Abstract

The quality of an extract is determined by the type and level of compounds contained therein. The solvent is one of the elements that influences the extract's quality. Therefore, extraction must be carried out using a solvent that can extract bioactive substances. This study was aimed at obtaining the optimal concentration of ethanol in extracting flavonoids, phenolics, and beta-carotene from pepino melon fruit. In this study, dried pepino melon fruit was extracted using ethanol of 50%, 70%, and 96%. The extracts were analyzed qualitatively for phenolic and flavonoid using thin layer chromatography (TLC), and quantitatively using a spectrophotometry. Meanwhile, beta-carotene levels were determined using high pressure liquid chromatography (HPLC). The data of level flavonoid, phenolic, and beta-carotene were statistically analyzed using multivariate analysis of variance. The results showed that 50%, 70%, and 96% ethanol solvent produced extracts with a yield value of 51.8%; 87.4%; 54.6%; total flavonoid content of 0.298 ± 0.04 mgQE/g; 0.559 ± 0.03 mgQE/g; 0.289 ± 0.01 mgQE/g; total phenolic content of 4.763 ± 0.08 mgGAE/g; 3.631 ± 0.12 mgGAE/g; 3.317 ± 0.10 mgGAE/g; beta-carotene level of 0.157 ± 0.02 mg/g; 0.910 ± 0.16 mg/g; 1.054 ± 0.13 mg/g, respectively. The statistical analysis showed that there was a significant difference in different ethanol concentrations in extraction with the content of total flavonoids, total phenolics, and beta-carotene (p<0.05). The optimal ethanol solvent for extracting flavonoids, phenolic compounds, and beta-carotene from pepino melon fruit was 70% ethanol.

Abstract

Vascular endothelial growth factors receptors (VEGFR) inhibitors play a vital role in the treatment of renal cell carcinoma. These are small molecules that predominantly exhibit anti-angiogenesis activity in conjunction with other anti-tumor effects. These drug therapies are approved for the use in patients as frontline agents or adjuvant therapy in renal cell carcinoma. However, VEGFR inhibitors are associated with undesirable adverse events, with some having a more manageable toxicity profile compared to others. As a result, choice of treatment poses a challenge for healthcare providers and patients. Nonetheless, these agents demonstrate improved disease/progression free survival (DFS/PFS) values and remain a critical component in the treatment of kidney cancer.

Abstract

Snakehead fish (Channa striata) is one of the natural ingredients that have the potential to heal wounds quickly. Snakehead fish extract ointment with a combination of kelulut honey, clove oil, golden sea cucumber extract, and green betel extract can accelerate wound healing. Standardization is a process carried out to ensure the stability, safety, quality, and efficacy of the natural ingredients used. This study aims to standardize simplicia (fish fillet) raw materials used for the manufacture of snakehead fish extract herbal ointment. The method used follows the Indonesian National Standard, samples were sent to LPPT UGM and Baristand Pontianak. The results of the simplicia (fish fillet) test for snakehead fish were 81.6% water content, 0.813% total ash content, 0.02% acid-insoluble ash content, 19.3% protein, 2.93% total fat, 0.01% iron mg/kg, zinc 0.002 mg/kg, and calcium 817 mg/kg and the alkaloid and tannin quality tests were negative. the results of the simplicia test (fish fillet) for snakehead fish E. coli 3 AMP/gr, ALT 3.2 x 104 colonies/g, and the Salmonella test also met the requirements. The results of characterization tests with various methods and parameters that have been carried out show that the simplicia (fish fillet) of snakehead fish (Channa striata) has good quality and quality to be processed into extracts.

Abstract

Fibrosis is common profound in diabetes condition, they characterized by accumulation of extracellular matrix in tissue then cause tissue injury. The study aim to investigate effect of monoculture, conditioned media, and coculture of NIH3T3 cells in LPS-high glucose media conditions on mRNA TGF-B and collagen secretion. The fibroblast NIH3T3 were cultured with various culture type there is mono culture NIH3T3, conditioned media from RAW264.7 culture media, and coculture with RAW264.7 cells (ratio 1:1 and 1:2)  in normal media (media containing 4500 mikro g/mL of glucose) and HG-LPS media (media containing LPS 0.25 mikro g/mL and D-glucose 5400 mikro g/mL). Our study shows that mRNA TGF-B and collagen secretion of fibroblast NIH3T3 cells in monoculture and conditioned media from inflammatory macrophages are not statistically different. On other hand, the TGF-B mRNA expression and the relative ratio of collagen secretion in RAW 264.7 and NIH 3T3 cell coculture media with a culture ratio of 1:1 were the highest compared to other models (p kurang dari 0.05). Our study conclude that RAW 264.7 and NIH 3T3 coculture cells (1:1) in LPS and high glucose media is appropriate for fibrosis in vitro models.

