Abstract

Propolis is known to have an antimicrobial activity and can prevent various diseases. Propolis consumption is feared to have negative impact on the activity of digestive lactid acid bacteria (LAB). The aim of this research was to examine the effect of propolis on the growth and lactic acid production of three LAB. Ethanol Extraction Propolis (EEP) concentrations examined were control, eep and X propolis 0% (control), 0.2%, 0.6%, 1.0% and X propolis at 0.4% concentration. The parameters analyzed were the growth of bacteria counted with Total Plate Count (TPC) method and lactic acid production using titrable acidity analysis. Propolis at 0,6% concentration stimulated the growth of Lactobacillus casei subsp. rhamnosus (LCR, 24.725x108 cell/mL), but inhibited the average lactic acid production (0.071%) lower than control (0,149%). Propolis did not affect the growth of Streptococcus thermophillus (STP), but propolis at 0,6% concentration stimulated lactic acid production (0.182%) higher than control (0.112%). Propolis inhibited the growth of Lactobacillus delbrueckii subsp. bulgaricus (LDB), but at 0,2% concentration, its population was still highest (3.775x108 cell/mL) and lactic acid production was stimulated (0.195%) higher than control (0.123%).

Abstract

Cancer is a deadly disease that ranks the second after heart disease. One of the causes of cancer is free radicals causing oxidative stress in humans. Antioxidant is compound that can reduce oxidative stress. The antioxidant activity is closely connected with anticancer activity. It is known that in addition to being an analgesic, aphrodisiac of Java chili also has potential as anticancer. This study examined the potential anticancer and antioxidant from the ethanol extract of Java chili from Pamekasan and Karang Asem. The method used in this study began with the extraction of Java chili using ethanol 70 %, phytochemical testing, determination of total phenol anticancer testing using MTT method on MCF-7 breast cancer cell line and antioxidant testing using DPPH. This study uses Java chili extract derived from Pamekasan and Karang Asem. IC50 value of the cytotoxic activity originating from the area Karang Asem amounted to 63.283 ppm and Pamekasan of 83.662 ppm. IC50 value of antioxidants samples derived from Karang Asem area of 285.613 ppm and are derived from Pamekasan amounted to 288.037 ppm.

Abstract

Melinjo (gnetum gnemon) peel extract is believed to have potential as antihyperuricemic agent, however, in vivo assays have yet to be conducted to prove the antihyperuricemic activity. The objectives of this research were to analyze antihyperuricemic activity of melinjo peel extract in Sprague Dawley male white rat. A total of 24 rats were divided into 6 groups, which are a normal, positive, and negative group, 70% ethanolic extract of melinjo peel dose 450 mg/kg BW (ET I) and dose 900 mg/kg BW (ET II), and water extract of melinjo peel dose 93.35 mg/kg BW (EA). The results show that antihyperuricemic potency of 70% ethanolic extract of melinjo peel (ET1 and ET2) was more effective than allopurinol dose 90 mg/kg BW with a percentage decrease of 50% and 54%. Ethanol extract of melinjo peel was able to reduce uric acid levels to be in the range of normal values (<5 mg / dL). Bodyweight gain of rats given ethanol extract of melinjo peel dose 450 mg/kg BW had values that were close to the normal group. Melinjo peel extract contained flavonoid, tannins, saponin, and triterpenoid.

Abstract

Lack of confidence in men and women, one of which is when there is dandruff on the hair and scalp. Hair loss is a further symptom of the presence of white scales on the scalp that cause itching. The presence of excessive microorganisms can aggravate this dandruff. Ingredients such as ketocanozole, zinc pyrithione, selenium sulfide and sulfur are compounds commonly used in shampoo formulas. Propolis is an alternative natural ingredient to treat dandruff. The purpose of this study was to determine the minimum growth inhibitory concentration (MIC) of propolis against dandruff-causing bacteria. Dandruff-causing microbes were isolated from dandruff hair and with the well method in a petri dish, the minimum growth inhibitory concentration of propolis was determined. The microbial colonies produced were very diverse with a round shape like grapes, yellowish white in color. The concentration of 6.25% is the MIC value of propolis extract against dandruff microbes. The effectiveness values against ketoconazole and propolis X from the tested propolis extract were 223.52% and 99.59%, respectively. Keywords: hair loss, dandruff, propolis, bacteria , Trigona

