Abstract

Fenugreek seed (Trigonella foenum-graecum L.) is one of Indonesian medicinal plant with many known activities such as antidiabetic, antihypercholesterolemia, phytoestrogen and anticancer. Fenugreek seed contains steroidal sapogenines (i.e. diosgenin, yamogenin, gitogenin, tigogenin and trigoneoside), alkaloid trigonellin and flavonoid (i.e vitexin, orientin). This experiment was carried out to investigate the cytotoxic activity and its potency to induce apoptotic on breast cancer cell line, MCF-7. Cytotoxic activity was carried out by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium) method. Fenugreek seed was prepared by extraction (methanolic and ethanolic extract) and fractionation using solvents with different polarity (hexane, ethylacetic, buthanolic and water fractions). The potency of apoptotic induction assay was carried out using flowcytometric method staining with propidium iodide (20μg/mL) on 100 ppm sample. Apoptotic potency was evaluated based on % population of cells in subG0/G1 phase. Results showed that ethylacetic fraction has the lowest IC50 (41.81 ppm) and significantly (p<0.05) increase % population of cells in subG0/G1 phase (20.87%) on 100 ppm concentration. It is concluded that fenugreek seed posses cytotoxic activity and potency of apoptotic induction on MCF-7 cells.

Abstract

Tujuan penelitian ini adalah memformulasi genistein menjadi nanoemulsi dan membandingkan penetrasinya dengan produk Gen90 Nano. Dibuat sebanyak 3 jenis formula dengan perbandingan komposisi antara genistein dan lesitin soya menggunakan metode emulsifikasi spontan. Hasilnya formula 3 merupakan formula terpilih yang menghasilkan nanoemulsi berukuran 191,7 nm dengan indeks polidispersitas 0,171 dan potensial zeta -47,5 mV. Uji penetrasi secara in-vitro menggunakan sel difusi Franz menunjukkan jumlah kumulatif terpenetrasi dari nanoemulsi genistein sebesar 18,29 ± 0,16 (μg/cm2) dibandingkan dengan produk Gen90 Nano sebesar 24,60 ± 0,57 (μg/cm2). Perbandingan ini dilakukan untuk melihat kualitas penetrasi dari formula nanoemulsi genistein, karena Gen90 Nano merupakan produk paten dari kompleks inklusi hidroksipropil siklodekstrin-genistein

Abstract

Parasitic plant [Dendrophthoe pentandra (L.) Miq.] on the masisin [Rhodomyrtus tomentosa (Aiton) Hassk.] was traditionally used for cancer treatment in Central Kalimantan. Maceration of dried Dendrophthoe pentandra using ethanol followed by ethyl acetate produced ethanol and ethyl acetate extracts which exhibit inhibitory activity against leukemia L1210 cells with IC50 of 17.3 and 20.9 µg/mL, respectively. Fractionation of ethyl acetate extract by column chromatography yielded 7 fractions (F1~F7). Inhibitory activity test on fractions F1~F7 against leukemia L1210 cells showed inhibitory activity with IC50 0f 19.9, 19.0, 20.7, 16.0, 13.7, 12.5, and 17.4 µg/mL, respectively. During evaporation of F5 by rotary evaporator, a yellowish precipitate was observed and identified as quercitrin (quercetin 3-O-rhamnoside). Inhibitory activity test on quercitrin against leukemia L1210 cell showed that the crystal has an IC50 of 14.7 µg/mL. Separation of F7 by semipreparative HPLC reverse phase column yielded isolate A and B which exhibited inhibitory activity on leukemia L1210 cells with IC50 of 15.0 and 14.4 µg/mL, respectively. Subsequent solvent partition of ethanol extract into ethyl acetate-water and separation of ethyl acetatesoluble portion by column chromatography produced 7 fractions (F8~F14). Further separation of F14 by column chromatography yielded quercitrin and isolate C which has not been identified yet.

