Abstract

A research on the isolation and identification of fungi Colletotrichum spp. PCS isolate as the cause of anthracnose desease on chili fruit (Capsicum annuum L.) in Bali. Fungi Colletotrichum spp. isolated from large chilies anthracnose symptoms were taken from three locations of chili planting in Bali i.e. Apuan Village, Tabanan (isolates APN), Village Belok Sidan, Badung (isolates BLS) and Village Pancasari, Buleleng (isolates PCS). To ensure that these three isolates of the fungus caused anthracnose on chili fruit, tested Koch's postulates and virulence test were used. Based on fungal virulence test, isolates PCS was the most virulent isolates than APN and BLS isolates, therefore the PCS isolates was further identified. Macroscopic  and microscopic identification of the fungus Colletotrichum spp. isolate PCS were conducted at the Laboratory of Biopesticides, Faculty of Agriculture, University of Udayana by observing the shape of the colony, colony color, growth rate, hyphae structure, spore shape, and size of the spores. The identification of the fungus Colletotrichum spp. isolate PCS was carried out in the Department of Molecular Biology, Faculty of Mathematics and Natural Sciences, Bogor Agricultural University based on the analysis of 18S rRNA genes by using ITS1 and ITS4 primers. Phylogeny analyzes were performed using the MEGA 6.0, maximum parsimony method (MP) with 1000x bootstrap. The results showed that the fungus Colletotrichum spp. isolate PCS was a fungal of Colletotrichum acutatum as a clade with sequences of fungus C. acutatum by 100% of Bootstrap Support (BS).

Abstract

The main objective of this research was to isolate and identify microbes residing in the rhizosphere zone of strawberry (Fragaria x ananassa Dutch.), cultivated at Pancasari village, Bedugul Bali. This was an explorative and descriptive research conducted from April to July 2018. Soil samples were collected from the rhizosphere of strawberry plants and microbes residing in them were isolated by applying dilution and platting method, purified, and identified at the Food Microbology Laboratory, Faculty of Agricultural Technology, Udayana University. Important characteristics of the isolated microbes were compared to those specified in references, such as Bergey’s Manual of Determine Bacteriology 9th Edition (Holt et al., 1994) for bacterial isolates and Fungal Identification keys (Barnett and Hunter, 1972; dan Pitt and Hocking, 2009) for fungal isolates. Microbes successfully isolated and identified in this research included fungi and bacteria belong to Aspergillus flavus, Aspergillus niger, Fusarium oxysporum, and Colletotrichum sp., Pseudomonas sp., and Bacillus sp. Abstract

The Lesser Sulphur-crested cockatoos (Cacatua sulphurea) are among the most endangered parrots in Eastern Indonesia. This bird was common species, but since 30 years ago, it was apparent that the decline of the species had been ‘dramatic’ and that it was ‘threatened throughout its range. The best chance to conserve the nominate race lines in Southeast Sulawesi where there is a population of perhaps 50-100 individuals remaining in Rawa Aopa Watumohai National Park [RAWNP]. Direct visualization could be carried out, employing a belt transect and point count method to construct a census. The field research on February until March 2011, founded 55 cockatoos from10 points of all 19 points different survey locations. The research method using scan sampling per 3 menit durations with recorded about 1110 points from 12 days of behavior observations. The daily activities result are: resting 68,9%; moving 21,1%; and eating 10%. On breeding process its begin from grooming or courtship 9 menits and mating 83 seconds. Cockatoos eating proportion: fruits 64,3%; flowers 14,3%; young leafs 14,3% and seeds 7,1%. The research founded eight nestholes trees, from Kolaka Parinarium carymbosum and Kuiya Alatonia scholaris. The cockatoo daily activites and habitat conditions are based data for supporting conservation habitat management in the future.

Abstract

This research was aimed to find out the genetic structures of three generations of Balinese population, in order to determine the best loci used for paternity testing among this population, and observed the mutation rate of these loci. The DNA samples were taken from the epithelium cell of 25 families which were collected from the children, father, mother, grandfather and grandmother of the children, from both mother and father sides (family with three generations). The DNA was extracted in Phenol-Chloroform method with modifications. DNA amplification was conducted in PCR method using pairs of primer 5, namely: FGA, D18S51, D2S1338, TPOX, and D16S539, and its products were electrophoresed and visualized in 10% of PAGE, stained in silver nitrate. The genetic structures of the three family generations showed 30 variants with different frequencies in each locus. The highest heterozygosity value was detected in FGA (8 alleles), then followed by D18S51 (7 alleles), TPOX (6 alleles), D16S539 (5 alleles), and the lowest was in D2S1338 (4 alleles). The highest value of heterozigosity and Power of Discrimination were found in FGA, followed by TPOX, D18S51, D2S1338, and the lowest was in D16S539. Therefore, it can be concluded that out of five loci tested, 4 of them can be recommended to be used for paternity testing of Balinese population, except D16S539. Abstract

