Abstract

Endophytic fungi Kabatiella caulivora var B, have been isolated from Alyxia reinwardtii which was collected at Purwodadi Botanical Garden, Pasuruan, East Java. The objective of this present work is to asses the antimicrobial activity from extracts of the endophytic fungi Kabatiella caulivora var B against Bacillus subtilis, Escherichia coli, Salmonella thypi, Pseudomonas aeruginosa, Staphylocooccus aureus and Candida albicans. Disc diffusion method was conducted in this project with two concentration extracts (10 mg/disc and 5 mg/disc). The results showed that the methanol and ethyl acetate extracts of Kabatiella caulivora var B both have antimicrobial activity against all microbial tested, but the methanol extract showed negative activity against Staphylocooccus aureus and Candida albicans at concentration 5 mg. The discovery of endophytic fungi that produces bioactives metabolites has opened a promising new approach to obtain drugs on a commercial scale from microbes that grow inside plants.

Abstract

Generally guajava is known as a plant, which all of its parts are used as adstringent. But research results shown that this plant has some efficacies, for instance us analgesic, antibacterial, antidiabetic, antispasmodic, anti inflamation etc. Recently the people also use the ripe guajava fruit juice, especially the red one, as a remedy for of dengue infection, which its scientific background is not known yet. Therefore many research activities should be done in order to know what efficacies are shown by any part of guajava plant.

Abstract

Psidium guajava leaves have been proven be antiallergic. From the results of previous research, the ethanol extract of leaves is included in the class materials "Practically Non-Toxic." However, repeated administration may cause toxic effects on body organs, one of which was gaster. Subchronic toxicity had been carried out on 20 male Wistar rats and 20 female wistar albino rats. The test materials were administered orally, every day for 28 days. There are 3 (three) doses of test materials which were 250 mg, 500 mg, and 750 mg/kg BW. Observation of the general toxic effect was performed everyday, then organ sampling was performed on day 29 to examined gaster histology and score using Barthel-Manja methods. The result showed that administration of Psidium guajava leaves extract for 28 days caused physical and behavioral changes, caused soft and mucous feces, and also respiratory disorder. But the extract at dose 250 mg, 500 mg and 750 mg/kg BW did not caused changes on gaster histology profile and score. In conclusion, the extract did not have subchronic toxicity effect on gaster rat, but might caused general toxic effect in the higher dose.

Abstract

This study evaluated heavy metal contamination (lead, copper and cadmium) of canned pineapple. The research was done towards some canned pineapples with different expiry date, one year, two years and three years using wet digestion and atomic absorption spectrophotometry. The result showed that the lead contamination values of 1, 2 and 3 years expiry date canned pineaplles are 0.93 ppm, 0.73 ppm and 0.23 ppm. The copper contamination values are 2.15 ppm, 1.56 ppm, 0.92 ppm while the cadmium contamination values are 0.02 ppm, 0.02 ppm, 0.01 ppm. The closer to the expiry date, the higher values of heavy metal contamination.

Abstract

Cempaka kuning (Michelia champaca) which is belongs to Magnoliaceae family, has been traditionally used for treatment fever. This research was conducted to evaluate antimalarial activity of Cempaka kuning stembark extract against Plasmodium falciparum 3D7 in vitro. Cempaka kuning stembark was extracted with n-hexane, chloroform and methanol yielded n-hexane extract, chloroform extract and methanol extract, followed by antimalarial activity assay of these extracts. Phytochemical screening showed that n-hexane extract contain volatile oil and terpenoids, chloroform extract contain Volatile oil, terpenoids and fiavonoids, then methanol extract contain volatile oil, terpenoids, flavonoids, tannins and glycosides. Result of this study also showed that n-hexane extract, chloroform extract and methanol extract were active against P. alciparum 3D7 in vitro, with an IC50 value of 0.36, 0.24 and 1.00 µg/mL. The findings indicate that Cempaka kuning stembark extracts possess strong antimalarial activity and are prospective to be developed as antimalarial.

Abstract

Neuropathy pain is a pain that caused by nerves injury. Nowadays, treatment for neuropathic pain change to drugs that works as GABA agonist and cause reimbalance between excitatory and inhibitory neurotransmitter in central nervous system (CNS). The present study was designed to investigate the effectiveness of gabapentin and baclofen in decreasing N-Methyl-D-Aspartate (NMDA) receptor NR2B subunit activity in neuropathic pain. Fourty mice were divided into 8 groups i.e sham, negative control, gabapentin (10, 30, 100 nmol) and baclofen (1, 10, 30 nmol). Neuropathic pain was induced by ligation of sciatic nerve with Partial Sciatic Nerve Ligation (PSNL) method). Treatments were administrated intrathecally once a day for seven consecutive days, at a week after induction. On day 15th, mice were sacrified and the spinal cord were removed quickly. The expression of NMDA receptor NR2B subunit were examined with imunohistochemistry and data were analyzed by one way anova. The result from this research was gabapentin and baclofen administration significantly decrease expression of NMDA receptor NR2B subunit in mice compared to sham group. The higher the dose, the more effective to decrease the number of neuron that express NR2B. The conclusion of this research was gabapentin and baclofen treat neuropatic pain by decreased the number of NMDA receptor NR2B subunit.

