Abstract

This research was conducted with aim to reveal the morphological changes on the stomach in thelong-tailed macaque (Macaca fascicularis) (Mf) during prenatal and postnatal development. Stomachtaken from seven fetuses (40, 55, 70, 85, 100, 120, and 150 gestation day) and two newborns (10 and 105day old) animals was used in this study. The stomach was studied at macroscopic for observation of thegeneral structure. Measurement conducted to stomach length, wide of stomach body, greater curvature,lesser curvature of the stomach in each of old age group. Perception result indicate that the stomach of Mf was red bean like in form with short minor kurvatura. These values increased along with the age of fetusand infant. The Lesser curvature very short. Greater curvature relative length (twice improvement) atpost natal (infant) compared to before birth (fetus). The morphological development of the stomachincreased with age, with very rapid further development after birth, which is possibly due to the age of thefetus and in addition, the start of digestive activity and optimalization of stomach function.Keywords: anatomy, digestion, primates

Abstract

Penelitian ini bertujuan mengetahui pengaruh medium maturasi terhadap kompetensi perkembangan in vitro oosit kambing bligon. Ovarium kambing diperoleh dari Rumah Potong Hewan, dibawa ke laboratorium dalam 0,9% NaCl pada suhu 31-34 C. Oosit diaspirasi dari folikel berdiameter 2-6 mm dengan menggunakan syringe 3 ml dan jarum 23 G. Oosit dengan minimal 2 lapisan sel-sel kumulus diproses untuk maturasi in vitro. Oosit dicuci 3 kali dengan dulbecco’s phosphate buffered saline (DPBS) dan tissue culture medium 199 (TCM) 199, kemudian didistribusikan ke dalam 50 μl medium maturasi yang berbeda, yaitu TCM 199 (tanpa suplementasi),

Abstract

Penelitian ini bertujuan mengetahui pengaruh injeksi media kultur embrio supernatan ke dalam uterus mencit 2 hari sebelum transfer embrio (TE) terhadap angka implantasi, yang diindikasikan oleh adanya embrio dan leukemia inhibitory factor (LIF) pada uterus. Mencit jenis Swiss Albino dibagi menjadi 2 kelompok yakni kelompok perlakuan dan kelompok kontrol, masing-masing terdiri atas 30 mencit. Kelompok perlakuan mendapat injeksi media kultur embrio supernatan (MKES) dalam uterus 2 hari sebelum TE sedangkan kelompok kontrol mendapat injeksi media kultur embrio (MKE) dalam uterus 2 hari sebelum TE. Dua hari setelah TE (h6 kebuntingan), mencit diperfusi dengan menggunakan larutan para formaldehid 4% dan diambil uterusnya. Uterus diproses untuk blok parafin dan dipotong dengan mikrotom setebal 5 µm. Preparat dideparafinisasi dan diproses imunohistokimia dengan kit ABC dan antibodi LIF. Preparat diperiksa di bawah mikroskop untuk melihat implantasi embrio, distribusi LIF, dan jumlah LIF di desidua dan non-desidua. Angka implantasi antara kelompok perlakuan dan kelompok kontrol masing-masing adalah 52,77 dan 40,88% (P0,05). Jumlah LIF desidua pada kelompok perlakuan dan kelompok kontrol masing-masing adalah 12,83 dan 8,83 (P0,05) sedangkan jumlah LIF di non-desidua antara kelompok perlakuan dan kelompok kontrol masing-masing adalah 8,00 dan 4,50. Dapat disimpulkan bahwa injeksi media kultur embrio supernatan 2 hari sebelum TE dapat meningkatkan angka implantasi pada mencit.

Abstract

This study was conducted to evaluate fertility grade of Friesian Holstein cows based on milk urea levels. A number of eight cows which fulfill the criteria of not pregnant and in second to sixth lactation, produced milk of 2.5 liters/day, and body weight ranging from 400-450 kg were used in this research. The cows were given fodder that consisted of 30 kg of forage and 10 kg of a mixture of concentrate (pollard, pulp out, and molasses), in the morning and afternoon. Milk and blood samples were taken once a week for five weeks in the morning before fed. Milk urea levels were measured using the urea FS (DiaSys) and blood samples were tested using enzyme linked immuno-sorbent assay (ELISA) with the progesterone hormone kit (DRG, Germany). The variables measured were pregnancy rate, service per conception (S/C), days open, and progesterone levels. Reproduction and milk urea levels data were analyzed by linear regression to determine the relationship between two variables. Progesterone data were analyzed descriptively. The results showed that the average calving interval, days open, and S/C were 607.5±170.27 days; 341.25±187.65 days; and 2.41±2.32 days, respectively. The average of milk urea levels was 14.16±2.55 mg/dL. Regression analysis showed a significant effect (P0.05) and negative correlation between milk urea level and S/C. The average of non-pregnant progesterone hormone level was 12.04±7.13 ng/mL. In conclusion, the fertility grade of Friesian Holstein cows decreased when the milk urea level at 14.16 mg/dL and non-pregnant progesterone at 12.04±7.13 ng/mL.

