Abstract

Kappa receptor is one opioid receptor subunit which when activated can stimulate analgesic effect, however with lower dependency risk compare to other opioid receptor subunit. (mu and delta). The objective of this experiment was to examine the effectivity of kappa opioid receptor agonist  in acute and chronic pain (inflammation and neuropathy), in order to find new strategy in pain management. Groups of ICR mice (n = 10) were treated to gain acute and chronic pain model. Acute pain was gain by hot stimulation through hot plate and tail flick. Inflammation model was made by CFA intraplantar injection. Neuropathy pain was induced by binding the sciatic neuron. To examine the pain-blocker effectivity of kappa receptor agonist,  trans-1S,2S]-3,4-dichloro-N-methyl-N-[2-(1-pyrrolidinyl)cyclohexyl]benzeneacetamide(-)U50,488H was inject subcutaneously 3 mg/kg - 20mg/kg body wight. Then hot plate and tail flick were conduct, morphine 10 mg/kg bw use as standard. From results can be concluded that   activation of kappa receptor can induce pain-blocker or analgesic effect as well, against acute or chronic pain, inflammation or neuropathy. ABSTRAK Reseptor kappa merupakan salah satu sub unit reseptor opioid yang jika diaktivasi dapat mempunyai efek analgesik tetapi dengan risiko dependensi yang lebih kecil dari pada sub unit reseptor opioid yang lain (mu dan delta). Penelitian ini bertujuan untuk menentukan efektivitas agonis reseptor opioid kappa pada keadan nyeri akut dan kronik (inflamasi dan neuropati), sehingga diharapkan didapatkan strategi baru dalam penanganan nyeri. Untuk pengujian efektivitas antinyeri dari agonis reseptor kappa, trans-1S,2S]-3,4-dichloro-N-methyl-N-[2-(1-pyrrolidinyl) cyclohexyl]benzeneacetamide   (-)U50,488H diinjeksikan secara subkutan mulai dosis 3 mg/kgbb hingga 20mg/kgbb kemudian dilakukan uji hot plate dan tail flick, sebagai pembanding digunakan morfin 10mg/kgbb. Pegujian dilakukan pada 15, 30, 45, 60, 90 dan 120 menit setelah penginjeksian (-)U50,488H. Aktivitas antinyeri dinyatakan dalam % MPE (maximal possible antinociceptive effect). (-)U50,448H memiliki aktivitas antinyeri sebanding dengan dosis pemberian baik pada keadaan nyeri akut, inflamasi maupun neuropati. Pada dosis 3 mg/kgbb menghasilkan 47% MPE, dosis 5,6 mg/kgbb menghasilkan 76% MPE, dosis 10mg/kgbb menghasilkan 88% MPE dan dosis 20 mg/kgbb menghasilkan 100% MPE. Waktu puncak dicapai pada 15 menit setelah injeksi. Dari penelitian ini dapat disimpulkan bahwa aktivasi reseptor kappa juga dapat memberikan efek anti nyeri, baik pada nyeri akut, inflamasi maupun neuropati.

