Abstract

Fungal endophytes have been known as sources for bioactives with high chemical structure variability. This study aimed to screen extracts of some endophytic fungi associated with plants from Enggano Island as antioxidant and antibacterial using thin layer chromatography (TLC) bioautography method. Antibacterial ativity was performed against Escherichia coli and Staphylococcus aureus. Seventeen out of 22 extracts showed growth inhibition activity against both bacteria, while 16 extracts were active as DPPH free radical scavenging agents. Further determination on IC50 value of four prominent extracts revealed that fungal endophytes AK3018-1, FC-1, KC-4, and SO-3 have IC50 value of 85, 84, 704, and 347 ?g/ml, respectively in DPPH radical scavenging method. Extracts of fungal endophytes CR-3, CS-2, and SM-2 showed prominent antibacterial activity among other extracts, indicated by wide clear white zone around the spot. Further evaluation on minimum inhibitory concentration (MIC) of those three extracts by microdilution method showed that CR-3, CS-2, and SM-2 have MICs value of 512, >512, and 64µg/ml, respectively against S. aureus.Fungal endophytes AK3018-1, FC-1, KC-4, SO-3, CR-3, CS-2, and SM-2 were respectively isolated from Dioscorea bulbifera tuber, Fibraurea chloroleuca twig, Knema cinerea twig, Smilax odoratissima stem, Cryptocarya sp. twig, Calophyllum soulattri twig, and Smilaxmacrophylla stem.

Abstract

Isolation and selection of Aphyllophorales as lignin degrading fiingus had been done in Bukit Bangkirai Forest, East Kalimantan. Aim of the research was to obtain Aphyllophorales fungi which have ability to degrade lignin. The fresh fruit bodies were collected from trunks, branches and litters of plants in the forest. The work was conducted at dry season. Fruit bodies were taken to base camp for isolation. The isolates were tested for their ability to degrade lignin. We found 24 isolates of Aphyllophorales fungi. Those isolates were grew in poly R-478 medium, 12 isolates were able to degrade lignin. Megasproporia sp. strain IB-24 had the highest ability in lignin degradation. The fungus can degrade poly R-478 8,05% during 90 minutes.

Abstract

A study on species composition and threat potential of invasive plant species was conducted in the Gunung Halimun-Salak National Park,Cidahu Resort, West Java. We developed two plots of different altitudes. The result showed that there were four invasive alien species possessing potential threat to the ecosystem and native species in the National Park, i.e. Piper aduncum (Piperaceae, with an Important Value of 20,70); Calliandra calothyrsus (Mimosaceae, IV = 9,11), Austroeupatorium inulaefolium (Asteraceae, IV = 18,77), and Clidemia hirta (Melastomataceae) as shrub. The threats of invasive plants happen in open forest area and could occur from the residential or public places.

Abstract

The aim of this research was to determine the optimum percentage of glycerol addition into the diets of  Nile Tilapia (Oreochromis niloticus) on growth performance, feed convertion ratio, and survival rate. The research was conducted using 15 aquaria with the size of 60 x 50 x 45 cm3, each containing 25 O. niloticus with average initial weight of 4.63 ± 0.15 g. The experiment was conducted using a complete random design with five treatments and three replications. Isonitrogenous (31%) and isocaloric (17 MJ.kg-1) diets were provided for 60 days of rearing period. Five diets were formulated with glycerol content of 0% (G0); 5% (G5); 10% (G10); 15% (G15), and 20% (G20). Fish were fed twice daily with experimental diet at satiation level. The result showed that addition of 10% glycerol in the diet (G10) presented the highest values for weight gain (105.85 ± 0.40%), specific growth rate (3.44 ± 0.01%), protein retention (27.75 ± 0.02%), protein efficiency ratio (2.53 ± 0.15%), lipid retention (60.44 ± 0.03%), and feed convertion ratio (1.59 ± 0.01%) (P < 0.05). Survival rate was not affected by dietary containing glycerol (P > 0.05). Therefore, addition of 10% glycerol into the diet gave a significant increase on growth performance and feed convertion ratio without adverse effect on survival rate of Nile Tilapia.

Abstract

A study on desiccation sensitivity of rambutan (Nephelium lappaceum L.)excised embryo has been conducted.Excised embryos were desiccated into target moisture content of 30%, 25%, 20%, 15% and 10% respectively.Results shown that the critical level for moisture content was 20% with 60% viability and 7.431% per etmal growth rate and 929ms electric conduction Further study is needed as to investigate on how long the embryo will withstand subsequent storage treatments.

Abstract

Flowering in jatropha (Jatropha curcas L.) was considered as one of major factors that contribute to its productivity. Small number of female flowers produced in each inflorescence was believed as the main cause of low seed production.Introduction of Hd3a flowering gene driven by rol C promoter was supposed to improve total number of flowers including female flower.The objective of this research was to optimize cell proliferation and regeneration medium in Jatropha transformation method mediated by Agrobacterium, to obtain transgenic Jatropha containing Hd3a flowering gene as well as to understand the effect of this transgene on jatropha flowering character.Callus induction medium containing 0.5 mg/1 IBA added with 3 g/1 PVP produced the highest frequency of shoot formation.We obtained 26.67% to 33.33% putative transgenic plantlets that were able to grow in 40mg/l hygromycin selection medium. PCR analysis revealed that seven out of 10 putative transgenic plantlets were positively transgenic.Extremely early flowering character that was confirmed by histological analysis was also shown by some transgenic plantlets.

