Abstract

Olopatadin HCl merupakan antihistamin generasi kedua dengan mekanisme kerja ganda, yaitu antagonis selektif reseptor histamin H₁ dan stabilisasi membran sel mast, yang efektif untuk terapi alergi seperti konjungtivitis dan rinitis alergi. Penelitian ini bertujuan memvalidasi metode spektrofotometri UV–Vis untuk penetapan kadar olopatadin HCl dalam sediaan obat tetes mata yang beredar di Indonesia. Metode dikembangkan dengan larutan standar pada rentang konsentrasi 20–80 ppm, penentuan panjang gelombang maksimum, serta pengujian parameter validasi meliputi linearitas, akurasi, presisi, dan sensitivitas. Hasil menunjukkan panjang gelombang optimum pada 299 nm dengan persamaan regresi y = 25262x + 91851 (r = 0,99). Nilai perolehan kembali sebesar 105,63% dan koefisien variasi 0,52% memenuhi kriteria AOAC, sedangkan batas deteksi (LOD) dan batas kuantifikasi (LOQ) masing-masing sebesar 0,05 ppm dan 0,16 ppm. Kadar olopatadin HCl dalam sampel diperoleh sebesar 0,095 ± 0,001%. Metode ini terbukti sederhana, ekonomis, dan andal, sehingga layak diterapkan untuk pengendalian mutu rutin di laboratorium penelitian maupun industri farmasi.

Abstract

This study aimed to evaluate the antioxidant activity and toxicity of the ethanolic extract of red mangrove leaves (Sonneratia caseolaris). Extraction was conducted by maceration using 96% ethanol. Antioxidant activity was tested using the DPPH and ABTS methods with vitamin C as a reference, while toxicity was determined through the Brine Shrimp Lethality Test (BSLT). The extract showed very strong antioxidant activity with IC50 values of 37.67 ppm (ABTS) and 4.25 ± 3.05 ppm (DPPH). LC-HRMS identified TEMPO, Choline, and Betaine, with TEMPO showing the highest antioxidant potential. The BSLT result indicated non-toxic properties (LC50 >1500 ppm). The extract shows promise as a safe and effective natural antioxidant source.

Abstract

Penelitian ini merupakan kajian literatur terhadap tiga puluh artikel yang membahas pemanfaatan spektrofotometer UV-Vis dalam berbagai penelitian berbasis jahe merah (Zingiber officinale var. rubrum). Hasil studi literatur menunjukkan bahwa instrumen ini umumnya digunakan untuk analisis kadar total fenolik, flavonoid, dan aktivitas antioksidan menggunakan metode Folin–Ciocalteu, AlCl₃, serta DPPH. Data yang diperoleh dari spektrofotometri berperan penting dalam mendukung berbagai temuan ilmiah yang baru, mulai dari validasi metode analisis hingga pengembangan formulasi inovatif seperti minuman fungsional dan nanoenkapsulasi bahan aktif. Dengan segala kemudahan dan kesederhanaannya, spektrofotometer UV-Vis memberikan kontribusi besar dalam menjembatani analisis kimia dasar dengan pengaplikasiannya di bidang farmasi, pangan, dan bioteknologi berbasis bahan alam.

Abstract

Metabolit sekunder adalah senyawa bioaktif krusial dalam berbagai industri yang memerlukan analisis mutu terstandar. Kompleksitas struktural senyawa ini menjadikan Kromatografi Cair Kinerja Tinggi (KCKT) sebagai metode standar emas untuk penetapan kualitatif dan kuantitatif. Namun hasil HPLC sangat bergantung pada optimasi kondisi instrumen dan matriks sampel. Tinjauan literatur ini bertujuan untuk membandingkan implementasi KCKT dalam analisis menyeluruh golongan metabolit sekunder. Kajian dilakukan secara sistematis terhadap jurnal ilmiah nasional dan internasional dalam 10 tahun terakhir (2015-2025). Hasil pengamatan menunjukkan KCKT terbukti efektif untuk menganalisis beragam senyawa metabolit sekunder pada matriks sampel yang luas, mulai dari ekstrak tumbuhan hingga produk komersial. Ditemukan variasi signifikan pada kondisi—seperti pemilihan kolom C18, komposisi fase gerak, dan panjang detektor gelombang—yang menegaskan perlunya optimasi metode yang spesifik untuk setiap analit. Tinjauan ini menyimpulkan bahwa KCKT adalah metode yang ampuh dan serbaguna, serta berfungsi sebagai panduan ilmiah bagi peneliti dalam menentukan kondisi analisis yang optimal.