Abstract

Government Regulation of The Republic of Indonesia number 51/ 2009 stated thatPharmacist must be the person that responsible for pharmaceutical wholesaler activity.Pharmaceutical wholesaler must implement Good Distribution Practices (GDP). There are79.045 types of registered pharmaceuticals that distributed by 2.821 pharmaceuticalwholesalers in 33 Indonesiaâ??s provinces. This research aimed to evaluate theimplementation of GDP on pharmaceutical wholesalers in Daerah Istimewa Yogyakarta(DIY) Province. This research conducted in July 2010 using questionnaire and interview to29 pharmaceutical wholesaler are willing to become respondents from 49 pharmaceuticalwholesaler listed in DIY. The products distributed by pharmaceutical wholesaler in DIYProvince which is the raw material of pharmaceuticals, vaccines, psychotropic, prescriptiondrugs, over the counter drugs, cosmetics, food, milk, and medical equipment. Theresponsible person in 83% pharmaceutical wholesaler is a woman, 38% chargepharmaceutical wholesaler 23-30 years old, pharmacist being the responsible person on31% pharmaceutical wholesaler, 52% had never been in charge GDP training. There are 3%pharmaceutical wholesaler which doesnâ??t have a Standard Operating Procedure, 21% donâ??thave the organizational structure, 59% didnâ??t have temperature control equipment, 34% donâ??thave humidity control equipment, 3% donâ??t carry out the documentation, and 10% didnâ??tconduct self inspections. ABSTRAKTerdapat 79.045 jenis sediaan farmasi yang berizin edar yang didistribusikan oleh 2.821PBF yang tersebar di 33 provinsi di Indonesia. Penelitian ini mengevaluasi implementasiCDOB pada PBF di Provinsi Daerah Istimewa Yogyakarta (DIY). Survei dilakukan bulan Juli2010 menggunakan kuesioner dan interview kepada 29 PBF yang bersedia menjadiresponden dari 49 PBF yang tercatat di Propinsi DIY. Hasil penelitian menunjukkan bahwaproduk yang disalurkan oleh PBF di Provinsi DIY yakni bahan baku farmasi, vaksin,psikotropik, obat keras, obat bebas, obat bebas terbatas, kosmetik, makanan, susu, danalat kesehatan. Sebanyak 83% PBF, penanggung jawabnya wanita, 38% penanggung jawabPBF berumur 23-30 tahun, 31% penanggung jawab PBF adalah apoteker, 52% penanggungjawab PBF belum pernah mengikuti pelatihan CDOB. Terdapat 3% PBF yang tidak memilikiStandar Operasional Prosedur, 21% tidak memiliki struktur organisasi, 59% tidak memilikialat pengontrol suhu, 34% tidak memiliki alat pengontrol kelembaban, 3% tidakmelaksanakan dokumentasi, dan 10% tidak melakukan inspeksi diri.

Abstract

The management of hypertension ideally requires collaboration between health professionals which includes in the three aspects: medical care, pharmaceutical care, and nursing care. The purpose of this study was to determine the impact of pharmacist counseling conducted on therapeutic outcomes in systolic blood pressure (SBP), diastolic blood pressure (DBP), body mass index (BMI), blood glucose levels (BGL) period, total cholesterol, and quality of life of patients were measured Short-Form Health Survey 36 (SF-36). This study was conducted at RSUD dr.R.M. Djoelham Binjai involved 60 primary hypertensive patients. Patients were divided into two groups: group counseling and with out conseling. Data were analyzed by independent sample t-test. The result of this study showed significant differences in SBP (p=0.048), BMI(p=0.014), BGL(p=0.002) and total cholesterol (p=0.000) and value of quality of life who received counseling (63.17%) and without counseling (54.80%) with p=0.001. But obtained no significant differences between DBP (p=0.068).Keywords : primary hypertension, pharmaceutical care, counseling.

Abstract

ABSTRACT : This research studied the potential and action mechanism of probiotic milk-guava leaf extract combination as an immunomodulator using mice as an experimental animal. The mouse were given the combination of probiotic milk-guava leaf extract for 7 days, then the levels of TNF-α in serum and lymphocites proliferation were measured. The results showed that the extract of guava leaf can increase levels of TNF-α in serum and the number of lymphoblast, either in single preparation or in combination with probiotic milk. However, administration of probiotic milk cannot improve the TNF-α in serum or increase the number of lymphoblast. It can be concluded that the immunomodulatory effects of the probiotics- milk-guava leaf extract combination was caused by the substances in the extract of guava leaf rather than by the probiotic milk. Keywords: probiotic milk, guava leaf extract, immunomodulator ABSTRAK: Konsorsium mikroba probiotik yang dikembangkan dalam bentuk sediaan susu probiotik telah dilaporkan memiliki berbagai aktivitas, antara lain sebagai imunomodulator dan antimikroba. Jus buah jambu biji telah digunakan secara tradisional oleh masyarakat untuk membantu meningkatkan jumlah trombosit penderita demam berdarah dengue. Kombinasi susu probiotik-ekstrak daun jambu biji telah terbukti mempunyai efek anti bakteri penyebab diare, namun mekanisme kerjanya belum diketahui. Dalam penelitian ini dilakukan kajian potensi dan mekanisme kerja kombinasi susu probiotik-ekstrak daun jambu biji sebagai imunomodulator. Sejumlah mencit diberi perlakuan kombinasi susu probiotik-ekstrak daun jambu biji selama 7 hari, kemudian diukur kadar TNF-α dalam serum dan kecepatan proliferasi limfositnya. Hasil penelitian menunjukkan bahwa ekstrak daun jambu biji dapat meningkatkan kadar TNF-α dalam serum dan dapat meningkatkan jumlah limfoblast, baik dalam bentuk tunggal maupun kombinasinya dengan susu probiotik. Pemberian susu probiotik saja tidak dapat meningkatkan TNF-α dalam serum ataupun meningkatkan jumlah limfoblast. Dari hasil penelitian ini dapat disimpulkan bahwa efek imunomodulator kombinasi susu probiotik-ekstrak daun jambu biji lebih disebabkan oleh kandungan zat zat berkhasiat dalam ekstrak daun jambu biji dari pada oleh susu probiotik. Kata kunci: probiotik, Lactobacillus acidophilus, jambu biji, Psidium guajava, imunomodulator.

