Abstract

Background: The specimen storage room serves as bioarchive for prospective research purposes. Cryopreservation or preservation at very low temperatures has been used to preserve research isolates for decades. A quality assessment must be conducted to ensure the isolates conditions. This study aimed to assess the quality of isolates by testing the level of viability and contamination from different cryopreservation periods.Methods: This was an experimental study with a total sample of 92 Mycobacterium tuberculosis (Mtb) isolates which were sampled randomly using cryopreservation, aged 8–10 years (in Tryptic Soy Broth media, TSB/) and aged 3–5 years (in Middlebrook 7H9) which was re-cultured in Ogawa Medium at the Tuberculosis Laboratory, Universitas Padjadjaran in May–November 2019. After observing confluent growths, the pure isolate was stained using the Ziehl-Neelsen (ZN) method to confirm the presence of Mtb growth or contamination. A Simple ratio was used to count the recovery rate as a viability parameter and contamination rate of each batch.Results: Test results showed a recovery rate of 66.67–100% of positive cultures stored despite they had been cryopreserved for 10 years. There was no significant difference in the percentage of positive cultures between preservation period groups.Conclusion: Mtb isolates can survive and remain viable after being stored for up to 10 years at -80 ° C in cryopreservation media.

Abstract

Background: Hand sanitizers generally contain alcohol. However, it can cause dry and irritated skin. Therefore, it is necessary to find antibacterial alternatives that are safe for the skin. One of the plants that has antibacterial activity is Terminalia mantaly (T. mantaly). This study aimed to investigate the antibacterial activity of ethanol extract and hand sanitizer gel of T. mantaly.Methods: The antibacterial activity test against gram-positive (Staphylococcus aureus and Streptococcus pyogenes) and gram-negative bacteria (Escherichia coli and Shigella dysenteriae) was carried out using the liquid micro dilution method. The phytochemical tests were also performed. This study was conducted at the Chemistry Laboratory, Faculty of Sains and Informatics, Universitas Jenderal Achmad Yani in April-June 2022.Results: The ethanol extract of T. mantaly stem contained secondary metabolites of alkaloids, tannins, quinones, saponins, steroids, and flavonoids. The minimum inhibitory concentration (MIC) values of T. mantaly stem ethanol extract against Escherichia coli, Shigella dysenteriae, Staphylococcus aureus, Streptococcus pyogenes were 0.63%, 0.63%, 0.16%, and 0.16%, whereas the hand sanitizer gel gave MIC values of 0.63%; 0.31%; 0.16%; and 0.16% respectively. The minimum bactericidal concentration (MBC) values of T. mantaly stem ethanol extract and hand sanitizer gel had the same results, namely 2.50%, 0.16%, 1.25%, and 1.25%. The physical stability of the hand sanitizer gel from the ethanol extract of T. mantaly stem met the physical stability standards of the gel. Conclusions: The ethanol extract of the T. mantaly stems has stronger antibacterial activity against gram-positive bacteria than against gram- negative bacteria.

Abstract

Background: Dyslipidemia occurs in 60% of hypertensive patients. Effective treatment of hypertensive patients with dyslipidemia can reduce the risk of cardiovascular disease by more than 50%. However, compliance with dyslipidemia medication in dyslipidemia patients is still low. Low compliance can be attributed to poor knowledge. This study aimed to analyze the relationship between knowledge and compliance using antidyslipidemic drugs in hypertensive patients with dyslipidemia.Methods:  A cross-sectional analytical study was conducted from August to September 2022 at the Pasirkaliki Public Health Center (Pusat Kesehatan Masyarakat, Puskesmas), Bandung, Indonesia. The consecutive sampling method was used to collect 64 hypertensive patients with dyslipidemia who were taking anti-dyslipidemic drugs. Primary data on knowledge and compliance were obtained through valid and reliable questionnaires. The Chi-square test was used to determine the relationship between the two variables.Results: The study results showed that 95.3% of respondents had good knowledge. Respondent compliance showed obedience of 78.1%. Respondents with good knowledge had an obedience rate of 78.1%. There was a relationship between knowledge and compliance to using anti-dyslipidemic drugs in hypertensive patients with dyslipidemia (p=0.001).Conclusions: There is a relationship between knowledge and compliance with the use of anti-dyslipidemia drugs in hypertensive patients with dyslipidemia. Almost all patients have good knowledge.  Continuous maintenance and improvement of patient knowledge is required, which can be achieved by conducting health education to better understand the relationship between diseases and the importance of taking medication.