Abstract

The Covid-19 pandemic in Indonesia, including Yogyakarta, has created a psychological impact on the community. One of the psychological impacts is depression. This issue can be overcome by early detecting depression using an instrument called the World Health Organization-Five Well-Being Index (WHO-5). Unfortunately, this instrument is currently unavailable in the Indonesian version. Therefore, this study aims to obtain a valid and reliable Indonesian version of the WHO-5, which can later be used by pharmacists as an instrument to early detect depression in Indonesia, especially in Yogyakarta. This research employed a cross-sectional design. The research subjects were people who visited Apotek UAD 1 and 2, who met the inclusion and were not included in the exclusion criteria. The WHO-5 questionnaire proceeded through a translation process from the English version to the Indonesian version. The original version of the WHO-5 questionnaire, which was the final translated version, was tested on 40 respondents to gain its validity and reliability. The analysis of the validity test using the Pearson Correlation of five questions on the WHO-5 questionnaire has revealed an average value of r = 1, indicating that the questionnaire is valid. The result of the reliability test with Cronbach Alpha is 0.853. In conclusion, the Indonesian version of the WHO-5 questionnaire meets the validity and reliability criteria so that the questionnaire can apply to the community.

Abstract

Typhoid fever is a common health problem in the community caused by Salmonella bacteria. The incidence rate of this infection will increase if a person's immune system is weakened. Plant extracts are generally considered to be potential immunomodulatory agents developed, which have smaller side effects. Research shows that Cnidoscolus aconitifolius leaves have medicinal properties including as hepatoprotective, antidiabetic, and anticardiovascular. The results of the antioxidant activity test show the results that Cnidoscolus aconitifolius leaves extract (CAE) has potential as an antioxidant with a very strong category. This study was conducted to determine the effect of giving Japanese papaya leaf extract on CD4+ and CD8+ expression in Babl/c mice induced by Salmonella typhimurium bacteria. The study was started by preparing 70% ethanol extract from Cnidoscolus aconitifolius leaves and preparing 30 Babl/C mice as experimental animals which were divided into 6 groups (healthy control, negative control, positive control and treatment group by giving CAE dose of 100 mg/kgbw, 200 mg/kgbw and 400 mg/kgbw). Induction was carried out by oral infection with Salmonella thypimurium bacteria. After 3 days the infected mice were treated orally once a day for 7 days. Evaluation of CD4+ and CD8+ expression was carried out using the flowcytometer method of the lymph organs. Data analysis was carried out by the Anova test followed by the post hoc test (Tukey) using the SPSS for Windows application. The results showed that giving CAE at doses of 100 mg/kgbw, 200 mg/kgbw and 400 mg/kgbw could increase the expression ratio of CD4+ and CD8+, whereas giving CAE at a dose of 400 mg/kgbw showed significantly different results (p<0.05) to the negative control. This shows that the CAE has potential as an immunomodulatory agent that can improve immune function.

Abstract

Dyslipidemia, is due to an increase in blood lipid levels, which include cholesterol, triglycerides and low-density lipoprotein. Dyslipidemia is expected to remain as a major risk factor for cardiovascular diseases, diabetic and atherosclerosis. Mangosteen is an antioxidant agent that can exhibit a potential free radical scavenging property and protected oxidation of low-density lipoprotein. The aim of this research was to determine hypolipidemic and antioxidant effect of mangosteen peel extract (MPE) in dyslipidemia rats. Lipid profile including cholesterol total (CHOL), triglyceride (TG), High Density Lipoprotein (HDL) and Low-Density Lipoprotein (LDL) of dyslipidemia rats were measured using photometric method. Rats were fed cholesterol for 4 weeks until rats were indicated dyslipidemia. After rats suffered dyslipidemia, the high cholesterol feed was stopped and rats were given mangosteen peel extract 1000; 500; 250 mg/Kg body weight (bw) daily for 14 days (first treatment) and 28 days (second treatment), negative control (normal feed), and positive control (dyslipidemia rats). MDA plasma level also was measured. The CHOL, TG, LDL and HDL of dyslipidemia rats remarkable decreased after treated by 1000 mg/Kg mangosteen peel extract. After 28 days of treatment, the CHOL, TG and LDL were critically declined by 1000 mg/Kg mangosteen peel extract. MDA plasma level showed decreased in all treatments after 28 days of treatment. The dyslipidemia rats treated by mangosteen peel extract showed hypolipidemic activity, according to decreased level of lipid profile including cholesterol, triglycerides and LDL. The MDA level can also be decreased by mangosteen peel extract by increasing the concentration.

