Abstract

Indonesia is a country with high birth rate and the rate continues to increase each year. Methods to reduce high birth rate are needed, especially herbal based contraceptives. This research aims to study the possibility of Calina papaya leaf ethanolic extract as an antispermatogenic agent which hopefully could be expanded into natural antifertiliy drug candidate. Research was done using 24 white male Wistar rats divided into four groups. Dosages were given as follows: 0 mg/Kg W (control group), 100 mg/Kg W (P1), 200 mg/Kg W (P2) and 300 mg/Kg W (P3). Treatment was given in 30 days period. Rats were sacrificed on Day 31 and the testes were taken. Testes were then made into histological specimen using paraffin block and stained using Haematoxylin-Eosin. The diameter and surface area of seminiferous tubules, lumen surface area, and spermatogenesis index were then observed. Results showed that there is a significant difference (p0.05) on lumen surface area and spermatogenesis index between control and treatment groups. It is also found that the optimum dosage given to attain the desirable effect is 300 mg/Kg W. There is no significant difference found on surface area and diameter of tubulus seminiferous between control and treatment group (p0.05). Based on the results, papaya leaf (Calina variety) ethanolic extract has the potential a natural antispermatogenic with the optimum dose of 300 mg/Kg W.

Abstract

This research aimed to characterize VirB4 protein of local isolate Brucella abortus with Western blotting method. The result showed that there were four protein bands with molecular weights of 64.61, 59.25, 21.63, and 16.70 kDa by triggering a reaction between the whole Brucella abortus and anti-Brucella abortus serum. The results also revealed that there was only one protein band with a molecular weight of 59.25 kDa triggering a reaction between the whole Brucella abortus and anti-VirB Brucella abortus serum. Finally, it can be concluded that VirB4 protein can affect the virulence factor of Brucella abortus, successfully characterized with the appearance of one band with a molecular weight of 59.25 kDa by using Western blotting method.

Abstract

Penelitian ini bertujuan mengetahui distribusi penyebaran golongan darah A, B, dan AB dan mengetahui kemungkinan hubungan antara golongan darah dengan kemunculan pola warna rambut pada kucing domestik di Indonesia. Pada penelitian ini digunakan 50 ekor kucing domestik yang berasal dari daerah Bogor dan Bekasi. Pemeriksaan golongan darah kucing dilakukan dengan identifikasi golongan darah dengan kit antibodi monoklonal Shigeta(Jepang). Hasil penelitian menunjukkan bahwa semua kucing domestik yang diteliti bergolongan darah A dan tidak diperoleh tanda-tanda adanya hubungan antara golongan darah dengan warna rambut. Terdapat sembilan kemunculan warna rambut yang terdiri atas warna tunggal, kombinasi dua warna, dan kombinasi tiga warna.

Abstract

The objective of this research was to investigate cytotoxicity effect of Haarlem oil (HO) on HL-60 cell line and Steinernema feltiae (S. feltiae). The test results using trypan blue method and CellTiter-Glo® assays revealed that HO showed higher cytotoxicity effect against HL-60 cell line especially at concentration of 1:10 and 1:5. Meanwhile, the MTT (3-(4,5-dimethythiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay revealed that the higher cytotoxicity effect showed at 1:5, with the cell viabilities were around 40-65%. The nematicidal assay discovered that HO at concentration of 1:5 showed higher activities compare to other concentrations with percentage viability of 70%. These studies have shown that HO shows cytotoxic effect against HL-60 cell lines, and moderate activity against S. feltiae as well.

Abstract

Tujuan penelitian adalah menentukan kemungkinan faktor-faktor risiko terhadap kemunculan toksoplasmosis. Sebanyak 132 sampel serum darah diambil pada kucing lokal di Yogyakarta. Data-data epidemiologis seperti asal usul kucing, ras kucing, jenis kelamin, umur, lokasi sistem pemeliharaan, jenis pakan, dan frekuensi diare diberi kode untuk mempermudah analisis, kemudian dimasukkan, disimpan, dan dianalisis dengan program Statistix Versi 7 (Analytical Sofware inc). Analisis data dilakukan secara bivariat (Chi-square (2), dan kekuatan asosiasi (OR), dan multivariat (regresi logistik). Hasil penelitian menunjukkan bahwa faktor-faktor yang berpengaruh terhadap adanya toksoplasmosis pada kucing berdasarkan analisis bivariat adalah pembersihan kotak pasir 1 kali sehari dan mandi 2-3 kali seminggu sedangkan faktor-faktor yang memiliki peluang meningkatkan seropositif toksoplasmosis berdasarkan analisis multivariat adalah pemeliharaan kucing yang bebas di dalam rumah, dimandikan lebih dari 1 kali seminggu, dan dimandikan lebih besar dari 1 bulan sekali.

