Abstract

The purpose of the research is to develop Microbiology Module on the topic "The Antagonism of P. chrysogenum against Escherichia coli, Staphylococcus aureus, and Bacillus subtilis" that have criteria valid, practice, and effective for improve the student’s critical thinking skills. This research type is a quasi-experiment research using a non-randomized pretest-posttest control group design. The learning model uses the ADDIE approach which is implemented with the Problem Based Learning (PBL) model. Data collection uses response questionnaires, pretest and post-test, and validation sheets. The effectiveness of the Microbiology Module in improving students' critical thinking skills can be known through the pretest and post-test scores analyzed through the One Way Ancova test. The research and development result proved that the validation score gives by media and teaching materials experts was 99%, material experts were 98%, and Biology education practitioners were 100% from the student responses were 93%. The p-level results showed 0.00<0.05, so the modules were effective to use in the learning process. The research results proved that a significant difference between the experimental class learned using the Microbiology Module compared to the control class PowerPoint without the Microbiology Module. The modules is valid, practical, and effective to use in improve the student’s critical thinking skills.

Abstract

Yogurt is one of the fermented dairy products produced by lactic acid bacteria from milk, which contains beneficial nutritional components for human health. The innovation of yogurt production with the addition of probiotic cultures and food ingredients containing bioactive compounds can enhance its quality. This research aims to investigate the effect of adding purple sweet potato extract and storage treatment at 4℃ on yogurt's antioxidant and antibacterial activities. Four treatments were conducted in yogurt production, namely without adding purple sweet potato extract and adding 5%, 10%, and 15% (v/v) extract. All treatments were fermented using Lactobacillus bulgaricus, Streptococcus thermophilus, and Lactobacillus plantarum S1K2T1 as starter cultures. Subsequently, the yogurt was stored at 4℃ for 6 days, and chemical parameters, antioxidant, and antibacterial activities were examined. The highest result was observed in the treatment with 15% addition, showing a total antioxidant activity of 70.98% and an IC50 value of 13.96 ppm, which is equivalent to 2.06 times that of 200 ppm ascorbic acid. The six days storage affected antioxidant activity, but it was not significantly different. The antibacterial test showed indications of inhibition against Staphylococcus aureus ATCC 25923, Salmonella typhi NCTC 786, and Escherichia coli ATCC 25922, although the effect was very weak.

Abstract

One of the global problems that often occurs in society is environmental pollution, one of which is water pollution. The impact of water pollution is in the form of ecosystem damage and decreased water quality. One of nature's efforts to prevent environmental pollution in waters is by having bioindicators in waters, one of which is microalgae. However, if the community is less sensitive to environmental sustainability, environmental pollution will continue to occur. Therefore, public awareness is needed to protect the environment, one way is to foster an attitude of environmental concern among high school students by creating teaching materials related to the environment. To make it easier for students to understand and use the teaching materials, teaching materials were made in the form of PBL-based electronic modules that integrate materials related to microalgae and water quality to improve the environmental concern of class X high school students in a valid, practical and effective manner. The subjects used in this study were four classes of class X students of SMAN 12 Makassar. The instrument used in this study was an environmental concern attitude questionnaire that had been validated by experts. The results of this study can be categorized as valid, practical and effective because the validity results obtained 100% from the material expert validator, 92.2% from media and teaching materials experts, and 89.3 from biology education practitioners. While for the practicality test, there are 3 categories, namely, for individual tests the results were 86.58%, small group tests of 87.42%, and field tests of 87.44%. The effectiveness test is divided into 5 tests, namely, normality test, homogeneity test, N gain test, T test and ANCOVA test, where all tests obtained the results of the electronic module effectively used as teaching materials for class X of high school. This shows that the electronic module has an influence on the environmental care attitude of high school students in class X.

