Abstract

The objective of this study was to determine the effect of ethanol extract of neem (Azadirachta indica) leaves on the number of spermatids and androgen receptor expression in the testicular tissue of New Zealand White (NZW) rabbits. Twelve adults male NZW rabbits with body weight (BW) 2-2.5 kg were divided into three treatment groups (n = 4), namely P1 (control group) given 0 mg/kg BW, P2 and P3 were given 200 mg/kg BW and 300 mg/kg BW of neem leaves ethanol extract respectively. The extract was administered orally using a gastric tube for 52 days. At the end of the treatment, the rabbits were terminated and the testes were collected to be processed into histological preparations and stained with Periodic Acid Schiff (PAS) staining to detect the number of spermatid and Immunohistochemical (IHC) staining to detect androgen receptor expression. The results showed that neem leaves extract administered at P3 rabbits significantly decreased the number of spermatids and androgen receptor expression in myoid peritubular cells and connective tissue P0.05), but not significantly different in the Leydig cells and blood vessels (P0.05) of testicular tissue. The conclusion of this study is that the ethanol extract of neem leaves at a dose of 300 mg/kg BW can be used as a contraceptive candidate compared to a dose of 200 mg/kg BW.

Abstract

The objective of this study was to determine the effect of commercial ration by the addition of ammonium sulfate and urea fermented feed ingredients on the blood fat levels of local male ducks. This study used a completely randomized design with 32 male ducks which weredivided into four groups namely R0, R1, R2, R3, and R4. Each group was given the following treatment: R0 only given commercial ration;R1 was given 80% commercial ration + 20% fermentation of EM-4 + 1.5% urea); R2 was given 80% commercial ration + 20% EM-4 + 1.5%ammonium sulfate fermentation; and R3 was given 80% commercial ration + 20% EM-4 + 1.5% urea + 1.5% ammonium sulfate. Variablesin this study were blood fat consisting of cholesterol, triglyceride, High-Density Lipoprotein (HDL), and Low-Density Lipoproteins (LDL).The data obtained were analyzed by using 5% real level and LSD. The results showed that the provision of commercial ration added withlocal feed fermented with ammonium sulfate and urea had a significant effect on cholesterol levels (P0.5), but not significant (P0.05) ontriglyceride, HDL, and LDL levels. Furthermore, the ration that gave the best effect on the blood fat levels of male ducks blood was the ration given to R3 group.

Abstract

The objective of this research was to know the contamination of Aspergillus flavus in ikan kayu and the number of aflatoxin B1 (AFB1) produced. Also the corelation the number of A. flavus with AFB1. Samples were taken from traditional markets in Banda Aceh and Lhokseumawe. The sample were used to isolation and identify A. flavus also detect the present of AFB1 by ELISA (Enzym Linked ImmunosorbantAssay). The result shows that all samples show the present of A. flavus with the concentration 1 x 102-61 x 102 Cfu/g and the concentration of AFB1 between 0.9-38.7 ppb. In conclusion the present A. flavus in ikan kayu has a corelation with the production of AFB1.

Abstract

Tujuan penelitian ini untuk mengetahui pengaruh induksi EGF terhadap lokalisasi Cx43 dan gap junction selama ekspansi sel kumulus. Hasil penelitian ini menunjukkan bahwa ekspansi sel kumulus mempunyai signifikansi dengan meningkatnya konsentrasi EGF dan waktu kultur. Ekspansi sel kumulus diindikasikan dengan perubahan bentuk sel menjadi memanjang dan menyebar. Ekspresi Cx43 meningkat pada kultur jam ke-5 dan semakin meningkat pada kultur jam ke-10 dengan semakin padatnya protein pada badan sel. Pada kultur jam ke-15 terjadi penurunan ekspresi Cx43 pada badan sel. EGF berpengaruh terhadap peningkatan ekspansi sel kumulus tetapi tidak pada ekspresi Cx43 dengan bertambahnya konsentrasi dan waktu kultur. Hal ini menunjukkan bahwa komunikasi antar sel kumulus menurun, karena jarak antar sel yang semakin jauh.

