Abstract

This study aimed to verify the effect of ethanol extract of jaloh leaves at various doses on parasitemia percentage of in mice (Mus musculus), wich inoculated Plasmodium berghei. This study used 12 male mice (Mus musculus) strains Balb / C with average body weight of 30.2 grams of 2-month-old. This research using Completely Randomized Design (CRD) with four treatments and three replications. All the mice were infected with Plasmodium berghei prior to treatment on three days post infected the mice in group 2,3 and 4 were given with extract ethanol jaloh leaves at dose 50, 100, 150 mg/kgbw inspectively. The mice in group 1 (K0) were not given anything and consident as control.Observations parasitemia percentage using microscopy on preparations stained thin blood smear Giemsa. The parasitemia percentage data were analyzed using one way ANOVA. On the average (±SD) percentage of parasitemia during the three days in four treatments (K0, K1, K2, and K3) on the first day were 24.87±1.86; 24.33±1.53; 24.20±1.06; and 24.93±1.01. On the second day were 22.67±2.52; 23.00±1.00; 22.80±1.20; and 24.13±2.01; and on the third day were 24.77±1.66; 22.97±3.76; 21.13±1.90; and 24.27±1.42. Statistical analysis showed that ethanol extract of the jaloh leaves for KO, KI, KII, and KIV respectively treatment did not affect the percentage of parasitemia in mice (Mus musculus) inoculated Plasmodium berghei.

Abstract

Penelitian ini bertujuan mengetahui efek inhibin B terhadap ekspresi protamine P2 di dalam kepala spermatozoa pada kauda epididimis. Penelitian ini menggunakan 24 ekor tikus jantan berumur 4 bulan yang dikelompokkan secara acak ke dalam 4 kelompok (KO, KI, KII, dan KIII), setiap kelompok terdiri atas 6 ekor tikus. Kelompok KO merupakan kelompok kontrol hanya diinjeksi dengan PBS. Kelompok KI, KII, dan KIII diinjeksi dengan inhibin B dengan dosis masing-masing adalah 25, 50, dan 100 pg/ekor. Tikus diinjeksi sebanyak 5 kali secara intra peritoneal dengan interval waktu pemberian 12 hari selama 48 hari. Injeksi pertama, inhibin B dicampur dengan 0,05 ml PBS dan 0,05 ml CFA. Injeksi kedua hingga kelima, inhibin B dicampur dengan 0,05 ml PBS dan 0,05 ml IFA. Pada hari ke-6 setelah injeksi inhibin B terakhir, hewan coba dikorbankan secara dislocatio cervicalis lalu jaringan kauda epididimis dikoleksi dan difiksasi dengan paraformaldehid 4%. Setelah melalui proses dehidrasi, jaringan blok di dalam parafin dipotong dengan ketebalan 6 mikron dan diwarnai secara imunohistokimia dengan menggunakan antibodi anti protamine P2. Pengamatan secara imunohistokimia menunjukkan adanya ekspresi protamine P2 di dalam kepala spermatozoa pada semua kelompok perlakuan. Akan tetapi, seiring dengan penambahan dosis inhibin B menyebabkan terjadinya penurunan tingkat ekspresi protamine P2 di dalam kepala spermatozoa pada kauda epididimis. Hasil penelitian menunjukkan bahwa injeksi inhibin B dengan dosis 100 pg/ekor menurunkan secara nyata jumlah ekspresi protamine P2 di dalam kepala spermatozoa pada epididimis (P0,05) dibanding KO.

Abstract

This study aimed to investigate the effectivity of goat milk fermentation as tyrosinase inhibitory with Lactobacillus rhamnosus TW 2. The examination of fresh milk contained the density, protein, and fat content. The culture of Lactobacillus rhamnosus TW 2 acid lactid bacteria starter was added with the concentration of 3, 4, and 5%, inoculated in pasteurized goat milk, then incubated at 37° C for 24 hours. Fermented milk were extracted by centrifugation, then supernatant was collected and was used for inhibition of tyrosinase enzymes activity on L-tyrosin and L-dopa substrate. The result showed that the density, fat content, and protein content of Etawah crossbred goat milk are 1.028, 3.73, and 5.45%, respectively. Re-identification of lactic acid bacteria showed similar morphology, physiology, and bio-chemistry with the isolated lactic acid bacteria. The growth curve of TW 2 was observed in 12 hours. The 5 % of Lactobacillus rhamnosus TW 2 was the best concentration to inhibit tyrosinase activity in L-Tyrosin substrate. In conclusion, fermentation of goat milk using Lactobacillus rhamnosus TW 2 at concentration of 5% as starter is effective to inhibit tyrosinase activity significantly.Key words: goat milk, fermentation, tyrosinase inhibitory