Abstract

Natural ingredients from plants are a major source of volatile compounds with biological activities such as antimicrobial, antioxidant, anti-inflammatory, and antifungal, particularly in essential oils. Testing of these compounds relies on reliable techniques such as gas chromatography-mass spectrometry (GC-MS), which separates and identifies components through retention time and mass spectra, with validation ensuring linearity, precision, accuracy, limit of detection (LOD), and limit of quantification (LOQ). This review article assesses the use and validation of GC-MS methods for the analysis of volatile compounds from natural ingredients, based on national and international literature over the past five years. The review approach included a structured search of databases such as PubMed, ScienceDirect, Google Scholar, and SINTA journals, with a focus on validation parameters and GC-MS instrument settings. The findings show that GC-MS stably detects key compounds such as monoterpenes and sesquiterpenes with 97-99% accuracy, and validates them according to AOAC standards (R² > 0.99; RSD < 8%; recovery 80-100%), where different extraction techniques affect their biological content and effectiveness. GC-MS validation is important to encourage innovation of environmentally friendly natural products in the pharmaceutical, food, and cosmetic sectors as a substitute for synthetic preservatives.

Abstract

Skin aging and impaired wound healing are complex biological processes involving chronic inflammation and excessive degradation of the extracellular matrix. Matrix metalloproteinases (MMP-1, MMP-3, and MMP-9) and cyclooxygenase-2 (COX-2) play crucial roles in these processes and are considered important molecular targets for anti-aging and wound healing therapies. Kaempferol is a natural flavonoid compound known for its antioxidant and anti-inflammatory activities; however, its molecular interaction mechanisms with these targets remain incompletely understood. This study aimed to evaluate the potential of kaempferol as an anti-aging and wound healing agent through a molecular docking approach targeting MMP-1, MMP-3, MMP-9, and COX-2. Molecular docking simulations were performed using Molegro Virtual Docker 6.0, while ligand-protein interactions were visualized and analyzed using BIOVIA Discovery Studio Visualizer v.21.1.0.20298. The docking results indicated that kaempferol exhibited favorable binding affinities toward all target proteins, with the strongest interactions observed for MMP-9 and COX-2. Kaempferol formed conventional hydrogen bonds as well as hydrophobic and aromatic interactions within the active sites of the proteins. In conclusion, kaempferol demonstrates potential as a multitarget natural compound for anti-aging and wound healing applications, although further in vitro and in vivo studies are required to validate its biological activity.

Abstract

Spektrofotometri UV-Vis ialah metode analisis yang banyak dipakai karena prosedurnya sederhana, biaya yang relatif rendah, serta sensitivitas yang memadai. Review artikel ini bertujuan untuk meninjau penerapan spektrofotometri UV-Vis dalam validasi metode analisis pada senyawa fenolik dalam berbagai sampel. Kajian dilakukan secara sistematis terhadap jurnal ilmiah nasional dan internasional yang dipublikasikan dalam sepuluh tahun terakhir (2015-2025) melalui penelusuran beberapa basis data elektronik dengan kriteria inklusi dan eksklusi. Artikel terpilih dianalisis secara kualitatif dengan fokus pada parameter-parameter validasi metode, yang meliputi linearitas; akurasi;presisi; batas deteksi (LOD) ; batas kuantitasi (LOQ). Hasil review bahwa spektrofotometri UV-Vis telah diaplikasikan secara luas dalam analisis senyawa fenolik serta menunjukkan pemenuhan terhadap kriteria validasi metode yang berlaku. Review ini diharapkan dapat memberikan gambaran mengenai keandalan spektrofotometri UV-Vis dalam validasi metode analisis senyawa fenolik.

