The climate change caused by air pollution could be overcome by creating green open spaces. Plants can absorb air pollution, provide shade, and be used as aesthetics and location boundary. Furthermore, plants' resistance to air pollution varies according to physiology and morphology. The objectives of this study were to assess the species diversity of vegetation and the tolerance index to air pollution of some plant species using the APTI (Air Pollution Tolerance Index) method in household, industry, and transportation areas in Eat Java. The observed plant species in the three sectors were the same. The result showed that the Shannon-Wiener diversity index value (H') was considered medium, and based on the APTI test, Pseuderanthemum reticulatum had the highest tolerance level with an APTI value of 27.59in the industrial sector (I0), 38.31 in the household sector (P1) and 31.73 in the transportation sector (T0). Meanwhile, Syzygium myrtifolium had the lowest tolerance level to air pollution with an APTI value of 12.64 in the industrial sector (I1), 11.59 in the household sector (P1) and 12.70 in the transportation sector (T0). This research provides information regarding the environmental conditions of the ecosystem in green open spaces in the industrial, household and transportation sectors and gives recommendations on the sustainable management of green open spaces in line with Sustainable Development Goals (SDGs) number 13 based on the tolerance level of plants.

Marker validation for salt tolerance in Indica rice. Biodiversitas 23: 4517-4523. The development of salt-tolerantrice variety is of importance with concern to the use of the coastal area for rice production and, consequently, the development ofarapid and accurate method of variety selection. Molecular markers associated with Quantitative Trait Loci (QTL) for salt tolerance in rice have been reported elsewhere. This research aimed to identify molecular markers associated with salt tolerance in rice. The research was carried out from September 2020 to February 2021 at Plant Breeding and Biotechnology Laboratory, Faculty of Agriculture, Jenderal Soedirman University, Indonesia. Nine Simple Sequence Repeat (SSR) markers and one Sequence Tagged Site (STS) marker were evaluated for their association with salt tolerance. The rice varieties consisted of three salt-tolerant varieties,  one salt-sensitive variety, and six commercial varieties. Nona Bokra PCR amplicon was used as a positive control for DNA band scoring. Binary data of the amplicon banding pattern were analyzed using Principal Component Analysis (PCA) to determine the grouping of the evaluated varieties about their tolerance to assigned markers. The result demonstrated that the evaluated rice varieties fell into four groups of marker resemblances. Among the evaluated SSR markers, RM 3412, RM 342, RM 8094, and Wn 11463 significantly contributed to the grouping. The present study confirmed that Wn 11463 is a strong marker for salt tolerance in rice. However, validation needs to be made on a broader range of genotypes to see the significance of RM 8094 and RM 3412 as salt tolerant markers.

Edamame protein hydrolysis using Lactococcus lactis, Lactobacillus bulgaricus and Lactobacillus paracasei produce short peptides with higher antioxidant potential. Biodiversitas 23: 3604-3613. The high demand for protein sources for functional vegetable foods, especially proteins that have ACE (Angiotensin-Converting Enzyme) inhibitory activity has encouraged efforts to meet this need through exploration of potential protein sources and the development of food processing technology to modify the protein into derivative compounds in the form of short peptides to promote higher bioactivity than the native protein. This research focused on the hydrolysis of edamame proteins into shorter peptides through a fermentation process using a group of Lactic Acid Bacteria (LABs), that is Lactococcus lactis, Lactobacillus bulgaricus and L. paracasei, to gain a higher functional activity of its protein hydrolysate. Edamame milk was fermented with that LABs as starter culture until 24 hours at 37ºC. The activity of the protease released by the LABs was confirmed using the agar diffusion method based on the emergence of a clear zone surrounding the culture, while the antioxidant activity was confirmed with the ABTS and hydroxyl radical scavenging methods. The results exhibited that the population density of starter culture in edamame milk reached 108 CFU/mL, and the pH decreased from 6.75 to 4.15. All LABs used in fermentation were able to hydrolyze protein as indicated by the increasing degree of hydrolysis and changes in protein profile of SDS-PAGE analysis. In addition, when compared to the unfermented edamame milk, the protein hydrolysate from LABs fermentation showed an increase of ABTS and hydroxyl radical inhibitory activity, in which the highest IC50 values ??of 8.96 and 13.33 g/mL of each were found in edamame milk fermented by Lc. lactis InaCC B187. An increase in ACE inhibitory activity was also observed in edamame milk fermented using this microbe (9.80 g/µL). These findings indicate that Lc. lactis InaCC B187, L. delbrueckii subspecies bulgaricus FNCC41, and L. paracasei InaCC B145 have the potency as an effective starter culture to increase bioactive peptides in edamame soybeans potential for functional food sources.

