Abstract

The purpose of this study was to examine the efficacy of a herbal combination of avocado, mung bean sprouts, and holy basil (Jamu ATOKE) in optimizing the reproductive health of adult female rats. Eighteen female Sprague Dawley rats aged 9-10 weeks old weighing around 180-250 g, and had not been pregnant previously were equally and randomly divided into 3 groups: one control group and two treatment groups (n= 6 rats per group). The treatment groups received Jamu ATOKE with a concentration of 2.5 and 5%, respectively. Jamu ATOKE was added to the drinking water and consumed by the rats for 30 days prior to pregnancy. Reproductivity (pregnancy success, estrogen and progesterone hormone concentrations), and health performance (body weight gain, food, and water consumption, motor activity, red blood cell (RBC) count, white blood cell (WBC) count and differentials, SGPT, SGOT, ureum, and creatinine) of rats were observed. The results showed that groups receiving the Jamu ATOKE had better reproductive and health performance compared to those of the control group. The administration of Jamu ATOKE can significantly increase the concentration of estrogen and progesterone in rats (P0.05). It can also improve the fertility and health of the female rats prior to pregnancy.

Abstract

The aim of this study is to evaluate the representation of erythrocytes, which is the count of erythrocytes; hemoglobin concentration, hematocrit value, and platelet count in piglet (Sus scrofa) after hypovolemic shock with normal and hypervolemia resuscitation of crystalloid fluid. This study uses nine male castrated piglets 6-8 weeks of age. The piglets are given treatments such catheter installation, shock induction, normovolemia resuscitation, and hypervolemia resuscitation. The fluid for resuscitation is crystalloid fluid, NaCl 0.9%. Blood sample is taken in baseline, normovolemia condition, and hypervolemia condition. The sample is analyzed using hematology blood analyzer and is tested using T-Paired. Total erythrocytes count after normovolemia resuscitation is 3.07x106/µL, while after hypervolemia resuscitation the erythrocytes count decrease until 2.86x106/µL. Hemoglobin concentration after normovolemia resuscitation is 9.4 g/dL, while after hypervolemia resuscitation hemoglobin concentration decreases to 8.64 g/dL. Hematocrit count from piglets after normovolemia resuscitation is 28 %, while it decreases after hypervolemia resuscitation to 25.89%. Platelet count after normovolemia resuscitation is 297.22x103/mm3 and decreases to 263.89x103/mm3 after hypervolemia resuscitation. Evaluation of the erythrocytes index shows that the piglets suffer anemia. Hypovolemic shock and normal, and hypervolemia resuscitation cause the significant decreases in all erythrocytes indexes except the platelet count.

Abstract

The experiment was conducted to study the effect of ethylene glycol (EG) on disturbance of spermatogenesis process of the male mice. Sixteen mice at the age of 2 months were randomly into 4 groups of 4 each. The mice of control group were only given drinking water without EG. Group KPI, KP2, and KP3 were given EG 5, 10, and 15 ml/liter drinking water, respectively. On day 31 of post treatment, all mice were killed for their testis. The testis were collected and processed for microscopic examination and haematoxylin-eosin staining. The result showed that EG given orally for 30 days on male mice have  negative effect on the process of spermatogenesis as the numbers of spermatogonia, spermatocyte and Sertoli cells decreased significantly (P0.01)

Abstract

Penelitian ini bertujuan mengetahui persentase leukosit polymorphonuclear (PMN) dalam preparat ulas darah sapi endometritis. Enam ekor sapi endometritis dibagi dalam dua kelompok perlakuan. Kelompok I (n=3) diterapi dengan 250 mg gentamisin/ekor, 250 mg flumequin/ekor, dan PGF2α sebanyak 12,5 mg/ekor secara intra uteri. Kelompok II (n=3) diterapi dengan menggunakan antibiotik dengan dosis dan cara pemberian yang sama seperti pada Kelompok I. Hasil penghitungan leukosit diferensial sebelum terapi menunjukkan persentase jumlah limfosit yang lebih tinggi dibandingkan bentuk leukosit lainnya pada Kelompok I dan II masing-masing adalah 62,50±1,17 dan 63,66±2,35, sedangkan persentase jumlah neutrofil pada Kelompok I dan II masing-masing adalah 29,33±0,94 dan 27,33±0,94. Setelah terapi, tidak ada perbedaan persentase (P0,05) bentuk leukosit antara kedua kelompok perlakuan. Terapi kombinasi antibiotik dan PGF2α pada sapi penderita endometritis tidak menghasilkan perubahan diferensial leukosit termasuk PMN.

