Abstract

Octahydroxanthene Compounds are bioactive substance which can be produced from benzaldehyde derivative. In this research, we have carried out transformation of vanilin from Indonesian natural product to octahydroxanthene derivatives compound at optimized reaction time and guanidine was used as a catalyst. Several spectroscopy methods such as IR, 1H-NMR and GC-MS were used to confirm the structure of product. The percent yield from optimation product obtained was 91% with 96% of peak area.

Abstract

Tanaman herbal merupakan warisan kearifan lokal dalam upaya pengobatan dan mempertahankan kesehatan masyarakat. Secara tradisional masyarakat Kalimantan tengah menggunakan tanaman aka kalesi (Sphatolobus ferrogenus) sebagai obat-obatan tradisional yang cukup luas. Penelitian ini bertujuan untuk mengisolasi senyawa metabolit sekunder yang terdapat dalam ekstrak metanol kulit batang aka kalesi (Sphatolobus ferrogenus). Isolasi dilakukan dengan tahapan ekstraksi, fraksinansi dan purifikasi. Hasil penelitian didapatkan dua isolat yang telah diuji kemurniannya dengan metode kromatografi lapis tipis. Isolat diidentifikasi dengan spektroskopi 1H-NMR dan 13C-NMR didapatkan senyawa gliseril monostearate dan sigmasterol. Kedua senyawa diuji sifat antidiabetes dengan metode inhibitor enzim α-glukosidase. Aktivitas senyawa gliseril monostearate tidak aktif, sementara sigmasterol menunjukan aktivitas yang lemah dengan IC50= 388 ppm.
