Abstract

Penelitian ini bertujuan membuktikan pengaruh alanin-glutamin dipeptida 7% terhadap perbaikan fungsi hipokampus melalui pengamatan beberapa parameter kinerja motorik seperti jarak tempuh (JT), waktu stereotipe (WT), waktu ambulatori (WA), dan waktu istirahat (WI). Penelitian ini dilakukan dengan memberikan alanin-glutamin dipeptida 7% selama 12 hari pada tikus umur 12 dan 24 bulan yang mengalami penuaan fisiologis dan penuaan akibat stres oksidatif. Pengamatan terhadap semua parameter kinerja motorik dilakukan dengan alat Opto-Varimex yang dikoneksikan dengan software Autotract. Hasil penelitian menunjukkan alanin-glutamin dipeptida 7% berpengaruh terhadap perbaikan kinerja motorik pada kondisi penuaan fisiologis dan penuaan akibat stres oksidatif.

Abstract

The objectives of this research were to examine the effect of Sechium edule ethanolic extract to repair beta pancreas cells damage and tomeasure nitrogen oxide (NO) concentration in streptozotocin-induced rats. Type I diabetic rats were obtained by intraperioneally injected therats with multiple low dose streptozotocin (MLD-STZ) for 5 days. In this research, rats were assigned into 5 treatment groups: untreated rats ingroup 1 were considered as negative control, while MLD-STZ induced type 1 diabetic rats in group 2 were considered as positive control. Inanother 3 groups, type 1 diabetic rats were orally treated with three doses of Sechium edule ethanolic extract (14, 28, 42 mg/kg bw) respectivelyfor 7 consecutive days. Repairing of beta pancreas cells were analyzed descriptively whereas concentration of NO were analyzed using one wayANOVA and continued with LSD test (α= 0.01). The result showed that the administration of Sechium edule ethanolic extract significantlydecreased NO concentration (P0.01) up to 67.9 µM and repaired beta pancreas cells damage. Dose of 42 mg/kg bw of Sechium edule ethanolicextract is the effective dose for diabetic rat therapy. Key words: Sechium edule extract, diabetic mellitus, rat, beta pancreas cells

Abstract

Penelitian ini bertujuan mengetahui efektivitas pengobatan endometritis dengan menggunakan kombinasi antibiotik (gentamicine, flumequine) dan analog prostaglandin F2 alfa (PGF2α) berdasarkan pengamatan dinamika ovarium dengan metode ultrasonografi (USG). Enam ekor sapi endometritis dibagi dalam 2 kelompok perlakuan. Kelompok I (K1, n= 3) diterapi dengan 250 mg Gentamicine, 250 mg Flumequine, dan 12,5 mg PGF2α secara intra-uterus. Kelompok II (K2, n= 3) diterapi menggunakan antibiotik dengan dosis dan cara pemberian yang sama seperti pada Kelompok I. Hasil pengamatan terhadap sapi-sapi endometritis K1 dan K2 memperlihatkan rataan panjang siklus estrus selama 18 hari. Hasil analisis statistik menunjukkan tidak ada perbedaan yang nyata (P>0,05) terhadap kelas folikel pada kedua kelompok perlakuan. Abstract

Leptospirosis is an infectious disease caused by bacteria from the genus Leptospira with various serovars and often presents with a wide range of clinical signs from mild to severe conditions, particularly it presents with a mild fever to asymptomatic infection-causing animal death. The purpose of this study was to identify the characteristics of canine leptospirosis through laboratory examinations supported by medical record data in a small animal practice at the drh Cucu K. Sajuthi Joint Veterinary Practice, in Jakarta. The study was conducted on serum samples of sixteen dogs from drh Cucu K. Sajuthi Joint Veterinary Practice patients, which presented varied clinical signs of leptospirosis, and which had been accompanied by hematological examination, blood biochemistry, and Microscopic Agglutination Test (MAT) against various serovars such as Bataviae, Icterohaemorrhagiae, and Javanica. The collected data was analyzed descriptively. The results showed that canine leptospirosis was more commonly derived from Bataviae serovar and was accompanied by clinical signs such as vomiting, jaundice, anorexia, abdominal pain, lethargy, pale mucosal membrane, diarrhea, dehydration, dyspnea, and polyuria/polydipsia. A predisposition of gender and age was commonly found in male dogs less than 5 years old. Abnormalities found from hematological examination were leukocytosis, granulocytosis (neutrophilia), anemia, thrombocytopenia, and lymphopenia, while blood chemistry showed azotemia, an increase in liver enzymes (AST, ALT, ALP), hyperbilirubinemia, hyponatremia, hyperphosphatemia, and hypoalbuminemia.

