Abstract

Indonesia is a country that is rich in flora and fauna diversity, one of the unique flora in Indonesia is Nepenthes. This bag serves to trap insects or other small animals, the Biology Education and Research Forest (HPPB) is located in the Andalas Limau Manis University campus area which is classified as lowland tropical rain forest which is the habitat of Nepenthes, in the Biology Education and Research Forest (HPPB) this type of pitcher plant, namely Nepenthes mirabilis. Nepenthes mirabilis is a plant that is able to digest insects trapped in pockets at the ends of its leaf tendrils, so it is classified as a carnivorous plant. Microorganisms play an important role in breaking down organic materials into inorganic ones available to plants. Types of bacteria and bacterial populations greatly determine the decomposition process of animals that have been trapped in the bag as a source of nutrients for Nepenthes plants. This study aims to explore the natural microflora of Nepenthes mirabilis fluids, determine the proportional presence of potential bacteria, and determine the in vitro potential of chitinolytic bacteria from Nepenthes mirabilis fluids. This study used a survey method which was analyzed descriptively. The results showed that Nepenthes mirabilis fluid contained the presence of Fermentative, Amylolytic, Proteolytic, Chitinolytic and Lipolitical bacteria. Nepenthes mirabilis liquid in open bags contained chitinolytic bacteria, the highest presence of chitinolytic bacteria was found in MBB samples (20,104 cfu/ml) and MBA samples (18,104 cfu/ml). .28), fermentative (2.33 and 2.00), amylolytic (1.42 and 1.28), proteolytic (2.00 and 2.00), lipolytic (1.75 and 1.28).

Abstract

Mangrove (Rhizophora mucronata) is a plant that can be used as an antibacterial because it contains several secondary metabolite compounds such as saponins and flavonoids which can inhibit bacterial growth. This research aims to determinehe antibacterial activity of mangrove leaves (Rhizophora mucronata) in inhibiting the growth of Helicobacter pylori bacteria. This research is an in vitro study using the agar dilution method for 2-5 days. The treatment groups consisted of negative, positive (co-trimoxazole), ethanol extract of mangrove leaves (Rhizophora mucronata) with concentrations of6.25%, 12.5%, 25% and 50%. The treatment was carried out by adding the Helicobacter pylori suspension to a petri dish containing agar media. Observations were made to see whether bacteria grew in the media. The data is then analyzed and broken down from the results of calculating the diameter of the inhibition zone. The test results showed that the ethanol extract of mangrove leaves (Rhizophora mucronata) had antibacterial activity in the weak category in inhibiting the growth of Helicobacter pylori bacteria. This antibacterial activity can be caused because mangrove leaves (Rhizophora mucronata) contain secondary metabolite compounds with an antibacterial mechanism of action, so this is in line with the results of this study.

Abstract

Mayonnaise is a semi-solid emulsion processed product that has a very high fat content because the basic ingredients consist of vegetable oil (70-80%) and egg yolk (15-20%). Generally, mayonnaise has a high fat and calorie content. High fat content if consumed excessively will cause various diseases such as cholesterol, coronary heart disease and the risk of obesity. The use of lemuru fish oil as a substitute for mayonnaise is still rarely done. The protein content in lemuru fish is high enough that can be used as a source of protein. In addition to protein, lemuru fish oil is also enriched by the PUFA content of DHA of 30.10%. The aim of this research was to determine the quality of mayonnaise based on the organoleptic value and stability of the lemuru fish oil mayonnaise emulsion after a shelf life test. Proximate fat analysis using the Soxhlet method, protein analysis using the Kjeldahl method, ash analysis using a furnace, crude fiber analysis using the gravimetric method, water content analysis by evaporating the water in the material using an oven, carbohydrate analysis using the differential method. Shelf life test using the Accelerated Shelf Life Test method. Organoleptic testing uses a hedonic scale (1-5), with a total of 30 panelists. Emulsion stability test using a centrifuge by calculating the volume of oil separated. This study used a factorial randomized block design with two factors, namely temperature (5, 25, 35°C) and time (7, 14, 21 days). Mayonnaise proximate results from all parameters have met SNI except for fat and calorie parameters which tend to be lower than SNI standards. The ANOVA test results showed significant differences in organoleptic color and aroma, but it did not show significant differences in organoleptic texture and mold. The best organoleptic results at one-week storage. The emulsion stability results showed significant differences due to temperature and storage time factors. The best emulsion stability results at one week storage at 5, 25 and 35°C with a value of 100%.

Abstract

Black betel (Piper betle L. var. Nigra) can be found in Indonesia and has been proven to have antimicrobial activity so it has the potential to be used as a medicinal plant. Tissue culture can be used to produce seeds that are the same as the parent plant in large, uniform quantities and to produce secondary metabolites from cell suspension cultures. The sterilization method is a very important initial stage for in vitro plant growth, so optimizing sterilization is the aim of this research. The planting materials used are black betel leaves and nodes. Optimization of the sterilization method was carried out using variations in materials and sterilization time duration. The ingredients used were dish soap, bactericide, fungicide, sodium hypochlorite solution, vinegar, alcohol, and Plant Preservative Mixture (PPM). The medium for leaf explants is MS with 2,4-D and for nodes was MS with BAP. Data on the number of contaminated explants were analyzed using the SPSS program. The results of analysis of variance (ANOVA) showed that the optimal sterilization of black betel leaves was using a fungicide solution for 30 minutes, 5% sodium hypochlorite solution for 10 and 15 minutes, vinegar for 7 minutes, 70% alcohol for 10 minutes and PPM for 1 minute with a 22.1% of explants contaminated. Sterilization by rubbing the nodes under running water and the use of vinegar to sterilize black betel nodia resulted in a significantly lower percentage of contamination (73.3%, P<0.05) compared to the method where the leaves were not cleaned by rubbing under running water and no vinegar.