Abstract

Transdermal transport was an alternate to propranolol HCl delivery to overcome the low bioavailability by oral route. Kinetics of transdermal transport can be predicted by software WinSAAM. The aim of this research was to know kinetics of propranolol HCl transdermal transport with the present of enhancer oleic acid, propylene glycol and iontophoretics. Propranolol HCl transdermal transport was examined through the hairless rat as membrane on the vertical diffusion cell in the in vitro permeation. Enhancement methode used was oleic acid in propylene glycol and iontophoretics at varying concentrations. The donor phase contained 5 mg/mL propranolol HCl in citrate buffer, and the acceptor phase contained phosphate buffer saline at pH 7.4. Results of propranolol HCl transdermal transport analyzed by WinSAAM software. Parameters of transdermal transport were the rate of mass transfer from donor compartement to skin (Ka), available dose to transport (AD), and the rate of mass transfer from skin to acceptor compartement (KR).The results indicated that propranolol HCl transdermal transport with the present of enhancer can be explained by three compartement model and first order kinetics. Theoretically, value of AD influenced by oleic acid, interaction of oleic acid-iontophoretics and interaction of propylene glycol-iontophoretics. Value of Ka inliuenced by iontophoretics and interaction of oleic acid-propylene glycol-iontophoretics. Value of KR induenced by iontophoretics.

Abstract

Isolation and identitication of secondary metabolites from root of Jatropha gossypyolia, and also its biological activity test toward microbes and tumor cells have been conducted, Isolation was carried out by using vacuum liqiud chromatography (VLC) method (adsorbent: silica gel, eluent: mixture of n-hexane, ethylacetate, chloroform, and methanol). The isolate molecular structure Was determined by spectroscopic technique i.e. FTIR, NMR-ID, 13C-DEPT, NMR-2D, comparing the data obtained with related data from literatures. Its biological activity was evaluated against bacteria (Acetobacter sp., Escherecia coli, Staphylococcus aureus, and Streptococcus sp.), fungi (Aspergillus nigcr, Pentcillium sp. [grey], Penicillium sp. [White] and Rhizopus sp.), and murine leukemia P-388 cells. The results showed that the isolate is a white needle crystal which is identified as jatrophone. Jatrophone is very active toward Acetobacter sp. (bacteria) and A. niger (fungi) with growth inhibition Zone of 3,6 cm and 4.4 cm, respectively. Meanwhile, jatrophone has low activity against murine leukemia P-388 cells with IC50, > 100 µg/mL.

Abstract

It has been proven that Centella asiatica (L.) Urb. herbs have an antioxidant activity that make it possible to used in preventing degenerative illness. Centella asiatica (L.) extract has been formulated to fast disintegrating tablet (FDT) dosage form. The aims of this study are to find the optimum formula of FDT of Centella asiatica (L.) Urb. The simplex lattice design method with 3 factors (which are mannitol as diluents, crospovidone as superdisintegrant and povidone as binder) and 2 levels of each was used. Centella asiatica (L.) Urb. extract was produce from maceration process using ethanol-water in ratio 75%:25% as a solvent. Direct compress method was used to produce the FDT. The results show that the optimum composition between excipients FDT formula could be reached, which fullfiled the physical properties parameter which are the hardness, friability and disintegration time of FDT.

Abstract

Trichoderma is well known industrial fungi producing many kinds of industrial enzymes and as biological control agent. The applications of modern biotechnology using Trichoderma or its derived metabolites. Considering to the variety of the species of Trichoderma and enzymes or metabolites produced by Trichoderma lead to create a fusan of Trichoderma. The first step toward protoplast fussion is protoplasm isolation. In this experiment has been conducted protoplasm isolation of Trichoderma harzianum, T. viride, T. aureoviride and T. pseudokningii using lysing enzyme in room temperature for 1- 5 hours incubation. The aim of this experiment is to find the best condition and time of incubation in producing protoplast optimally. Trichoderma mycelia (1 mg) sink in osmotic stabilizer solution (pH 5.8), crushed and add 1 ml lysing enzyme 2%, then incubate in shaker incubator (room temperature) for 5 hours. Observation of the protoplast is conducted using haemacytometer for 1 until 5 hours incubation. After 5 hours incubation in room temperature showed the T. harzianum protoplast is the highest number of protoplast (40,150 protoplast/ml), followed by T. pseudokoningii (10,375 protoplas/ ml), T. viride (15,075 protoplast/ml) and T. aureoviride (10,050 protoplas/ml).
