Abstract

Penelitian ini bertujuan mengkaji tingkat stres sapi yang dipotong di Rumah Potong Hewan (RPH) Yogyakarta. Dalam penelitian ini digunakan 20 ekor sapi yang dibagi menjadi dua kelompok, yakni kelompok sapi yang dilakukan restraint dengan restraining box tipe Mark 1 (K1, n=13) dan kelompok sapi yang dilakukan restraint dengan metode konvensional (K2, n= 7). Masing-masing kelompok diambil darahnya sebelum dan saat dipotong untuk diperiksa kadar kortisol dengan enzyme-linked immunosorbent assay (ELISA). Rata-rata kortisol sapi betina sebelum vs setelah restraint pada K1 dan K2 masing-masing adalah 55,64±52,13 vs 21,23±20,63 ng/ml dan 127,48±150,19 vs 106,28+75,39 ng/ml. Rata-rata kortisol sapi jantan sebelum vs setelah restraint pada K1 dan K2 masing-masing adalah 40,28±38,12 vs 24,21±17,21 ng/ml dan 49,51±38,67 vs 69,62±63,98 ng/ml. Rata-rata kadar kortisol tanpa memperhatikan jenis kelamin pada K1 dan K2 masing-masing adalah 79,50±88,50 vs 95,80±69,69 ng/ml (P>0,05). Disimpulkan bahwa tidak terdapat perbedaan kadar kortisol sebagai indikator stres pada sapi-sapi yang dipotong di RPH Yogyakarta yang mengalami restraint dengan restraining box tipe Mark 1 dan restraint metode konvensional.

Abstract

The purpose of this study was to analyze how the effect of each increase in Body Mass Index (BMI) on the increase in blood pressure (BP). The research design was a laboratory experiment with 52 non-obese and obese rats as subjects (non-obese, n=14 and obese, n=38). Rats were induced obesity using a high-fat diet for four weeks. BMI used is based on the Lee Index with obesity indicators if 300 g/cm. Data analysis using independent sample t-test, simple linear regression test, and Pearson correlation test; (p0.05). The results: BP in non-obese rats was lower than obese rats (89.00±7.38 vs 190.11±4.42; p=0.001*), BMI was positively associated with BP (p=0.001*); an increase in BMI will increase BP (p=0.001*), there is a strong correlation between BMI and BP (r=0.977; p=0.001*). The regression equation (Y = 707.42 + 2.75 X) indicates that every 1 g/cm increase in BMI will be followed by an increase in BP of 2.75 mmHg. The analysis of R-square = 0.942 shows that the accuracy of the linear equation model is 94.2%. In conclusion, BMI has a strong positive correlation with BP, an increase in BMI will increase BP; obesity induces high blood pressure in rats.

Abstract

The aim of this study was to investigate the apolipoprotein-B (apo-B) gene in atherosclerosis mice which were orally given curcuminoid extract of Curcuma xanthorriza. A total number of 30 white mice were split into 6 groups, the first group considered as control (without any treatment), second group as atherogenic feed control, the third group as extract control, while the fourth, fifth and sixth groups as atherogenic feed and curcuminoid Curcuma xanthorriza extract group treated with 5 mg/mouse, 10 mg/mouse and 15 mg/ mouse, respectively for three months. The blood samples were taken from all six groups for the deoxyribonucleic acid (DNA) analysis using total DNA isolation, DNA amplification with polymerase chain reaction (PCR), and DNA sequencing. The data analysis showed that 374 bp nucleotide sequence gen of apo-B from Rattus norvegicus in groups B, C, D, E, and F did not cause any changes in genes. The analysis showed the sequence of apo-B Rattus norvegicus gene in the treatment group was apparently identical with that of Rattus norvegicus group A as the control group without treatment. As conclusion, the administration of curcuminoid zanthorrizza to atherosclerosis mice did not change the gene structure of apo-B 100.

