Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak vesikula seminalis terhadap kualitas spermatozoa dan konsentrasi testosteron tikus putih (Rattus norvegicus). Dalam penelitian ini digunakan 20 ekor tikus putih jantan dewasa, galur Wistar, berumur 3 -4 bulan, berat badan 250-300 g dan dibagi menjadi 4 kelompok (K1, K2, K3, dan K4), masing-masing kelompok berturut-turut diberikan 0,2 ml NaCl fisiologis, 25 μg cloprostenol, 0,2 ml ekstrak vesikula seminalis (EVS), dan 0,4 ml EVS secara intraperitoneal. Pada akhir perlakuan, seluruh tikus dikorbankan secara dislocatio cervicalis. Selanjutnya, kauda duktus epididimis dinekropsi untuk dikoleksi spermatozoanya. Pemeriksaan kualitas spermatozoa, meliputi motilitas dan konsentrasi spermatozoa. Pemeriksaan konsentrasi testosteron serum darah dilakukan menggunakan teknik enzymelinked immunosorbant assay (ELISA). Motilitas dan konsentrasi (x10 6 ) spermatozoa pada kelompok K1; K2; K3; dan K4 masingmasing adalah 3,00±0,00; 3,20±0,28; 2,00±0,86; dan 3,20±0,28 dan 146,60±71,90; 187,80±60,80; 124,20±64,70; dan 129,40±27,07. Konsentrasi testosteron pada kelompok K1; K2; K3; dan K4 masing-masing adalah 5,53+1,75; 4,68+4,56; 2,51+1,33; dan 2,40+1,60 ng/ml. Hasil penelitian menunjukkan bahwa pemberian EVS mampu meningkatkan motilitas spermatozoa tetapi tidak memengaruhi konsentrasi spermatozoa dan testosteron serum darah tikus putih.

Abstract

The study to evaluate changes in body weight, muscle weight, muscle protein content and total DNA and RNA on breast muscle of selectedlocal chicken due to different levels of fermented feed containing Kiapu (Pistia stratiotes L.) was conducted using a factorial randomized designfor 7 weeks. This study used 72 selected local chickens, which were separated into 2 groups (male and female) and fed on 3 types of feedtreatment, namely P0 (control treatment with 100% commercial feed), P1 (80% commercial feed + 20% fermented Kiapu feed), and P2 (70%commercial feed + 30% fermented Kiapu feed). Parameters were body weight, muscle weight, muscle protein content and total DNA and RNAon breast muscle of chicken. Total protein, DNA and RNA content were analysed by enzymatic reactions of prokaryotic cell and measured with aUV-Spectrophotometer according to the specified OD (optical density). The results showed that feeding Kiapu fermented 20-30% increased bodyweight and protein content of breast muscle of female chicken by change in DNA content. However, the feeding treatment had no changes inmuscle weight and RNA content of both in male and female chicken. On the other hand, substitution of 20-30% Kiapu fermented feed causednegative changes in DNA level of breast muscle. From this study, it was concluded that substitution of fermented feed containing 20-30% Kiapuassociated with the growth process of breast muscle protein is suitable for female but not suitable for male chicken.

Abstract

A pair of cross-sectional studies was conducted to determine prevalence and risk factors of Strongyloides spp. (threadworm) infections in beef cattle in Kasiman Subdistrict, the Regency of Bojonegoro. A total of 263 and 270 of local beef cattle with various age were selected  for faecal sample during dry season (August-October) and wet season (February-March), respectively. Feces were processed for counting the number of eggs per gram feces (EPG) using McMaster method. The prevalence of strongyloidosis was 4.56% and 7.04% in the dry season and the rainy season, respectively. The factors that associated with prevalence and intensity infection were age and sex of cattle. During the dry season and the rainy season, the calf  and weaner group (1 year) was more susceptible for Strongyloides infection than cattle with older age and showed higher intensity of infection (based on EPG values). The bulls showed a higher risk and intensity of infection than cows.