Abstract

Mangrove forests sequestrate and store a lot of carbon and are important to tackle climate change. However, much of these forests have been cleared dramatically and such clearings destroyed carbon sinks and released carbon into the atmosphere as carbon dioxide. The international climate agreements highlight Reduced Emissions from Deforestation and Forest Degradation (REDD+) as a key and effective option for mitigating climate change. To help making REDD+ a reality, an alternative approach is needed to measure above ground carbon stock quickly and accurately. Here, we link ground-based data collected from field measurement using belt transect method with satellite image data of the ALOS AVNIR-2. The objectives are to identify the characteristics of mangrove vegetation, to estimate the amount of above ground carbon stock, and to examine capability of vegetation indices of NDVI, SR, dan SAVI from the ALOS AVNIR-2 to estimate the amount of above ground carbon stock in mangrove. Results showed that the mangrove vegetation in Gugus Pulau Pari was relatively diverse and this forest contained above ground carbon between 4694.35 and 42841.25 kg/m2. The statistics analyses showed that there was no significant correlation between the vegetation indices of NDVI, SR, and SAVI with the amount of above ground carbon stock in tropical mangrove forest. In short, the vegetation indices of NDVI, SR, and SAVI from the ALOS AVNIR-2 were not sufficient to estimate the amount of above ground carbon stock in tropical mangrove forest.

Abstract

Mini rose (Rosa hybrida L.)is one of the favorite ornamental plants. To get a new appearance of this mini rose, two experiments were conducted at Cipanas and Bogor, from April 1999 to March 2000.In the first experiment, the treatment was the dosage of gamma ray irradiation, i.e. 0, 1, 2, 3, 4, 6, 8, 10 and 12 krad.In the second experiment, the treatment was the concentration of callus induced mediumi.e. 2,4-dichlorophenoxyacetic acid (2,4-D) (0.5 and 1 mg/1) + chinetine (1, 3 and 5 mg/1).Randomized Block Design was used with 5 replications. Mini rose Romantica Meilandina (pink color) and Prince Meilandina (dark red color) was used as a source of plant material.The results showed that the irradiation dosage from 1 until 3 krad gave the best appearance compared with the other treatments.Considering the observance to plantlet in the same dosage, the color of the flower was changed from the natural color to white and red color.The combination of 0,5 mg/12,4- dichlorophenoxyacetic acid + 3 mg/1 chinetine was the best medium to the callus growth.

Abstract

Zingiber zerumbet (L.) J.E. Smith, usually called as lempuyang gajah is belonging into Zingiberaceae. This plant has been used as traditional medicine for curing coach, asma, headache, diarrhoe and disentry. Objective of study was to determine the effect of pretreatment and growth media on the productivity of Z. Zerumbet. Research was conducted in Treub Experimental Garden,Research Centre for Biology-The Indonesian Institute of Science (LIPI), Bogor. The experiment using a Factorial Completely Randomized Design with two main factors in five replications. The first factor was pretreatment of rhizome (no treatment and pretreatment on growth chamber); the second factor was growth media i.e. soil, soil+sand+cow manure (1:1:2) and soil+sand+cow manure+compost (1:1:2:2.). The result showed that pretreatment of rhizome had significant effect on number of shoots, no pretreatment of rhizome sicnificantly had higher value. Growth media had significant effects on growth and yield which had the highest value on all parameters observed. There was an interaction effect of rhizome pretreatmnent and growth media. The best result was on the combination treatment of no pretreatment rhizome and mixed media of soil+sand+cow manure+compost(1:1:2:2.) which had the highest dry weight of rhizome.

Abstract

Growth and biochemical characters of thirty-three Nipponbare-based rice mutant lines independently harboring activation-tag in salt stress condition were investigated. Rice cv Nipponbare wild type, IR29 and Pokkali were used as isogenic, susceptible, and tolerant cultivars, respectively. Plants were grown in Yoshida nutrient solution containing NaCI (6 g/1) as the stress treatment and in Yoshida nutrient solution without NaCI as the control. Evaluation of salt tolerance was conducted at 21 days after salinization.The results showed that there was a significant difference in growth characters among genotypes in reduction of the plant height,root length, shoot fresh weight, root fresh weight, shoot dry weight, as well as in the root dry weight. Biochemical characters of the genotypes also showed significant difference in their reduction of chlorophyll-a and chlorophyll-b concentrations,total carotenoids and proline accumulation. Based on multivariate growth and biochemical characters,T3.PMO.VI.63.5a.30.9,T3.PMO.VI.30.1a.21.1, T3.PMO.V1.63.5a.33.7F3.PUR.IX.49.1d.l.6.5,T3.PMO.VI.81.3a.4.4, F3.PUR.IX.49.1d.l.6.5,and F3.PUR.VIII.5.1f.l.4.8 were assumed as potential salt-tolerant lines, respectively, while mutant lines T3.PMO.VI.30.1a.l07.7,T3.PMO.VI.63.5a.20.6,T3.PMO.III.4.4c.7.2,andT3.PMO.VI.30.1a.51.1 were assumed as salt-susceptible lines respectively.