Abstract

Tujuan penelitian adalah mengidentifikasi Eschericia coli (E. coli) O157:H7 pada susu dan lingkungan peternakan sapi perah. Sampel berjumlah 77 yang terdiri atas 27 sampel susu dan 50 sampel lingkungan. Sampel susu berasal dari ambing sapi (14), milk can peternak (6), milk can tempat penampungan susu (4), dan cooling unit di koperasi (3). Sampel lingkungan berupa feses (14), air sumber dan air tandon (12), pakan (6), serta swab tangan sebelum dan sesudah pemberian minyak pelicin (17), dan tanah (1). Isolasi E. coli dari sampel menggunakan media pemerkaya kaldu brilliant green lactose bile Broth (BGLB), media selektif agar eosin methylene blue (EMB), dan agar sorbitol MacConkey (SMAC). Koloni bakteri yang tidak memfermentasi sorbitol pada SMAC (colorless) diidentifikasi dengan uji aglutinasi lateks O157 dan antisera H7. Identifikasi bakteri dari sampel susu menunjukkan 7,41% (2/27) sampel teridentifikasi E. coli O157. Susu tersebut berasal dari ambing sapi dan milk can peternak. Bakteri E. coli O157 yang teridentifikasi dari sampel lingkungan (sampel pakan) sebanyak 2% (1/50). Hasil uji aglutinasi antisera terhadap tiga sampel positif O157 menunjukkan bahwa ketiganya tidak memiliki antigen H7 dan disimpulkan bahwa tidak ada sampelsusu dan lingkungan yang tercemar E. coli O157:H7.

Abstract

This study aims to evaluate and compare the effects of the combination of ketamine-medetomidine and ketamine-acepromazine anesthesia on pig electrocardiogram (ECG) images. The study was conducted to see the ECG Leads II in six pigs which were divided into two groups. Group I (K1) was given a combination of ketamine (10 mg/kg BW) and medetomidine (0.08 mg/kg BW), while Group II (K2) was given a combination of ketamine (22 mg/kg BW) and acepromazine (1.1 mg/kg BW) intramuscularly. ECG recording was performed after the pigs were anesthetized at the surgical stage by attaching the recording electrodes to the front wall of the chest, front left and right ankles, and back right and left back ankles. The ECG recording used 1 voltage (1 cm = 1mV) with a speed of 25 mm/sec. The parameters observed were heart rate frequency, heart rhythm, P duration, P amplitude, PR interval, R amplitude, QRS interval, QT interval, ST segment, T wave, and Mean Electrical Axis (MEA). Data obtained from this study were analyzed by T-test. The results showed that K1 had an average heart rate of 100 x/minute, regular heart rhythm, P duration of 0.07 sec, P amplitude of 0.27 mV, PR interval of 0.17 sec, R amplitude of 0.75 mV, QRS interval of 0.05 sec, QT interval of 0.20 sec, ST segment of 0.17sec, T wave of 0.17 mV, and MEA of 83.60°; meanwhile, K2 had an average heart rate of 122 x/minute, regular heart rhythm, P duration of 0.06 mm/sec, P amplitude of 0.23 mV, PR interval of 0.14 sec, R amplitude of 0.80 mV, QRS interval of 0.04 sec, QT interval of 0.16 sec, ST segment of 0.14 sec, T wave of 0.12 mV, and MEA of 68.60. The ketamine-medetomidine combination produced good quality of anesthetics for the cardiovascular system.

Abstract

This research aimed to study the effectiveness of ethanol extract of binahong leaves on blood glucose, insulin, blood chemical profiles (serum glutamic pyruvate transaminase=SGPT, serum glutamic oxaloacetic transaminase=SGOT, ureum, and creatinine), and skin histopathology in diabetic rat. A total of 20 male Wistar rats aged 3 months (± 250 gram) were divided into five groups, with four rats in each group. Group I (non-diabetic control) was injected with 0.1 M sodium citrate buffer, while group II (diabetic control), III, IV, and V were injected with single dose of Streptozotocin (STZ) at dose 40 mg/kg intraperitoneally (IP). One week after the injection, the dorsal skin of the rats were excised. Group I and II were given cream topically and 1% NaCMC orally, group III was given 50% ethanol extract of binahong leaves (EEB) topically and 1% NaCMC orally, group IV was given cream topically and EEB 300 mg/kg orally, and group V was given 50% EEB topically and EEB 300 mg/kg orally. These treatments were continued for 14 days. Blood samples were obtained at the end of study to examine blood glucose, insulin, and blood chemical profiles (SGOT, SGPT, ureum, and creatinine). Examination of skin histopathology and leukocyte count were also done. The result showed that blood glucose, insulin, SGOT, SGPT, and ureum level of diabetic rats given topical or oral EEB did not significantly different from diabetic control group, even though blood glucose, insulin, SGOT, SGPT, and ureum level of diabetic rats given topical and oral EEB were found lower compared to diabetic control group. Administration of EEB 300 mg/kg orally in diabetic rats could lower creatinine level significantly (P0.05). Histopathological examination of dorsal skin of diabetic rats which were given EEB topically showed the decrease of fibroblast proliferation, leukocyte infiltration, and hemorrhage in dermis area. Leukocyte count on skin tissue was significantly lower (P0.05) in diabetic rats given EEB. In conclusion, topical or oral administration of EEB can help healing process in diabetic wound.

