Abstract

This study aimed to analyze the muscle microstructure of longissimus dorsi Brahman cross (BX) cattle slaughtered at the slaughterhouse of North Aceh District. The meat samples for the study were taken from three male BX cattle aged 18-30 months with body weights ranging from 400 kg to 500 kg and the average body condition scores (BCS) of 3-4. The muscle used was the longissimus dorsi muscle. The mean (±SE) diameter and cross-sectional area (CSA) of muscle fibers and the percentage of fast fibers in the longissimus dorsi BX muscle were 71.69±1.23 µm, 4263.43±305.69 µm2 , and 48.17±8.81%, respectively. This study concluded that the longissimus dorsi muscle of the BX has relatively large muscle fibers, some of which in each fascicle are composed of fast muscle (fast fiber/type II).

Abstract

Newcastle disease (ND) is an acute poultry disease caused by Paramyxovirus group. It has characteristic neurological symptoms, called torticollis. The molecular assay to find out the presence of viral genes in the brain can be an option in detecting ND virus infections since it penetrates brain barrier system. The purpose of this study was to identify ND viruses in the brain of chickens with torticollis symptoms, to analyze its phylogenetic and to characterize its virulence genetic code. Samples used were 12 dead chickens with historically had torticollis symptoms, obtained from poultry farms at several areas in West Java and Banten. Chicken brains were prepared for reverse transcriptasepolymerase chain reaction (RT-PCR) test. All positive samples then sequenced to obtain its nucleotide sequences from some of Fusion (F) genes analyzed its phylogenetic by comparing with Indonesian ND isolate virus from GenBank using Mega X software. The results of RT-PCR test showed that only one sample (Virus MSL.03) contained genes of ND virus. Based on homology tests and phylogenetic analysis, the virus belonged to subgenotype VIIh with an identical level of 95.34-95.86% when compared to several isolates from Indonesia. The MSL.03 ND virus has 112RRRKRF117 pattern in F0 indicatest its virulent category.

Abstract

This research purpose was to study the phenotype characters of body weight and pectoralis thoracicus muscle performance of hybrid chicken (F1) derived from crossbreed between parent stock broiler strain Cobb-500 and Pelung (Gallus gallus gallus). The chicks were divided into 3 groups and each group consists of 5 days-old chicks (DOC). Group 1 (T1) was used as control involving DOC derived from crossbreed between female and male of broiler Cobb-500 strain. Group 2 (T2) involving DOC derived from crossbreed between female and male Pelung, while Group 3 (T3) was DOC derived from crossbreed between female broiler strain Cobb-500 and male Pelung. The chicken was grown up to 49 days to evaluate the body weight, the pectoralis thoracicus weight, the pectoralis thoracicus area, and myofiber area. Data were statistically analyzed using one way Anova. The results showed that the average of body weight in T1; T2; and T3 were 2,170±86.02; 506.2±20.02; and 1,238±68.25 g, respectively (P0.05). The average of pectoralis thoracicus muscle weight in T1; T2; and T3 were 165.04±11.97; 21.26±2.44; and 68.74±1.94 g, respectively (P0.05). The average of pectoralis thoracicus muscle area in T1; T2; and T3 were 97.0±9,7; 61±6,4; and 63,4±1,8 cm2 , respectively (P0,05). The average of pectoralis thoracicus myofiber area in T1; T2; and T3 were 14.72±1.04; 4.66±2.1; and 13.13±1.3 µm2 , respectively (P0.05). It was concluded that crossbreed between broiler strain Cobb-500 and Pelung improved the body weight and pectoralis thoracicus muscle performance of hybrid chicken (F1).