Abstract

Caesalpinia major Dandy ex Excell is one of the traditional medicinal plants from Nusa Tenggara Timur Province (Flores island, Ruteng) which the local people called as “Dekar". The root of this plant is used as anthelmintic, anti rheumatic, back-ache and also as tonic. Chemical structure elucidation of isolated compound from C. major caesaldekarin A has been done and it's based on ultraviolet, infrared, nuclear nagnetic resonance and mass spectrometry: To know the correlation between proton and proton, proton and carbon, carbon and carbon, NMR two dimensional (2D) have been also interpreted by the data of 'H-'H COSY, 13C-'H COSY, COLOC, HMBC and INADEQUATE. The stereochemistry of the relative structure of caesaldekarin A is determined by 2D NMR NOESY.

Abstract

Gold nanoparticles (AuNP) are potentially developed as nanomedicine because AuNP is easily synthesized, functionalized, and biocompatible. With gum arabic as a stabilizer, vincristine was conjugated with gold nanoparticles. As a reducing agent, it used 0.02 M Natrium Boro Hidrat (NaBH4) solution. Gold nanoparticles (AuNP) coated with conjugated gum Arabic (GA) and vincristine (VCR) were successfully synthesized and characterized. The conjugation of GA-VCR and AuNP displayed a narrow hydrodynamic particle size distribution with average size < 100 nm by TEM and PSA (particle size analyzer). We investigated the cytotoxic activity of conjugated vincristine-gum arabic-gold nanoparticle by tetrazolium salt assay (MTT) using cancer cell line CCR-CEM. Cytotoxic activity of conjugated VCR-GA-AuNP before and after purification by Size Exclusion Chromatography (SEC), against leukemia cell line CCRF-CEM, was described by IC50 value. All formulation had a cytotoxic of activity with IC50 <20 μg/ml. The IC50 of samples against CCRF cell line were 1,026 μg/mL and 2,607 ug/mL, respectively.

Abstract

Gold sea cucumber extract (Stichopus hermanni) is rich in collagen and unsatturated fatty acid also known omega 3 such as docosahexaenoic acid and eicosapentaenoic acid that highly effective as an antioxidant. Oligomeric proanthocyanidin is the main content in grape seed oil and the most powerful antioxidant agent that can protect collagen. The aim of research was to determine the effect of gold sea cucumber extract concetration on antioxidant activity of grape seed oil nanoemulsion. The three formulas with results test of antioxidant activity with DPPH method using 15% Brij CS12® and 30% propylene glycol has done which the oil phase was dispersed in the aqueous phase.Variation that have been done is to use the antioxidant activity (IC50) of gold sea cucumber extract for 2, 4, and 6 times. The resulting preparations was conformed of organoleptic, nanoemulsion type, clarity, globule size, viscosity and rheology, pH and freez thaw test. Antioxidant activity of gold sea cucumber extract and grape seed oil were (IC50= 53.79 and 17.41 μg/mL respectively). The nanoemulsion was analysed using two-way ANOVA method. Antioxidant activity at storage for 0; 2 and 4-weeks of formula I, II, III were (IC50= 49.15; 49.45, 51.15 µg/mL), (IC50= 44.12, 47.48, 48.14 µg/mL), and (IC50= 42.14, 43.45, 45.33 µg/mL) respectively. Can be concluded that the higher concentration of gold sea cucumber extract, the antioxidant activity of nanoemulsion is higher, but it decreased during the storage.

Abstract

Chitosan is a dietary iber that has been used as an antihyperglycemia agent. Irradiation of chitosan resulted in 1,4-β glycosidic chain termination which shortened the chitosan chains, lower its molecular weight, and minimize steric effects. This is a preliminary experimental study to ind the effective irradiation dose and chitosan type in lowering blood sugar levels. Fifty two mice were used in this research and divided into various treatment groups which included: normal control, negative control, positive control (Acarbose), non-irradiated chitosan and irradiated chitosan. Chitosan solution were given for 15 days and then oral glucose tolerance test were conducted using 1.5 g/kg bw of glucose. Blood sugar levels were monitored at minute 0, 30, 90, 120, and 150; and Area Under the Curves (AUC) were calculated. The results showed that the irradiation of chitosan can lower blood sugar levels greater than the non-irradiated chitosan; and chitosan irradiation of 100 kGy dose I and II can lower blood sugar levels better than irradiated chitosan and positive control. It was concluded that although the 100 kGy irradiated chitosan appears to have greater ability than the positive control group in lowering blood sugar levels. But after LSD test towards the AUC value, it appears that both groups have the same effectivity.