INTISARI Berat Bayi Lahir Rendah (BBLR) adalah salah satu kejadian yang membuat angka kematian bayi meningkat di Indonesia. BBLR dapat berisiko bayi mengalami komplikasi beberapa penyakit, kecacatan dan bahkan gangguan tumbuh kembang. Angka BBLR di Daerah Istimewa Yogyakarta mencapai tingkat 10% pada tahun 2013 Penelitian ini bertujuan untuk mengetahui jumlah kejadian BBLR dan mengetahui faktor risiko apa saja yang berhubungan dengan kejadian BBLR di RSKIA Sadewa pada tahun 2016. Jenis penelitian ini adalah kuantitatif berdasarkan studi observasional analitik dengan desain cross-sectional. Sampel sebanyak 1200 terdiri dari semua bayi BBLR dan bayi normal yang ada di RSKIA Sadewa Sleman pada tahun 2016 di periode bulan Januari 2016 sampai Desember 2016. Teknik sampling yang digunakan adalah purposive sampling. Penelitian ini menggunakan data sekunder dari rekam medis untuk mendapatkan data yang dibutuhkan dan dilakukan analisis korelasi dengan uji Chi Square. Hasil penelitian didapatkan 319 BBLR dari 1200 bayi yang dilahirkan di RSKIA Sleman. Ada faktor risiko yang memiliki hubungan signifikan (p < 0,05) dengan kejadian BBLR yaitu umur Ibu, umur kehamilan Ibu, paritas, pekerjaan ibu, dan kontrol kehamilan (ANC). Didapatkan juga tingkat keeratan hubungan dari faktor yang memiliki hubungan yaitu umur ibu (r = 0,373), umur kehamilan ibu (r = 0,416), paritas (r = 0,114), pekerjaan ibu (r = 0,112), dan kontrol kehamilan (ANC) (r = 0,079). Berdasarkan hasil yang diperoleh didapatkan bahwa faktor umur kehamilan ibu adalah faktor yang memiliki tingkat keeratan hubungan tertinggi terhadap kejadian BBLR dengan nilai keeratan yang sedang (r= 0,416). Disimpulkan bahwa angka kejadian BBLR sebesar 319 dari 1200 kelahiran di RSKIA Sadewa Sleman DIY. Umur ibu, umur kehamilan ibu, paritas, pekerjaan ibu, dan kontrol kehamilan (ANC) memiliki hubungan yang signifikan terhadap kejadian BBLR dan umur kehamilan menjadi faktor yang memiliki hubungan paling erat. Abstract

Penelitian tentang karakteristik sarang tempua Ploceus Philippinus di area persawahan Gurun Aur, Kecamatan Banuhampu, Kabupaten Agam, Sumatera Barat, telah dilaksanakan dari bulan Maret sampai Mei 2016 dengan metode survey langsung. Tujuan dari penelitian ini adalah untuk mendeskripsikan karakteristik dan material penyusun sarang. Dari hasil penelitian ini diketahui bahwa bentuk sarang burung tempua secara umum menyerupai pendulum di pelepah daun pohon kelapa dengan rata-rata tinggi sarang 7,81 m dari permukaan tanah. Sarang mengantung di pelapah pohon kelapa dengan pintu masuk dari bawah.Bentuk sarang yang belum lengkap juga ditemukan jika burung gagal mendapatkan pasangan. Sarang ini menyerupai kubah berbeda dengan sarang yang telah lengkap berbentuk seperti kantung dengan variasi lonjong dan bentuk ginjal. Sarang juga memiliki nilai yang bervariasi pada ukuran panjang, berat dan tinggi gantungan. Bahan yang digunakan burung tempua membangun sarang sebagian besar dari potongan rumput (Graminae) dan malai tumbuhan Imperata cylindrica untuk alas chamber (ruang telur).

