Abstract

Background: Penetrating neck injuries are relatively rare but pose significant challenges due to the concentration of vital structures in this area. This case report presents a unique in-stance of a 20-year-old male with a penetrating neck injury caused by iron splinters, resulting in the rupture of the internal jugular vein and tension pneumothorax.Case Presentation: The patient, a 20-year-old male, presented with restlessness, chest pain, rapid breathing, and a penetrating wound on the left side of the neck. Initial management included needle decompression for tension pneumothorax and radiological assessments to localize the foreign body. C-arm revealed that the foreign body had penetrated the upper left thoracic region and lung parenchyma. The surgical intervention involved repairing the internal jugular vein and performing pulmonary resection to remove the iron splinters. Conclusions: This case highlights the importance of a multidisciplinary approach in managing complex penetrating neck injuries. Advanced imaging techniques and timely surgical interventions are crucial for optimizing patient outcomes. Future research should focus on refining diagnostic protocols and exploring minimally invasive surgical techniques to improve the management of such injuries.

Abstract

Background: No treatment can eliminate keloids. Thymoquinone (TQ) is hypothesized to play a pivotal role in treating keloids by modulating cellular processes such as proliferation, migration, and apoptosis. However, the existing studies investigating its effects on these mechanisms in keloid fibroblasts are limited and require further exploration. Objective: This study aims to investigate the effects of TQ on the proliferation, migration, and apoptosis of keloid fibroblasts in vitro. Methods: This experimental study was conducted using keloid fibroblast cultured in vitro. Cells were seeded in a 96-well plate at a density of about 5x103 cells per well with 100 μl of culture medium, and cells were cultured for 24, 48, and 72 hr for each concentration of TQ. Cell proliferation was assessed using a CCK-8 Kit, measuring optical density with a microplate reader. Apoptosis was measured using the TUNEL assay. Cell migration following TQ treatment was evaluated using the Scratch assay. The statistical test used a one-way analysis of variance (ANOVA) followed by the least significant difference (LSD) test. Results: TQ inhibited the proliferation of keloid fibroblasts at a dose of 5 µM after 48 hours of incubation and 10 µM after 24 hours of incubation. The inhibitory effect of TQ on fibroblast proliferation increased in a dose- and time-dependent manner. Treatment at 5 and 10 µM doses increased apoptosis in keloid fibroblast cultures. The TQ5 µM group achieved 60% closure, while the 10 µM group showed 55% closure. Migration was significantly inhibited in the 25 µM and 50 µM groups, with only 30% and 10% closure, respectively, at 72 hours. Conclusion: Thymoquinone inhibits the proliferation and migration of keloid fibroblast cells while promoting apoptosis. These properties suggest that TQ could be developed as a potential treatment for keloid-related skin issues.

Abstract

Background: The first 1,000 days are crucial for a child's development. Indonesia has limited research on responsive caregiving and early learning opportunities, especially for children with nutritional challenges. While studies have addressed general child nutrition, few have explored comprehensive nurturing care interventions. This gap underscores the need for further research. Educational videos, particularly animated ones, effectively capture attention and improve understanding, making them valuable for enhancing maternal knowledge and child growth.Purpose: This study aimed to assess the impact of nurturing care educational videos on mothers' knowledge and child growth in children aged 6-23 months with nutritional challenges. Methods: A quasi-experimental design with non-equivalent and untreated control groups was employed. Seventy-three mothers and their children aged 6-23 months, facing nutritional challenges, were recruited through consecutive sampling. The intervention group received nurturing care educational videos. Statistical analyses included dependent t-tests, independent t-tests, and repeated measures ANOVA.Results: The intervention group showed significant improvements in nurturing care knowledge (p = 0.012), responsive caregiving (p = 0.018), and safety and security (p = 0.006). A significant difference in safety and security was observed between groups at posttest 1 (p = 0.020). Both groups exhibited significant increases in children's body weight and height: the intervention group (body weight, p = 0.005; body height, p < 0.001) and the control group (body weight, p < 0.001; body height, p < 0.001).Conclusion: Nurturing care educational videos had a significant positive impact on enhancing mothers' knowledge and promoting child growth. Notable improvements were observed in nurturing care knowledge, responsive caregiving practices, and safety and security measures among caregivers exposed to the intervention.

Abstract

Background: In the examination of blood type using the tube method, a test cell containing 5% erythrocytes of the solution volume is required. The blood type test using the tube method is carried out in hospital blood banks and in the educational world during practical activities. The obstacle in making a 5% erythrocyte solution is that erythrocytes are lysed so that it is made every day. In addition, making a 5% erythrocyte solution takes a long time so it is not efficient. Purpose: This study aims to find a solution that can maintain erythrocytes for a long time and detecting blood type.Method: This experimental study involved the preparation of 120 tubes containing 5% erythrocyte suspensions from blood types A, B, AB, and O. The samples were diluted in 10 different test solutions, including CPD, ACD, EDTA 5%, Heparin 3%, NaCl 0.9% + glucose (0.025%, 0.05%, 0.1%, 0.2%, and 0.3%), and NaCl 0.9% Daily lysis observations were performed using vortex mixing, centrifugation, and color assessment. The solution with the most extended stability was further tested for ABO blood grouping using the tube method to analyze agglutination patterns. Results: The test solution that can maintain erythrocytes for a long time is the citrate phosphate dextrose (CPD) solution. This solution can prevent erythrocytes for a long time and does not damage erythrocyte antigens. It can also detect blood types well, making blood type examination time more efficient. Conclusion: CPD solution successfully prevents erythrocyte lysis for a longer time compared to other solutions and functions in detecting blood type antibodies.
