Abstract

Malaria infection during pregnancy can cause accumulation of infected-red blood cells in placental intervillous space and induces placental tissue inflammation and hypoxia. This condition triggers endoglin expression, and release of soluble endoglin which can interfere TGF-β binding to its receptor. The aim of this study was to investigate the correlation between placental endoglin expression and TGF-β level with low fetal weight (LFW) in malaria-infected mice. Nine pregnant mice infected with Plasmodium berghei on the day 9th post mating (malaria infected group) and eight normal pregnant mice (non-infected group) were used in this study. The mice were sacrificed on the day 18th post mating, and all fetal body weights were measured by analytical scale. Enzyme Link Immunosorbent Assay (ELISA) was done to determine the level of placental TGF-β while immunohistochemical staining was performed to examine endoglin expression in placental tissue. The mean of fetal body weights of malaria infected group was significantly lower than non-infected group (p=0,002), while the expression of placental endoglin in malaria infected group was significantly higher than non-infected group (p=0.003). The level of placental TGF-β in malaria infected group was also higher than non-infected group but the difference was not significant (p=0.064). Pearson correlation test showed that there were significant negative correlations between fetal body weights with the level of placental TGF-β (p=0.017, r=-0.568) and the expression of placental endoglin (p=0.002, r=-0.694). Malaria infection in pregnant mice will increase both TGF- β and endoglin in placenta tissue and correlate with low fetal weight.

Abstract

The prevalence of diabetes increased from year to year. Diabetic animal model was developed to study the pathogenesis and treatment in diabetes. The aim of this study was to determine the profile of diabetic blood glucose levels using a stratified dose streptozotocin (STZ-SD) and multi-low dose streptozotocin (MLD-STZ) induction methods. 27 Wistar rats were divided into three groups, control group, SD-STZ group, and MLD-STZ group. The induction of experimental animal was using the method of SD-STZ and MLD-STZ for 5 consecutive days intraperitoneally. SD-STZ method using stratified dose of 40, 35, 30, 25, and 20 mg/Kg of body weight, while MLD-STZ method using a dose of 20 mg/Kg of body weight for 5 times. The measurement of blood glucose level (BGL) and body weight (BW) in animal was made on 2, 4, 7, and 14 days after STZ induction. The results of this study indicate that the diabetic conditions (BGL ≥ 300 mg/dl) on the SD-STZ induction method was more stable, and no deaths occurred in experimental animals.Keywords: Streptozotocin (STZ), diabetes, blood glucose level (BGL).

Abstract

This paper reports a screening results of the secondary metabolites composed in Spathodea campanulata Beauv stem bark, evaluate inhibiting activity of malondialdehyde (MDA) on rats cancer model exposed with benzopyrene, and the histology of its lung. The secondary metabolite of the stem bark fraction consisted of alkaloids, flavonoids-phenolic, terpenoid and steroid compounds. The isolated fraction contained of these metabolites significantly indicate bioactivity by reducting of malondialdehyde (MDA) level, and also histology appearance of the lung tissue prepared from the benzopyrene-exposed rat indicated a curative activity.

Abstract

Despite their importance for the rain forest ecosystem, still not much known about the community composition of small frugivorous mammals in West-Malaysian forests. Therefore, we conducted a mark and recapture study in four 100 m x 100 m sized plots in a lowland Dipterocarp forest of Peninsular Malaysia for 4000 trap nights. Plots hosted either Dipterocarp forest or alluvial fresh water swamp vegetation. We assumed that small mammal assemblages varied between these two vegetation types. In total 249 different individuals of 14 species (9 genera) were caught, comprising rodents and treeshrews. The most abundant species was the common tree shrew (Tupaia glis, 25.8 individuals ha-1), followed the plantain squirrel (Callosciurus notatus, 10.25 individuals ha-1) and the grey tree rat (Lenothrix canus, 10.0 individuals ha-1). These three species accounted for 77.8% of the total small mammals biomass per hectare. Sex-ratio of treeshrews and squirrel species was found at 1:1, whereas Muridae showed male-biased sex-ratios. Shannon-diversity indices (H) of all trapping plots ranged at mediocre level. Plot comparison (Bray-Curtis indices) showed that plots with similar vegetation types were not more similar in their small mammal community structure. Other factors, such as distance to the forest edge and joining oil palm plantations seemed to influence species abundances. Small mammals belong to the less studied species in local rain forests, thus studying their ecology is vital in order to act towards general conservational issues.

Abstract

Ceplukan (Physalis minima L.) has long been used to treat various conditions in traditional medicine. This study aims to demonstrate the anxiolytic effects of Methanol Extract of Ceplukan Leaves (MECL) in the Elevated Plus Maze (EPM) test and correlate to IL-6 level of ovariectomized rat brain. Total of 24 Wistar rats were divided into six groups: one normal group, one group of 1 month ovariectomized (ovx), one group of 2 months ovx, three groups of 2 months ovx (each given with MECL 500; 1500 and 2500 mg/kg doses for 1 month). The anxiety-like behavior level was measured by EPM test. After EPM test, the brain was removed to measure level of IL-6 by ELISA. The data were processed and analyzed by one-way ANOVA and Pearson correlation. We found that the MECL-treated rats enter the opened-arm higher than the control rats. It indicates that the MECL-treated rats are less anxious than the control rats. The results also show the decreased of IL-6 level in MECL-treated rats.

