Abstract

Anemic patients often have low hemoglobin and red blood cell level, which on the slide method and ABO system blood type test is used to obtain low degree of agglutination. Addition of treatment of a centrifugation was used to obtain a stronger degree of agglutination. The purpose of this research was to determine the difference degree of agglutination between centrifuged and uncentrifuged blood specimens in anemic patients. Quasi experimental research with non-equivalent control group design was conducted in March 2022 at the Hematology Laboratory, Universitas Muhammadiyah Semarang. The research sample were 16 specimens of anemic patients for slide method and ABO system blood type test. All samples were checked for blood type testbefore being centrifuged, then re-checked after the centrifugation there only concentrated red blood cell deposit were taken with only a small amount of plasma. The data from the test were analyzed using the Kruskal Wallis method. Comparison anti-A, anti-B, and anti-AB between centrifuged and uncentrifuged blood specimens there was a significant difference (p value < 0,005 means p = 0,002). The result showed that there were effect by centrifugation for the degree of agglutination improved better in centrifuged if compared with uncentrifugedanemicpatients blood specimens on blood type test slide method.

Abstract

A room that uses an Air Conditioner (AC) is one of the breeding grounds for air microorganisms, therefore it is necessary to have a number of germs in the room so as not to cause airborne disease. The purpose of this study was to determine the effect of temperature, humidity, and wind speed on the number of germs in an air-conditioned room. This type of research is an experimental consisting of 3 dependent variables and 1 independent variable with a cross-sectional research design. Laboratory room air samples were taken by the BRAB method, 12 times in a row. Sampling of the air using an air sampling pump. The results of the study showed that there was a fairly close relationship and a significant effect on temperature with the number of germs (p value = 0.005), with a temperature effect of 20.9%. There is a relationship and influence between humidity and germ numbers (p value = 0.000), with a humidity effect value of 31.9%. There is a relationship and influence between wind speed and germ numbers (p value = 0.009), with a wind speed effect of 18.5%. There is a simultaneous effect of temperature, humidity, and wind speed on the germ number of 33.6%.

Abstract

Health care is one industry that cannot escape from the touch of technology. Improvement of service quality, effective operational management, and customer satisfaction are the main objectives in the use of technology. One ofthe technology products that are important in clinical laboratories is the laboratory information system(LIS). Clinical laboratories produce and manage a lot of data as information to support medical diagnosiswith no exception in clinical laboratories at the Puskesmas (public health centre). Puskesmas as the spearhead of health services required to give the best performance and the use of LIS in clinical laboratory units can help improve laboratory operational performance. The purpose of this study was to determine the needs of the laboratory for LIS and to design a website-based LIS application according to the needs of the Puskesmas in Bandar Lampung City. This research method is a survey with data collection from the interviews with MLTs at 13 Puskesmas in Bandar Lampung City from June to December 2021. The design of a website-based SIL application is carried out using the data flow diagram (DFD) method. The results of the research is application features that can be applied in the design of making applications, namely ordering laboratory examinations, inputting patient data with a complete identity format, inputting laboratory results, and monthly results reports. These features are then designed in an application called the Orange Laboratory Information System (O-SIL).

Abstract

Black tea and chocolate are plants that contain caffeine. Chocolate is a commodity that generates foreign exchange for the country and is the main ingedient for making chocolate which also contains caffeine. Caffeine damages the liver by inhibiting the gowth of connective tissue in liver cells. The purpose of this study was to determine the histopathological effect on the liver organs of white rats given black and chocolate tea steeping and the damage that occurs when given in a predetermined dose and time. The research was conducted experimentally at the Anatomical Pathology Laboratory of the Sekolah Tinggi Ilmu Kesehatan Nasional. The study was divided into 7 treatment groups. Liver organs were observed macroscopically and microscopicallyfrom the results of histopathological preparations with Hematoxylin Eosin staining. The results of macroscopic observations there were no differences in the control and treatment goups. The results of microscopic observations in the black tea steeping goup 3 and 6 mg/200 g/day, and chocolate steeping 3, 6, and 12 mg/200 g/day there was degeneration, black tea steeped 12 mg/200 g/day there was necrosis. The results of the Mann Whitney test showed a significant difference in the black tea steeping goup at the three doses and 12 mg/200 g/day chocolate steeping, while 3 and 6 mg/200 g/day chocolate steeping did not show a significant difference. Abstract