Abstract

 The purpose of the research was to study a tissue cyst formation time Toxoplasma gondiiexperimentally. A number of 84 mice were divided randomly into four groups. Each group consisted of 21mice. The mice of the group I were infected with 101, II with 102 and III with 10 tachyzoites respectivelyintraperitoneally, whereas the group IV as a control (not infected with tachyzoites). All infected micewere treated with sulfadiazine, 15 mg/mouse per oral diluted in drinking water, for 5 days. On first untiltwenty first day after treatment one mouse of each group was necropsied. Liver, lymph, kidney, lung,heart, brain, or diaphragm muscle were then taken for histological preparations. Data on tissue cystformation time was analysed descriptively. The research revealed that innoculation with tachyzoites 103cyst could be found on day 14th after infection of liver, 102 cyst was found on the 6 day of liver, in day7th in heart and brain on day 10th of after infection, 103 cyst was found on day 4th inheart and brain in day 7thth in liver, day 6 after infection, while in the control dosage there is no formation similar to cyst found.Keywords: cyst, tissue, T. gondii, mice th1

Abstract

Tujuan penelitian ini adalah mengetahui gangguan fungsi hati yang terjadi akibat pemberian karbon tetraklorida (CCl4) pada berbagai tingkatan dosis terhadap induk bunting. Karbon tetraklorida dosis tunggal 0,014 dan 0,14 ml/20 g bobot badan mencit diberikan secara intraperitoneum pada mencit bunting, lalu diamati kerusakan yang terjadi pada hati. Kerusakan hati ditandai dengan peningkatan kadar enzim alanin transaminase (ALT), aspartat transaminase (AST) dalam serum, dan gambaran histopatologi hati. Dibandingkan dengan kont rol, pemberian CCl4 dosis 0,014 ml/20 g bobot badan mengakibatkan peningkatan ALT dan AST, bahkan pada dosis 0,14 ml/20 g bobot badan mengakibatkan induk mati dalam waktu kurang dari 24 jam setelah perlakuan. Gambaran histopatologis sel-sel hati pada kelompok mencit bunting yang mendapatkan CCl4 0,014 ml/20 g bobot badan menunjukkan terjadinya steatosis. Dapat disimpulkan bahwa pemberian karbon tetraklorida pada induk bunting menimbulkan kerusakan seiring peningkatan dosis yang diinduksikan, bahkan berakibat kematian .

Abstract

The research purpose was to evaluate the influence of date extract supplementation in lactate ringer-egg yolk extender with 0.025% sodium dodecyl sulfate (LREYS) on post-thawing quality of Pelung chicken spermatozoa. Completely randomized design with 5 treatments and 4 replications was used in this study. Semen was collected from three Pelung chickens once in three days using the dorsal-abdominal massage method. Semen was divided into 5 treatment groups of date extract supplementation that were 0% as control (LREYSDE0), 0.5% (LREYSDE1), 1% (LREYSDE2), 1.5% (LREYSDE3), and 2% (LREYSDE4). The post thawing liquid semen was observed for sperm motility, viability, plasma membrane integrity and acrosome integrity. The result showed that the motility, viability, membrane plasma integrity and acrosomal integrity of spermatozoa in LREYSE extender with 1.5% date extract supplementation (30.44±1.02%; 49.83±0.91%; 43.26±1.02%; and 45.33±0.72% respectively) was significantly higher (P0.05) than others treatment that were 0% (18.21±0.43%; 35.62±0.51%; 30.54±0.60%; and 31.49±0.71% respectively), 0.5% (20.45±0.72%; 40.72±0.87%; 36.81±0.55%; and 38.56±0.63% respectively), 1% (25.68±0.93%; 46.27±0.75%; 40.52±1.02%; and 42.83±1.09% respectively) and 2% (22.55±0.92%; 43,71±0,74%; 38,84±0,71%; and 41,39±0,86% respectively) of date extract supplementation. It is concluded that the 1.5% date extract supplementation in extender can maintain the best post-thawing sperm quality.

