Abstract

This study was conducted to investigate the histological characteristic, type, and distribution of connective tissue in Sunda porcupine skin. The investigation was carried out in three adult of sunda porcupines at microscopic level using hematoxylin eosin, Masson thrichrome, Verhoeffs van Gieson, alcian blue pH 2.5 and periodic acid Schiff staining methods. Skin consists of epidermis, dermis hypodermis, and subcutaneous muscle. Quill follicles were the main and dominant structure as well as the specific characteristic on Sunda porcupine skin. The connective tissue was distributed well in basal membrane, dermis, quill follicle, and hypodermis with various intensity and density. The collagen was the main fiber found in the skin while the elastin fiber was not observed. The acid carbohydrate was found distributed well in the skin while the neutral carbohydrate was not detected in this study. In addition the fibers of connective tissue associated with the adipose tissue which found plentifully in quill follicles and hypodermis. The present results showed that the wide distribution of connective tissue might have an important role on the wound healing physiology of Sunda porcupine skin.

Abstract

Penelitian ini bertujuan isolasi dan purifikasi sel Leydig dengan menggunakan gradien Nycodenz untuk meningkatkan jumlah perolehan sel Leydig yang hidup setelah purifikasi dan kultur. Isolasi dan purifikasi sel Leydig dilakukan dengan perlakuan gradien Nycodenz 3 kolom (5, 10, dan 15% sebagai Nycodenz I) dan 5 kolom (4, 8, 10, 12, dan 15% sebagai Nycodenz II) serta gradien Percoll 5 kolom (21, 26, 34, 40 dan 60%) sebagai kontrol. Parameter yang diamati adalah konsentrasi, kemurnian, dan viabilitas sel Leydig setelah isolasi dan purifikasi serta kultur in vitro. Hasil yang diperoleh menunjukkan bahwa isolasi dan purifikasi sel Leydig dengan gradien Nycodenz (I dan II) secara nyata (P0,01) menghasilkan konsentrasi sel yang lebih rendah dibandingkan dengan gradien Percoll. Namun penggunaan gradien Nycodenz II c enderung menghasilkan kemurnian sel yang lebih tinggi (91,40%) dibandingkan dengan Nycodenz I (85,53%). Hasil tersebut tidak berbeda nyata dibandingkan dengan gradien Percoll (92,20%). Viabilitas sel Leydig pada semua perlakuan hampir sama yaitu 98%. Namun demikia n, setelah dikultur selama 3 hari, konsentrasi sel Leydig pada perlakuan Percoll (2,44x106 sel/ml) dan Nycodenz I (3,21x106 sel/ml) secara statistik (P0,05) lebih rendah dan dibandingkan dengan Nycodenz II (3,88x10 6 sel/ml), sedangkan viabilitas sel Leydig setelah dikultur pada gradien Nycodenz I (90,00%) dan Nycodenz II (91,17%) secara sangat nyata (P0,01) menghasilkan viabilitas yang lebih tinggi dibandingkan dengan gradien Percoll (82,30%). Berdasarkan hasil tersebut dapat disimpulkan bahwa penggunaan gradien Nycodenz II efektif untuk mempurifikasi sel Leydig dan setelah dikultur menghasilkan konsentrasi dan viabilitas sel yang lebih tinggi dibandingkan dengan gradien Percoll.