Dengue Hemorrhagic Fever is a viral infectious disease that often causes high morbidity and mortality in Indonesia every year. Platelet count is one of the parameters that can be used as an indication in the diagnosis of this disease. In its development, severe dengue infection often causes proteinuria because there are no abnormalities in plasma, this condition will continue as the severity of the disease increases. This study aims to find out whether there is a relationship between thrombocytopenia and the incidence of proteinuria in dengue infection. This type of research is a retrospective analytic with a cross-sectional design, the data were taken from medical records at the Vita Medika Clinic laboratory in March – June 2020, including data on examination of thrombosis and examination of proteinuria in dengue patients. Data were analyzed by Spearman correlation test. The results of the study showed that there was a relationship between the number of platelets and the incidence of proteinuria in dengue infection with a value of p=0.002 (p<0.05). This is indicated by the mean, median, maximum and minimal platelet counts in respondents with proteinuria which were lower than respondents without proteinuria.

Abstract

Sheep blood agar is the standard medium for culturing pathogenic bacteria, as an alternative, human blood agar media is used as an alternative from transfusion. This study aims to observe the differences in the characteristics of Staphylococcus aureusbacteria on sheep blood agar media and human blood agar media which was carried out at BBLK Palembang. This type of research was conducted experimentally with a total sample of 32 and analyzed statistically. Bacterial suspension derived from the pure strain of Staphylococcus aureus ATCC 25923. The colony characteristics were observed macroscopically including colony morphology, colony number and colonydiameter. The average value of the number of colonies of Staphylococcus aureus bacteria on sheep blood agar and human blood agar media were 173 and 148, while the colony diameters were 2.5 mm and 2 mm, respectively. The results of the research data on the number of colonies were tested with an unpaired T test with p value = 0.001 and colony diameter data was tested with the Mann Whitney alternative test with p = 0.031, which means that there is a statistically significant difference between the characteristics of the Staphylococcus aureusbacteria colony on the media. sheep blood agar and human blood agar media.

Abstract

The ICU and operating rooms are places where sterility must be maintained. The purpose of this study was to determine the pattern of bacterial contaminants and their resistance in the ICU and operating rooms of several hospitals in Bandar Lampung. This type of research is descriptive, with a cross sectional design. Held in September – November 2019, at three hospitals which are BPJS referral hospitals in Bandar Lampung City, Indonesia. Samples were taken in the ICU and operating rooms by swab on walls, floors, beds, officers' clothes, equipment, and air samples. Sample examination includes microscopic examination, isolation, identification andantibiotic sensitivity testing. The results showed that from 54 samples examined, 61 isolates of contaminant bacteria were grouped into 11 bacterial species. The patterns of gram negative contaminants were Proteus morganii (19.7%), Pseudomonas aeroginosa (11.5%), and Proteus mirabilis (9.8%). The Gram positive bacteriawere dominated by Staphylococcus aureus (13.1%). Proteus mirabilis bacteria were resistant (16.7% - 100%) to 9 antibiotics used in this study. Klebsiella pneumonia, E. coli and Staphylococcus aureus, on average were 56.82%, 54.55% and 45.45% resistant to antibiotics. Some bacterial species are still sensitive to ciprofloxacinand meropenem. The conclusion is that pathogenic bacteria are still found on the surfaces of objects in the ICU and operating rooms, and these bacteria are resistant to antibiotics.