Abstract

Background: Sleep quality is influenced by numerous factors, including external factors, such as the sleep environment. One aspect of the sleep environment that can influence sleep is lighting. Light exposure emitted by lamps during sleep can impair sleep quality. The aim of this study was to investigate the association between lamp light during sleep and sleep quality in preclinical medical students.Methods: This cross-sectional observational analytic study was carried out on 386 preclinical medical students who were determined using the cluster sampling method. Data was collected in November 2022. Lamp light during sleep and sleep quality among students were measured using the Indonesian version of the Pittsburgh Sleep Quality Index. The association between lamp light and quality of sleep was analyzed by the Chi-Square test.Results: There were 26.68% of students who used lights when sleeping. Poor sleep quality was detected in 46.37% of students. The association between lamp light during sleep and quality of sleep yielded a p-value of 0.002.Conclusion: There is a significant association between lamp light during sleep and sleep quality in preclinical medical students. Thus, improving sleep quality in preclinical medical students may be achieved by modifying the lamp light during sleep.

Abstract

Background: Diabetic wounds are difficult to treat, causing persistent infections and often leading to limb amputation. One convenient way to manage diabetic wounds is by applying wound dressings. This study aimed to develop a wound dressing in form of a gel containing brown algae and binahong leaf extract for diabetic wound dressings.Methods:  This study was an experimental study conducted in the Organic Chemistry Laboratory of Universitas Pendidikan Indonesia from June to September 2022. As many as 33 mice induced by alloxan were divided into 9 groups consisting of application of brown algae extract gel, binahong lead extract gel and a mixture of brown algae, and binahong leaf extract gel with the concentration of 2.5%, 5%, and 10% respectively. All treatments were given within 7 days. Laboratory data collected included phytochemical screening, total phenolic compound, antioxidant activity and organoleptic test.Results: Brown algae and binahong leaf extracts had a high total phenolic content of 331.25 mg GAE/g and 207.01 mg GAE/g, respectively with antioxidant activity of IC50 = 327.33 µg/mL and 209.30 µg/mL, respectively. It was found that brown algae and binahong leaf extracts could accelerate wound closure in a diabetic mouse model. Gel formulation with 10% w/w brown algae extracts presented 91.66% of wound closure, two times greater than treatment with commercial drugs.Conclusion:  The developed gel containing brown algae and binahong leaf extracts can be a promising wound dressing for healing diabetic wounds.

Abstract

Background: Recently, there has been a significant increase in new HIV infections in several regions in Indonesia, one of which is Pati Regency, Central Java. The considerable barrier in preventing HIV/AIDS is the widespread stigma against people living with HIV (PLHIV). Innovative interventions are required to eliminate HIV stigma, particularly among young people. This study evaluated the effectiveness of the Pandang Dengar Orang Dengan HIV (ODHIV) campaign in reducing HIV stigma among youth in Pati Regency, Indonesia.Methods: This was an experimental study with a pretest-posttest control group design with randomization. The residents of Pati Regency aged 15 to 29 years who were active users of Instagram and WhatsApp were involved. The Pandang Dengar ODHIV campaign was conducted using Instagram Reels four times a week for three weeks in the intervention group. The instruments used were the Global Stigma and Discrimination Indicator Working Group (GSDIWG) and the Strive research consortium. The T-test was used to analyze the data with a significance level of 95%.Results: The results revealed a decrease in HIV stigma in both the intervention and control groups (p=0.001). After the intervention, the mean stigma score in the intervention group was lower than the mean in the control group (mean difference of 0.044), however, was not statistically significant (p-value=0.961).Conclusions: The Pandang Dengar ODHIV campaign is effective in reducing HIV stigma among youth in Pati Regency, Indonesia. Further research is needed to demonstrate the effectiveness of campaign in wider audiences and other areas.