Abstract

The aim of this research was to find out the effect of probiotic on blood cholesterol in laying hens. Materials used were laying hens (Isa Brown strain) 43 weeks old and ration using probiotics. This research was conducted by using completely randomized design (CRD). The advanced research was done with testing the local probiotic with various concentration treatment, they are P0 (ration without probiotic), P1 (ration + 1% local probiotic), P2 (ration + 2% local probiotic), and P3 (ration + 3% local probiotic) with 5 time repetitions and in every repetitions consists of 2 chicken. Variables that were observed in the advance research were laying hens’s blood cholesterol that consists of total cholesterol, high density lipoprotein (HDL), low density lipoprotein (LDL), triglycerides, and total leucocyte. The level of total cholesterol on P0, P1, P2, and P3 were 129.00±38.26, 116.00± 26.53, 121.00±33.72, and 137.00±52.53 mg/dL (P0.05) respectively; the HDL level were 50.60±16.47, 35.80±11.28, 59.40±19.74; 56.80±30.56 mg/dL (P0.05); the LDL level were 100.20±49.53, 39.20±17.14, 104.60±51.23, and 96.80±66.22 mg/dL (P0.05), respectively; the trigycerides level were 862.00±355.5, 533.00±146.1, 829.00±242.6, and 810.00±304 mg/dL (P0.05), respectively; and leucocyte count were 22.00±1.72, 21.00±3.79, 31.00±3.99, and 34.00±3.55 x103 mm3 (P0.01), respectively. In conclusion, the administrtion of local probiotic do not effect the blood cholesterol of layer chicken.

Abstract

Penelitian ini bertujuan mendeteksi brucelosis pada sampel susu sapi dengan uji polymerase chain reaction (PCR) dan membandingkan tingkatsensitivitas dan spesifisitasnya dengan metode milk ring test (MRT). Sebanyak 24 sampel susu sapi yang dikoleksi secara aseptik dari lapang diuji PCR dan MRT. Hasil pengujian menunjukkan bahwa 79,17% (19/24) sampel susu positif brucelosis dengan uji PCR dan 83,33% (20/24) dengan uji MRT. Sensitivitas dan spesifisitas PCR mendeteksi brucelosis masing-masing sebesar 75 dan 100% dibandingkan dengan uji MRT.

Abstract

This study aimed to develop the use of thermographic analysis to evaluate wound healing for laparotomy surgery in animals. The experiment used nine adult dogs 5 males and 4 females, bodyweight 5-10 kg) that had undergone the same general anesthesia and laparotomy procedures. Evaluation of wound healing was performed at 24, 48, and 72 hours after surgery and 7 days after operations using a digital thermal camera. During each thermographic evaluation period, blood samples were taken for analysis of total leukocytes and leukocyte differential. The results of thermal imaging were compared with the values of inflammatory cells and the clinical condition of wound healing. Comparison of thermographic analysis with inflammatory status was evaluated using the regression equation and showed a strong correlation coefficient (Y= -0.4847 + 40.14, R2= 0.867), that is, the higher the temperature, the lower the inflammation. The conclusion of study is that thermographic analysis can be used to evaluate wound healing of laparotomy surgery in dogs.

Abstract

The aim of this study was to identify proteolytic bacteria from the ruminal and reticulum fluids of aceh cattle based on the 16S rRNA gene. Samples used were ruminal and reticulum fluids of aceh cattle slaughtered in Abattoir of Aceh Besar. Samples were diluted and cultured into Skim Milk Agar medium at 39 C for 48 hours. The morphology of bacterial colonies growth in the medium was observed. Colonies resulted in the largest clear zone were isolated and used for Deoxyribonucleic Acid (DNA) isolation, 16S rRNA gene amplification and sequencing. Theresults showed that morphology of dominant colonies was yellowish white color, round shape, position on the agar surface. The results ofphylogenetic analysis of RS1 and ReS2 isolates isolated from rumen and reticulum fluids of aceh cattle respectively had a close familialrelationship and belonged to the bacterial group of Kosakonia. Sequence homology showed isolate RS1 and ReS2 are probably either newEnterobacteriaceae species or unconfirmed species. Halo zone produced by ruminal bacteria had a wider diameter (25 mm vs 20 mm) than thatcaused by reticulum bacteria). Based on the results, RS1 (bacterium in the rumen) and ReS2 (bacterium in the reticulum) belong to similar type, namely Kosakonia sp. with a proteolytic activity. Presumably, these bacteria originate from the rumen that enters the reticulum with degraded feed.