Abstract

This research is a research and development of Microbiology Handout on the topic "Antibacterial Test on Bacterial Growth Inhibition" which uses natural materials, namely star fruit extract (Averrhoa carambola L.), which will be used as teaching materials in the learning process to help improve students' science process skills. The purpose of developing Microbiology Handout is to produce teaching materials that are valid, practical and effective for use in learning Microbiology subjects. Microbiology Handout development uses the ADDIE development model. Validation of Microbiology Handouts was carried out by learning media and teaching material expert validators, Microbiology material experts, and Microbiology learning practitioner expert validators. The Microbiology Handout trial was conducted through three stages of trials, namely a one-to-one trial of 3 students, a small group trial of 10 students, and a field trial of 15 students. The effectiveness of Microbiology Handout in improving students' science process skills can be seen through pretest and posttest scores analyzed using the IBM SPSS version 26 application through the One Way Ancova test. The results of research and development show the acquisition of validation scores by learning media and teaching material experts of 100%, Microbiology material experts of 94.3%, education practitioner experts of 99.64%, student response results of 91.43% and efficiency results of 0.028

Abstract

Padina australis, brown macroalgae, has potential bioactive compounds that possess remarkable bioactivities including antimicrobial. The present study extracts macroalgae P. australis using Ultrasound assisted extraction (UAE) with n-hexane solvent, followed by screening phytochemical compounds. Moreover, the antimicrobial activity and MIC value of the n-hexane crude extracts of Padina australis was tested against Shigella sonnei, Escherichia coli, Salmonella typhi, and Bacillus cereus by using the disc diffusion method. This result demonstrates the optimum time of UAE process is at 10 minutes that yields 0.117 %. The phytochemical screening of n-hexane crude extracts of Padina australis macroalgae revealed the presence of secondary metabolites such as alkaloids, flavonoids, saponins, tannins, steroids and phenols. Antibacterial activity of this extract against Shigella sonnei, Escherichia coli, Salmonella typhi, and Bacillus cereus at 100 ppm concentration with inhibitory diameter range of 10.97 to 13.86 mm. Then, the n-hexane crude extracts of P. australis is potent against Bacillus cereus with MIC value of 6.25 ppm.

Abstract

The 21st century is recognized as the age of knowledge, marked by technological advancements, where students are expected to possess strong critical thinking skills. However, in the realm of science education, students' attitudes towards critical thinking remain relatively low. This is primarily due to the passive reception of information, with students merely memorizing it for examinations or tests. Thus, there is a pressing need for solutions to address these challenges. This research aims to determine the effectiveness of the Cloud-assisted PjBL model on students' critical thinking skills on the topic of Environmental Pollution. The method used was a quasi-experimental. The research instruments were a test for critical thinking skills, a questionnaire for environmental care attitudes, and for the Cloud-assisted PjBL learning response questionnaire. The outcomes indicated that the N-Gain data for the experimental class yielded a value of 0.75, indicating a high category, while the control class obtained a value of 0.49, indicating a medium category. The average result for environmental care attitudes among students in the experimental and control class is good. The conclusion in this research is that the Cloud-assisted PjBL model is effective on students' critical thinking skills and can improve students' environmental care attitudes.

Abstract

This research is motivated by the low level of student enthusiasm towards entrepreneurship due to the lack of activities that foster entrepreneurial personality. The research aims to examine the influence of biotechnology-based bio entrepreneurship on students' enthusiasm and entrepreneurial personality. A quasi-experimental design with a non-equivalent control group was used, involving class X students at SMAN 1 Cireunghas, Sukabumi district. Questionnaires were administered to collect data. Results showed that in the experimental group, 81% of students demonstrated a high level of enthusiasm with an N-Gain score of 0.81, increasing from a pretest average of 73.13 to a posttest average of 93.7. Regarding entrepreneurial personality, 79% of students exhibited high scores, with an N-Gain of 0.79, improving from a pretest average of 82.97 to a posttest average of 97.3. Hypothesis testing revealed significant differences between the experimental and control groups, with a significance level of 0.003 for enthusiasm and 0.020 for entrepreneurial personality (p < 0.05). Thus, biotechnology-based bio entrepreneurship positively influences both the enthusiasm and entrepreneurial personality of students.