Abstract

ABSTRACT : Antioxidants are nutrient that can prevent the oxidative damage in the body. Arabica coffee and pandan is most common species of plant found in Indonesia. Arabica coffee and pandan known as plants with antioxidant activity. This study aimed to determine and compare antioxidant activity of water and ether fraction from methanolic extract of Arabica coffee leaves, pandan leaves and both combination by DPPH method. Vitamin C was used as positive control. The result show that Arabica coffee leaves have antioxidant activity better than pandan leaves and also gives big donation of antioxidant activity in their combinations. Statistical analysis showed that antioxidant activity from all sample were significanly different (p<0,05).

Abstract

Traditional medication have been long enough been conducted by our ancestors since long ago and hereditary endowed from generation to generation. Research of useful plant for traditional medication of Sasak society who live in West Lombok have been done. Data collected by interview with chosen informan like the figure or tribe-head, and follow some of community daily activity and also field observation. The result showed that there were not less than 25 plant species exploited to cure of diseases. Useful plant and ethnobotanical aspect will be discussed in this paper. ABSTRAK Pengobatan tradisional sudah lama dilakukan oleh nenek moyang kita sejak jaman dahulu dan diwariskan secara turun temurun dari generasi ke generasi. Penelitian mengenai pemanfaatan tumbuhan untuk pengobatan tradisional suku Sasak di Lombok Barat telah dilakukan. Data dikumpulkan melalui wawancara dengan informan terpilih seperti kepala adat, dukun kampung dan mengikuti sebagian aktivitas harian penduduk serta observasi lapangan. Tercatat tidak kurang dari 25 jenis tumbuhan yang dimanfaatkan untuk mengobati berbagai macam penyakit. Cara pengolahan dan aspek etnobotani lainnya akan dijelaskan dalam makalah ini.

Abstract

We investigated and compared the cytotoxic activity of ethanolic extract of the rhizome of 3 species of Zingiberaceae plants that grow abundantly in Indonesia, i.e. Zingiber ottensii, Zingiber zerumbet, and Nicolaia speciosa in an in vitro tetrazolium salt assay using human breast cancer cell line MCF-7. The results showed that 2 out 3 extracts showed significant cytotoxic activity. i.e. Zingiber ottensii and Zingiber zerumbet with IC50 value respectively 60 and 50 ug/mL, while the ethanolic extract of Nicolaia speciosa showed considerable larger IC50, i.e. 625 ug/mL. From the results it can be concluded that the rhizome of Zingiber ottensii and Zingiber zerumbet have a good prospect to be further investigated and developed as raw materials for new cancer drug. ABSTRAK Telah dilakukan penelitian untuk membandingkan daya sitotoksik ekstrak etanol rimpang 3 jenis tumbuhan suku Zingiberaceae yang banyak tumbuh di Indonesia, yaitu bengle hantu (Zingiber ottensii L.), lempuyang gajah (Zingiber zerumbet L.), dan kecombrang (Nicolaia speciosa Horan) menggunakan galur sel kanker payudara MCF-7. Pertumbuhan sel dievaluasi menggunakan metode garam tetrazolium (MTT). Hasil penelitian menunjukkan bahwa 2 di antara 3 ekstrak rimpang yang diuji menunjukkan daya sitotoksik yang kuat, yaitu ekstrak etanol bengle hantu dan lempuyang gajah dengan nilai IC50 masing-masing sebesar 60 dan 50 ug/ml, sedangkan ekstrak etanol rimpang kecombrang memiliki IC50 yang jauh lebih besar, yaitu 625 ug/mL. Dari hasil penelitian ini dapat disimpulkan bahwa ekstrak etanol rimpang bengle hantu dan lempuyang gajah memiliki potensi besar untuk diteliti dan dikembangkan lebih lanjut menjadi sumber bahan baku obat anti kanker baru.