Abstract

This investigation is intended to find a simultaneous assay method for glucocorticosteroids declared as doping compound by the International Olympic Committee. Experiments were carried out in the determination of dexamethason and methylprednisolon in urine matrix by gas chromatography-mass spectrometry (GC-MS) using various derivatisation reagents, reaction temperature and time. Selection of the best derivatisation reagent was based on the separation of total ion chromatogram (TIC) and appearance of three characteristic ions, m/z 305, 345,680 or m/z 206,456, 662 for dexamethasone; and m/ z 279, 369, 459 or m/z 147, 424, 644 for methylprednisolone. Dexamethasone and methylprednisolone derivates in the GC-MS spectra were identified by characteristic ions of m/z 305 (dexamethasone) and m/ z 279 (methylprednisolone). The best derivatisation yield was achieved by a combination of TMSI - [BSTFA-TMCS (99:1)](2:1) reacted at 90°C for 90 minutes, which produced a linear correlation between concentration (0-150 ng/ml ) and ratio of quantitation-ion abundance of standard compound/ion abundance of D3-testosterone internal standard with r= 0.9937 for dexamethasone and r=0.9823 for methylprednisolone; high precision CV=12.14% (dexamethasone) and CV=13.87%(methylprednisolone); high recovery for both dexamethasone and methylprednisolone, and LOD values of 4,44ng/ml (dexamethasone) and 1.80ng/ml (methylprednisolone). The method can be used for simultaneous quantitative assay of glucocorticosteroids, and has been validated for ten other glucocorticosteroid compounds.

Abstract

Diabetes mellitus (DM) is a metabolic disorder caused by a defect in insulin secretion or insulin action. Number of diabetic patients will increase if strategies are not prepared for preventing and controlling diabetes. This study aimed to evaluate the outcome of pharmacist education on the level of knowledge, level of medication adherence and diet, as well as the A1C value of type 2 diabetic patients in Dr. Sardjito Hospital Yogyakarta. This research was conducted prospectively using Nonrandomized Concurrent Control Trial. The number of subjects recruited were based on a three-months period and subjects were taken by consecutive sampling method. There were two groups: one group received education intervention for 3 months, and the other group as control did not receive education. Data were analyzed descriptively and quantitatively using Independent T and T-Paired test. The results showed that patients in the control group have developed an increase in the level of knowledge and adherence by themself, on the other hand patients in this group showed a decrease in A1C values, but the decrease was not as big as patients in the intervention group. Patient’s adherence to diet did not improve in either control nor intervention group. It can be concluded that education given by pharmacist can increase knowledge and adherence towards medication and decrease the A1C values of DM type-2 patients.

Abstract

Teknik mutasi dengan iradiasi telah terbukti meningkatkan kandungan artemisinin dalam daun . Pada penelitian ini dilakukan uji aktivitas antimalaria ekstrak daun galur radiasi 10, 20, dan 40 Gy serta optimasi dosis terapi bertujuan untuk mendapatkan ekstrak daun yang mempunyai aktivitas antimalaria yang terbaik dan dosis terapi yang optimum. Penelitian Tahap Optimasi dosis radiasi dilakukan secara in vivo dengan menggunakan mencit yang diinfeksi dengan 1 x 107 P. berghei yang di bagi menjadi 4 kelompok yaitu kontrol positif, dosis 10 Gy, dosis 20 Gy, dosis 40 Gy. Pemberian artemisinin dilakukan selama 3 hari setelah derajat parasitemia telah mencapai 5%. Pengamatan parasitemia dilakukan setiap hari hingga hari ke-7 paska terapi. Hasil penelitian diketahui artemisinin 10 Gy memberikan hasil yang terbaik karena pertumbuhan parasitnya paling rendah dan mencit bertahan hidup lebih lama. Tahap optimasi dosis terapi dilakukan dengan metode yang sama dengan variasi dosis terapi 100, 200, dan 300 mg/kg BB. Hasil penelitian menunjukkan ekstrak daun galur mutan bisa menghambat pertumbuhan P.berghei secara signifikan (p <0,05) terhadap kontrol dengan Efektif dosis (ED50) dari 238,7 mg /kg BB. Kesimpulan ekstrak daun galur mutan 10 Gy yang terbaik dengan efektif dosis terapi (ED50) 238,7 mg/kg BB.