Abstract

ABSTRACTThe present study aims to determine the effect of fermented diet containing probiotics and digestive enzymes with different feed levels on growth, fat and cholesterol contents of  broiler chickens. Evaluating the effectiveness of probiotics (P) and digestive enzymes (DE) in fermented diet on broiler was observed by 4 treatments: P0=control (fermented diet without P and ED) EP), P1(fermented diet contained 0.5% P only)), P2 (fermented diet contained 0.5% DE only), P3 (fermented diet contained 0.5% P and 1% DE) and P4 (fermented diet contained 1% P and 0.5% DE). A hundred DOCs of broiler chickens strain CP 707 were used (unsex) and it designed by a completely randomized design with 5 treatments, 4 replications and each replication consisted of 5 DOCs. The data obtained were analyzed by means of variance and Duncan's multiple range test. The results of present study showed that broiler fed on fermented diet containing P and/or ED with different levels had a significant effect (P0.05) on final body weight, weight gain, carcass weight, fat content on thigh muscle. Broiler fed on fermented diet contained 0.5% DE only (P2) resulted in better growth and weight gain than other treatments. However, fermented diet contained probiotics and digestive enzymes had no effect on fat and cholesterol contents in breast muscle. It was concluded that the addition of digestive enzymes in fermented diet formulated by local raw materials was very effective than probiotics in increasing body weight gain, carcass weight and prevented the increase in fat and cholesterol contents both in breast and thight muscles of broiler chicken.

Abstract

The purpose of this study was to determine the macro minerals concentration in serum of repeat breeding (RB) Aceh cattle. In this study 16 Aceh cattle were examined; they consisted of 7 fertile Aceh cattle (P1) and 9 Aceh cattle with RB (P2), all of which were 3-8 years old with abody condition score (BCS) of 3-4. Serum collection was carried out for examining the level of serum minerals, including magnesium, phosphorus, sodium, potassium, chloride, and calcium. Data was analysed using T-test. The levels of magnesium, phosphorus, sodium,potassium, chloride, and calcium in P1 vs P2 were 2.18±0.60 vs. 2.20±0.34 mg/dL; 6.18±1.34 vs. 6.48±0.74 mg/dL; 142.71±5.09 vs.142.44±2.29 mmol/L; 4.81±0.76 vs. 4.76±0.51 mmol/L; 104.57±4.35 vs. 107.67±7.36 mmol/L; and 9.07±0.45 vs. 9.90 ± 0.60 mg/dL (P0.05), respectively. It was concluded that the concentration of serum macro minerals do not affect the incidence of RB in Aceh cattle.

Abstract

This research was aimed to know histopathology change of white mouse ovary as result of EG intoxication. Eight white mice randomly divided into three treatment groups and one control group. Treatment of P1 group was given 10 ml EG/L drinking water, P2 was given 20 ml EG/L drinking water, and P3 was given 30 ml EG/L drinking water for two weeks. Control group was given aquadest. Then, white mice were euthanized and ovaries sample were taken for histology observation. The result for treatment groups (P1 and P2) had shown ovary hyperemic (congest). But, the change is worst in P2 rather than P1. Changes in P3 treatment group had shown ovary hyperemic (congest) and leukocyte infiltration in follicular antrum and follicular necrosis causing lost of ovum cell (cariolysis). Poisoned of EG in white mouse can cause inflammation and necrosis of ovary follicle thus the formed of ovum cell was failed (ovogenesis).

Abstract

Penelitian ini bertujuan melakukan studi filogenetik virus AI tipe A subtipe H5N1 isolat asal Provinsi Aceh yang dapat memberikan informasi secara molekuler tentang kekerabatan antar isolat virus AI tipe A subtipe H5N1. Sebanyak 11 isolat virus AI asal Provinsi Aceh yang dikoleksi oleh Laboratorium Virologi Balai Penyidikan dan Pengujian Veteriner (BPPV) Regional I di Medan, Sumatera Utara selama kurun waktu 2007-2008 digunakan dalam penelitian ini. Metode penelitian meliputi preparasi sampel isolat virus, ekstraksi RNA, amplifikasi gen H, elektroforesis DNA hasil amplifikasi, sekuensing, dan analisis data hasil sekuensing dengan software MEGA 4.0. Hasil sekuensing diketahui bahwa daerah yang teramplifikasi sebesar 191 bp. Hasil allignment dari nukleotida 191 bp ditemukan urutan nukleotida yang menyandi 5 asam amino yang berbeda pada posisi asam amino ke-493, 498, 499, 504, dan 515 antara isolat virus AI subtipe H5N1 asal Provinsi Aceh selama tahun 2007-2008 dengan isolat dari pembanding dari gene bank. Kontruksi pohon filogenetik menunjukkan bahwa virus AI subtipe H5N1 asal Provinsi Aceh tahun 2007-2008 mengelompok secara terpisah dari isolat Indonesia yang lain, tetapi masih dalam klaster virus AI subtipe H5N1 Indonesia.

Abstract

This research was conducted in order to examine the goblet cells proliferation in duodenum, jejunum, and ileum of laying hens due to exposured with protein of excretory/secretory (ES) of Ascaridiagalli adult worm. Thirty heads of laying hens were divided in to two groups. The first group was treated with 4,000 infective larva (L2) of A. galli and the second group was immunized with 380µg of ES andfour hours later was challenged with 4000 L2. All treatments were given orally using stainless steelcanule directly to the oesophagus. Data was taken on the 3, 6, 9, 12, and 15 days post immunization(p.i.). The goblet cells were determined by Periodic Acid Schiff (PAS) staining. The result showed that immunization was able to increased goblet cells proliferation significantly at 12 and 15 day p.i. on theduodenum, and at 9, 12, and 15 day p.i. on the jejunum, but goblet cells proliferation did notsignificantly on the ileum. From this result we suggested that ES would beneficial in the strengthen thehost’s defence mechanisms in the intestinal mucosa.Keywords: Ascaridia galli, excretory/secretory, goblet cells