Abstract

Timolol maleat is one of the active substances of the non selective beta blocker, which is widely used for therapy or profilaxsis  aims of angina tectoris, hypertension and myocardial infarc. Transdermal drug delivery system of timolol maleat is to avoid first pass effect metabolism in the liver, that is will compose non active metabolits  as much as 50 %. The influence of addition of ethanol, propylen glycol, and polysorbates 20 as an penetrance enhancer towards timolol maleat diffusion from hydroxypropylmethyl cellulosa gel preparation had been studied in vitro . Spangler membran and diffusion cell are used for diffusion test  with distilled water at pH 7,0 as reseptor medium and at 37 ±1oC for 180 minutes. Quantitative analysis of timolol maleat was determined by spectrophotometry at 295 nm. The result showed that the diffusion significantly increased, i.e. after the addition of 30 % w/w of ethanol was 353,8 ±11,9 μg/cm2, of  30 %w/w of propylene glycol was 203,9 ±1,9 μg/cm2 , and of 5 % w/w of polysorbates 20 was 311,5 ±8,0 μg/cm2 . ABSTRAK Timolol maleat sebagai salah satu zat aktif golongan beta bloker non selektif telah banyak digunakan sebagai obat untuk terapi ataupun untuk tujuan profilaksis penyakit angina pektoris, hoeprtensi dan infark miokardial. Penghantaran obat timolol maleat secara transdermal untuk menghindari metabolisme lintas pertama dihati, yang akan terbentuk metabolit tidak aktif sebesar 50 %. Telah diteliti pengaruh penambahan etanol, propilenglikol dan polisorbat 20 sebagai peningkat penetrasi terhadap difusi timolol maleat dari sediaan gel hidroksipropilmetil selulosa secara in vitro. Digunakan membran Spangler dan sel difusi untuk uji difusi selama 180 menit dengan medium penerima air suling pH 7,0,  pada suhu 37 ±1oC.  Analisis  kuantitatif timolol maleat dilakukan dengan metode spektrofotometri pada panjang gelombang 295 nm. Terjadi peningkatan difusi timolol maleat secara bermakna pada formula dengan penambahan etanol 30 % b/b, propilenglikol 30 % b/b, dan polisorbat 20 5 % b/b dengan jumlah timolol maleat yang terdifusi dari masing-masing formula sebesar 353,8 ±11,9 μg/cm2, 203,9 ±1,9 μg/cm2, 311,5 ±8,0 μg/cm2 .

Abstract

The effect of aethanolic extract of mahakaan's (Gynura pseudochina (L) DC tuber on shortening bleeding and coagulation time and trombocytes cell of the white female mice has been studied. The overal doses used were 30,100 and 300 mg/kg BW. The effect was observed on 1st, 7th, 14th and 21st days by using the modified cutting tail method, slide method and using hemositometer. As a comparator used vitamin K with dose of 0,026 mg/20g BW was given. The result indicated that the extract has ability to shorten bleeding and coagulation time at all doses, and the dose of 300 mg/kg BW showed a stronger effect on shortening bleeding time compared to vitamin K 0,026 mg/20g BW (p<0,01). The doses of 100 and 300 mg/BW showed a stronger effect on shortening coagulation time. The thrombocytes was not influenced by the all doses, and neither was the lenght of the administration (p>0,01). keywords : Gynura pseudochina (L.) Abstract

ABSTRACT:Didymella bryniae strain MA71 endophytic fungus was isolatedfrom twigs of Biduri plant (Calotropis gigantea L.Dryand). It has anti yeastactivity and anti bacterial activity against gram positive and negative bacteria.Secondary metabolites were isolated via fermentation process using PDYmedia for 12 hr. Anti microbial activity of these fermentation product was assessedvia agar diffusion method using disk paper. Test result demonstratedthat extracellular secondary metabolites which was extracted using EtOAcwere active towards Staphylococcus aureus ATCC 25923, Escherichia coliATCC 25922 and Bacillus subtilis ATCC 6633. On the other hand, these EtOACextracted metabolites were also active against Candida albicans ATCC 10231.Its inhibitory zone were 17.3 mm, 17.2 mm, 17.41 mm and 11.6 mm respectively.Chloramphenicol was used as positive control for bacteria and nystatinfor yeast. Overall, the se condary metabolites produced by fermentation of endophyticfungi Didymella bryniae strain MA71 have broad spectrum of antibacterial and yeast activity.Keywords: endophytic Fungus, shaked fermentation, secondary metabolites,antimicrobials, and Calotropis gigantea L.