Abstract

The purpose of this research was to study the effect of different level of protein addition in goat ration on feed addition in the ration on feed intake and physiological responses of Saburai goats (rectal temperature, respiratory rate, and heart rate). Twenty male Saburai goats were randomly divided into 4 treatments group with 5 goats each. Goats in group 1 (PO) were fed with forage, while goats in group 2 (P1), 3 (P2), and 4 (P3) were fed with forage + concentratre with 13%, 16%, and 19% crude protein, respectively. The treatments were P0= feeding forage, P1= feeding forage + concentrate with 13% crude protein (CP), P2= feeding forage + concentrate with 16% CP, and P3 feeding forage + concentrate with 19% CP. The results showed that the rectal temperature and heart rate did not significantly affected (P0.05) in all treatment, however the heart rate was significantly (P0.05) affected among the treatments. The conclusion is the addition of different protein level in the ration has no effect on rectal temperature and respiratory rate, but significantly increases the heart rate frequency.

Abstract

Penelitian ini bertujuan mengidentifikasi dan karakterisasi Staphylococcus aureus (S. aureus) yang berasal dari kasus bumblefoot dan arthritis. Dalam penelitian ini digunakan 10 isolat S. aureus yang terdiri atas lima hasil isolasi asal kasus bumblefoot dan lima hasil isolasi asal kasus arthritis pada ayam broiler. Identifikasi S. aureus dilakukan melalui uji fermentasi manitol salt agar (MSA), pewarnaan Gram, uji koagulase, clumping factor, dan uji katalase. Karakterisasi fenotipe S. aureus yang dilakukan meliputi sifat hemolisis pada plat agar darah, sifat hidrofobisitas, dan kemampuan hemaglutinasi. Hasil penelitian menunjukkan bahwa semua isolat S. aureus mampu memfermentasi MSA, positif uji koagulase, clumping factor, dan katalase. Staphylococcus aureus mampu menghemolisis plat agar darah dengan memperlihatkan sifat α-hemolisin (80%), β-hemolisin (10%), and γ-hemolisin (10%). Semua isolat S. aureus (100%) bersifat hidrofil dan mampu mengaglutinasi sel darah merah kelinci.

Abstract

The aim of this study is to investigate the contraceptive effect of the extract methanol of the leavesHydrocotyle javanica Thumb and its effect threat of implantation, the quantity and condition of fetuses,histology appearance of the ovary and uterus and rate of pregnancy. The female mice were given orallyeither methanol extract of leaves, for 10 days in various doses: 20, 40, 60, and 80 gram/kg BW. On day20, the mice were mated and when pregnancy occurred, treatment was stopped. On day 17 of pregnancy,the mice were sacrificed: uterus and ovaries were examined histologically. Abstract

Penelitian ini mengetahui pengaruh pemberian tepung atau ekstrak alkohol daun katuk terhadap perubahan histologis testis kambing kacang lokal jantan. Dalam penelitian ini digunakan 15 ekor kambing dengan umur sekitar 1,5 tahun. Semua hewan penelitian terlebih dahulu diaklimatisasi selama 2 minggu terhadap lingkungan penelitian. Hewan-hewan tersebut dibagi menjadi 3 kelompok yang masing-masing terdiri atas 5 ekor kambing. Kelompok P0 (kontrol) telah hanya diberikan air distilasi. Pada kelompok P1, diberikan perlakuan berupa suplementasi tepung daun katuk dan untuk kelompok P2 diberikan ekstrak alkohol daun katuk. Semua perlakuan diberikan secara oral sebanyak dua kali per hari (pada pagi dan sore hari) selama 35 hari berturut-turut. Setiap kambing percobaan diberikan pakan yang terdiri atas daun-daunan dan rumput, serta disediakan akses untuk memperoleh air minum secara ad libitum. Pada akhir waktu penelitian, seluruh hewan dikastrasi dan bagian tubulus seminiferus dari testis diambil dan diproses lebih lanjut untuk evaluasi secara histologis. Hasil yang diperoleh menunjukkan bahwa terdapat indikasi peningkatan produksi spermatid dan spermatosit pada Kelompok P1 dan P2. Dengan demikian disimpulkan bahwa pemberian suplemen daun katuk dapat meningkatkan produksi spermatid dan spermatosit kambing kacang lokal jantan.

