose of 104,37 mg/200 g BW is effectivest in decreased bleeding and blood coagulation time with ability to stop bleeding time 85,45% and blood coagulation time 70%.

Abstract

Diabetes mellitus is a disease that affects many people. Dewandaru (Eugenia uniflora L.) leavesis a traditional medicinal plant contains saponins, flavonoids, tannins which one of the benefits to treat diabetes mellitus. Aimedof this study to identify and seek the most effective combination dose of 70% ethanol extract dewandaru leaves in combined with metformin to decrease blood glucose levels in Wistar male rats. Dewandaru leaves macerated with 70% ethanol solvent, condensed macerate obtained was tested in each group of test animals. Thirty rats were divided into 6 groups. Group I was given 70% ethanol extract of dewandaru leaves 300 mg/200 gBW, the second group (negative control) was given Na2EDTA 14 mg/200 gBW, group III (positive control) was given metformin 4.5 mg/200 gBW, group IV, V and VI was given the extract doses of 75 mg / 200 gBW, 150 mg/200 gBW and 300 mg/200 gBW combine with metformin 4.5 mg/200 gBW repectively. The treatment conduct for 12 days, 3 days induced with Na2EDTA, and then for 9 days given treatment according to each group. The glucose levels were determinated day 0, the 3rd, 6th, 9th and 12th. Then the data obtained were analyzed by one way ANOVA with a level of 95%. The result showed a decrease in blood glucose levels significantly between the treatment group 70% ethanol extract of dewandaru leaves dose of 75 mg, 150 mg and 300 mg/200 gBW in combine with metformin. 70% ethanol extract of dewandaru leaves dose of 75 mg/200 gBW in combine with metformin iseffective dose that coulddecrease blood glucose levels with three days theraphy.

Abstract

Malaria is a parasitic disease caused by protozoa of plasmodium genus. Mundu (Garcinia dulcis Kurz.) is one of the potential with the chemical compound xanton and triterpenoid for antiplasmodial. The aim of this experiment was to find out the effect of mundu (Garcinia dulcis Kurz) cortex n-hexane extract on leucocytes amount and hemoglobin level in mouse infected with Plasmodium berghei, and to know the dose of the most effective n-hexane extract as antiplasmodium. Mundu extract made by maceration use n-hexane. The animal test mice before treatment with extract induced by Plasmodium berghei to the number of erythrocytic in the blood of 107 and the concentrate extract was tested in each group of animals test with a dose of 50 mg / kg bw mice, 100 mg / kg bw mice and 150 mg / kg bw mice, negative control group were given 0.5% CMC and the positive control were given chloroquine be suspention in CMC solution. The anti-plasmodium effect was observed by calculating the leucocytes amount and hemoglobin level. Data was prosessed with one way ANOVA statistical test followed by tukey test. The result of the experiment showed that n-hexane extract of mundu cortex had the anti-plasmodium effect by decrease the leucocytes amount and increase hemoglobin on the mouse induced with Plasmodium berghei. The most effective dose of mundu cortex n-hexane extract was 100 mg/kg BW.

Abstract

Noni (Morinda citrifolia L.) has long been known as a nutritious plants. Noni has known contain ascorbic acid and β - carotene as an antioxidant function. The purpose of this research is to create a cream preparation with qualified physical properties and stability of extracts of noni as an antioxidant. The noni fruit extract was made cream in three formula with concentrations of 5%, 7,5%, and 10%. The physical properties and stability testing of the cream were viscosity, dispersive power, pH and viscosity changes. Antioxidant activity of noni fruit extract cream was tested by DPPH method. The results showed that the noni fruit extract could made cream until concentration of 10%. The noni fruit extrac creams had antioxidant activity. Formula III contained noni fruit extract was the best formula which had antioxidant activity with IC50 value of 69,61 mg/mL.

Abstract

Diabetes mellitus is a metabolic disorder characterized by hyperglycemia and related with abnormality metabolism of carbohydrate, lipid, and protein cause a decrease of insulin secretion or insulin sensitivity decrease or both. Yakon leaves is traditional medicinal plants that one of benefits could treat diabetes mellitus. The experiment was purpose to find antihyperglycemic activity of combination of yakon leaves ethanolic extract and metformin in mice induced alloxan. Test animals were divided in 6 groups each group consist of 5 mice. Groups I was given CMC 0,5%, group II was given metformin (65 mg/kg BB), group III was given yakon leaves ethanolic extract (840 mg/kg BB), group IV was given combination of yakon leaves ethanolic extract (420 mg/kg BB) and metformin (32,5 mg/ kg BB), group V was given combination of yakon leaves ethanolic extract (630 mg/kg BB) and metformin (16,25 mg/ kg BB). The treatment were given during 14 days, with measurement of glucose levels at 7 days and 14 days, then it was statistically tested by one way analysis of variance at 95% confidence continued with Tukey test. The result showed that the combination of yakon leaves ethanolic extract (630 mg/kg BB) and metformin (16,25 mg/ kg BB) was better than metformin or yakon leaves ethanolic extract.

Abstract

Sukun leaves is one plant that used for the treatment of diabetic. This study was aimed to determine antihyperglycemic activity of aqueous extract of sukun leaves, determine the extract dose which affect on blood glucose level, and determine the effect increased in dose of antihyperglycemic activity rats by alloxan induction. The method was used alloxan diabetic. Test animal were divided into five groups, consists of five male white rats: group I was diabetic control (CMC 0,5%), group II was comparative control (glibenclamide) at a dose of 0,09 mg/200 g BB rats, and group III, IV, V were given doses of aqueous extract of sukun leaves of 7 mg/200 g BB rats, 14 mg/200 g BB rats, 28 mg/200 g BB rats. Alloxan were induced on day 0 (after fasting 16 hours) intraperitoneally. Blood sugar levels were determined on days 4, 8 and 12 after administration of the test preparation. The results showed that aqueous extract of sukun leaves has antihyperglycemic activity. The aqueous extract sukun leaves affect by lowers blood glucose level with an effective dose of 7 mg/200 g BB. The greater the dose of aqueous extract of sukun leaves (7 mg/200 g BB rats, 14 mg/200 g BB rats, 28 mg/200 g BB rats) nothing with increased antihyperglycemic effect in significantly.