Abstract

Analisis genetika menggunakan random amplified polymorphic DNA (RAPD) untuk mengetahui kemiripan genetika antara kambing peranakan Ettawa (PE) hasil inseminasi buatan (IB) dan kawin alam (KA). Hasil penelitian menunjukkan bahwa dari 20 primer OPA hanya 10 primer yang bisa digunakan untuk mengamplifikasi fragmen deoxyribonucleic acid (DNA) dalam kelompok kambing PE, yang terdiri atas 21 kambing hasil KA dan 19 kambing hasil IB. Berdasarkan profil DNA yang diperoleh dan hasil analisis dengan software numerical taxonomy and multivariate analysis system (NTSYS) disimpulkan bahwa hubungan genetika antara kambing PE hasil kawin alam dengan inseminasi buatan adalah sebesar 48% dan dikategorikan rendah.

Abstract

This study aims to determine the effectiveness of Areca catechu Crude Aqueous Extract (ACCAE) as an anthelmintic, the number ofAscaridia galli worms in the duodenum, jejunum, and ileum after treatment, and an economic analysis. In this study, 50 female chickens wereused. The chickens were divided into 5 groups: Treatment 1, negative control (P1), which was given aquadestylates; treatment 2 (P2), which wasgiven infusion of ACCAE at a dose of 26 mg/mL; treatment 3 (P3), which was given infusion of ACCAE at a dose of 53 mg/mL; treatment 4(P4), which was given ACCAE with a dose of 79 mg/mL; and treatment 5 (P5) (positive control with pyrantel pamoat). Each group consisted of10 chickens. An examination was carried out that included weighing the population of adult worms according to their predilection 14 days aftertreatment. Weighing was carried out at the beginning before treatment, on the 7th day after treatment and on the 14th day after treatment. Thechickens were then euthenized, the intestinal worms were counted, and the economic analysis was calculated. Based on the results of the study, itcan be concluded that the best dose for removing A. galli worms is 79 mg/mL, and the location of the predilection of the intestine where the mostworms are found is the jejunum and the least is in the ileum. Economically, the highest income was obtained from treatment 4, namely the groupwith infusion of areca nut at a dose of 79 mg/mL.

Abstract

Dogs and cats are often kept as pets, thus they stay close to humans. Parasitic/zoonotic diseases in humans were often caused by ectoparasite in cats and dogs. This study aimed to determine the morphological differences between cat fleas and dog fleas and the prevalence of cats and dogs infested by fleas around the home environment in Ilir Barat 1 District, Palembang City. The method used was descriptive exploratory method with field observation techniques and specimen collection. The study was conducted in November - December 2018 in five Sub-districts, (Bukit Lama, Bukit Baru, Lorok Pakjo, Demang Lebar Daun, and 26 Ilir 1). The results showed that cat fleas (Ctenocephalides felis) and dog fleas (Ctenocephalides canis) had different color and some morphological differences, such as the shape of the head, the number of spines in the genal comb, the number of notches in the tibia, and the number of seta in the metapisternum. The highest prevalence of fleas infestation in cats was in Bukit Lama and 26 Ilir 1 (33.3%), while the highest prevalence of infestation in dogs was in Demang Lebar Daun Village (36.6%). Only in Lorok Pakjo Sub-district that no dog fleas infesting dogs are found. __________________________________________________________

