Abstract

Streptomyces merupakan bakteri dari kelompok Actinomycetes. Genus ini diketahui dapat memproduksi berbagai senyawa aktif diantaranya antibiotik, antiviral, dan enzim. Enzim yang dapat ditemukan pada bakteri dan jamur yaitu salah satunya enzim kitinase. Enzim kitinase yang diproduksi oleh bakteri lebih baik dibandingkan kitinase dari sumber lain karena dengan waktu yang relatif singkat dapat melakukan proses perkembangbiakan. Bakteri kitinolitik merupakan agen pengendalian hayati terhadap jamur patogen maupun serangga hama yang sangat potensial karena aktivitas kitinase yang dimiliki. Penelitian ini bertujuan untuk mengisolasi, mengetahui karakteristik Streptomyces spp. dari lumpur selokan dan uji aktivitas enzim kitinase. Aktivitas enzim kitinase diketahui melalui zona bening yang terbentuk pada media uji. Hasil penelitian diperoleh lima spesies Streptomyces spp. dari sampel lumpur selokan yang diisolasi dengan karakteristik yang berbeda-beda. Hasil penelitian juga menunjukkan bahwa dari lima spesies Streptomyces spp., dua spesies yaitu Streptomyces sp.3 dan Streptomyces sp.5 mampu memproduksi enzim kitinase, sementara tiga spesies Streptomyces lainnya tidak memproduksi enzim kitinase. Streptomyces sp.3 memproduksi enzim kitinase dengan terbentuknya zona bening sebesar 1,85±0,07 cm dan pada Streptomyces sp.5 terbentuk zona bening sebesar 0,65 ±0,14 cm . Berdasarkan data tersebut Streptomyces sp.3 dan Streptomyces sp.5 memiliki potensi sebagai bakteri kitinolitik yang memiliki manfaat sebagai agen pengendalian hayati.

Abstract

This study aims to determine the diversity of insects and plants used as a habitat in unburned and previously burned peat land forests in Palangkaraya, Central Kalimantan. Insects were collected by beating the branches of plants, aerial sweeping, pitfall traps and light traps. Plants were identified in situ or plant samples were collected and identified later in the Lab. The diversity of insects were compared between unburned and previously burned peat forests by analyzing its index diversity (H’) and index of similarity (IS). In total, the insects collected from unburned peat forest were 551 individual, belongs to 12 order and 51 families, and 431 individual insects were collected from previously burned peat forest which belongs to 10 order and 38 families. The family of insects that most frequently found at both areas was Formicidae (Hymenoptera). The insects diversity of both forests were still high, that is H' = 3,45 of unburned peat forest and H '= 3,11 of previously burned peat forest, with the similarity index IS > 50% of both peat forests. The number of plant species found was higher in unburned peat forest (38 species) than in previously burned peat forest (9 species). The previously burned forest was dominated by Acacia plants, while in unburned peat forest the plants seem to evenly spread.

Abstract

Serangan island is part of Ngurah Rai Forest Park. Fifty five species of birds, belong to 30 families have been found in Serangan island. Based on earlier information that Phalacrocorax melanoleucos was commonly shown in lagoon and mangrove. This study aimed to investigate the habitat condition and the behavior of P. melanoleucos in Serangan Island. The study has been conducted from November 2016 to May 2017, in lagoon area of Serangan Island in two observation points of 8º44.647? S 115º13.332? E and 8º44.769? S 115º345? E. Scan sampling technique was applied to collect the data of the bird behavior and the data of species of plant being used by P. Melanoleucos was collected by tracking methods. P. melanoleucos was commonly found foraging in the morning (45 % ± 0.15 %). Similar pattern of behavior was observed during the day and in the afternoon, but the amount of time spent for perching during the day and afternoon were slighly lower, 42 % ± 0.02 % and 41 % ± 0.02 % respectively. Agonistic behaviour was rarely observed during the study (5 % ± 0.06 %). Casuarina equisetifolia and Soneratia alba plants were commonly used by birds for perching. Abstract