Abstract

This research was aimed to find out the relationship between the diameter of follicle and thedifferent of follicle number on oocyte quality of local goat. The oocyte originated from the ovary was takenfrom the Slaughterhouse in Banda Aceh. The ovary was aspirated in the Histology Laboratory ofVeterinary Medicine Faculty of Syiah Kuala University, Banda Aceh. This experiment consisted of twostages, the follicles were selected and divided into 3 groups according to their diameter size. The smalldiameter size 2 mm, medium size 2-5 mm, and big size 5 mm. The second, the ovaries were dividedinto three groups based on number of follicle, i.e; 1(≤5; 2(6-10); and 3 (≥11). The diameters of oocytetaken from the three groups of follicle size were measured by using eyepiece micrometer, while the oocytemorphology was categorized into 4 groups. The result of regression data indicated that the biggest folliclediameter yielded the oocyte that had bigger diameter, too. The model of regression is y = 0.955 + 0.157x, y is the diameter of oocyte and x is the diameter of follicle. The close relationship (r) between the diameter offollicle and the diameter of oocyte was 0.967. There are significant differences (P0.05) between the size offollicle and the morphology of oocyte. The oocytes originated from larger follicle tend to have bettermorphological appearance than the oocyte originated from smaller follicle.Keywords: oocyte, follicle

Abstract

Penelitian ini bertujuan mendapatkan prosedur atau metode kromatografi cair kinerja tinggi (KCKT) yang valid dan optimal dalam analisis deltamethrin sebagai senyawa yang berpotensi menjadi residu dalam produk hewan. Alat utama yang digunakan dalam penelitian ini adalah satu set KCKT Shimadzu 6.1, dengan kolom C-18 (30° C), panjang gelombang detektor UV-vis 236 nm. Fase gerak yang digunakan dalam penelitian ini adalah asetonitril 80% dalam akuabides yang dialirkan dengan laju 1,25 ml/menit. Hasil penelitian menghasilkan kromatogram yang terlihat menunjukkan peak area yang nyata terpisah dari senyawa lain. Batas deteksi diketahui pada konsentrasi 0,1 μg/ml, sedangkan batas kuantifikasi pada konsentrasi 0,5 μg/ml. Rerata luas area untuk konsentrasi 0,5; 1; 1,5; 2; 5; dan 10 μg/ml masing-masing adalah 18.255,33; 47.142,00; 55.587,00; 64.181,33; 204.269,00; dan 395.918,00 dengan persamaan garis linier y= 39.866x-1.719,5 (R= 0,99). Hasil analisis juga menunjukkan presisi dan akurasi hasil yang baik. Hasil penelitian dapat disimpulkan bahwa metode yang dikembangkan pada penelitian ini mempunyai validitas yang baik dan optimal untuk analisis deltamethrin, yang merupakan senyawa potensial menjadi residu pada produk asal hewan.

Abstract

Tujuan penelitian ini adalah mendapatkan informasi dasar mengenai struktur genetika menggunakan marka melokuler DNA mikrosatelit gen bovine lymphocyte antigen (BoLA) dan hubungannya dengan kualitas semen sapi bali. Data diambil dari 8 ekor sapi jantan yang ada di Unit Pelaksana Teknis Daerah (UPTD) Inseminasi Buatan, Baturiti, Tabanan, Bali. Hasil pemeriksaan kualitas sperma menunjukkan bahwa volume semen sapi bali 3,0-6,5 ml, konsentrasi spermatozoa 800-11.000 juta sel/ml, dan persentase spermatozoa motil 70-75%. Hasil penelitian pada sapi bali menunjukkan bahwa ketiga lokus mikrosatelit yang digunakan teramplifikasi pada sapi bali dan jumlah alel pada lokus RM185 dan BM1815 adalah 2 sedangkan jumlah alel pada lokus DRB3 adalah 1. Heterozigositas per lokus berkisar 0-0,533. Nilai PIC per lokus antara 0-0,375. Dari hasil penelitian ini dapat disimpulkan bahwa ukuran alel pada ketiga mikrosatelit berpotensi sebagai marka molekuler yang berperan terhadap kualitas semen pada sapi bali.