Abstract

Antioxidant compounds have the ability to inhibit or prevent oxidative damage to target molecules. Oxidative damage is one of the agents that play a role in the development of cancer cells. It is interesting to conduct research to find other anti-cancer compounds in order to minimize the side effects of these anti-cancer drugs.This study aimed to examine the antioxidant activity and cytotoxicity in vitro of acetone extract of cocoa pods (Theobroma cacao L.) on WiDr cells. This research is an experimental study, to see the effect of giving cocoa pod peel extract on the inhibition of free radicals, and WiDr cell death. Samples were extracted using 80% acetone. The antioxidant activity test was carried out using the DPPH method. The anti-cancer activity test was carried out using the MTT assay method. The test results obtained the IC50 value which indicates the cell death rate related to the concentration of the extract using linear regression analysis, as well as the percentage of antioxidant inhibition. Cocoa pod peel extract has an IC50 of 15.41 ppm and is classified as a very strong antioxidant. The IC50 value of the extract for vero cells was 528.13 ppm, while the IC50 value of the extract for WiDr cells was 47.47 ppm. This study showed that cocoa pod peel extract was classified as a very strong antioxidant component and was active in inhibiting the growth of WiDr cells and not actively inhibiting the growth of normal vero cells.

Abstract

Infeksi bakteri, jamur maupun virus dapat terjadi pada seorang penderita diabetes melitus. Penderita dengan metabolisme yang buruk lebih rentan terhadap infeksi bakteri yang berulang. Kerusakan gigi dapat terjadi pada penderita diabetes melitus sebab adanya peningkatan kadar glukosa dalam cairan salivanya, terkhusus pada penderita yang tidak memperhatikan kebersihan gigi dan mulut sehingga memicu pola bakteri. Tujuan penelitian ini adalah  untuk melihat gambaran pertumbuhan Streptococcus spp. pada mulut penderita diabetes melitus di kota Makassar. Jenis penelitian ini adalah penelitian deskriptif dengan melakukan observasi langsung secara laboratorik dengan 24 sampel penelitian yang merupakan penderita diabetes melitus di RSUP Dr. Tadjuddin Chalid Makassar, Rumah Perawatan Etn Center Indonesia, dan Klinik Azka Nadhifah. Penelitian ini dilaksanakan pada Mei –  Juni Tahun 2022 di Laboratorium Mikrobiologi Teknologi Laboratorium Medis Poltekkes Kemenkes Makassar dengan teknik pengambilan sampel secara Consecutive Sampling. Hasil penelitian menunjukkan dari 24 sampel terdapat 15 sampel teridentifikasi Streptococcus spp., 2 sampel teridentifikasi Pseudomonas spp., dan 7 sampel teridentifikasi Enterobacter spp.,. Disimpulkan terdapat pertumbuhan Streptococcus spp  pada mulut penderita diabetes melitus.

Abstract

Udara adalah campuran gas yang terdapat pada lapisan yang mengelilingi bumi. Komponen yang konsentrasinya paling bervariasi yaitu uap air dan CO2, kegiatan yang berpotensi menaikkan konsentrasi CO2 seperti pembusukan sampah tanaman, pembakaran atau sekumpulan massa manusia di dalam ruangan terbatas yaitu karena proses pernapasan.Sumber pencemaran yang utama berasal dari transportasi, dimana hampir 60% dari polutan yang dihasilkan terdiri dari karbon monoksida dan sekitar 15% terdiri dari hidrokarbon. Tujuan penelitian ini mengetahui Pengaruh Lamanya bekerja,lamanya kontak langsung operator, dan penggunaan APD petugas operator di SPBU terhadap Kandungan Oksigen di dalam darah Karyawan.Metode penelitian ini observasional analitik yaitu mencari hubungan antar variabel resiko dan efek yang analisisnya untuk menentukan ada tidaknya hubungan antar variable. Hasil penelitian ini Terdapat hubungan yang cukup erat serta adanya pengaruh yang signifikan antara lamanya bekerja petugas operator SPBU dan penggunaan APD pada operator SPBU dengan kadar oksigen dalam darah operator SPBU sebesar 23,4% dan 9,9%, sedangkan untuk lama nya kontak petugas operator SPBU tidak terdapat hubungan yang signifikan terhadap kadar oksigen dalam darah.