Spatial distribution of mangrove health index on three genera dominated zones in Benoa Bay, Bali, Indonesia. Biodiversitas 23: 3407-3418. A study of mangrove forest stratification was conducted in Benoa Bay which experienced high coastal development pressures during the last decade. The study aimed to determine mangrove health index distribution (MHI) and forest community structures along three genera-dominated zones which were Sonneratia, Rhizophora and Bruguiera. Random forest method was applied to classify the areas of distributed genera along the bay. Forest structure was assessed with 54 plots. The forest was mainly composed of Rhizophora and Sonneratia-dominated zones in a respective forest area proportion at approximately 51% and 45%. Those zones were dominated by R. mucronata and S. alba with an importance value index (IVI) at 266.35% and 145.57%, respectively. A narrow Bruguiera zone composed of B. gymnorrhiza domination was found in the most landward area, only covering 4% of the mangrove area. Overall accuracy and a kappa coefficient indicated high accuracy of forest classification at 97% and 0.94 respectively. We found that 47.44% of mangrove areas could be classified into the highest healthiness category indicating that the mangrove forest in Benoa Bay is in excellent condition. The Rhizophora zone made a significant contribution to the entire forest state since the excellent category coverage in this zone was approximately 73.80%.

Abstract. Khoo DH, Shivanand P, Taha H. 2022. Short Communication: Determination of crude oil degradation efficiency of biofilm producing bacteria isolated from oil contaminated site. Biodiversitas 23: 4138-4143. Microbes have gained much attention for their application in the bioremediation of petroleum contaminated areas, and biofilm producing bacteria are considered potential candidates for this purpose. The aim of this study was to screen six oil degrading bacterial strains for their ability to form biofilm, and to measure the crude oil degradation efficiency of selected strains. It was found that only two strains, Micrococcus sp. UBDBH15 and Pseudomonas sp. UBDBH26 were categorized as strong biofilm producers, with the latter showing the highest amount of biofilm formed. Under the conditions tested, Pseudomonas sp. UBDBH26 was also found to have the highest degradation rate (0.0047 g/day) with a significant degradation of 7.07 ± 3.02 and 11.38 ± 2.93 % of crude oil after 7 and 14 days respectively. Micrococcus sp. UBDBH15 had a lower rate of degradation (0.0033 g/day), resulting in a non-significant degradation. However, in comparison with a non-biofilm producer, Enterobacter sp. UBDBH06, this study suggested that biofilm might enhance the degradation of oil, but further studies are needed to confirm this assumption.

Abstract. Hassan FM, Mahdi WM, Al-Haideri HH, Kamil DW. 2022. Identification of new species record of Cyanophyceae in Diyala River, Iraq based on 16S rRNA sequence data. Biodiversitas 23: 5239-5246. The biodiversity and water quality of the Diyala River require screening water in terms of biological contamination, because it is the only water source in Diyala City and is used for many purposes. This study aimed to identify a new species record of Cynaophyceae and emphasize the importance of using molecular methods beside classic morphological approaches, particularly in the water-shrinkage-aqua system. Five different sites along Diyala River were selected for Cyanophyceae identification. Morphological examination and 16S rRNA sequence analysis was conducted, and the phylogenetic tree was constructed using Mega 6 Programme. The morphological examination of samples showed a total of 28 species corresponds to Cyanophyceae, including one species of Spirulina. In our study of 28 identified species, three new species record were identified in Diyala River. The newly recorded species were confirmed by 16S rRNA and the phylogenetic tree construction. The species are registered in the National Centre for Biotechnology Information (NCBI) with the following accession numbers: Arthrospira indica (MW854667.1), Arthrospira platensis (MW854665.1), and Limnospira fusiformis (MW854666.1). Most notably, Arthrospira platensis is not listed in the checklist of Iraqi algae. Thus, these species are considered as a new record of Iraqi algal flora. The identification of new species record in Diyala River reflexes the impact of climate change on this river, and the necessity to use 16S rRNA to identify microalgae in all ecosystems.

Biodiversitas 23: 4040-4046. Nitrogen (N) is a vital macronutrient that can be a limiting factor for rice plant growth. Nitrogen-efficient plants can be assembled by modifying nitrogen-related metabolic pathways to improve Nitrogen Use Efficiency (NUE) in plants. T0 generation of transgenic Mekongga rice plants containing the LeAlaAT gene was obtained. In transgenic plant research, zygosity test was desirable to ensure the integration and inheritance stability of transgene. This study aimed to assess zygosity and nitrogen use efficacy of two transgenic Mekongga rice lines, namely M41 and M50. This research involved three main activities, i.e. detection of flanking sequence for T0 generation by TAIL-PCR method, zygosity test for T1 generation, and efficacy test for T2 generation of Mekongga transgenic lines in response to different N fertilizer rates of 0, 60, 90, and 120 kg ha?1. Zygosity analysis showed that six out of 14 M41 and three of 13 M50 transgenic lines were homozygous for LeAlaAT transgene. The efficacy test showed that homozygous transgenic lines had higher tiller numbers, grain numbers, and biomass compared to non-transgenic ones under the same nitrogen level. The notable discrepancies of the transgenic plants can also be recognized in the absorption, agronomical, and grain NUE of the studied plants.