Abstract

The objectives of this study were to assess the diversity of Newcastle Disease Virus (NDV) isolates; to detect and isolate NDV from poultry; and to identify and characterize NDV by serological and molecular assays. A total of 84 cloacal-oropharynx isolates of poultry was collected from privately owned poultries and poultry markets from 12 districts in Aceh Besar and Banda Aceh. Screening was performed by real time reverse transcriptation-polymerase chain reaction (rRT-PCR) to 15 isolates of poultry. Selected isolates were inoculated in 9-11 days old embryonated specific pathogen free (SPF) eggs and showed positive hemagglutination (HA). Characterization was performed through hemagglutination inhibition (HI) test using Komarov and Hitchner B1 antisera, elution test, RT-PCR and realtime RT-PCR fusion (F). All isolates had a higher affinity to Komarov antisera (titer up to 4 log), indicating virulent strain. This was supported by elution test which showed that 93.66% isolates were virulent and 6 % non-virulent. In conclusion, RT-PCR can detect Matrix gene from 15 isolates (100%), while Fusion gene only detected from 11 isolates (73.3%). rRT-PCR is more capable of detecting antigenic diversity compared to RT-PCR.

Abstract

Penelitian ini bertujuan mengetahui efek injeksi inhibin B terhadap penurunan konsentrasi follicle stimulating hormone (FSH) di dalamserum pada tikus putih (Rattus norvegicus). Dalam penelitian ini digunakan 24 ekor tikus putih berjenis kelamin jantan dengan strain Wistar berumur 4 bulan dengan bobot badan 150-200 g. Tikus-tikus dikelompokkan secara acak ke dalam 4 kelompok, yaitu KK0, KP1, KP2, dan KP3, masing-masing kelompok terdiri atas 6 ekor. Kelompok KK0 merupakan kelompok kontrol hanya diinjeksi dengan phosphate buffer saline (PBS), sedangkan kelompok KP1, KP2, dan KP3 diinjeksi dengan inhibin B dengan dosis berturut-turut 25, 50, dan 100 pg/ekor. Injeksi inhibin B dilakukan secara intraperitoneum sebanyak 5 kali selama 48 hari dengan interval waktu 12 hari. Injeksi pertama inhibin B dilarutkan dengan0,05 ml PBS dan 0,05 ml Freud’s complete adjuvant (FCA). Injeksi kedua sampai kelima, inhibin B dilarutkan dengan 0,05 ml PBS dan 0,05 ml Freud’s incomplete adjuvant (FICA). Pada hari ke-6 setelah injeksi inhibin B terakhir, tikus dikorbankan secara dislocatio cervicalis,lalu darah dikoleksi langsung dari jantung dan didiamkan hingga didapatkan serum untuk pemeriksaan konsentrasi FSH dengan menggunakan metode enzyme-linked immunosorbent assay (ELISA). Hasil penelitian menunjukkan bahwa injeksi inhibin B dengan dosis 100 pg/ekor menurunkan konsentrasi FSH secara nyata (P0,05) bila dibandingkan dengan kelompok kontrol. Berdasarkan hal tersebut, inhibin B berpeluang untuk dikembangkan sebagai kandidat kontrasepsi pria hormon berbasis peptida.