Abstract

Penelitian ini bertujuan untuk melihat tingkat toksisitas subkronis ekstrak etil asetat kapang endofit daun sirsak pada tikus Sprague Dawley terhadap glukosa darah, kolesterol, dan trigliserida selama 90 hari. Pemberian ekstrak etil asetat sebanyak 1 mL secara oral pada tikus betina perlakuan. Kelompok kontrol normal tidak diberi ekstrak etil asetat kapang endofit daun sirsak. Dalam penelitian ini digunakan tikus betina sebanyak 40 ekor termasuk kontrol normal. Ekstrak diberikan dengan dosis 20 mg/kg berat badan, 80 mg/kg berat badan, dan 240 mg/kg berat badan. Hasil penelitian diperoleh bahwa konsentrasi glukosa darah tikus betina secara berturut-turut adalah 119,68-152,53 mg/dL, 66,20-89,66 mg/dL, dan 118,95-149,32 mg/dL pada hari ke-30, ke 60 dan ke 90. Konsentrasi kholesterol darah tikus betina secara berturut-turut adalah 52,6-120,0 mg/dL, 94,40-119,0 mg/dL, dan 53,83-79,25 mg/dL pada hari ke-30, ke 60 dan ke 90. Konsentrasi trigliserida darah tikus betina secara berturut-turut adalah 108,50-171,40 mg/dL, 112,20-118,10 mg/dL, dan 96,33-120,70 mg/Dl pada hari ke-30, 60 dan ke 90. Pemberian ekstrak etil asetat selama 90 hari dengan dosis sampai 240 mg/kg BB tidak menyebabkan kelainan nilai gula darah, kolesterol, dan trigliserida tikus Sprague Dawley. Semua parameter pada hari ke-90 tidak menunjukkan perbedaan secara statistika dibandingkan dengan kelompok normal. Ekstrak etil asetat kapang endofit daun sirsak pada penelitian ini tidak menimbulkan efek toksik terhadap konsentrasi glukosa, kolesterol, dan trigliserida selama uji subkronik hari ke-90.

Abstract

Xanthan gum merupakan polisakarida ekstra selular yang dihasilkan dari fermentasi dekstrosa oleh bakteri Xanthomonas campestris. Xanthan gum banyak digunakan dalam berbagai bidang industri sebagai stabilizer, pengental, dan pengemulsi. Dalam industri minyak, xanthan gum digunakan  untuk mengentalkan lumpur pengeboran dan membawa potongan padatan akibat pengeboran kembali ke permukaan. Proses fermentasi dilakukan pada biofermentor dimana substansinya diletakkan pada waktu bersamaan. Berdasarkan pengamatan, diperoleh xanthan gum pada komposisi limbah padat tapioka tertinggi 50 g / L dan urea 5 g / L. Optimasi dengan menggunakan Metode Permukaan Respon (RSM) dengan dua variabel yaitu limbah padat tapioka sebagai sumber karbon dan urea sebagai sumber nitrogen. Berdasarkan hasil perhitungan RSM, dapat disimpulkan bahwa setiap 16 jam respon maksimal mendekati titik pusat (0,0). Model memprediksi produksi xanthan gum optimal pada komposisi konsentrasi limbah padat tapioka sekitar 30 g / L dan urea 5 g / L. Hasil antara nilai prediksi dan nilai sebenarnya menunjukkan model dalam penelitian ini layak dan efektif. Hasil uji reologi menunjukkan karakteristik produk yang baik yang didukung oleh uji FTIR yang menunjukkan spektrum yang sama antara xanthan gum dengan xanthan gum komersial.

Abstract

This study aimed to isolate the endophytic fungi of soursop leaves that have the potential as anti-yeast. Soursop leaves were collected from 3 locations in West Java, Indonesia. The secondary metabolites produced by the endophytic fungi were extracted with ethyl acetate and tested for anti-yeast activity against Saccharomyces cerevisiae and Candida tropicalis. Profile of extracts contents characterization was carried out by using thin-layer chromatography (TLC). Twelve monoculture soursop leaves endophytic fungi isolates from Sukabumi (3 isolates), Cianjur (3 isolates), and Garut (6 isolates). It showed different abilities to inhibit C. tropicalis and S. cereviseae. The best extract is isolat from Cianjur (Sir-C1) and Sukabumi (Sir-S1). The stain patterns seen between the nine endophytic fungi extracts were similar. This indicates that the compounds in them may also be the same. But the extracts have a different absorbance for anti-yeast activities. This indicates that the total metabolites of each isolate are different. TLC results showed that ethyl acetate extraction produced almost the same metabolite components from nine endophytic fungi extracts, and all extracts had an anti-yeast activity with various absorbance. There are differences in the ability of 12 soursop leaf endophytic fungi to inhibit yeast model C. tropicalis and S. cerevisiae. Some extracts have similar TLC profiles, but their anti-yeast activity is not similar