Abstract

Vaginal infections causing leukorrhea (fluor albus) are a major reproductive health issue affecting many women, with the prevalence of Candida albicans fungal infections reaching 25–50%. Areca nut (Areca catechu L.) seeds are known to possess antimicrobial properties. This study aimed to develop a film soap formulation using 96% ethanol extract of areca nut seeds and to evaluate its antimicrobial effectiveness. The research was conducted experimentally in the laboratory, including sample collection and identification, preparation of 96% ethanol extract of areca nut seeds, and testing of antimicrobial activity against Candida albicans using the disk diffusion method. The results showed that the 96% ethanol extract of areca nut seeds contained several phytochemical compounds, including alkaloids, flavonoids, saponins, tannins, phenols, triterpenoids/steroids, and glycosides. Evaluation of the film soap included organoleptic tests, homogeneity, thickness measurement (0.13–0.4 mm), pH determination (4.67–5.00), and foaming ability (1.3–1.8 cm), all of which met the required standards. However, water content (36.45–43.40%) and weight uniformity (0.26–0.39 g) did not meet the requirements, indicating the need for further optimization. The antimicrobial activity test of the film soap showed average inhibition zone diameters at extract concentrations of 0%, 2.5%, 3%, and 3.5% were 10.4 mm, 15.3 mm, 17.4 mm, and 20.8 mm, respectively. These values indicated moderate to strong antimicrobial activity. The optimal concentration of antimicrobial film soap from areca nut extract was found at 3.5% (F3), as demonstrated by the inhibition zone diameter of 20.8 mm. This study concludes that the 96% ethanol extract of areca nut seeds can be formulated into film soap with antimicrobial activity comparable to that of the positive control.

Abstract

Validasi metode merupakan suatu tindakan penilaian terhadap parameter tertentu, berdasarkan percobaan laboratorium untuk membuktikan bahwa parameter tersebut memenuhi persyaratan penggunaannya. Review artikel ini bertujuan untuk menilai tingkat mutu baik suatu bahan ataupun sediaan farmasi dengan menggunakan parameter validasi metode pada alat Kromatografi Cair Kinerja Tinggi (KCKT) untuk mencari senyawa Kurkumin. Kajian ini diterapkan dengan melakukan pemilihan dan pencarian literatur dengan menelusuri beberapa jurnal penelitian dari skala nasional maupun internasional yang dibatasi pada publikasi selama 10 tahun terakhir (2016-2025) melalui basis data elektronik seperti Google Scholar, Pubmed, ScienceDirect, Mendeley berdasarkan kriteria inklusi dan eksklusi dengan menggunakan kata kunci yang relevan dengan topik penelitian. Artikel yang sudah dipilih dianalisis menggunakan data hasil analisis kualitatif dan kuantitatif dengan fokus parameter validasi metode, meliputi akurasi, spesifisitas, presisi, linieritas, batas deteksi (LOD), batas kuantitasi (LOQ). Hasil review artikel ini menunjukkan bahwa Kromatografi Cair Kinerja Tinggi (KCKT) mampu memisahkan kurkumin dari matriks yang rumit dengan nilai akurasi tinggi dengan rentang pemulihan sebesar 98% - 102% serta presisi yang bagus dengan nilai %RSD ≤ 2%, sehingga sangat cocok digunakan untuk pengendalian kualitas produk yang mengandung kurkumin.

Abstract

This study aims to isolate arbutin compounds from 70% ethanol extract of breadfruit (Artocarpus altilis) leaves and stems, determine the levels of arbutin in the extract, and evaluate the inhibitory activity of tyrosinase enzyme. Isolation was carried out through extraction and fractionation methods using n-hexane, ethyl acetate, and butanol solvents. Identification of arbutin was carried out by High Performance Liquid Chromatography (HPLC) based on the suitability of retention time to the standard. The results showed that the level of arbutin in 70% ethanol extract of breadfruit leaves was 14.84 mg/L and breadfruit stems was 14.81 mg/L. Tyrosinase activity test showed that ethyl acetate fraction of breadfruit leaves had the highest effectiveness as tyrosinase inhibitor with IC₅₀ value of 355.24 ppm, compared to crude extract and other fractions. Although the activity has not matched the positive control kojic acid (IC₅₀ = 39.28 ppm), these results indicate the potential of breadfruit leaf and stem extracts as a source of natural active ingredients for skin lightening