Abstract

Orangutan is a great ape that has anatomical and physiological aspects similar to those of gorillas and chimpanzees, although orangutan is more arboreal than other great apes. This research aims to study the morphofunctional characteristics of the lower hindlimb and foot muscles of Bornean orangutan and its relation to the type of locomotion and daily behavior. This research was conducted on the right leg of a male orangutan by observing the morphology of the lower hindlimb and foot muscles and observing the behavior of the Bornean orangutan through video observations. The results showed that digital flexor muscles are more developed than plantar flexor muscles as an adaptation towards arboreal activities that require finger flexor movements such as gripping tree branches. Orangutans have rudimentary hallux, which is related by the less developed intrinsic muscle and indicated by the way orangutan grips tree branches dominantly using digits II-V. At a certain time, male Bornean orangutans are active on terrestrial that causes the digital extensor muscles to have more developed as an adaptation to terrestrial activity. It can be concluded the morphology of the lower hindlimb and foot muscles is in accordance to its function and behavior adaptation in arboreal and terrestrial.

Abstract

The aims of this study were to detect Jembrana antigen with polyclonal antibody and to describe antigen distribution in the Bali cattle organ that positively infected with Jembrana disease. Spleens, lungs, and livers were harvested from 10 naturally infected Bali cattle whose infection was confirmed through positive Polymerase Chain Reaction (PCR) result for Jembrana virus. Immunohistochemistry test was performed using polyclonal antibodies produced in rabbits. The results showed that cells infected by Jembrana viruses displayed positive reaction with a reddish brown color. Immunohistochemistry methods using polyclonal antibodies can detect Jembrana antigens in the spleen, liver, and lung with the highest average detection score (P0.05) was found in spleen, followed by liver and lungs. There was significant difference in the distribution of Jembrana antigens between the spleen, liver, and lungs with spleen having the highest antigen density.

Abstract

This research was aimed to find out a substance that was useful in early pregnancy diagnosis of Friesian Holstein cows. Pregnancy Specific Protein B (PSPB) was isolated, purified and partially characterized from cotyledon Friesian Holstein cows. Characterization of PSPB protein was conducted using SDS-PAGE and Western Blot. Antisera were developed against PSPB and by immunohistochemical techniques the protein was localized to the binucleated cells of the cotyledon. The estimated molecular size of Friesian Holstein cows PSPB was 59.88 kDa with the concentration of PSPB protein in cotyledon was 6480 ng/ml. PSPB protein could induce anti-PSPB antibody with the value of optical density (OD) were 0.179±0.0102 (before immunization); 1.466±0.3288 (3rd week after immunization); 1.936±0.4754 (1st week after booster;) and 2.256±0.4842 (2nd week after booster).

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian antiserum inhibin terhadap kinerja reproduksi kambing lokal. Dalam penelitian ini digunakan 12 ekor kambing betina lokal yang dibagi dalam 2 kelompok, kelompok kontrol dan perlakuan (KI dan KII), masing-masing terdiri atas 5 dan 7 ekor kambing. Seluruh kambing diinjeksi dengan 0,5 ml PGF2α (LutalyzeTM) secara intramuskulus, 2 kali injeksi dengan interval 10 hari. Pada kelompok perlakuan (KII), kambing diinjeksi dengan 500 μg antiserum inhibin pada hari ke-9 siklus dan diikuti penyuntikan 0,5 ml PGF2α 48 jam kemudian. Inseminasi dilakukan 10 jam setelah awal berahi dan diulang 12 jam kemudian. Parameter yang diamati adalah persentase berahi, kebuntingan, kelahiran, kelahiran kembar, jumlah anak total, dan jumlah anak per kelahiran. Hasil penelitian menunjukkan semua kambing (100%) pada kedua kelompok memperlihatkan berahi. Persentase kebuntingan pada KI dan KII masing-masing adalah 60,0 dan 57,1%. Angka kelahiran KI dan KII masing-masing adalah 100% dan persentase kelahiran kembar masing-masing adalah 0,0 dan 50,0%. Total jumlah anak pada KI dan KII masing-masing adalah 3 dan 6, dan rata-rata jumlah anak per kelahiran masing-masing adalah 1,0 dan 1,5 ekor. Perlakuan dengan antiserum inhibin dapat meningkatkan kinerja reproduksi kambing lokal.