Abstract

Damage to cells caused by stress can be reduced by the presence of antioxidants, one of which is Superoxide dismutase (SOD). The role of the active ingredients of black tea and turmeric will be studied using the in-silico method to identify the active compounds as components in SOD activation. The bioavailability and toxicity of the active compounds of black tea and turmeric were studied and followed by molecular docking and virtual games. The parameters studied are Gibbs's free energy (∆G) and binding site similarity (BSS). The results were analyzed using Gibbs's free energy (∆G) and binding site similarity (BSS) parameters. It was found that those that could increase the activity of Cu/Zn SOD enzymes were Epicatechin gallate (black tea) and curcumin (turmeric), with values ​​of -9.5 and -7.4 Kcal/mol and the same BSS value of 81.8%. The control ligand used was beta amyrin. According to Lipinski's rules, Epicatechin gallate and curcumin compounds can be absorbed well and are safe for consumption. This study concludes that Epicatechin gallate, an active compound of black tea, and curcumin, an active compound of turmeric rhizome, have the best potential to increase the activity of Cu/Zn SOD enzymes based on the results of virtual screening and molecular docking. Epicatechin gallate and curcumin are predicted to be well absorbed by the body because they qualify Lipinski's rules and are not toxic and safe for consumption.

Abstract

Propolis merupakan salah satu bahan alam yang dimanfaatkan oleh masyarakat, antara lain sebagai antibakteri, hepatoprotektor, anestesi lokal, antikanker dan antioksidan. Penelitian ini bertujuan menentukan tingkat toksisitas subkronis ekstrak etanol 70% propolis (EEP) pada tikus Sprague Dawley terhadap glukosa, kolesterol dan trigliserida darah selama 28 hari. Ekstrak propolis diberikan sebanyak 1 ml secara oral pada tikus jantan dan betina sebanyak 60 ekor dan 10 ekor sebagai kontrol. Ekstrak diberikan dalam dosis 0,3 mg/kg bobot badan (BB), 2,1 mg/kg BB, 14,7 mg/kg BB, 102,9 mg/kg BB, dan 720,3 mg/kg BB sebanyak 5 kali dalam seminggu selama 28 hari. Konsentrasi glukosa darah tikus jantan dan betina pada hari ke-28 adalah 79,85 – 97,49 mg/dL dan 61,88 – 90,26 mg/dL. Konsentrasi kolesterol darah tikus jantan dan betina pada hari ke-28 adalah 100,42 – 116,25 mg/dL dan 118,63 – 132,78 mg/dL. Konsentrasi trigliserida darah tikus jantan dan betina pada hari ke-28 adalah 128,64 – 142,37 mg/dL dan 110,95 – 128,58 mg/dL. Pemberian ekstrak etanol 70% propolis sampai dosis 720,3 mg/kg bb selama 28 hari tidak mengakibatkan abnormalitas terhadap nilai glukosa, kolesterol dan trigliserida darah tikus Sprague Dawley.  Semua parameter pada hari ke-28 tidak menunjukkan perbedaan yang signifikan secara statistika dibandingkan dengan kelompok normal.  Ekstrak etanol 70% propolis pada penelitian ini tidak menimbulkan pengaruh toksik pada konsentrasi glukosa, kholesterol dan trigliserida selama pengujian subkronis 28 hari.  Uji ANOVA pada hari ke-28 tidak menunjukkan perbedaan nyata dibandingkan dengan kelompok normal.

Abstract

Tujuan dari penelitian ini adalah menentukan efek farmakologis propolis dan nanopropolis untuk pengobatan penyakit hati pada tikus betina yang diinduksi senyawakarsinogenik 7,12 - dimetilbenz(?)antasena (DMBA). Penelitian dilakukan dengan mengamatihistopatologi dan makroskopik hati pada 28 ekor tikus betina galur Sprague - Dawley. Tikuspercobaan dibagi menjadi 7 kelompok perlakuan dengan 6 kelompok yang diinduksi DMBA(Kelompok I- VI ) dan 1 kelompok sebagai kontrol normal. Kelompok I sebagai kontrolnegatif diberi 1 ml NaCl secara injeksi intraperitoneal (ip). Kelompok II - IV diberinanopropolis 8; 32 dan 56 ppm ip. Kelompok V diberi ekstrak ethanol propolis 233 ppm ip,kelompok VI sebagai kontrol positif diberikan doxorubixin ip dan kelompok VII sebagaikontrol normal diberi penyediaan akuades. DMBA diinduksi selama 11 minggu danpengobatan dilakukan 15 minggu. Setiap minggu tikus ditimbang bobotnya dan diperiksaterhadap inisiasi tumor. Hasil penelitian menunjukkan bahwa ekstrak etanol propolis 233 ppmdan nanopropolis konsentrasi 32 dan 56 ppm dapat mempertahankan kondisi optimal hatitikus. Efeknya adalah setara dengan kontrol normal.
