Abstract

Background: Enterococcus faecalis is one caused bacteria of root canal infections. ChKM is mostly used as sterilization agent in endodontic treatment. Stichopus hermanii extract has been reported to have antibacterial effects for gram-negative and gram-positive bacteria, so could be potentially developed as a root canal sterilization agent. Purpose: The aim of this study was to determine the inhibitory effect of Stichopus hermanii extract on Enterococcus faecalis bacteria. Materials and Methods: This study was an experimental study with post test only control group design and were tested by diffusion methods with 4 groups concentration of 2,5%, 3%, 3,5%, 4%, and 2 controls groups using DMSO 1% as negative control, and ChKM as positive control, each group consisted of 7 samples. The inhibition effect were examined by measure the diameter of the clear zone around the disc. Data were analyzed by one way ANOVA test and followed by LSD test. Results: Results showed that the mean of inhibition zone at concentrations of 2,5% (7,3543 mm), 3% (8,1086 mm), 3,5% (8,7286 mm), 4% (9,4029 mm), DMSO 1% (6,0257 mm) and ChKM (25,49 mm). It had been proved that. Stichopus hermanii extract could inhibit the growth of Enterococcus faecalis (p<0,05). The largest diameter of the clear zone was in the concentration of 4%. Conclusion: Stichopus hermanii extract could inhibit the growth of Enterococcus faecalis and the most effective inhibitory concentration is 4%.

Abstract

Background: Periodontitis is caused by the occupation of gram-negative bacteria (mixed periodontopatogen) in oral cavity. Despite causing resistance, antibiotics are still used as a therapy for periodontitis. Sand sea cucumber (Holonturia scabra) extract hold somepotentials, if developed, to become the alternative therapy for periodontitis due to its antibacterial effects against gramnegative bacteria. Purpose: To determine sand sea cucumber extract (Holonturia scabra) in inhibiting the growth of mixed periodontopatogen bacteria. Materials and Methods: This study used a research design the post-test only control group design. 40 samples of BHI agar plates, contained mixed periodontopatogen bacteria samples, were divided into 5 groups samples. K1 (negative control) was given distilled water containing filter paper, K2 (positive control) was given the antibiotic tetracycline disc 30 g, while P1, P2, and P3 were given a filter paper containing sand sea cucumber extract with a concentration of 150 g/ml, 300 g/ml, and 450 g/ml. All sample from each group were anaerobically inoculated for 2x24 hours. After inoculation, the area aroundthe filter paper will appear to be clear and it will be measured with digital calipers (mm). The acquired data will be analyzed using the Kruskal-Wallis and Mann-Whitney test. Result: The inhibition rate of K1 (6,00 ± 0,00), K2 (28,975 ± 1,20049), P1 (6,8525 ± 0,30156), P2 (7,6237 ± 0,51281), and P3 (8,75 ± 0,3525). Conclusion: Sand sea cucumber extract (Holonthuria scabra) can inhibit the growth of Mixed periodontopatogen bacteria.

Abstract

Background: Bone graft materials have been widely used to regenerate bone defects, which are caused by trauma, tumor resection, pathological processes, and congenital bone defects. Type of bone graft is classified into several types, autograft, allograft, xenograft, and alloplast PRP and PRF derived from autologous blood which sends a high concentration of growth factors on bone defect area. Purpose: The aim of this study is to investigate the effect of PRF with xenograft and alloplast to osteoblast in bone New Zealand rabbits. Materials and Methods: The experiment was held by Post Test Group design. Twenty seven male New Zealand Rabbits were divided into three group. The first group performed the treatment on the right hind limb be treated with Alloplast and PRF treated, The second group performed the treatment on the right hind limb be treated with xenograft and PRF, and The third group was the control group performed the treatment on the right hind limb was given PRF. After treatment the rabbits were sacrificed. Osteoblast of each group was measured by EDTA method All data experiment were analyzed by ANOVA and LSD test (p<0,01). Results: The result of this study showed that the results of osteoblasts between PRF with alloplast compared with prf with xenografts, significant differences as a result of the process of bone formation to see the results of better bone formation. Conclusion: In PRF with alloplast has better results compared with PRF with xenografts from to increase osteoblasts expression.