Abstract

The study was done to investigate the glycogen distribution in the stomach of long tailed macaques (Macaca fascicularis). The study used eight individuals of long tailed macaques consist of 6 fetuses (55, 70, 85, 100, 120, and 150 days pregnancy) and 2 infants (10 and 105 days). The gastrict tract were histologically stained using Hematoxyllin-Eosin (HE), and histochemically stained using Periodic Acid Schiff (PAS), to detect glycogen. The results revealed that the histogenesis pattern of stomach glandullar cells are developed in line with the age of the fetuses and infant. We also found that glycogen existance are increase in line with the age in the glandullar stomach. From the present study we concluded that the glycogen distribution in gastrict glandular cells were influenced by the age in the fetal period, while in the infant was under influence of the digestion activities, and the activities of the gastrict glandular cells was already seen at 55 days gestation.

Abstract

Penelitian ini bertujuan mengamati stabilitas ekspresi protein AdhF36 Salmonella typhi pada perlakuan osmolaritas dan pH dengan metode eksperimen laboratorium-eksploratif. Hasil penelitian ini menunjukkan perbedaan ekspresi protein AdhF36 Salmonella typhi pada perlakuan osmolaritas dengan pH. Hasil SDS-PAGE menunjukkan bahwa pita protein AdhF36 tetap terekspresi pada perlakuan osmolaritas 50-350 mM, sebaliknya pita protein ini tidak terdeteksi pada perlakuan pH 4,5-6,0. Hal ini didukung oleh hasil uji Western Blot yang berhasil mendeteksi protein AdhF36 hanya pada perlakuan osmolaritas. Hasil ini membuktikan bahwa Salmonella typhi lebih toleran terhadap perubahan osmolaritas dibandingkan dengan pH.

Abstract

The purpose of this study was to examine the molecular basis of Tursiops sp. originated from Java sea based on gene sequences ofCytochrome C Oxidase sub-unit II. Samples of blood were collected from five male bottlenose dolphins from PT. Wersut Seguni Indonesiacaptivity. Total deoxyribonucleic acid (DNA) was isolated from blood samples, then amplified by polymerase chain reaction (PCR), sequenced,and the data were analyzed using MEGA version 5.1 program. The amplification result of the PCR product obtained 824 base pairs (bp), andDNA sequencing obtained 684 nucleotides constituent of COX II genes that encode 228 amino acids making up the COX II. Nucleotide andamino acid sequence of COX II bottlenose dolphins originated from Java sea has a very high homology to Tursiops aduncus that are 99.13%and 100% respectively. Phylogram by neighbour joining method based on the nucleotide sequence of COX II gene indicating bottlenose dolphinsfrom Java sea belongs to group of Tursiops aduncus.Key words: bottlenose dolphin, COX II gene, nucleotide, Jawa Sea

Abstract

Tujuan penelitian ini mengetahui kualitas mikrobiologi susu kambing mentah yang diambil langsung dari ambing. Dalam penelitian ini digunakan 50 sampel susu kambing peranakan Ettawa (PE). Sampel dianalisis terhadap total plate count (TPC), jumlah Staphylococcus sp., jumlah koliform, Escherichia coli (E. coli), dan Salmonella sp. berdasarkan reaksi biokimia. Rerata untuk TPC; Staphylococcus sp.; total koliform masing-masing adalah 1,65x10 3 ; 5,75x10 3 ; 1,3x10 cfu/ml, sedangkan E. coli dan Salmonella sp. adalah negatif. Berdasarkan Standar Nasional Indonesia (SNI) No 01-6366-2000 tentang persyaratan susu segar, maka TPC, koliform, E. coli, dan Salmonella sp. memenuhi standar, sedangkan Staphylococcus sp. melebihi ambang batas. Hasil penelitian menunjukkan bahwa susu kambing mentah yang diambil langsung dari ambing masih layak konsumsi.