The role of insects in papaya plants is as pollinators, pests, parasites and predators of other insects. Referring to the role of insects, this research was conducted to determine the insects on an papaya plantations. Traping method using methyl eugenol and ripe papaya fruit as well as the manual method with insect net used in this research. Each method was repeated three times with an interval of sampling is one week. Insect netting done on the same day with traping method. Samples were preserved and then taken to the laboratory for identification. Environmental factors were measured are: temperature, humidity, latitude and altitude. Insects were identified by reference: Borror et al. (1992), CSIRO I & II (1991) and also Peggie and Amir (2006). Five species belonging to the genus Bactrocera found on methyl eugenol traps and five species belonging to the genus Drosophila was found in ripe papaya fruit traps as well as 16 other species found using insect netting around the papaya plant. In addition, there is also the genus Paracoccus on a papaya plant. Bactrocera papayae and genus Paracoccus is an insect pest on papaya plants.Keywords: diversity, insect, papaya plantation, Sanur

Abstract

Taman Nasional Way Kambas Lampung (TNWK) berlokasi di Ujung Selatan Pulau Sumatera dimana Gajah Sumatera dikonservasi. Gajah Sumatera (Elephas maximus sumatranus) merupakan satwa yang dilindungi dan terdaftar dalam red list book International Union for Conservation of Nature (IUCN), dengan status terancam punah. Penelitian ini bertujuan untuk mengetahui perilaku harian anak Gajah Sumatera (Elephas maximus sumatranus) di Pusat Konservasi Gajah tersebut. Pengambilan dan pengumpulan data perilaku harian dilakukan dengan metode “fokal animal scan sampling” yaitu masing-masing individu hewan di luar kandang diikuti selama satu hari dan data diambil dengan interval 5 menit. Pengambilan data dilakukan pada pagi hingga malam hari pukul 08:00 - 21:20 WIB. Penelitian dilakukan dari tanggal 25 Januari 2016 hingga 3 Maret 2016. Dari hasil pengamatan didapatkan bahwa sebagian besar waktu anak Gajah Sumatera digunakan untuk makan, mencari makan dan bergerak, dan hanya sebagian kecil waktunya untuk beristirahat, bermain, minum, mandi, buang air kecil, dan buang air besar. Misalnya, 34,2% waktu harian anak gajah digunakan untuk makan, 21,8% untuk bergerak dan hanya 4,8% untuk beristirahat. Jenis makanan yang paling sering ditemukan dimakan oleh gajah adalah rumput ilalang (Imperata cylindrica).

Abstract

The purpose of this study was to determine the effect of habitat on the morphometric and gonadal development and to know the range of index values ??gonad maturity sea eel (Macrotema caligans). The results showed that habitat mainly the population of seagrass is related to morphometry sea eel. At five sites studied showed an average size morphometry highest in the sea eel living in Semawang Beach, which is the average size body length of 24.96 ± 0.95 cm. This study also shows morphometry females sea eel larger than males. Gonadal development of eel female is formed through the five stages. The first stage, characterized by underdeveloped oogonia,  the second stage, a massive increase gonads up to three times the original size. The third stage, lipid vacuoles are formed inside the egg with an increase in cell volume. The fourth stage, marked by the maturation of oocytes and the fifth stage, oocytes undergo atresi. Values ??range of gonad maturation index sea eel (Macrotema calligans) on the entire study was 0.16% to 9.75%.

Abstract

This research was aimed to find out the genetic structures of three generations of Balinese population, in order to determine the best loci used for paternity testing among this population, and observed the mutation rate of these loci. The DNA samples were taken from the epithelium cell of 25 families which were collected from the children, father, mother, grandfather and grandmother of the children, from both mother and father sides (family with three generations). The DNA was extracted in Phenol-Chloroform method with modifications. DNA amplification was conducted in PCR method using pairs of primer 5, namely: FGA, D18S51, D2S1338, TPOX, and D16S539, and its products were electrophoresed and visualized in 10% of PAGE, stained in silver nitrate. The genetic structures of the three family generations showed 30 variants with different frequencies in each locus. The highest heterozygosity value was detected in FGA (8 alleles), then followed by D18S51 (7 alleles), TPOX (6 alleles), D16S539 (5 alleles), and the lowest was in D2S1338 (4 alleles). The highest value of heterozigosity and Power of Discrimination were found in FGA, followed by TPOX, D18S51, D2S1338, and the lowest was in D16S539. Therefore, it can be concluded that out of five loci tested, 4 of them can be recommended to be used for paternity testing of Balinese population, except D16S539. Abstract