Abstract

Background: Osteomyelitis is an bone infection caused by bacteria of Staphylococcus aureus. The use of clindamycin within long term may cause of resistance. A compound in Nannochloropsis oculata’s extract contain antibacterial, antiinflamation, antioxidant that can be used as therapy of osteomyielitis. Purpose: Knowing comparison of the effectiveness between the combination of clindamycin and Nannochloropsis oculata’s extract to increasing the density of collagen on the mandibular osteomyelitis. Materials and Methods: The experimental research’s type with a design post test only control group. Fourty male wistar rats as model of mandibular osteomyelitis has made by inducing bacteria of Staphylococcus aureus inserted in socket after an extraction of left incisive teeth on lower jaw and waiting for 28 days. The rats are divided into 5 group: negative control (K1), clindamycin (K2),clindamycin with 50mg Nannochloropsis oculata’s extract (K3), clindamycin with 100mg Nannochloropsis oculata’s extract (K4), clindamycin with 200mg Nannochloropsis oculata’s extract. After 7 days, all of group are euthanazed and then the density of collagen measured using histopathology slide used Masson’s Trichom and 100x enlargement, so then analyzing the density of collagen by Kruskal-Wallis and Mann-Whitney. Result: There were significant differences in collagen between K1 and K2 (p=0,037), K1 and K3 (p=0,013), K1 and K4 (p=0,002), K1 and K5 (p=0,000). and there were significant differences in collagen between K2 and K5 (p=0,013). But there were no significant differences in collagen between group K2 and K3 (p=1,000), K2 and K4 (p=0,288). Conclusion: The combination of clindamycin with 200mg Nannochloropsis oculata’s extract having an effect on increasing number of the density of collagen significantly on the healing of mandibular osteomyelitis.

Abstract

Background: Indonesia is the country with the biggest sea cucumbers world’s producer. So far, sea cucumbers are only used as a side dish alone. Indonesia can provided added value and high economic value of gold sea cucumber through right procedure. A lot of gold sea cucumbers active substance that are suspected to have an influence on wound healing. For the best result, extract gold sea cucumber must be prepare with correct procedure. Water extract is one of a safe procedure with a small risk that may damage parts of the active substance. Purpose: The aim of this study is to know the characterization of water extract gold sea cucumber (Stichopus hermanii). Materials and Methods: Gold sea cucumbers were collected, homogenized with distillated water and shaken with water-bath shaker. Then extract was freeze dried, the result then characterized using spectrophotometer. Result: Thelargest component to fewest component of water extract gold sea cucumber is total protein 76,82%, essential amino acid 48,11%, non essential amino acid 28,70%, glycoprotein 4,62%, collagen 4,05%, GAG’s 1,62%, proteoglycan 1,13%, heparin sulfat 1,02%, calcium 59%, saponin 56%,heparin 38%, hyaluronat acid 29%, calcium 0,59%, Zinc 0,01%. Conclusion: There is many active substance of water extract gold sea cucumber that involed in wound healing process.

Abstract

Background: Maxillary expansion device causing mechanical pressure to open the maxillary suture which causes inflammation and bone remodeling around the area. Stichopus hermanii is known to contain flavonoid, chondroitin sulfate, omega 3 (EPA and DHA) and dermatan sulfate which is used to inhibit inflammatory process, increase bone metabolism and mineralization, and bone remodelling. Purpose: The aim of this experiment is to know the effect of Stichopus hermanii on maxillary suture remodeling expansion to palatal width using cephalometric analysis. Materials and Methods: Experimental units used was 24 male Caviacobaya divided into 4 groups (n=6). Group K(-) is negative control. Helical spring was applied to and activated in the other 3 groups for 10 days. Group K(+) was given 2% NaCMC gel, both group P1 and P2 was given 3% Stichopus hermanii gel. Group P2 werealso given retention period for 10 days after activation period. Cavia cobaya were then decaputated and observed for the palatal width changes. The data was analyzed with One Way ANOVA test continued with LSD test. Result: The result of ANOVA test showedsiginficant differences among the treatment groups p=0,000 (p<0,05). LSD test showed significant differences between group K(-) and K(+) (Sig. 0,000), K(-) and P1 (Sig. 0,000), K(-) and P2 (Sig. 0,000), K(+) and P2 (Sig. 0,020) and group P1 and P2 (Sig. 0,036).Conclusion: 3% Stichopus hermanii gel has effect on maxillary suture remodeling expansion to palatal width use cephalometric analysis and combination of Stichopus hermanii with 10 6days retention period resulting the widest palatal width.

Abstract

Background: Ulcers are common and cause pain, difficulty in speaking, eating and swallowing. Avicennia alba role in the process of wound healing and has been shown to accelerate the healing process of gastric ulcer. It contains flavonoid, saponin, NSLcomponents, minerals and vitamin C, which plays a role in wound healing. Purposes: To prove the effect of ethanol extract of Avicennia alba consentration 10%, 20%, 40% against traumatic ulcer. Materials and Methods: The subjects of this research were 30 wistar rats that were randomized into 5 different groups; K0, K1, P1, P2 and P3. (K0) control, (K1) were given hyaluronic acid, (P1) were given extract of Avicennia alba 10%, (P2) were given extract of Avicennia alba 20%, and (P3) were given extract of Avicennia alba 40%. The subject was wounded using amalgam stopper that has been heated before. Subject was given topical application once daily. The ulcer’s diameter was measured at the second day until the day cured using digital caliper. Data were analyzed using One way ANOVA test. Result: The result showed signification of p<0,05, showed that there were difference diameters betweentwo groups. The average diameter differences of traumatic ulcer among rat are: 0,75 mm (K0), 1,51 mm (K1), 1,01 mm (P1), 2,02 mm (P2), and 1,70 mm (P3). The results showed statistically significant value of p>0.05 which there was no difference in effectivenessAvicennia alba extract in the K1 group. Conclusions: Ethanol extract of Avicennia alba 10%, 20%, and 40% have effect in the healing of traumatic ulcer. The effectiveness of those were the same as giving a 0.2% hyaluronic acid. The concentration 20% of Avicennia alba was effective to the healing of traumatic ulcer