Abstract

Cadmium (Cd) is one of the most toxic heavy metal. Numerous tissues both in vitro and in vivo suggest that Cd induced lipid peroxidation. The presence of antioxidant can inhbit this process. This present study was designed to determine the effects of mango, papaya, and banana juices on on malondi aldehyde levels (MDA, product of cellular lipid peroxidation) during Cd exposure in vitro. Significant differences was observed between three selected fruits juices and MDA levels and also between concentration of fruits juices and MDA levels. The highest correlation was found between banana juices and MDA levels with IC50 1,12, and followed by papaya and mango juices with IC 50 1,37 and 3,87 respectively. The findings of this study suggest that the three selected fruits juices may exert its protective action against Cd-induced lipid peroxidation in liver homogenate in vitro possibly through its antioxidant mechanism.Key WordCadmium, Lipid Peroxidation, Antioxidant Activity and Fruits Juices

Abstract

Preeclampsia has major symptoms of hypertension and proteinuria and is a cause of significant maternal and infant mortality in the world. The slow development of preeclampsia research possibility caused by the difficulty in acquiring animal preeclampsia. Many existing animal model have been developed, but most of them are expensive to do. The purpose of this study was to determine the effects of intraperitoneal injection of pregnant patients serum with high TNF-α levels toward sFlt-1 serum concentration and blood pressure of pregnant mice. Pregnant patients serum with high TNF-α levels (>20 pg/mL) was injected intraperitoneally to pregnant mice at gestational age 13 and 14 days. At 18 days of gestation, the blood pressure was measured, then the mice were dissected and the serum was taken to measure serum sFlt-1 concentration using ELISA kit(Bioassay Technology Laboratory, E0611Mo). The results showed there was a significant increase in blood pressure (p = 0.000) and the sFlt-1 levels (p = 0.002) of pregnant mice group that injected by pregnant patient serum with high TNF-α levels compare to control group. From these data it can be concluded that intraperitoneal injection of pregnant patients serum with high TNF-α levels to pregnant mice can increase blood pressure and sFlt-1 serum concentration of mice.

Abstract

Exploration of Mekongga national park resulted in novel finding of anticancer property from Tetrameles nudiflora. The investigation of toxic features of T. nudiflora is vital for further studies of its pharmacological activities. Acute toxicity test was done on methanolic extracts of T. nudiflora in DDY mice. Animals were divided into 5 groups: Group 1 was given 1 ml solution of 2.5% Tween 80 in a single oral dose; Group 2, 3, 4 and 5 were orally administrated a single dose of 1, 2, 4 and 8 g/kg body weight T. nudiflora, respectively. Toxic effects of the extract were evaluated on the basis of behavioral observations in the form of locomotor activity; curiosity; defecation; urination and also animal mortality. Observations were carried out for 14 days. No significant changes on body weight, and behavioural activities were recorded. Mortality was recorded up to 22% of the male group, and 11% within the female group. The T.nudiflora extracts tested for toxicity against brine shrimp had 50% lethal concentration (LC50) values of 46.67 μg/ml. It can be concluded that methanol extracts of T. nudiflora are potential to be explored as anticancer (LC50=46.67 μg/ml). The extract is slightly toxic in male mice with Lethal Dose 50 (LD50) 12.6 g/kg body weight, and practically non toxic for female (LD50>15 g/kg body weight).Key words : Tetrameles nudiflora, brine shrimp lethality test, acute oral toxicity, LD 50.

Abstract

The present study was undertaken to evaluate the effect of a topical application of ethanolic extracts of Mauli banana stem on buccal mucosa wounds in rats (Rattus novergicus). Three sets of experiments with 3 groups male rats each consisted of 9 animals were used for studying wound oxidative stress status. Group 1 (P0) as the negative control was left to heal spontaneously; group 2 (P1) as the postive control were treated with alocair topically; and group 3 (P2) as treatment group were treated with ethanloic extracts of Mauli banana stem topically, respectively, 24h after wound creation for 3 days. The oxidative stress status was evaluated by monitoring the SOD, CAT activity, MDA and CC levels. The effect of ethanolic extracts of Mauli banana stem on oxidative stress status revealed significant increased in SOD activity, decreased in MDA levels, and no significant change both in CAT activity and CC levels compared to negative control. These results showed that The ethanolic extracts of Mauli banana stem might affect the oxidative stress status during wound healing process. Abstract

This research aims to assess the environmental factors that affect the abundance of scales populations on the apple crop. The study was conducted in July 2012 to January 2013. The study was conducted at two sites with different altitude. Bumiaji village's altitude is ±900 m and Tulungrejo village's altitude is ±1,515 m asl. Multiple linear regression analysis was conducted to determine the environmental factors that affect the abundance of scales populations on the apple crop. Based on the results of the regression analysis, air temperature and humidity affect the abundance of scales populations in Binangun while rainfall and long solar radiation have no effect. Scales population abundance of the apple crop in Binangun was influenced by air temperature and humidity one week before. Based on the results of the regression analysis, air temperature, air humidity and rainfall affect the abun dance of scales population in Tulungrejo while long solar radiation has no effect. Scales population abundance ofthe apple crop in Tulungrejo was affected by air temperature two weeks before, while humidity and rainfall were affected one week before. The higher air temperature scales, flea population level will increase. And the higherrelative humidity, scales population levels will increase.