Abstract

The use of incomplete or imbalanced data in ecological modelling and conservation planning can lead to inaccurate predictions due to limitations in detection methods. Therefore, obtaining comprehensive and precise empirical data on species presence and availability, particularly prey species is essential. This study aims to: (1) identify the presence of Javan leopards and their potential prey species, and (2) estimate the relative abundance indices (RAI) of each prey species. Using camera trap data from the Meru Betiri National Park (MBNP) survey conducted in 2024, we recorded 24 species, including the Javan leopard, and identified seventh potential prey species. We estimated relative abundance of each potential prey species was determined by dividing the number of individuals recorded in independent photos or videos by the total number of trap days and multiplied by 100. The result of RAI analysis indicated that the most abundant prey species in the study area was the long-tailed macaque, with an RAI value of 44.16 individuals per 100 trap-days, banteng was the least frequently detected species, appearing in only three independent images, with an RAI value of 0.23. The presence of Javan leopards, a charismatic flagship species, highlights the ecological significance of this area and provides valuable data for biodiversity management, serving as a foundation for future research and conservation efforts.

Abstract

Microgreens of sunflower (Helianthus annuus L.) are small and nutritious vegetables consumed at the early stages of growth. The growing process dictates the microgreen quality, such as planting media and watering frequency. This study aims to determine the interaction of various planting media with the watering on the microgreen growth parameters. This study conducts an experiment using 9 total treatments replicated 3 times. The research followed a randomized group design (RGD) and utilized analysis of variance (ANOVA) with a significance level of 5%, as well as Duncan's further test at a 5% significance level. The research parameters include plant height, hypocotyl length, epicotyl length, number of leaves, wet weight, consumable weight, and dry weight. The results showed that the interaction of planting media and watering frequency had a significant effect (P<0,01) on the growth parameters of sunflower microgreens. The best treatment combination is M1P3, Rockwool with watering three times a day with a plant height of 7,67 cm, hypocotyl length of 7,09 cm, epicotyl length 0.36 cm, number of leaves 2,53, wet weight 6,67 g, consumable weight 20.67 g and dry weight 1,22 g. Rockwool is sterile and has a structure that absorbs and stores water sufficiently for the growth of microgreens. The results of this study contribute significantly to improving the growth of sunflower (Helianthus annuus L.) microgreens through a combination of proper growing media and watering frequency.

Abstract

Salinity is one of the main obstacles in plant cultivation, especially during the germination and early growth stages. High salt concentrations can cause osmotic stress and ionic toxicity, thereby hindering water absorption and disrupting the physiological processes of the plant. This study aims to examine the effects of various concentrations of salt solution (NaCl) on the germination and early growth processes of water lettuce (Lactuca sativa var. angustana) in a hydroponic system. Soil salinity is a major problem in agriculture that can cause osmotic stress and ionic toxicity in plants, especially at the early growth stages such as germination. This condition hinders water absorption by the seeds and disrupts the balance of essential ions that play a role in cellular activities, thereby reducing the efficiency of physiological processes crucial for the formation of uniform and healthy plants. The research was conducted using a Randomized Complete Block Design. Complete with five treatments of NaCl concentration (0, 25, 50, 75, and 100 mM), each repeated three times. Seeds were sown on cotton media and watered with the solution according to the treatment for 14 days. The observed parameters include germination percentage, plant height, leaf count, and wet weight. The results show that the increase in NaCl concentration is inversely proportional to all growth parameters, where the treatment without NaCl yielded the best results, while the 100 mM concentration provided the lowest values. Statistical analysis confirmed the significant effect of salt concentration on the reduction of seed viability and vegetative growth of watercress. In conclusion, high salinity negatively impacts the physiology and early development of watercress through osmotic stress and ion toxicity mechanisms. These findings are important for the development of plant cultivation techniques in high-salinity areas and support the development of varieties that are more resistant to environmental stress, particularly in intensive agricultural systems such as hydroponics.