Abstract

The aims of this study were to investigate the freezing capability of Pasundan bull spermatozoa in Tris-egg yolk (TEY), Tris-soy (TS), and AndroMed® as diluents. Semen were collected twice a week from four Pasundan bulls aged 3-5 years old using an artificial vagina and evaluated macro- and microscopically. Semen had ≥70% sperm motility, ≥800x106/mL sperm concentration, and less than 20% sperm abnormalities were divided into three parts and each of them diluted with TEY, TS, or AndroMed®. After an equilibration step at 5°C for four hours, diluted semen were packaged in 0.25 mL straw, frozen in liquid nitrogen for ten minutes and kept in liquid nitrogen container until examination. Motility test on fresh, diluted, equilibrated, and after-thawed semen was done using Androvision®. The results showed that after thawing motility of sperm diluted in AndroMed® (58.64±0.72%) was higher than in TEY (49.45±1.22%) and TS (39.34±6.33%). Sperm motility of Pasundan bulls diluted in these three diluents reduced around 33.27±2.45% during freezing process.

Abstract

Penelitian ini bertujuan menentukan konsentrasi dan berat molekul protein  ekskretori/sekretori larva (L3) Ascaridia galli (A. galli). Larva L3 diperoleh dari usus halus 100  ayam tujuh hari setelah pemberian dosis 6000 L2 melalui esofagus ayam. Sebanyak 5–10  L3 dikultur secara in vitro  dalam setiap ml medium Rosswell Park Memorial Institute (RPMI 1640), pH 6,8, tanpa merah fenol dalam inkubator pada temperatur 37 0C dan 5% CO2 selama 3 hari. Ke dalam medium ditambahkan 100 unit ml-1 penisilin G, 100 µg ml-1 streptomisin, 5 µg ml-1 gentamisin dan 0,25 µg ml-1 kanamisin. Ekskretori/sekretori dipreparasi dari produk metabolisme L3 yang dilepaskan ke dalam medium kultur. Untuk mendapatkan protein ekskretori/sekretori, medium kultur dipekatkan dengan vivaspin 30.000 MWCO, dan kuantitas protein dihitung dengan metode Bradford. Berat molekul protein ekskretori/sekretori divisualisasikan dengan sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS PAGE). Hasil penelitian  menunjukkan bahwa konsentrasi protein ekskretori/sekretori adalah 0,595 mg/ml dengan berat molekul 28 kDa.

Abstract

The aim of this research was to determine the effect of coffee on creatinine and plasma malondialdehyde (MDA) level in rats fed on high purine diet. A total of 24 male Wistar rats divided into 4 groups of 6 rats each. Beef broth were given to 3 groups (K1, P1, P2) for thirty days. Beef broth administration were followed by giving coffee (caffeinated= P1 or by a decaffeinated= P2, 144 mg/200 g BW). Serum creatinine levels and plasma MDA were examined periodically on day 0, 15, and 30. Data were analyzed by multivariate analysis of variance or MANOVA test (CI= 95%, P0.05). Coffee administration (caffeinated and decaffeinated) may decrease the serum creatinine (day-30, K1= 3.17+ 0.69; P1= 1.63+0.11; P2= 1.14+0.08) and MDA levels (day-30, K1= 9.8+0.28; P1= 5.80+0.55, P2= 3.87+3.26) after 30 days of treatment (P= 0.000 for creatinine and MDA levels in K1, P1, P2 groups). A lower serum creatinine and MDA levels can be found in decaffeinated coffee grup. Coffee consumption for 30 days (especially decaffeinated) may decrease serum creatinine and plasma MDA in rat induced by a high purine diet.

Abstract

This study aimed to calculate the total bacteria and to identify the proteolytic bacteria in rumen and reticulum of local Kacang goat. The samples used were the rumen and reticulum fluids of five goats at abattoirs in Banda Aceh. Isolation of proteolytic bacteria was carried out using skim milk agar with pour plate method and incubated at 39° C for 48 hours. The bacterial colonies morphology was observed and the total bacterial count was recorded. DNA of the widest proteolytic index colony was isolated, amplified, and sequenced. The results showed that the dominant colonies morphology was white with position inside the agar. The average of total proteolytic bacteria in goat rumen fluid and reticulum fluid were 6.416x106 CFU/mL and 2,382x107 CFU/mL, respectively. Isolates with the widest proteolytic index was Ru3 (2.5 mm) in the rumen which homology and phylogenetic tree analysis of 16S rRNA showed 93% sequence similarity with Bacillus subtilis, while in reticulum was Re1 (2.0 mm) which has 92% sequence similarity to Tatumella. It is concluded that the number of proteolytic bacteria in reticulum is greater than the number of proteolytic bacteria in the rumen of local kacang goat. Homology analysis in this study proved that the Ru3 and Re1 isolate were probably either a new species or unconfirmed species.