Abstract

Anggrek Dendrobium sp. merupakan anggrek yang paling populer untuk tanaman hias pot dan bunga potong. Permintaan dari dalam negeri masih belum terpenuhi, dan masih mengimpor dari negeri tetangga. Salah satu usaha untuk memenuhi permintaan anggrek dalam dan luar negeri yaitu dengan perbanyakan vegetatif dengan menggunakan batang anggrek. Penelitian ini bertujuan untuk mengetahui pengaruh ukuran batang dan konsentrasi zat pengatur tumbuh Zeatin terhadap pertumbuhan vegetatif anggrek Dendrobium sonia. Penelitian dilaksanakan pada November 2015 hingga Maret 2016 di shade house Puri Candra Asri, Batubulan, Gianyar – Bali. Rancangan penelitian yang digunakan adalah Rancangan Acak Kelompok (RAK) dengan dua faktor yaitu konsentrasi Zeatin (0 mg/L, 0,1 mg/L dan 0,2 mg/L) dan panjang batang (0-10 cm, 10-20 cm dan 20-30 cm. Total ada sembilan kombinasi perlakuan dan masing – masing kombinasi perlakuan terdiri dari 7 ulangan. Data kuantitatif berupa jumlah akar, panjang akar, jumlah tunas, panjang tunas dan jumlah daun pada 16 minggu setelah tanam (MST). Data jumlah akar, panjang akar, panjang tunas dan jumlah daun dianalisis dengan Analysis of Variance (ANOVA) kemudian ilanjutkan dengan uji Duncan’s multiple range test (DMRT) pada taraf 5%. Pertumbuhan panjang akar dan jumlah daun terbaik yaitu pada perlakuan Z2B2 (Zeatin 0,1 mg/L + batang 10-20 cm), sedangkan pertumbuhan jumlah akar, jumlah tunas dan panjang tunas terbaik yaitu pada perlakuan Z2B3 (Zeatin 0,1 mg/L + batang 20-30 cm).

Abstract

This study aims to prove that supplementation of red bean extract (Phaseolus vulgaris L) decreased malondialdehyde (MDA) levels of stressly induced activity of male wistar rats (Rattus norvegicus). This research was an experimental with pre-test post-test control group design. The rats were randomly divides into 3 groups: negatif control group (P0), positive control group (treated with overtraining and aquades 2ml/P1), and treatment group (treated with supllementation of red bean extract 650 mg/200 g body weight and overtraining/P2). The rats were swam for 45 minutes everyday. Data were analysed by one way anova test. The study showed that MDA level pretest of the each group was 69,70±0,52 mmol/L; 70,12±0,54 mmol/L and 69,59±0,51 mmol/L respectively. MDA level posttest of the each group was 69,66±0,27 mmol/L; 90,08±0,20 mmol/L and 62,85±0,50 mmol/L respectively. From the result of this study, it can be concluded that suplementation of red bean extract (Phaseolus vulgaris L) decreased malondialdehyde (MDA) levels of stressly induced activity of male wistar rats (Rattus norvegicus). Abstract

The purpose of this research was to study the antioxidant activity from ethanol extract of horstail (Equisetum debile L.) in the free radical scavenging in mice blood plasma. The antioxidant activity test was conducted with the DPPH method and measuring the MDA concentration in mice blood. The antioxidant activity test with the DPPH results showed that the ethanol extract of horstail (Equisetum debile L.) had the antioxidant activity of IC50 which was 1.604 mg/mL or 1,604 ppm. The statistical analysis result of the MDA blood plasma in mice with various doses (125 mg/kgBM, 250 mg/kgBM, 375 mg/kgBM and 500 mg/kgBM) showed that the 500 mg/kgBM dose extract was able to reduce the MDA concentration in mice blood which given the most amount of exercise.

Abstract

This study aimed to examinine the physiological condition of Baung's blood (Hemibagrus nemurus Blkr.) on the Batang Ombilin River, West Sumatra. This research uses quantitative descriptive method where quantitative data in the form of physic value is presented through comparison table average. The results showed the physiology of blood of Baung fish (H. nemurus Blkr.) where the average amount of erythrocytes was similar in both sites where in Ombilin, the erythrocyte count had an average value of 2,268 x 106 cells/mm3 and in Sawahlunto 2,267 x106 cell/mm3. While for the average number of leukocytes of Baung fish (H nemurus Blkr.) In Sawahlunto is greater than the average number of fish leukocytes in Ombilin where in Sawahlunto with an average leukocyte count of 3,5x104 cells / mm3 and in Ombilin is 2,7x104 cells/mm3. And the blood hematocrit value of Baung fish. In Ombilin obtained a value of 38,8% and in Sawahlunto of 38,6%. As for the calculation of hemoglobin levels in both sites, hemoglobin levels were found in low levels of 8,5 g/dl in Ombilin and 5,4 g/dl in Sawahlunto. The levels of erythrocytes, leukocytes and blood hematocrit of Baung fish are at normal ranges. While hemoglobin levels at both sites were under normal conditions.