Abstract

Background: Actinobacillus actinomycetemcomitans is a major periodontopathic bacterium with multiple virulence factors, including LPS. LPS is a potent stimulator cytokines proinflammatory such as TNF-α. Nannochloropsis oculata is a green micro algae that hasanti-inflammatory effects. Purpose: The aim of this study was to prove the effect of Nannochloropsis oculata extracts to decreased levels of TNF-α which induced by Actinobacillus actinomycetemcomitans bacteria. Materials and Methods: The experiment was held post test only control group design. 48 male wistar rats divided into 6 groups. K1 group was negative control group without any treatment, K2 group induced by Actinobacillus actinomycetemcomitans bacteria, K3 group treated with minocycline antibiotics 0.1%, and the other groups K4, K5, K6 were treated with Nannochloropsis oculata extract on various concentration that is 2.375%, 2.5%, 2.625%. After treatments on the 37th day, all groups of rats were euthanized and measured the levels of TNF-α in gingival tissue by ELISA. All of thedatas were analyzed by Kruskal Wallis dan Mann-Whitney U test. Result: This study showed the levels of TNF-α K2 group (36.9444) was significantly higher than K1 group(27.8889). K3 group (29.2778) showed no significant decreased compared to K2 group (36.9444). K4 (35.2778), K5 (33.1111) and K6 (29.2778)groups showed no significant decreased levels of TNF-α compared to K2 group (36.9444). Conclusion: Nannochloropsis oculata extracts can not reduce the levels of TNF-α in rats which induced by Actinobacillus actinomycetemcomitans bacteria.

Abstract

Background: Molecular weight is one of the chitosan characteristic that affects the effectiveness application for wound healing of dental extraction. Chitosan could improve wound healing process because of its anti inflammation property and improve proliferation phase. Lymphocyte cell is one of the main cells that act during inflammatory phase. Purpose: the aim of this experiment is to account lymphocyte cell in wound healing of dental extraction using chitosan with different molecular weight. Materials and Methods: 54 Male Rattus Norvegicus were divided into 3 groups. Group I is control group (without chitosan). Group IIwas given low molecular weight chitosan. Group III was given high molecular weight chitosan. Rats were sacrificed by decapitation on day 5 and day 7 post extraction then they were examined histopatologically to see the change in lymphocyte cell number. Result: Data were statistically analyzed with One Way ANOVA and LSD with degree of significant p< 0,05 showed significant difference between high molecular weight chitosan group and low molecular weight chitosan group after 5 and 7 days observation. Conclusion: High molecular weight chitosan was found more effective to decrease lymphocyte cell amounts more than low molecular weight chitosan in wound healing of dental extraction process.

Abstract

Background: Recurrent aphthous stomatitis (RAS) is common painful mucosal condition affecting the 5%-60% world’s population and become the third largest oral disease in Indonesia. Despite their high prevalence, RAS treatment is currently still dominated by the use of chemicals that scientifically have adverse side effects. Therefore, it is necessary to develop new approach using herbal product with no side effects. This literature review summarizes the potential of kersen leaf extract (Muntingia calabura L.), its dosage forms, and the possibility using nanotechnology that can speed up the drug delivery process. Objective: to determine the possibility effect of nanoparticle cherry leaf extract in gel form on decreasing number of macrophages and stimulating collagen in stomatitis. Methods: literature search using the PRISMA method with Science Direct, PubMed, Scielo, NCBI, Research gate, and Google Scholar databases of 1107 articles with keywords “Muntingia calabura L. leaves extract”, “nanogel”, “flavonoid”, “quercetin”, “fisetin”, “macrophage”, “collagen”, “soft tissue healing process” and “recurrent aphthous stomatitis”. The results of the literature selection found 22 articles. Results: flavonoids in cherry leaf extract can reduce the number of macrophages and increase collagen stimulation. The mechanism of action begins by inhibiting the production of pro-inflammatory cytokines, inhibiting macrophage migration and adhesion, stimulating the production of growth factors by macrophages for collagen synthesis, and decreasing collagenase. Nano gel form may increase retention time and drug penetration. Conclusion: Nanogel based cherry leaf extract may reduce the number of macrophages and stimulating collagen in minor RAS.