Abstract

Tujuan dari penelitian ini adalah untuk mendeteksi keberadaan virus Newcastle disease (VND) dan mengkaji keragaman dari virus terisolasi. Sampel penelitian berupa usapan kloaka dan orofaring dari 177 ekor ayam kampung yang diambil dari unggas pekarangan dan pasar unggas di 12 kecamatan dalam wilayah Kabupaten Aceh Besar dan Kota Banda Aceh. Penapisan virus dilakukan pada sampel pool dengan real-time reverse transcriptation-polymerase chain reaction (rRT-PCR) dengan target gen matriks. Inokulasi 309 sampel representasi 157 ayam asal pool positif matriks pada telur ayam berembrio spesifik pathogen free (SPF) menghasilkan 69 isolat yang berasal dari 51 ekor ayam. Sebagian besar (45,09%)ayam mengeluarkan virus melalui orofaring, 25,39% melalui kloaka dan orofaring, serta 19,61% melalui kloaka. Karakterisasi keragaman isolat dilakukan dengan uji hemagglutination inhibition (HI) menggunakan serum Komarov dan Hitchner B1, rRT-PCR gen fusi dan uji elusi. Adanya keragaman epitop permukaan virus ditunjukkan dengan titer HI yang bervariasi antar isolat, perbedaan mencapai 4 log dengan serum Komarov, dan 3 log dengan B1. Sebagian besar isolat mempunyai afinitas yang lebih tinggi terhadap serum Komarov yang mengindikasikan kecenderungan kepada galur virulen. Penentuan patogenisitas menggunakan rRT-PCR menunjukkan 73,95% isolat termasuk ke dalam galur virulen (mesogenik/velogenik), sementara dari uji elusi menunjukkan 72,46% isolat termasuk galur velogenik, 20,29% mesogenik dan 7,25% dari galur lentogenik.

Abstract

The objective of this study was to evaluate the effect of exercise levels on the hematological and metabolical status, and carcass quality of Garut sheep. In total 24 of Garut sheep used in this study. The levels of exercise contain three treatments namely non exercise, semi exercise  and exercise. Blood collection through the jugular vein of sheep samples was carried out at the end of the study, using a 5 mL EDTA tube to measure the haematological condition of the animal sample. Likewise, collecting muscle samples to determine the physical quality of meat.The data was analyzed by analysis of variance and continued by Duncan test for a post hoc test. The results showed that the models of exercise were not significantly (P0.05) affected to hematological level and also carccas quality including juiceness, tenderness, drip loss and water holding capacity. Organoleptic sensory  using semiexercise was more favourable meat  compare to exercise and non exercise. While for the colour of meat, non exercise treatment was significantly (P0.05) affected to the colour of meat compare to exercise and semi exercise. Furthermore the semi exercise  was the best treatment to produce fresh taste meat with good sensory characteristic.

Abstract

This study aimed to improve reproductive condition of postpartum anestrus of Etawa crossbreed (PE) goats using PGF2α hormone or progesterone (controlled internal drug release/CIDR-G) and improve pregnancy success from artificial insemination (AI) with human chorionic gonadotrophin (hCG) hormone administration. Administration of progesterone increased estrus by 100%, which was higher than the 70% produced from prostaglandin F2 alfa (PGF2α) administration. Estrus onset in CIDR group (35.80±13.33 hours) was faster than PGF2α group (45.6±9.2 hours). Clinical signs of vulva (redness and swollen) were more intense in CIDR-G group (80% and 70%, respectively) compared to PGF2α group (70% and 57%, respectively). Estrus duration in PGF2α group (52.17±3.34 hours) was longer than CIDR-G group (49.61±3.56 hours). Pregnancy success in group given hCG was 66.7% while in non-hCG group was 50%. In conclusion, CIDR-G and PGF2α administration is effective to induce estrus of postpartum anestrus PE goats. Administration of hCG at the onset of estrus is effective to improve pregnancy success.