Abstract

Penelitian ini bertujuan mengetahui profil biokimia darah pada sapi perah yang mengalami kawin berulang. Sapi yang digunakan dalam penelitian ini adalah dua puluh ekor sapi perah peranakan Friesian Holstein, berumur 3-8 tahun sudah pernah beranak minimal satu kali, mempunyai siklus reproduksi normal, dan kondisi tubuh sehat. Seluruh sapi dibagi ke dalam dua kelompok yang masing-masing terdiri atas 10 ekor sapi. Kelompok I adalah sapi fertil sedangkan kelompok II adalah sapi yang telah diinseminasi lebih dari tiga kali tetapi belum atau tidak bunting. Profil biokimia darah yang diukur adalah kadar total protein, total kolesterol, glukosa, dan kalsium. Data dianalisis menggunakan t-test. Hasil penelitian menunjukkan bahwa total protein; total kolesterol; glukosa; dan kalsium pada kelompok I vs kelompok II masing-masing adalah 6,77±0,38 vs 6,82±0,821 g/dl (P0,05); 166,08±37,06 vs 125,95±38,108 mg/dl (P0,05); 68,40±9,60 vs 48,58±6,675 mg/dl (P0,01); dan 9,90±1,43 vs 9,23±0,94 mg/dl (P0,05). Disimpulkan bahwa sapi-sapi perah yang mengalami kawin berulang mempunyai total kolesterol dan kadar glukosa yang lebih rendah dibanding sapi-sapi perah yang fertil.

Abstract

The aimed of present study is to investigate the morphological and histochemical of mandibular glands of the cave swiflet (Collocalia linchi). The study used 24 adult wallet linchii paired mandibular gland located in the ventral of the mandible. They were ovoid in form and whitish in color. The gland consisted of mucous acinar cells and waspositive with PAS but negative with AB (pH 2,5). The result suggested that the acinar cells of the mandibular gland contained only neutral mucopolysaccharides and no acid mucopolysaccharides. Staining with 7 biotinylated lectins, Con-A, DBA, WGA, RCA, PNA, SBA, and UEA which represent carbohydrates with galactosa-, Nacetylgalactosamine,sialic acid, 2-5 N-acetylglucosamine, á-D- mannose, showed various positive reaction in the secretion of the acinar cells depends on the type of lectin and sampling period. The result suggested possiblecorrelation between receptor gonadal hormone with the activity of reproductive and nesting period of walet linchi.Keywords: lectin, Collocalia linchi,salivary gland.

Abstract

Penelitian ini bertujuan mengetahui gambaran involusi uterus kambing kacang berdasarkan pengamatan dengan ultrasonografi (USG) transkutaneus. Hewan yang digunakan adalah 1 ekor kambing kacang betina pascapartus dengan status melahirkan normal dan melahirkan satu ekor anak. Kambing diperiksa dalam posisi berbaring (lateral recumbency). Pengamatan involusi uterus dilakukan setiap hari, dimulai dari hari pertama pascapartus sampai tidak ada lagi pengurangan diameter lumen kornua uterus. Pada hari pertama sampai hari ke-7, gambaran karunkula (hypoechoic), lokia (anechoic), lapisan miometrium, dan endometrium (hypoechoic) yang dipisahkan oleh lapisan pembuluh darah (anechoic) terlihat jelas dengan diameter lumen kornua uterus menurun dari 87,6 menjadi 52,8 mm. Pada hari ke-8 diameter lumen 45,4 mm menurun menjadi 38,4 mm pada hari ke-14. Lokia dan lapisan miometrium masih terlihat sedangkan lapisan pembuluh darah dan karunkula sudah tidak terlihat. Pada hari ke-15 diameter lumen 35,5 mm menurun menjadi 19,3 mm pada hari ke-20 dengan lapisan uterus, lokia, lapisan pembuluh darah, dan karunkula sudah tidak terlihat. Ukuran diameter lumen kornua uterus mengalami penurunan setiap hari dan berhenti mengalami penurunan pada hari ke-20 dengan diameter 19,3 mm. Penelitian ini memperlihatkan bahwa proses involusi uterus kambing dapat diamati dengan metode USG transkutaneus.