Abstract

This study aims to determine changes related to epididymal sperm parameters in mice given cogon grass root ethanol extract (CGREE). The experiment was carried out by dividing three groups of mice aged 4-week old (early pubertal), 8-week old (adult), and 24-week old (old) as control and treatment groups. The treatment groups were given 115 mg/kg body weight of CGREE for 14 days. The mice were sacrificed on day 15 to asses reproductive organ indexes and quality of the epididymal sperm. Index of epididymis and ductus deferens indices significantly reduced in the 4-week old treatment group compared to control (0.23 vs 0.33; 0.08 vs 0.16,  respectively), and vesical seminal significantly reduced in 8- week old treatment group compared to control (0.76  vs  0.68). The epididymal sperm quality significantly decreased in all treatment groups, but the sperm concentration in early pubertal group showed an increase. These results indicate that CGREE can enhance several semen parameters in prepubertal and adult mice but this effect is not apparent in old mice.

Abstract

The study aimed to identify genetic variation of cDNA sequence from low density lipoprotein receptor (LDLR) gene of hyporesponder cynomolgus macaques. The animal used in this study was one hyporesponder cynomolgus macaque obtained from selection result in Primate Research Center-Bogor Institute of Agriculture (PSSP IPB). Amplification of cDNA from LDLR gene was performed using polymerase chain reaction (PCR) method with 4 pairs of walking primer. Alignment of amplification result sequence from 4 pairs of walking primer generated target sequence of 2353 bp which located on position 188-2540. Analysis of target sequence alignment on reference sequence in GenBank found 8 single nucleotide polymorphism (SNP), namely c408CT; c.1200CT; c.1497CT; c.1644TC; 1791TC; 1804AG; 2088CT; and 2377GA. Of 8 SNPs, c.1804AG and 2377GA. Two SNP (c.1804AG; dan 2377GA) caused changing of amino acids composition namely p.K602E (lysine → glutamate) and p.V793I (valine → isoleucine). This result proved the potential use of genetic variation of cDNA sequence from LDLR gene as genetic marker for selection of hyporesponder cynomolgus macaques.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian kombinasi ekstrak jaloh dan kromium terhadap pembentukan heat shock protein-70 (HSP-70) dan kadar kortisol ayam yang mengalami cekaman panas. Dalam penelitian ini digunakan ayam broiler sebanyak 12 ekor yang dibagi atas empat perlakuan: Perlakuan I (EC), cekaman panas + dan kombinasi 1.000 mg ekstrak jaloh dengan 1.000 μg kromium per liter air minum, perlakuan II (E), cekaman panas + 1.000 mg ekstrak jaloh per liter air minum, perlakuan III (KD), cekaman panas + 0,0 mg ekstrak jaloh dan 0,0 μg kromium, dan Perlakuan IV (KL), tanpa cekaman panas dan 0,0 mg ekstrak jaloh dan 0,0 μg kromium. Masing-masing perlakuan terdiri atas tiga ulangan. Metode pemberian cekaman panas dilakukan dengan meningkatkan suhu dalam kandang pada 33±1° C selama lima jam per hari dalam waktu lima belas hari. Pemberian kombinasi ekstrak jaloh dan kromium dilakukan dengan cara melarutkannya dalam air minum dan diberikan selama dua jam (pemberian pukul 10.00) sebelum suhu di dalam kandang mencapai 33±1° C. Pengambilan sampel serum dan jaringan paru dilakukan pada hari ke-15 pelaksanaan penelitian (ayam umur 36 hari). Pengambilan sampel darah dilakukan sebelum ayam dipotong dan jaringan organ paru diambil setelah ayam dipotong. Pada sampel serum dilakukan pemeriksaan kortisol dengan enzymelinkedimmunosorbantassay (ELISA) dan deteksi HSP-70 di dalam jaringan paru dilakukan menggunakan metode imunohistokimia. Hasil penelitian ini menunjukkan bahwa pemberian cekaman panas pada suhu 33±1° C selama lima jam per hari dapat meningkatkan pembentukan HSP-70 dalam paru dan kadar kortisol dalam serum. Pemberian ekstrak jaloh secara tunggal lebih efektif menurunkan jumlah HSP-70 pada jaringan paru dibandingkan jika dikombinasi dengan kromium.
