Abstract

Pasak bumi is popular as an aphrodisiac in the traditional medical herb. Boiled drink water of the root powder of pasak bumi is believed can increase the secondary reproduction characters such as libido and the duration of erection. This article is part of the research report that focus on: the distribution of hypophysis acidophil and basophil cells caused of the pasak bumi optimal dose treatment. Research carried out in an experiment using Hemaktosilin Eosin(HE) staining method to study micromorphological distribution of the acidophil and basophil cells of the male rat hypophysis after pasak bumi treatment (the dose of 18 mg pasak bumi/200 g body weight during three days, and the control of 1 ml aquadest). Result of the research showed that: Basophyl cells increased significantly in the third days of pasak bumi administered. Meanwhile acidophyl cells decreased in the third days of pasak bumi administered.

Abstract

Inhibition of α-glucosidase and hypoglycemic activity are two effects commonly used to identify bioactive compounds with potential to treat diabetes. The objectives of this study were to analyse and compare the bioactive compounds and α-glucosidase inhibitory effect of four different types of Swietenia mahagoni seed extract, and to analyse the hypoglycemic activity of the greatest inhibition of α-glucosidase-extract in rats. The extracts were obtained using two different solvents (aqueous and ethanol) and two different methods: maceration and reflux methods. This resulted in four types of extract varying by solvent and extraction method.  Testing of these extracts for α-glucosidase inhibitory effect was carried out in vitro using spectrophotometer. Testing for hypoglycemic activity was carried out in vivo using rats. A total of 40 male Sprague-Dawley rats were divided into eight groups: (1) the negative control group, received an oral dose of aquadest only, (2) the positive control group, was given 90% sucrose orally without S. mahagoni seed extract, and five treated groups (3-7), were given 90% sucrose followed by the best extract-ethanolic S. mahagoni seed extract in doses of 100, 200, 300, 400, and 500 mg/kgBW, and (8) the acarbose group, was given 90% sucrose orally followed by acarbose. Glucose levels in each animal were measured at 0, 30, 60, 90, and 120 min after treatment. The results showed the greatest inhibition of α-glucosidase in ethanolic extract, using maceration methods. This ethanolic-maceration S. mahagoni seed extract also showed hypoglycemic effects in hyperglycemic rats at dose from 100 to 500 mg/kgBW. Ethanolic extract of S. mahagoni seed, using maceration method, can be proposed as potential antidiabetic agent.

Abstract

Indonesia merupakan negara dengan tingkat eksploitasi biawak V. salvator terbesar di dunia yang sebagian besar untuk melayani permintaan perdagangan kulit. Tingginya permintaan kulit biawak di Indonesia mengkhawatirkan menyebabkan turunnya populasi satwa tersebut. Penelitian ini bertujuan untuk mempelajari anatomi organ reproduksi jantan biawak air asia (Varanus salvator) (Reptil: Varanidae). Dua ekor biawak jantan dewasa digunakan dalam penelitian ini. Hewan dianestesi, dilakukan exanguinasi, dan difiksasi dengan larutan paraformaldehida 4% secara perfusi. Pengamatan dilakukan terhadap situs viscerum, morfologi, dan morfometri organ reproduksi mulai testis sampai hemipenis. Secara makroskopis, organ reproduksi jantan V. salvator terdiri atas testis, ductus epididymidis, ductus deferens dan hemipenis yang masing-masing berjumlah sepasang. Posisi testis menggantung di dinding dorsal coelom melalui mesorchium. Dari bagian dorsal testis terdapat ductus epididymidis yang panjangsampai di ujung kaudal ginjal. Ductus deferens, berupa saluran kecil, lurus dan berakhir di ujung hemipenis yang terletak di dalam pangkal ekor. Pada bagian kranial hemipenis ditutupi oleh papillae berbentuk konikal. Di kaudal dari hemipenis ditemukan otot retraktor yang memanjang ke arah ekor, dan diduga berperan menarik hemipenis ke dalam setelah kopulasi. Organ reproduksi jantan biawak secara umum mirip dengan reptilia lain khususnya ular dan kadal, dengan karakteristik adanya sepasang hemipenis.Kata kunci: Varanidae, Varanus salvator, organ reproduksi jantan, hemipenis, otot retraktor. (Anatomy of The Male Reproductive Organ of Water Monitor, Varanus salvator (Reptil: Varanidae))Indonesia is a country with high levels of exploitation of Varanus salvator that mainly serve the demand of leather trade. The high demand of lizard leather in Indonesian was alarming, cause a decline population of these animals. To improve our understanding on reproduction organs of the animal, we conduct this anatomical study. The study was used two adult male lizards. The animals were anesthetized, exanguinated and fixated in 4% paraformaldehyde by tissue perfusion method. Observations were performed to the visceral site, morphological and morphometrical of the male reproductive organs, from testes to hemipenes. Macroscopically, male reproductive organs of V. salvator were a pair of testes, epididymidis ducts, deferens ducts and hemipenes. The testis attached to dorsal wall of the coelom and fixed by the mesorchium. The epididymidis duct was long tubes that located in the dorsal of testes, winding up at the caudal end of the kidney. The deferens duct was a small duct, running straight and last at the end of each hemipenis, located at the base of the tail. The cranial part of each hemipenis was covered by conical shaped papillae. Furthermore, at the caudal of each hemipenis was found the retractor muscle that extends toward the tail, and is thought to contribute to the retracting hemipenis after copulation. The male reproductive organs of V. salvator are generally similar to the other reptiles, especially snakes and lizards, with peculiar a pair of hemipenes.Keywords: Varanidae, Varanus salvator, male reproductive organs, hemipenes, retractor muscles.

