Abstract

The aims of this research were to determine PGF2? concentration the produced by bali cattlesendometrial and seminal vesicle monolayer cell culture and in vitro luteolytic ability on luteal monolayercell culture. The endometrial and seminal vesicle epithelial cell of bali cattle were cultured in tissueculture medium (TCM) 199 growth medium supplemented with 10% fetal calf serum and 10% EstrusMare Serum. The cells were cultured at 1.9 x 106 density per ml medium. Then Followed by incubation at38.50 C in 5% CO2 atmosphere for 12 days. The level of PGF2? in the cell culture medium were assayed byRadioimmnuassay (RIA) technique. The luteal cells were cultured in 9 days incubation and divided into 2groups. Group I were added with 10% of cell culture product and group II were added with 1,25 mgdinoprost/ml. The level of progesterone produced by luteal cell culture was measured at day 9th and 11thincubation. The result showed concentration of PGF2? cell product of seminal vesicle cell culture wassignificantly higher (P < 0.05) compared to endometrial cell culture. There was no significant difference(P>0.05) in luteolytic ability between PGF2? cell culture product and dinoprost. In conclusion, the PGF2?could be produced by monolayer cell culture of bali cattle is endometrial and seminal vesicle epithelialcells more over they have similar ability with dinoprost in luteolytic ability.

Abstract

A study on effect of various concentrations of turmeric juice on pathological changes mice liver hasbeen carried out. As many as 25 mice were used in this study and they were divided into 5 groups, i.e.: groupK-: 0.2 ml vegetable oil without CCl4; group K+: CCl4 without turmeric extract; group T1: CCl4 with 10%turmeric extract; group T2: CCl4 with 20% turmeric extract and group T3: CCl4 with 30% turmeric extract.All mice, except the K- group, were treated with 0.2 ml CCl4 orally for 7 days. On day-8 to day-29, the CCl4treated mice were given various concentrations of turmeric extract, i.e.: 10%, 20% and 30%; whereas, the KandK+ groups were given drinking water adlibitum. On day-30, all mice were sacrificed and their livertissues were collected and then processed to histopathological staining with haematoxylin-eosin. Theresult showed a clear evidence to suggest that turmeric extract can induce a better recovery compared tonon-treated mice. The improvement of pathological condition was characterized by reduction of pathologicalchanges found in the liver cells of mice, such as: hidrophic degeneration. fatty degeneration, and necrosis.More importantly, treatment with 30% of turmeric juice induced a better recovery than those with 10%and 20%.

Abstract

Morphology, distribution and relative frequency of endocrine cells in the pancreas Bullfrog (Ranacatesbeiana) were studied using conventional and immunohistochemical methods. Samples pancreas takenfrom ten adult Bullfrogs (five males and five females). In general, pancreas of the Bullfrog consists ofexocrine portion, endocrine portion (Langerhans islets) and ducts. The Langerhans islets were distributedamong the exocrine portion of pancreas. Endocrine cells in the pancreas of Bullfrog were polimorph, round,oval or triangular in shapes with bipolar cytoplasmic granules. Glucagon cells were distributed mainly inthe peripheral, insulin cells in the center while the somatostatin cells in the area between glucagon andinsulin cells of Langerhans islet. The number of the glucagon cells were higher compare to the number ofinsulin and somatostatin cells.

Abstract

Dengue Haemorrhagic fever (DHF) is one of fearsome diseases in society. Incidence of the disease isincreasing. Dengue fever is caused by dengue virus and transmitted by Aedes aegypti mosquito vector.Various chemical controls have been conducted to prevent the spread of the disease, but active contents ofthe chemical controlling substances are suspected causing many negative effect, in environment, such asvector resistance, death of non target living creatures, and environmental contamination.This researchobjective was to find an alternative solution in order to control the dengue vector by using entomopathogenicfungi as biological control agent. This research was conducted by isolation and identification of fungiinfecting mosquito larvae. Macroscopic observation revealed that one of the nine isolation products wasLagenidium giganteum. The effectiveness test in laboratory showed the zoospore LD50 to Ae.aegypti larvaeof instar 2nd was 2,35 x 106 zoospore/ml, while the LD95 value was 1,35 x 107 zoospore/ml. The oosporeeffectiveness test showed LD50 was 6,7 x 102 oospore/ml and LD95 was 1,94 x 103 oospore/ml. Using LPCBdye and blue tolouidin 2,5%, the infection mechanism of L.giganteum fungi in Ae.aegypti mosquito larvawas detected. The research is concluded that the entomophatogen fungi L. giganteum was very prospectiveto be used as a biological control agent against vector of DHF.

Abstract

An experiment has been conducted to investigate the effect of multy enzym (optizyme enzyme) andyeast addited to caccao-pod in the ration to reduce 6 weeks broiler meat cholesterol. This research used acompletely randomized design (CRD) with four treatments: as a control ration (0% caccao-pod); 15%caccao-pod; 15% cacao-pod + 0,20% optizyme enzyme and 15% caccao-pod + 0,20% yeast, with six replicates.There were five broilers 2 weeks similar on the same weight in each replicate. The experimental diets forthe 6 weeks finishing period were formulated to 20% crude protein and Energy Metabolish (2900 ME kcal/kg) in mash formed diet while drinking water was given ad libitum. The experiment was done 4 weeks.The result there was a significant decreas of cholesterol on the diet content 0,20% optizyme enzyme or0,20% yeast, were reduced distribution body fat and meat cholesterol.

