Abstract

This research aimed at determining of the effects of the induction of paracetamoland kopyor coconut on the activity of superoxide dismutase (SOD) and pathologicanatomy of rats liver. Previous research shows that paracetamol caused negative effectto the body while kopyor coconut has the ability to improve human health, related to itscapacity to prevent oxidative damage of human body. In this research, the positiveeffect of kopyor is approached through measurement of body weight and the activity ofthe total SOD enzyme of animals tested, which are induced by paracetamol and fed withkopyor. Rats administered with temulawak were positive control. The result showedthat paracetamol treatment using toxic dose was found to reduce body weight of testedanimals. Meanwhile, both kopyor and temulawak supplements were able to counter thenegative effect of paracetamol, indicated by the increase in the body weight of testedanimals. Other results showed that treatments with temulawak and 5 times-dose ofkopyor supplement demonstrated a significantly high total SOD activity in the liver ascompared to negative control. Furthermore, pathologic observation of the testedanimals liver showed that 5 time-dose of kopyor treatment was confirmed to improve the recovery of the liver caused by negative effect of paracetamol, better than the negative control.Key words: kopyor coconut, superoxide dismutase, free radical, pathologic

Abstract

Curcuminoids and xanthorizol are active components that may act as anticancer in the rhizome of Curcuma xanthorrhiza. The purpose of this study was to determine the content of the class of compounds in curcuma extract and essential oil using the phytochemical screening test method, the Folin-Ciocalteu method to determine the total phenolic content and the AlCl3 method for the total fentuconoid content. In addition, the activity of curcuma in inhibiting the influence of DMBA on the growth of breast cancer in rats was determined. Curcuma extract used is the result of extraction using 70% ethanol on an industrial scale. Total phenolic content and flavonoid extract of curcuma were determined by spectrophotometer. Inhibition testing of tumors formed due to DMBA induction was carried out by administering temulawak extract at doses of 35, 70, 140, 210 and 280 mg/kg body weight. Based on the phytochemical analysis, the ethanol extract of curcuma contained terpenoids, alkaloids, phenolic compounds and flavonoids. The results of the analysis of the ethanol extract of curcuma contained total phenols and flavonoids with concentrations of 42.96 mg GAE/g and 3.96 mg QE/g, respectively. The group that was given curcuma extract as the control group (p<0.05) had a significant difference. Based on the Mann-Whitney analysis, the third group with a dose of 140 mg/kg BW had the highest effect on tumor inhibition. The hope is that the development of curcuma extract into a standardized herbal medicine so that it can be used for the treatment of breast cancer can use the results of this research.

Abstract

Soursop (Annona muricata L.) is a plant with many health benefits. Soursop leaves are widely used as traditional medicine by Indonesian citizen for wound infection healing. The antibacterial activity of soursop leaves cannot be separated from the presence of endophytic microbes that live inside. Endophytic bacteria live in plant tissues and have beneficial for their host plants. Endophytic microbes are able to produce same secondary metabolites as their host such as producing antibacterial compounds. Thirty one bacterial endophytes were isolated from soursop leaves that from 3 different regions: Garut, Cianjur, and Sukabumi. Morphologically, endophytic bacterial isolates from the three regions had different diversity. Bacterial isolates were tested for their activity against 4 test pathogenic bacteria, such as Pseudomonas aeroginosa, Escherichia coli, Staphylococcus mutans, and Streptococcus aureus. Antibacterial test using disc diffusion method with chloramphenicol 500 m/mL as positive control. Based on the antibacterial activity test, isolate G42 was able to inhibit all types of test bacteria. G42 isolates forms a zone of inhibition up to 17 mm in P. aeroginosa. Based on 16S rRNA, G42 isolates had similarity with Pantoea wallisii up to 93.24%. This study shows that endophytic bacteria isolated from soursop leaves have the potential to be developed into antibacterial agents.

Abstract

Soursop is a plant that is used as traditional medicine. Antioxidants are compounds that are able to delay, slow down, or inhibit oxidation reactions. This study aims to determine the effect of variations in extraction time and the comparison of variations in the amount of simplicia with aquadest solvent on the high pressure extraction method on antioxidant activity. The study made 13 treatments using the Response Surface Methodology (RSM) design with the fit of the Central Composite Design (CCD) model. The results showed that from the 13 treatments, the extraction time had an effect on the percent yield with the equation Y = -51.9692 + 24.2030 X1 + 1.2984 X2. The extraction time (X1) has a value greater than the ratio of simplicia powder to the volume of distilled water (X2) as well as the IC50 value as shown in the equation Y = 171,983 + 13,828 X1 – 4,110 X2. Extraction time (X1) has a greater value when compared to simplicia with aquadest solvent volume (X2). The results of the analysis of the percentage of extract yield and the optimal IC50 value using RSM analysis were thought to have an extraction time of 6.3 minutes and the ratio of simplicia powder to the volume of aquadest solvent was 1:61,2. The results of the antioxidant activity obtained that the optimal IC50 value was estimated at 4.3 minutes of extraction time and the ratio of simplicia powder to the volume of aquadest solvent was 1:78.2.