Abstract

The objective of this research was to detect a minimum concentration of the probes that could be used for dot blot hybridization analysis. The method required labeled DNA probes. In this study a non-radioactive label of Digoxigenin-11-dUTP was used for labeling the Sag1 and the Bag1 of Toxoplasma gondii DNA probe. Labeling method for the probes was done according to the random primed labeling technique. The result showed that 0.67 pg/µl Sag1 probe and 0.58 pg/µl Bag1 probe could be detected by anti-Dig-antibody. It could be concluded that 0.67 pg/µl Sag1 probe and 0.58 pg/µl Bag1 probe could be used to diagnose toxoplasmosis by dot blot hybridization method.

Abstract

This study aimed to evaluate the effectiveness of dietary synbiotic at different giving frequencies on growth, immune responses, and resistance of white shrimp infected by infectious myonecrosis virus (IMNV). Synbiotic used in this study was combination of probiotic Vibrio alginolyticus SKT-b and prebiotic oligosaccharides extracted from sweet potatoe (Ipomoea batatas L). Doses of probiotic and prebiotic used were 1% and 2% (w/w), respectively. The white shrimps (0.493±0.035 g) were divided into five treatments consisting of A and B (without supplementation of synbiotic: (A) positive control; (B) negative control), C (daily synbiotic supplementation), D (twice a week synbiotic supplementation), and E (weekly synbiotic supplementation). After 30 days of feeding trial, white shrimps were infected by IMNV (except negative control). The results showed that daily growth rate of white shrimp on all synbiotic treatments (C, D, and E) ranged from 6.93±0.025-6.97±0.019% and had higher values than controls (A and B) (P0.05). Meanwhile, feed conversion value in C and D (1.54±0.142 and 1.58±0.117) were lower than controls (P0.05). Supplementation of synbiotic with different frequencies also affected survival rate of white shrimp after the challenge test with IMNV; daily synbiotic supplementation (C) resulted in a 50% higher survival rate than positive control (P0.05). This was associated with immune responses parameters values of synbiotic treatment (before and after the challenge test) which were better than positive control. In conclusion the addition of synbiotic in feed resulted in higher growth performances, immune responses,and resistance of white shrimp to IMNV infection.

Abstract

Tujuan penelitian ini mengetahui karakteristik pedagang daging ayam (umur, tingkat pendidikan, pengalaman berjualan, pelatihan, dan statuskepegawaian) di pasar tradisional di Kota Jakarta dan hubungan antara karakteristik tersebut dengan pengetahuan terkait higiene dan sanitasiketika berjualan daging ayam. Sebanyak 217 pedagang daging ayam di pasar tradisional di DKI Jakarta digunakan sebagai responden. Metodepenentuan sampel dengan cluster random sampling dan penentuan sampel pasar dengan probability proportional to size (PPS). Data dianalisissecara deskriptif dan hubungan antara peubah diuji dengan uji Gamma. Hasil yang diperoleh adalah hampir seluruh responden (n= 216; 99,5%)berumur di atas 21 tahun, sebagian besar responden (n= 105: 52,3%) lulus SMP dan SMA, dan kebanyakan responden memiliki pengalamanberjualan 3 tahun. Tingkat pengetahuan responden termasuk dalam kategori sedang sampai dengan baik (n= 209; 96,3%). Hasil analisis datamenunjukkan terdapat hubungan nyata antara pendidikan dengan pengetahuan (P0,05) dengan kekuatan korelasi sedang (r= 0,357), serta antara pengalaman dan pengetahuan (P0,05) dengan terdapat hubungan nyata antara pendidikan dengan pengetahuan (P0,05) mempunyai korelasi sedang (r= 0,357), serta antara pengalaman dan pengetahuan (P0,05) mempunyai korelasi lemah (r= 0,296).