Abstract

Penelitian ini bertujuan mengevaluasi pengaruh bubuk tempe instan terhadap kadar malonaldehid (MDA) pada serum tikus hiperglikemik. Sebanyak 25 tikus jantan Spraque Dawley umur dua bulan digunakan dalam penelitian ini. Tikus dibagi atas lima kelompok perlakuan yaitu A (normal + diet standar), B (hiperglikemik + diet standar), C (hiperglikemik + 20% bubuk tempe segar), D (hiperglikemik + 20% bubuk tempe instan), dan E (hiperglikemik + 35% bubuk tempe instan). Tikus dikondisikan menjadi hiperglikemik dengan induksi streptozotocin dosis tunggal 45 mg/kg bb intraperitoneum. Diet tempe diberikan selama 8 minggu. Hasil penelitian memperlihatkan bahwa rata-rata kadar MDA kelompok A; B; C; D; dan E masing-masing adalah 2,16; 3,67; 3,13; 3,08; dan 2,98. Substitusi 35% bubuk tempe instan pada diet standar dapat menurunkan kadar MDA serum tikus hiperglikemik yang paling tinggi.

Abstract

Increasing in ambient temperature inside the cage could lead to heat stress in broilers. This research was conducted to find out effects of heat stress on body weight gain, heterophile-lymphocite ratio and body temperature in chicken broiler. Twenty broilers aged 20 days (strain Cobb) were randomly divided into 2 groups. The first group was treated with no heat stress, the second one was caged in 33±1 0C temperature for 4 hours per day for 14 days. The results indicated that heat stress reduced body weight gain, increased body temperature, and changed behavior, but no effect on feed conversion ratio (FCR) and heterophile-lymphocyte ratio. It suggested that the heat stress caused detrimental effects on broiler chicken.

Abstract

Tujuan penelitian ini adalah mengetahui perubahan histopatologis dan klinis ayam herbal setelah ditantang dengan virus avian influenza (AI) H5N1. Seluruh ayam dibagi atas 3 kelompok perlakuan Kelompok kontrol, Kelompok I (I-1, I-2, dan I-3), dan Kelompok II (II-1, II-2, dan II-3). Masing-masing kelompok terdiri atas 15 ekor ayam sehingga jumlah seluruh ayam yang digunakan adalah 105 ekor. Ayam Kelompok I dan II masing-masing diberi herbal I (sambiloto, temu ireng, adas bintang, sirih merah) dan herbal II (sambiloto, adas bintang, sirih merah) selama 3 minggu sebelum ditantang virus. Ayam kelompok perlakuan secara keseluruhan tidak ada yang hidup hingga hari ke-8. Hasil penelitian menunjukkan bahwa 7 ekor ayam KII-3 (46,7%) masih hidup sampai hari ke-4 setelah uji tantang virus. Perubahan histopatologis sistem pernafasan ayam kelompok perlakuan menunjukkan pembendungan, edema, dan kerusakan sel epitel mukosa. Sistem limfoid juga menunjukkan pembendungan, deplesi folikel limfoid, dan fibrosis limpa dan bursa Fabrisius. Analisis imunohistokimia mengindikasikan partikel virus AI telah menyebar di organ atau jaringan sistem pernafasan dan sistem pertahanan.

Abstract

The aim of this study was to isolate and identify the strain of methicillin-resistant Staphylococcus aureus (MRSA) from cow’s milk in Surabaya and to determine the fragment Spa-Gene in MRSA strains. There were 50 samples of cow’s milk obtained from four dairy farms. From 50 tested samples, 19 samples (38%) were positive for Staphylococcus aureus. Antibiotic sensitivity test using oxacillin and erythromycin showed that 6 samples were resistant to the antibiotic oxacillin and 6 samples were resistant to erythromycin. MRSA confirmation tests that were conducted on 6 samples revealed 5 positive results for the MRSA strain. Electrophoresis of Polymerase Chain Reaction (PCR) product showed that 4 out of 5 samples were positive for the presence of the Spa-Gene fragment. The research results showed that there were 3 models of Spa-Gene fragments; the first had a length of 90 bp and 140 bp, the second had a length of 140 bp, and third had a length of 90 bp. This study revealed the nature of Spa-Gene polymorphism of MRSA strains isolated from milk samples. It was concluded that the Spa-Gene can be used as a molecular epidemiological marker of the MRSA strain.