Abstract

This study was aimed to test the growth of bacteria Escherichia coli (E. coli) and Bacillus sp. which were exposed to magnetic fields. In the first stage of the study, the effect of magnetic fields on the growth of E. coli and Bacillus sp were observed. The futher study was aimed to evaluate the effect of magnetic field on antibiotic sensitivity againts the growth of E. coli and Bacillus sp. The magnetic fields treatments were 0.0 mT (control), 0.1 mT, 0.2 mT, 0.3 mT which were exposed for 10 hours for each treatment. Five antibiotics (trimethoprim, ampicillin, nalidixic acid, streptomycin and chloramphenicol) were used for each bacteria. The result showed that the magnetic field did not influence the colony growth of E.coli, but in Bacillus sp. was seen the increasing of colony area in magnetic field of 0.1 mT and 0.2 mT compared with control treatment. Antibiotic of trimethoprim, nalidixic acid, and ampicillin increase the growth inhibition of E. coli when the bacteria have been exposed to 0.1 mT; 0.2 mT; 0.3 mT magnetic field for 10 minutes. The inhibition by streptomycin and chloramphenicol antibiotic on E. coli did not affected by magnetic field exposure. The inhibition of Bacillus sp by trimethoprim and ampicillin increased when the bacteria have been exposed to 0.2 mT and 0.3 mT magnetic field for 10 minutes. The inhibition of nalidixic acid, streptomycin and chloramphenicol to Bacillus sp. did not affected by the magnetic field exposure.

Abstract

Penelitian ini bertujuan mengetahui respons antibodi dalam serum ayam petelur terhadap ekskretori/sekretori, dan ditantang dengan telur infektif Ascaridia galli (A. galli) Sebanyak 12 ekor ayam dibagi dalam empat kelompok. Kelompok pertama adalah ayam yang tidak diimunisasi dan tidak diinfeksi (kontrol), kelompok kedua adalah ayam yang diimunisasi dengan dosis 260 µg ekskretori/sekretori larva A. galli, kelompok ketiga adalah ayam yang diinfeksi dengan dosis 1000 telur infektif A. galli, dan kelompok keempat adalah ayam yang diimunisasi dengan dosis 260 µg ekskretori/sekretori dan satu minggu kemudian ditantang dengan dosis 1000 telur infektif A. galli. Respons antibodi pada masing-masing kelompok dianalisis dengan uji enzymelinkedimmunosorbantassay (ELISA) setiap satu minggu selama 10 minggu pascainfeksi. Hasil penelitian menunjukkan bahwa imunisasi dan atau infeksi dapat memicu peningkatan titer antibodi serum secara signifikan (P0,05) selama 10 minggu pascainfeksi. Titer tertinggi adalah 2,63±1,20 OD (optical density) dicapai pada minggu ke-3 pascainfeksi dan titer terendah adalah 1,51±0,48 OD pada minggu ke-0. Ekskretori/sekretori dapat memicu respons antibodi serum ayam petelur terhadap A. galli.

Abstract

Three intestinal bacteria isolates (namely C1, C2, and C3) from common carp (Cyprinus carpio) were isolated and identified as new potential probiotics. This research aimed to study the effect of three isolates potential probiotics addition on growth performance, survival rate and feed efficiency of common carp. Completely randomized design was assigned to fourth diet treatments with five replicates. As a control, we used the feed without any supplemention of potential probiotic. The result showed that the addition of potential probiotic bacteria (C1, C2, and C3) gave significant effect on daily growth rate, survival rate and feed efficiency and could be used in aquaculture industry to reduce feeding cost.
