Abstract

Green and red spinach is a vegetable that has a lot of nutritional content and is needed by the body. Besides containing many nutrients that are beneficial to the body, spinach vegetables also contain compounds that are toxic, can harm the body such as nitrite (NO2-) and cause methemoglobinemia. The purpose of this study was to analyze the levels of nitrites in green and red spinach with a visible spectrophotometric method and to determine whether the nitrites contained were still within safe limits of consumption in accordance with the ADI (Acceptable Daily Intake) value. Before the content analysis, a validation method was conducted with parameters including linearity with r = 0.9994, BD = 0.0499 µg / mL, BK = 0.1664 µg / mL,% recovery of day 1 = 1.5597 to 2 = 1.7547, the 3rd = 1.0126 and accuracy at a concentration of 0.3 μg / mL = 88.800%, 0.5 μg / mL = 95.332% and 0.7 μg / mL = 107.410%. Nitrite levels in fresh green spinach obtained an average of 23.4084 mg / kg, and 12.7272 mg / kg in red spinach. Furthermore, the green spinach after boiling yields an average of 16.8392 mg / kg and red spinach was 5.3359 mg / kg. Nitrite levels from all green and red spinach samples are still within safe limits for consumption in accordance with the ADI (Acceptable Daily Intake) value of 0.07 mg / kg of a predetermined human body weight.

Abstract

Optimization research has been done time on the production of antibiotics from bacterial isolate from Bajoe Distric Bone waters with nitrogen sources of variation, the first stage of the selection of isolates that have the greatest obstacle zone and which inhibits microbial isolates most of the isolates tested ERB 3 which inhibited Staphylococcus aureus, Salmonella typhi, Pseudomonas aeruginosa, Vibrio sp, Candida albicans and Aspergillus niger. Determination of the growth curve of the isolates with inoculate into a nutrient broth medium and incubated for 2 times 24 hours. Each interval of 2 hour was measured transmittance on spectrophotometer to determine the stationary phase. Results from growth curve obtained from the stationary phase at the 20th until the 48th hour. Determination of optimum fermentation conditions by varying the nitrogen source is peptone and tryptone. ERB 3 isolate was inoculated into the production medium and fermented using the medium of production in which every interval of 4 hours is making fermented products ranging from hour to hour-20thto48th. Activity was tested using agar diffusion method in the medium Glucose Nutrient Agar (GNA) against Staphylococcus aureus, Streptococcus mutans and Salmonella typhi. The results showed that the optimum time to produce antibiotics is the 24, 40 and 48hour and nitrogen source tryptone better than peptone. Keywords : Optimasi, Isolat, Antibiotika

Abstract

Chitosan is the result of deacetylation process from chitin which it can be found on Crustacean outer shell such as crabs. Chitosan can bind fat if it was consumed by human. The fat-binding ability of chitosan depends on the deacetylation degree. The research have been made into two phases. The first phase, chitosan was made from crab shell using NaOH. The deacetylation degree from chitosan that was made from the earlier process was analyzed with FTIR. The deacetylation degree result of the research was 59.39% using NaOH 50%. The second phase was the process of adsorbing triglycerides using chitosan in 10, 30, 45, and 60 minutes which was analyzed using spectrophotometer UV-Vis. The result of this research showed that the optimum triglycerides with 0.5, 1, 3 gram of chitosan was 2.99%, 3.14%, and 3.36%.