Abstract

virus was isolated from suspected Newcastle disease (ND) chicken in backyard farm at Sibang Village, Badung Regency, Bali Province. The isolates were then propagated and confirmed for NDV serologically. RNA isolation was performed by standard Trizol method. Phylogenetic tree analysis of NP gene (nt1020-1561) and HN gene (nt7019-7754) were performed using sequence of Badung-02/AK/14 and selected NDV strains from GenBank. Based on the NP gene sequence, the newly isolate closely related with other NDV strains belong to genotype VII that previously isolated such as Banjarmasin-010/10, Bali-1/07, and Cockatoo/90 with genetic distance 0.2%, 12.6%, and 18.0% respectively. The genetic distance with LaSota/46 virus (genotype II) is 19.6%. Based on HN gene sequence, genetic distance of the Badung-02/AK/14 with other viruses belong to genotype VII such as Banjarmasin-010/10, Bali-1/07, and Cockatoo/90 are 0.4%, 3.7%, and 4.2% respectively. The genetic distance with LaSota/46 virus (genotype II) is 9.0%. There were no difference between the result of nucleotide and amino acid sequence analysis, both in NP and HN gene (P0.05).

Abstract

Adhesi merupakan tahap inisiasi dari proses kolonisasi bakteri. Kemampuan adhesi ini diperantarai oleh keberadaan hemaglutinin pada permukaan bakteri. Tujuan penelitian ini adalah melihat kemampuan hemaglutinin pada Escherichia coli dalam proses adhesi. Uji adhesi pada sel epitel pipi dilakukan dengan menggunakan metode Velentine-Weiggand. Hasil penelitian menunjukkan bahwa Escherichia coli yang memiliki hemaglutinin menunjukkan kemampuan adhesi yang jauh lebih baik yakni rata-rata 67,6 bakteri/sel dibandingkan dengan   yang tidak memiliki hemaglutinin rata-rata 7,2 bakteri/sel. Kemampuan adhesi ini berkaitan dengan keberadaan hemaglutinin di permukaan bakteri.

Abstract

Penelitian ini bertujuan mengetahui ekspresi protein AdhF36 strain Salmonella Typhi, sebagai kajian kandidat vaksin berbasis molekul adesin. Strain yang digunakan pada penelitian ini bersumber dari Rumah Sakit Hasan Sadikin Bandung, Laboratorium Kesehatan Sulawesi Tengah, Palu, Rumah Sakit Udayana Denpasar, dan Rumah Sakit Saiful Anwar, Malang. Hasil penelitian menunjukkan bahwa sekitar 12 strain Salmonella Typhi tersebut mengekspresikan dan memiliki protein AdhF36. Hal ini memberikan gambaran umum bahwa protein AdhF36 potensial dikembangkan sebagai kandidat vaksin.

Abstract

This research aims to evaluate the protective potency of freeze-dried egg yolk powder on Garut ram sperm during the liquid semen preservation process. Semen with good sperm quality was divided into three groups and diluted using the following Tris diluents: fresh egg yolk-Tris (FEY-Tris), commercial egg yolk powder-Tris (CEY-Tris), and freeze-dried egg yolk powder-Tris (DEY-Tris). Semen that had been diluted was observed every 12 hours until sperm progressive motility was 50%. Sperm quality was tested for progressive motility percentages, viability, intact plasma membrane (IPM), and intact acrosome (IA). Fresh egg yolk had a particle size of 14460.00±330.76 nm, with polydispersity index value (PI) and zeta potential (ZP) being 1.00±0.11 and -9.22±0.30 mV, respectively. The particle size of commercial egg yolk powder was 877.90±168.86 nm with PI value 0.34±0.04 and ZP-28.7±1.24 mV. The particle size of freeze-dried egg yolk powder was 1296.00±86.73 nm, with PI value 0.70±0.04 and ZP -34.5±0.64 mV. Progressive motility percentage showed that DEY-Tris diluent managed to survive to 51.11±6.06% for 168 hours, compared to FEY-Tris (51.25±6.74%) which survived for 156 hours, although no longer than CEY-Tris diluent (53.47±6.33%) which survive for 180 hours (P0.05). Higher sperm motility was also supported by viability percentage, IPM, and IA, which were all higher (P0.05). In conclusion, freeze dried egg yolk powder mixed with Tris buffer was able to maintain sperm quality during the liquid semen preservation process.