Abstract

Parkinson's disease is a chronic neurological illness that may be caused by a decrease in dopaminergic neurons in the brain. Myristicin, eugenol, and flavonoids, which are bioactive compounds in nutmeg (Myristica fragrans Houtt.), may be able to treat Parkinson's disease. The goal of this study was to find out the antiparkinsonian effect of nutmeg in haloperidol-induced Parkinson's disease mice. Induction with haloperidol was carried out intraperitoneally for 7 consecutive days. Treatment with nutmeg uses three different doses (5, 10, and 20 mg/kg) for 7 days orally. Behavioral testing is carried out using the Rotarod test and the cylinder test. The results showed that nutmeg at a dose of 20 mg/kg was able to show significant improvement (P < 0.05) in the motor condition of test animals induced by haloperidol.

Abstract

Cervical cancer is one of the second-leading causes of death in women. The discovery of cancer drug candidates continues to be carried out due to the resistance that occurs in cervical cancer therapy. Plant metabolite compounds are one of the sources used to explore new drug candidates. Red ginger rhizome is a candidate plant that has anti-cervical cancer activity. This study aims to determine the cytotoxicity of an ethanol extract of red ginger rhizomes on the growth of HeLa cancer cells and predict anti-proliferative activity via the jak2/stat3 and hedgehog pathways. The sample (red ginger rhizome simplicia) was extracted by remaceration using 75% ethanol. The MTT assay method is used to test the cytotoxicity and anti-proliferation of metabolite compounds using Autodock 4.2 software. The receptors used in the jak2, stat3, and smo pathways were obtained from the Protein Data Bank with the codes 6VGL, 6NUQ, and 5L7I, respectively. The ethanol extract produced is a thick yellowish brown extract with an aromatic smell and spicy taste, with an extract yield of 18.63% w/w. 75% ethanol extract of red ginger rhizomes has cytotoxic activity in HeLa cancer with an IC50 of 104.22 ± 6.18 µg/mL and an IC50 of cisplatin of 38.61 ± 3.66. Prediction of antiproliferative activity via the jak2 pathway shows a binding energy and Ki value of -7.47 kcal/mol, -7.48 kcal/mol, and 3.33 uM, 3.27 uM, as shown by alpha-cedrol and beta-eudesmol compounds. The highest inhibition on the stat3 and smo pathways was shown by the beta compound eudesmol, with binding energy and Ki values of -6.05 kcal/mol, -7.57 kcal/mol, and 36.48 uM, respectively; 2.81 uM.

Abstract

This study aimed to determine the effect of increasing the concentration of the condensed extract of kaffir lime fruit peel in a gel dosage form on its physical quality and effectiveness. The condensed extract was obtained by maceration with 95% ethanol as a solvent. The dosage form chosen is a hydrophilic-based gel with a modification of the addition of a penetrant enhancer (IPM) and a solubilizer (Glycerin). The concentration of the condensed extract of kaffir lime fruit peel used were F1 (10%), F2 (15%), and F3 (20%). Gel preparations were tested for physical quality and effectiveness, consisting of antioxidant activity (IC50) and antibacterial (Zone Inhibition). Furthermore, the One Way ANOVA statistical method will analyze data from parametric experiments between batches and between formulas. If there is a significant difference in the statistical analysis between the formulas, then the test is continued using the Tukey Post-Hoc test method. The experimental results showed that increasing the concentration of kaffir lime peel extract (Citrus hystrix) affected the pH value, viscosity, dispersion, and effectiveness as an anti- acne and antioxidant. It was concluded that the anti-acne gel of extract F1, F2, and F3 were physically stable during storage, and based on the physical quality and effectiveness test it was known that F3 had the greatest zone inhibition (ZI): 13.87 ± 0.18 mm and IC50 :14.9049 mg/mL