Abstract

The present study was carried out to investigate the effects of tapak dara (Catharanthus roseus) onblood glucose level and insulin profile in hyperglicemic rabbits. Fifeteen local male rabbits were used forthis study. The rabbits were randomly divided into five groups. Group 1 (K-), a control negative group;group 2 (K+), a control positive hipergliccemia; group 3 (KT1) and group 4 (KT2), were groups hiperglicemiaand treated with water extract of tapak dara doses 1 and 2 g/kg bw, respectively; and group 5 (KO), a grouphiperglicemia that treated with glibenclamide 2 mg/kg bw. The result showed water extract of tapak daradose 1 g/kgbw could not decrease the blood glucose level in hyperglycemic rabbits, while dose 2 g/kg bwcould decline blood glucose level in rabbits. This decline had no significantly difference compared withglibenclamide treatment (P> 0.05). Immunohistchemistry result indicated that water extract of tapakdara could stimulate beta cells pancreas to produce insulin.

Abstract

Long lasting insecticidal nets (LLIN) is one effective way to prevent malaria. Permethrin treatedLLIN is one type of LLIN which is recommended by WHO. Several studies have shown that these types ofLLIN requiring heat assisted regeneration after washing to enhance the biological activity of insecticidethat contained in the LLIN fibers. This study aimed to compare the incidence rates of malaria in childrenunder five years old who live in the intervention area (where the heat assisted regeneration on LLIN afterwashing was applied) and control area (where the heat assisted regeneration on LLIN after washing wasnot applied). Data of malaria cases was collected from laboratory log book at all health centers in BangkaDistrict, in the period of June June 2007 until July 2008. Data were analyzed with Poisson regressionmodels. The results showed that the incidence rate of malaria in children under five years old was notsignificantly different between the treatment and control areas.

Abstract

The aim of this research was to study the safety application of xylazine-ketamine and xylazinepropofolrecurrent dosage combination as anesthesia for laparo-histerotomy surgery in cat. Thisresearch used 10 female cats, 12-18 months of age, followed randomly divided into two groups, P1:atropine 0,04 mg/kgBW/SC + xylazine 2 mg/kg BW/IM + ketamine 20 mg/kg BW/IM; P2 : atropine0,04mg/kg BW/SC + xylazine 2 mg/kg BW/IM + Propofol 20 mg/kg BW/IV. The blood of the allgroups was taken from vena femuralis at 0 minute (before treatment), 15, 30, 45 and 60 minutesduring anesthesia for measurement of blood gas value pH, pCO2 and HCO3. After all animals wereanesthetized, the animals were treated laparo-histerotomy surgery. The data were analyzed byusing Randomized Complete Block Design (RCBD). The result showed both of groups were notsignificantly difference (p>0,05) to blood gas values for pH, pCO2 dan HCO3. Besides, both groupsanaesthetic agent perfectly caused metabolic acidosis with respiratory alkalosis compensationperfectly, therefore it is relatively safe to use as anaesthetic agent for surgery that needs long timeprocedure, as laparo-histerotomy.

Abstract

Research and development of health science require an animal model which has been known for itsorigin and characteristics. One of experimental animal model which is commonly used is albino rat. Thepurpose of this study was to investigate weight of organs (kidney, liver, spleen, and lung), hematologicalvalues (hemoglobin, hematocrit, erythrocyte and leucocyte), and the values of the biochemical blood (SGPT,SGOT, glucose and total protein) of the albino rat at different ages. This study used 60 rats, which weredivided into 3 groups based on age namely 1, 2, and 3 months and groups based on sex that each groupconsisted of 10 males and 10 females. Samples of age and sex groups of rat were taken randomly. Eachcage consisted of 5 rats with the same ages and sex. Those rats fed and tap water ad libitum. The rats weresacrificed anaesthetically by with ether to take their blood and measure their organ‘s weight. Data wasanalyzed using one way ANOVA test, except data from heart organ which was analyzed using nonparametrictest (Friedman Test). To find out the increase of rats body weight age 1 -3 months, it was usedregression linier test. Results of statistic showed that there were significantly difference (p < 0,05) in bodyweight change, average of hematological values, blood biochemical values and organs weight in accordancewith increasing of rats age in all age groups. In contrast, average of hematocrit values had no significantlydifference (p > 0,05). Generally, male rats were bigger than female rats but there was no difference in all

Abstract

Tarsius is an endemic species in Indonesia but is is getting threatened. In-situ and ex-situ conservationof this species would yield better results when its genetic information and diversity determined. Theobjective of this ressearch was to study the genetic diversity of ND3 gene of Tarsius sp. Based on sequencingof PCR products using primer ND3F and ND3R resulted in 437 nts base sequence. Results of ND3 fragmentssequencing were put on multiple alignment with other primates from Genbank using of software ClustalW, and then were analyzed using MEGA program version 4.1. A different nucleotide site was found(nucleotide no. 24). The genetic distance based on nucleotide ND3 was calculated using Kimura 2-parametermodel. The genetic distance showed the smallest genetic distance 0%, the biggest 0,03% and average0,01%. The phylogenetic tree using neighbor joining method based on the sequence of nucleotide ND3 genecould not be used to differentiate among T. Dianae (from Central Sulawesi), T. Spectrum (from NorthSulawesi) and T. bancanus (from Lampung, South Sumatera).