Abstract

Background: Indonesia as an agricultural country has more than 33 million workers in the agriculture. Pesticides have an important role in agriculture because they function as powerful plant pest exterminators. However, its use is considered to cause various toxic effects, one of which is on the nervous system. This study aimed to evaluate the impact of pesticide exposure on cognitive impairment by assessing the butyrylcholinesterase biomarker in the blood of rats as experimental animals.Methods: This was a true experimental study conducted during September–November 2022 at the Pharmacology and Biochemistry Laboratory, Faculty of Medicine, University of Jember with a post-test only approach using Wistar rats which were divided into 1 control group and 4 treatment groups treated with chlorpyrifos 5 mg/kg orally. Serum butyrylcholinesterase enzyme levels were measured using the kinetic photometric method and then analyzed with a regression test in IBM SPSS® Statistics software.Results: The average value of butyrylcholinesterase was 920.23±145.27 (Control group); 904.48±91.6 (7 days after chlorpyrifos administration); 889.81±95.2 (14 days after chlorpyrifos administration); 457.84±69.5 (28 days after chlorpyrifos administration); 575.82±194.25 (56 days after chlorpyrifos administration). The results of the One-Way ANOVA test showed significant differences between groups as indicated by a significance test of 0.024 (p <0.05).Conclusions: There is a relationship between the duration of exposure to chlorpyrifos and butyrylcholinesterase levels in Wistar rats. This fact shows that the longer an organism is exposed to low doses of chlorpyrifos pesticides, the higher the level of neurological system toxicity that occurs in that organism.

Abstract

Background: In order to fulfil the requirements of the national immunization program and sustain the production capacity of the monovalent Hepatitis B vaccine, this study aimed to assess the safety and immunogenicity of the recombinant Hepatitis B Vaccine (Bio Farma) using the new Hepatitis B bulk. Methods: This study was an experimental, randomized, double-blinded, and controlled Phase I clinical trial, with 100 healthy subjects divided into 50 adults and 50 adolescents. Subjects were randomly assigned to receive either the Bio Farma registered Recombinant Hepatitis B Vaccine (group A) or a new source of Hepatitis B bulk (group B). Subjects received one or three doses of vaccine, depending on the baseline anti-Hbs titer. Subjects were given diary cards to record solicited and unsolicited adverse events for 28 days following vaccination. Vaccine immunogenicity was assessed by measuring the level of HBsAg antibody titer elevation.Results: No serious adverse events were reported during clinical trials. The frequencies of adverse events were not significantly different between the two vaccine-randomized groups. The most immediately observed local reaction was local pain, reported by 35.7–42.8% of adults and 24.0–26.3% of adolescents, without any systemic reactions. Seroconversion in adults in group B reached 100% and 78.5% in group A, meanwhile in adolescent subjects in both groups it reached 100%. A substantial increase in geometric mean titer (GMT) was observed in the majority of subjects after immunization.Conclusion: Recombinant Hepatitis B Vaccine with a new source of HBsAg B bulk is safe, well tolerated, and highly immunogenic.

Abstract

Background: Indonesia has the second-highest tuberculosis prevalence in the world. Moreover, Indonesia is among the 30 countries with the highest burden of multidrug-resistant tuberculosis (MDR-TB). This study aimed to determine the pattern of second-line anti-tuberculosis drug resistance in MDR-TB patients.Method: This study was a descriptive cross-sectional using data from MDR-TB patients aged 18 years and older, diagnosed with drug-resistant TB at Dr. Hasan Sadikin General Hospital from December 2021 to June 2022. Total sampling was used. Data on age, gender, history of previous antituberculosis drug treatment and second-line antituberculosis drug susceptibility test results were collected. Resistance distribution patterns were identified using the Line Probe Assay (LPA) and the Mycobacteria Growth Indicator Tube (MGIT) test.Results: Of 134 data retrieved, only 82 data were complete. The median age of the patients was 42 years (range 27-51 years), predominantly female (53.7%), without a history of antituberculosis drug treatment (52.4%). The highest number of resistances was resistant to high dose isoniazid (43.9%), followed by low dose fluoroquinolone (14.6%). Among patients who were resistant to low dose moxifloxacin, 16.67% of patients were still sensitive to high dose moxifloxacin. There was no resistance to bedaquiline.Conclusions: Almost half of the patients are resistant to high dose isoniazid, followed by resistance to low dose fluoroquinolone. These finding are expected to be taken into consideration by clinicians in making decisions on the diagnosis or management of MDR-TB patients and can further serve as input for the government in implementing MDR-TB control programs in Indonesia.
