Abstract

This study was aimed to perform phenotypic and genothypic characterization of Escherichia coli (E. coli), particularly VTEC strain isolated from cattle faeces. In this study, 25 E.coli isolated from faeces specimens and faeces base fertilizer of dairy and beef cattles were used. Examination were carried out using phenotypic and genothypic characterization which is specified for E coli VTEC strain. The result showed that 20 % samples of fresh faeces specimens were detected as VTEC strains and none of isolate was detected from faeces base fertilizer samples. From VTEC strains, could detect 16 % VT1 gene, 12 % VT2 genes and 8% of both. Detection on gene pyelonephritis-associated pilli (pap), S fimbrial adhesion (sfa), and afimbrial adhesion (afa) were found about 60%, 80% and 80%, respectively.

Abstract

Tujuan dari penelitian adalah menetapkan daya tahan hidup takizoit Toxoplasma gondii galur RH dalam susu kambing setelah dipasteurisasi pada suhu tinggi dalam waktu singkat. Dalam penelitian ini digunakan metode in vivo dan mencit diinfeksi dengan takizoit Toxoplasma gondii galur RH secara intraperitoneal dengan konsentrasi 2,76x106 takizoit/ekor. Mencit dibagi atas tiga kelompok perlakuan, yaitu susu pasteurisasi dan takizoit yang dipanaskan pada suhu 72 °C selama 15 detik (P), susu pasteurisasi dan takizoit tanpa dipanaskan sebagai kontrol positif (KP), dan  susu pasteurisasi tanpa takizoit sebagai kontrol negatif (KN). Hasil penelitian menunjukkan bahwa tidak ditemukan takizoit di dalam cairan peritoneal pada P dan KN. Takizoit Toxoplasma gondii galur RH ditemukan pada KP yang mempunyai jumlah konsentrasi hampir sama sebelum dan setelah infeksi.

Abstract

Catechin and their derivatives have been studied to have antiviral potential against COVID-19 through in silico method “pharmacokinetics screening and molecular docking”. Pharmacokinetics and toxicity profiles were obtained through the ADMETSAR server and SwissADME server. Then proceed with the prediction of affinity through the method molecular docking using the software application MOE 2007.09. The testmaterial is in the form of a 3D catechin structure and its derivatives as well as several control ligands downloaded via Pubmed. While templatethe Receptor M pro protein and the Spike glycoprotein are downloaded from pdb.org (PDB ID: 6LU7 and 6LXT). The pharmacokinetic profile of catechins is relatively better than all control ligands with the lowest toxicity. Molecular docking results also show that catechins and theirderivatives have a stronger affinity than control ligands. This research proves that catechin has antiviral potential through inhibition of Mpro protein and Spike glycoprotein COVID-19 virus.

Abstract

The aim of this research was to find out the total erythrocyte count, hematocrit value, and hemoglobin concentration in broiler chicken treated with turmeric, garlic, and zinc supplementation. One hundred broiler chickens were divided intofive groups by randomized and four replications of each. Feed supplement combinations were R0 (basal), R1 (garlic and turmeric), R2 (garlic and zinc), R3 (turmeric and zinc), and R4 (turmeric, garlic and zinc). Blood sample was taken at the age of 6 weeks after treatment to determine total erythrocyte count, hematocrit value, and hemoglobin concentration. The result showed that total erythrocyte count in group of R3 (turmeric powder 1.5% and zinc oxide 120 ppm) were significantly higher than R0 (P0.05) were 3.32±0.73x106/ìl. Total erythrocyte count, hematocrit value, and hemoglobin concentration groupof R2 and R3 were tended to be higher than R0, meanwhile group of R4 did not different from R0. Feed supplemented with hebal and zink would increase the total erythrocyte count, hematocrit value, and hemoglobin concentration in broiler chicken.