Abstract

This study aimed to examine the mucolytic activity of roselle calyces extract on cow intestinal mucus in vitro. The extraction of rosellecalyces was performed by the subsequence maceration method using different polarity solvent (n-hexane, ethyl acetate, and methanol). Mucolyticactivity of each extract was evaluated by measuring the viscosity of mixture of cow intestinal mucus and phosphate buffer pH 7 compared toacetylcysteine as the standard mucolytic drug. The result showed that the decreasing of cow intestinal mucus and phosphate buffer pH 7 mixturesviscosity was indicated the increasing of mucolytic activity. The n-hexane, ethyl acetate, and methanol extract of roselle calyces exhibit dosedependentmucolytic activity. Mucolytic activity of roselle calyces ethyl acetate extract with dose of 0.8% and roselle calyces methanol extractwith dose of 0.6% showed no significant difference to mucolytic activity from acetylcysteine with dose of 0.1% (positive control). An increaseddose of roselle calyces extract will decrease the viscosity of cow intestinal mucus and phosphate buffer pH 7 mixtures, indicating an increase inmucolytic activity.

Abstract

Penelitian ini bertujuan menginterpretasikan sonogram dengan kelainan organ hepatobiliari pada anjing. Pemeriksaan dilakukan pada 17 ekor anjing dengan menggunakan brightness mode dan color flow Doppler ultrasonografi. Berdasarkan hasil interpretasi pada sonogram, ditemukan 7 kasus abnormalitas hati dan 10 kasus abnormalitas kantung empedu. Abnormalitas yang ditemukan pada hati, terdiri atas tumor, hepatitis, dan kongesti hati. Kasus tumor hati ditandai dengan hepatomegali, tekstur hati tidak homogen, dan adanya m assa pada parenkim hati. Sonogram kasus hepatitis ditunjukkan oleh adanya hepatomegali dan peningkatan akti vitas vaskularisasi hati. Sonogram kasus kongesti hati ditunjukkan oleh pembesaran diameter dan peningkatan echogenicity dinding pembuluh darah hati. Abnormalitas kantung empedu yang ditemukan antara lain cholecystitis, cholelithiasis dan mucocele. Penebalan dinding dengan atau tanpa edema merupakan citra yang ditemukan pada kasus cholecystitis. Sonogram kasus cholelithiasis menunjukkan adanya massa hyperechoic di dalam lumen kantung empedu disertai acoustic shadowing. Sonogram kasus mucocele menunjukkan adanya massa hypoechoic yang terdapat di dalam kantung empedu.

Abstract

ABSTRACT The study aimed to find out the effect of Eurycoma longifolia Jack on testosterone level and spermatozoa quality of Etawa crossbreed goat. Goats were divided into two treatment groups, three goats each. Group 1 (K1) was control group which given orally 20 ml of distilled water every morning at 9:00 am for six days, and group 2 (K2) was administered with Eurycoma longifolia with the dose of 90 mg/kg bw in 20 ml of distilled water orally every morning at 9:00 am for six days. The concentration of testosterone was measured on day 1, 3, and 6 using the method of enzyme-linked immunosorbent assay (ELISA). The quality of sperm consisted of concentration and percentage of life sperm. The results showed that level of testosterone on K2 increased started from day 1 (9.36 ng/ml) to day 6 (12.43 ng/ml) (P0.05). The percentage of life spermatozoa increased higher on K2 compare to K1 on day 3 to day 6, that was 88.6 and 89.8% on K1 to 91.0 and 92.7% on K2. In conclusion, the administration of pasak bumi with the dose of 90 mg/kg bw in 20 ml of distilled water for 6 days able to increase testosterone levels and percentage of life sperm of Etawa crossbreed goat.