Abstract

Penelitian ini bertujuan untuk mengetahui perkembangan folikel sapi PO dara yang distimulasi menggunakan PMSG dosis rendah (dosis non-superovulasi) dan perolehan folikel dominan pre-ovulatori sebagai informasi dasar untuk penerapan intrauterine programming pada sapi. Sapi PO dara berjumlah sembilan ekor dibagi ke dalam tiga kelompok dosis PMSG yaitu kontrol (NaCl 0.9% sebagai placebo), dosis PMSG 0.5, dan 1.0 IU/kg BB. Injeksi PMSG dilakukan pada awal gelombang folikel ke-2 berdasarkan perubahan dinamika ovari yang dikonfirmasi menggunakan USG diikuti injeksi PGF2α 48 jam kemudian. Perkembangan folikel sejak injeksi PMSG hingga terbentuknya pre-ovulatori folikel (POF) diamati dan dipetakan menggunakan USG. Jumlah POF tertinggi terdapat pada dosis PMSG 1.0 IU/kg BB (4.67+1.67) secara signifikan (P<0.05), sedangkan POF yang terbentuk pada dosis PMSG 0.5 IU/kg BB berjumlah satu, sama dengan kelompok kontrol (P>0.05). Meskipun jumlah POF yang terbentuk berjumlah satu, namun rataan diameter dan volume kelompok sapi dosis 0.5 IU/kg BB lebih besar 16.13% dan 57.14% secara berurutan dibandingkan kelompok kontrol, diikuti oleh dosis 1.0 IU/kg BB dengan nilai tertinggi secara signifikan (P<0.05). Berdasarkan hasil tersebut dapat diketahui bahwa PMSG 0.5 IU/kg BB dapat terkontrol sehingga dapat digunakan sebagai metode pendekatan intrautering programming pada hewan monotokus.

Abstract

Telah dilakukan penelitian pada kancil (Tmgulus javanicus) di Kebun Binatang Ragunan - Jakarta dan Kebun Binatang Wonocolo - Surabaya untuk mendapatkan jenis makanan yang disukai, dan aktifitas makan, minum dan ruminasi dari kancil yang dikandangkan.Pada penelitian ini, disediakan 10 jenis makanan dan empat jenis diantaranya yaitu pisang, kacang panjang, kangkung dan pepaya lebih disukai. Aktifitas makan dilakukan pada siang dan malam hari, dan pada saat istirahat kancil melakukan aktifitas ruminasi. Perilaku makan dan ruminasi ini berbeda dengan perilakunya di habitas asalnya. Sedangkan aktifitas minum, jarang dilakukan, hal ini mungkin disebabkan oleh tingginya kadar air di dalam makanan yang disediakan. Aktifitas makan dari kancil yang dikandangkan telah berubah menjadi diurnal dan nokturnal.

Abstract

Currently, diabetic case as a degenerative disease has widely known that it can be cure by insulin.  A remain puzzle is proliferation do not found in rest Beta cells, so insulin injection has to be applied entire life of diabetic patient. So far, there is no diagnostic approach that expresses such of thing which may indicate an early anomaly on glucose metabolism process. The aim of this research to get case model of hyperglycemic rat that has long period in process.

Abstract

Gosipol is a substances contained in extracted cotton seed which is thought to have the antifertility ability therefore it is often used as a herbal contraceptive. The aim of this study were to assess the folliculogenesis in mice after administrated with cottonseed extract. 60 female mice strain DDY which was 14-15 weeks old and 30-35 g body weight were divided into five groups and given cottonseed extract each 0; 1,5; 2,1 and 2,7 g/kg BW for 5, 10, 15, 24, and 24 + 10 days (without cottonseed treatment). At the end of the treatment period, mice was euthanasia to observe follicular development histomorphology (each three mice of each treatment). Mice estrous status were evaluated based on the description of the vaginal smear cells with Giemsa staining. The results showedthat the number of developing follicles was low (P < 0.05) compared with control after 5 days cottonseed extract administration at dose 2,7 g/kg BW that were 23 ± 3,6. At dose 1,5 and 2,1 g/kg BW the number of follicles was low after 24 days that were 25 ± 10,4 and 27 ± 3,5. Recovery effects of follicle number after cottonseed extract administration for 24 days was the best at a dose of 1,5 g/kg BW. Prolonge of estrous cycle occured in mice which were administrated the cottonseed extract of at all dose treatment. In conclusion, although the decrease in the number of developing follicles and prolonge of estrous cycles occurred after cottonseed extract administration, but these effects are reversible after the administration ended.