Abstract

Penelitian ini bertujuan mengetahui respon estrus pada kambing Peranakan Ettawa (PE) dengan Body Condition Score (BCS) 2 dan 3 terhadap implant Controlled Internal Drug Release (CIDR) jangka pendek dikombinasikan dengan injeksi prostaglandin F alpha (PGF a). Sebanyak 10 ekor kambing PE betina dibagi 2 2 dalam 2 kelompok perlakuan berdasarkan BCS, yaitu kelompok I dengan BCS 2 (kondisi kurus), berat badan antara 25-30 kg (n=5) dan kelompok II dengan BCS 3 (kondisi ideal) berat badan antara 35-40 kg (n=5). Semua kelompok perlakuan disinkronisasi dengan implant CIDR (berisi 1,3 g progesteron) secara intravaginal jangka pendek selama 10 hari dan 48 jam sebelum pencabutan CIDR, kambing diijeksi dengan PGF a. Deteksi estrus dilakukan setiap hari dengan interval 6 jam dimulai dari pencabutan CIDR sampai 60 2 jam setelah pencabutan CIDR. Data onset dan durasi estrus dianalisis menggunakan independent sample T-test sedangkan tanda-tanda visual estrus dianalisis secara deskriptif. Respon estrus semua kelompok perlakuan adalah 100%. Onset dan durasi estrus antara BCS 2 dan 3 masing-masing adalah 21,60±1,47 vs 13,20±1,20 jam dan 27,60±3,06 vs 32,40±1,47 jam. Dari hasil penelitian dapat disimpulkan bahwa kombinasi implant CIDR jangka pendek dan injeksi PGF a efektif untuk sinkronisasi estrus pada kambing PE dengan 2 BCS 2 dan 3.

Abstract

The purpose of this study was to determine the prevalence, serovar, and risk factors of leptospirosis on cattle in Progo watershed. A total of 330 cattle samples were clinically examined and blood was collected for Leptospira examination using microscopic agglutination test (MAT). Results of MAT were used as dependent variable (Y), while breeders, cattle, shed, and feed factors were used as the independent variable (X). Univariate analysis, bivariate analysis with chi square, odds ratio, and multivariate with logistic regression were implemented to analyze the data. The results showed that all cattle were cinically health, 13.03% (43/330) cattle were positively leptospirosis which 37 out of 193 (9.17%) cattle belongs to the farmer. Mostly, leptospirosis cases were caused by hardjo serovar (38.0%). Risk factors influencing the prevalence of leptospirosis were caused by some factors such as the presence of rats in cage (OR 2.7), the distance of waste disposal which should be less than approximately 5 meters (OR 2.1), feed concentrates, grass and rice straw (OR 0.4), grazing (OR 0.2), and concentrate feed and rice straw (OR 0.1). It can be concluded that leptospirosis infection on cattle in Progo watershed is 13.03% (at livestock level) and 19.17% (at farm levels). Factors of rat in the cage and the distance of waste disposal (5 m) increase the risk of leptospirosis cases. Factors of feed combine with concentrate and rice straw as well as grazing reduced the risk of leptospirosis in cattle, but the addition of grass in concentrate feed and rice straw provides a greater risk of leptospirosis infection compare to concentrate feed and rice straw alone.Key words: beef cattle, leptospirosis, prevalence, risk factors

Abstract

Penelitian ini bertujuan mengetahui gambaran darah merah pada tikus yang diimplan porous tantalum berlapis hidroksiapatit. Dalam penelitian ini digunakan 12 ekor tikus jantan, galur Sprague Dawley, umur ±3 bulan yang dibagi dalam tiga kelompok yaitu kelompok kontrol tanpa implant (K1), kelompok implan porous tantalum (K2), dan kelompok implan porous tantalum berlapis hidroksiapatit (K3). Pengambilan darah dilakukan melalui vena pada ekor di hari ke-0 sebelum pemasangan implan, ke-14, dan ke-30 setelah pemasangan implan. Pengamatan data parameter darah merah meliputi jumlah sel darah merah, kadar hemoglobin (Hb), dan persentase hematokrit. Hasil perhitungan jumlah sel darah merah, kadar Hb, dan hematokrit menunjukkan perbedaan yang tidak nyata (P>0,05) pada masing-masing kelompok perlakuan kecuali jumlah sel darah merah pada kelompok tikus yang yang diimplan dengan porous tantalum tanpa lapis meningkat pada hari ke-30 dan menunjukkan perbedaan dibandingkan kelompok lainnya (P