Pollination is the most important live processes for plant especially for seedling plant. Some previous research results show that insects play an important role in helping the pollination process both in wild plants and in productive plants. The eggplant (Solanum melongena L.) is one of the important agricultural commodities. However, the eggplant production in the local farming system today often experiences obstacles when its production is not in sufficient quantities. The potential of Apis cerana L. as a pollinator is a chance solve the productions problems. The purpose of this study was to determine the role of honey bees (Apis cerana L.) in increasing yield of eggplant (Solanum melongena L.).This study obtained the eggplant productivity data. All data was collected and analyzed with SPSS 17 statistical program. Before analysis, all data normality were tested and it was analyzed by One-way ANOVA and when significant differences were found, analysis was continued with the Tuckey Test to find out the difference within treatment with a significance value ???? <0.05 .The results of this study note that bees have a certain amount of time to carry out their activities and the addition of bees colony to the experimental field produced two times more of yield compared with the other groups that were not treated with he colony. Abstract

Desa Kesiut sebagai Kawasan Pedesaan Prioritas Nasional (KPPN), menyimpan sumber daya dan kearifan lokal desa meliputi bidang pertanian, perkebunan, peternakan, perikanan dan industri rumahan. Berbagai program bantuan usaha ekonomi kreatif dilaporkan cukup efektif meningkatkan kesempatan kerja dan pendapatan rumah tangga miskin di Kecamatan Kerambitan. Kegiatan KKN PPM di Desa Kesiut bertujuan untuk pemberdayaan masyarakat dan pengembangan sumber daya kearifan lokal yang ada di desa melalui program kerja pokok. Metode penyelesaian program kerja pokok meliputi; (1) penyuluhan peningkatan produktivitas ternak babi dan sapi, (2) pelatihan inseminasi buatan pada ayam buras, (3) penyuluhan budidaya ikan air tawar dan pembuatan pakan ikan alternatif, (4) pelatihan biochast, (5) pelatihan pembuatan pupuk organik dan pestisida alami, (6) penyuluhan pengendalian hama pengganggu tanaman, (7) peningkatan produksi dan diversifikasi produk olahan pertanian serta pemasaran pasca panen. Melalui kegiatan KKN-PPM Universitas Udayana Tahun 2018 di Desa Kesiut, telah dilaksanakan pemberdayaan masyarakat yang partisipatif dan upaya pengembangan potensi lokal desa. Abstract

Catharanthus roseus is one plant recognized as medical potential which can decrease cholesterol. Thepresent study was carried out to evaluate the effect of aqueous leaf extracts of C. roseus on plasma lipidprofil levels in rats cholesterol rich diet and atherogenic index. This study was carried out on 15 SpraqueDawley male rats randomly distributed into five groups (n=3). Rats hypercholesterolemic wereadministrated cholesterol rich diet containing 1% (w/w). Normal control group with normal diet whitoutextracts (K1), positive control hypercholesterolemic group (K2) with cholesterol rich diet whitout extracts,and others three groups (K3-K5) were feed high cholesterol and 1 mL of aqueous leaf extracts of C. roseuswith a dose of 20% (w/v), 40% (w/v) and 80% (w/v) respectively, administered twice daily 1 mL orally.Treatment was given for 28 days. After treatment, the plasma lipids profile such cholesterol total, highdensity lipoprotein (HDL), low density lipoprotein (LDL), triglyceride (TGA) level were measured. Theresults showed that during the period of treatment with cholesterol rich diet have produced under condition hypercholesterolemic of rats with average cholesterol levels of 124.33 ± 4.04 mg/dL (K2 group). Treatmentwith extract dose of 80% showed the best results among the other doses in reducing levels of total cholesterol,triglycerides, LDL and raise HDL levels. Extract treatment dose of 80% up to four weeks resulted incholesterol total (79.33±3.51 mg/dL), TGA (72.33±6.65 mg/dL), HDL (55.00±3.60 mg/dL), and LDL(9.87±5.34mg/dL). In addition, value ratio of cholesterol:HDL was 1.4: 1 and atherogenic index value was0.44. These results suggest that extracts of leaf C. roseus optimum doses 80% (w/v) has hipolipidemicactivity in hypercholesterolemic rats and it has low atherogenic index value.