Abstract. Despal, Faresty C, Zahera R, Toharmat R. 2022. Title The feeding behavior of dairy cattle under tropical heat stress conditions at smallholder urban farming. Biodiversitas 23: 3771-3777. Dairy cattle (Bos taurus) feeding behavior influences feed intake and affect animal welfare and disease management. This study observed dairy cattle feeding behaviors under tropical smallholder urban farming environments. Thirty-six cattle at the Kebon Pedes area of Bogor City were observed during 12 h feeding (6 am-6 pm). These traits included eating, ruminating, resting duration, and frequencies, measured using a continuous method. Temperature, humidity, temperature-humidity index (THI), feed and nutrient intake, milk production, and components were measured. The results showed that the cattle were in mild to severe heat stress. The average cow daily eating, ruminating, and resting frequencies were 6.61, 7.61, and 22.33 times, while the average eating, ruminating, and resting durations per cow per 12 h were 129.64, 105.67, and 484.69 minutes, respectively. The dry matter intake was 11.17 kg with a roughage to concentrate ratio of 0.73:1. The protein, ash, ether extract, and crude fiber intake were 10.25, 6.62, 3.67, and 22.08% DM, respectively. The average milk productions and fat contents were 11.01 L/head/d and 5.44%. The results showed that eating and ruminating levels correlated positively with feed, and CF intakes while resting correlated negatively. It was concluded that the cattle were in heat-stressed (THI of >78) condition, reduced DM intake, and deficient in energy and CP, which led to lower milk production but higher milk components, specifically milk fat. It is recommended to improve forage quality that promotes eating and ruminating and reduces resting durations, improving milk production.

Biodiversitas 23: 3507-3512. Fuelwood still contributes as an essential energy source for domestic uses especially in rural communities including in West Papua, Indonesia. While several development programs are implemented to enhance the life quality of rural indigenous communities, their implication for fuelwood use is not clear. This study aimed to identify fuelwood tree species used by communities in three villages in West Papua and to, assess its availability prior to the empowerment program and compare it to nowadays after the program based on the community's perspective. A descriptive method with qualitative and quantitative data components was used in this study. Data were collected at predetermined villages representing three ecological zones (i.e., Sakumi, Jamboi and Watitindau) using purposive sampling and semi-structured interviews with community representatives, tribal chiefs and leaders. The results showed that there were 5-7 plant species used as fuelwood, namely Piper aduncum, Casuarina equisetifolia, Rhizophora sp., Gliricidia sepium, Intsia bijuga, Pometia pinnata, Dodonaea viscosa, Homalium foetidum, Syzygium sp. and Muntingia calabura. These all-wood species have a good flame. Some high commercial tree species such as I. bijuga, P. pinnata and H. foetidum are only used in their branches and twigs for firewood. The local communities still use firewood despite the empowerment program since alternative fuel sources are not available or lacking, while the availability of firewood is plentiful. Nonetheless, in the long run, the continuous and extensive use of firewood will put pressure on the remaining trees and forest. Therefore, we recommend sustainably using firewood and replanting several species frequently used as firewood.

Abstract. Boonman N, Chutrtong J, Wanna C, Boonsilp S, Chunchob S. 2022. Detection of Staphylococcus aureus from contact surfaces of public buses in Bangkok and metropolitan area, Thailand. Biodiversitas 23: 3395-3400. The purpose of present investigation was to determine the prevalence of different species of genus Staphylococcus on the contact surfaces of public buses in Bangkok and metropolitan area. A total of 180 samples were collected from handrails, seats, and window frames of each bus of 2 bus terminals, Bangkhen and Morchit, divided into 15 non-air-conditioned buses and 15 air-conditioned buses from each terminal. The samples were cultivated and identified by Gram’s stain and biochemical tests. The results showed that 76.7% from Bangkhen and 80% from Mochit were contaminated with Staphylococcus spp. Based on the type of vehicles, Staphylococcus spp. was found in 93.3% of air-conditioned buses and 63.3% of non-air-conditioned buses. Considering the sampling location, Staphylococcus spp. was found in 28.3% of handrails, 50% of seats and 51.7% of window frames. Four isolates of Staphylococcus aureus were discovered among 294 isolates in a total of Staphylococcus spp., including 3 isolates from Bangkhen and 1 isolate from Mochit. Only one isolate was contaminated on the non-air-conditioned bus, whereas 3 isolates were on the air-conditioned buses. Two isolates of them were found from seats and 2 isolates from window frames. Disk diffusion susceptibility test showed that 3 isolates of S. aureus resisted fusidic acid and fosfomycin. The results suggested that Staphylococcus spp. and S. aureus were prevalence in public transport systems which were a source of infection to the persons. Therefore, effectively controlling their distribution is necessary to reduce the risk of bacterial infections in public bus users.