Abstract

The purpose of this study was to evaluate the characteristics of carcass yields and internal and external organs of the broilers fed the diets containing the fermented moringa leaf meal (FMOL) + yellow corn (YC), the moringa-corn (MC) diet formulated to replace parts of the commercial diet (CD). As many as 100 DOC of broiler chicken strain CP 707 were reared for up to 5 weeks to feed experimental diets. A Block Randomized Design (BRD) with subsamples consisted of 4 treatments, 5 blocks, and 2 subsamples were applied in the recent study. The results of studies indicated that removing parts of the CD diet to substitute with the mixture of FMOL + YC caused a significant effect (P0.05) in declining the weights of the whole carcass and the cut-ups but in percentage no significant differences. The weights and percentages of all examined organs were not significantly affected (P0.05). It was concluded that using the moringa mixed with yellow corn reduced the weight of the whole carcass and cut-up carcasses but it could use up to 7.5% in the diet without detrimental effects on the internal and external organs of the broilers.

Abstract

The purpose of this study was to evaluate effect of Coleus amboinicus (CA) stem extracts on uric acid-induced nephropathy by comparingthe levels of Macrophage CD-68 expression and concentration of serum Cystatine C (CYS C ) in Wistar rats. Twenty-four male Wistar rats(Rattus norvegicus) with a body weight (bw) of 200-250 g, were allocated into three groups, with eight animals per group. The rats in controlgroup (PO) received 0.1% carboxymethyl cellulose (CMC) solution orally The rats in group 2 (P1) were orally induced with uric acid (UA) (500mg/kg) and oxonic acid (OA) (750 mg/kg.) and the rats in group 3 (P2) received uric acid (500 mg/kg), oxonic acid (750 mg/kg), and 500 mg/kgof the CA stem extracts for 35 days. Bloods were collected for analysis of serum CYS C expression and concentration of serum creatinine andblood nitrogen urea (BUN). The rats in all groups were sacrificed for kidney tissue extractions for macrophage CD-68 identification andhistopathology analysis. The levels of CYS C concentrations were analyzed by Avidin-Horseradish Peroxidase (HRP) Sandwich-ELISA. Theresults showed that Coleus amboinicus stem extract at dose of 500 mg/kg bw can significantly reduce BUN and creatinine levels (P≤0.05), whileCys C levels were not different. In the treatment group (P2) compared with group (P1). CD-68 (ED-1) macrophage activity decreasedsignificantly (P≤0.05) in the treatment group (P2) compared to the control group and (P1). Nephrophaty induction using UA and OA causessevere kidney lesions characterized by degeneration, necrosis and inflammation of the renal tubules and glomerulus in the treatment group.

Abstract

The aim of this research was to analyze the anti-oxidant activities of ethanol extract of mahogany seeds on the liver tissues of diabetic experimental rats to overcome oxidative stress condition. This study used male Rattus norvegicus strain Sprague Dawley which were divided into 5 groups: (i) negative control group (K-), (ii) positive control group/diabetes mellitus (DM) (K+), (iii) DM group treated with 500 mg/kg BW ethanol extract of mahogany seeds (EM), (iv) DM group treated with acarbose (KO), and (v) non-DM group treated with 500 mg/kg BW ethanol extract of mahogany seed (KE). Alloxan with dose of 110 mg/kg BW was used to induce diabetes in rats. The treatments were conducted for 28 days. At the end of treatment, the liver tissues were collected and analyzed for malondialdehyde (MDA) content using thiobarbituric acid reactive substance (TBARS) method and anti-oxidant cooper zinc superoxide dismutase (Cu,Zn-SOD) content using immunohistochemical technique. The result showed that level of MDA were significantly different (P0.05) in K-; K+; EM; KO; and KE with values were 0.81±0.17; 1.19±0.26; 1.10±0.04; 0.95±0.13; and 0.92±0.0 µmol/gram, respectively. The anti-oxidant content (Cu, Zn-SOD) of rat liver tissue in K+ lower than in K-, while anti-oxidant content in EM and KE were higher compared to K+. The study concluded that ethanol mahogany seeds extract decreased blood glucose levels and increased the anti-oxidant status in the liver tissues of diabetic rats.
