Abstract

Leptospirosis is a zoonotic infectious disease caused by the spirochete bacteria Leptospira, with a high risk of transmission in flood-prone areas such as Pontianak, West Kalimantan. Health undergraduate students play a critical role in preventing and managing infectious diseases, including leptospirosis. Hence, sufficient knowledge is necessary. This study aimed to describe the knowledge of health undergraduate students in Pontianak regarding leptospirosis. A descriptive study with a cross-sectional approach was conducted involving 172 respondents selected through proportionate stratified random sampling. Data were collected using an 18-question questionnaire covering definitions, etiology, epidemiology, clinical manifestations, transmission mechanisms and pathogenesis, diagnosis, prevention, and management. The results show that most health undergraduate students at Universitas Tanjungpura, Pontianak had poor knowledge (57.5%), while 35.5% had moderate knowledge, and only 7% demonstrated good knowledge. Electronic media or the internet was the most frequently used source of information (51.74%), followed by lecture materials (44.18%), journals (22.67%), textbooks (14.53%), and printed media (11.04%). This study concludes that the majority of students lack knowledge of leptospirosis. Improving health literacy and access to accurate information, particularly through electronic media, is essential to enhance their understanding of the disease.

Abstract

Irrational use of antibiotics is currently still a major health problem in the world. Worldwide around 50% of medicines are sold, distributed, and given to patients inappropriately. Irrational use of antibiotics can trigger antibiotic resistance. Knowledge and behavior regarding antibiotic stewardship are important in preventing antibiotic resistance. This cross-sectional research was conducted to determine the relationship between knowledge and behavior regarding antibiotic stewardship among healthcare workers. This study's 58 respondents were general practitioners/dentists, specialist doctors/dentists, and pharmacists. The instrument used in this study was a valid and reliable questionnaire. The data analysis used was univariate analysis and Spearman analysis with IBM SPSS Statistics 26 and significance set as p0.05. In this study, the results showed that the majority of respondents worked as specialist doctors/dentists (53.44%), aged in the range 26-35 years (36.20%), of the type female (58.62%), have work experience for more than 10 years (39.65%) and all of the surveyed respondents have good knowledge and behavior (100%). Spearman’s rank analysis showed no significant relationship between knowledge and antibiotic stewardship behavior (p0.05). The high level of expertise of respondents does not significantly affect the level of behavior because it can influence several factors, both internal and external. Further study is needed to identify other factors influencing antibiotic stewardship and rational use of medicine.

Abstract

Extended-spectrum beta-lactamase (ESBL)–producing Escherichia coli is a major cause of multidrug-resistant infections, posing significant challenges to antibiotic therapy. The search for natural antibacterial alternatives has led to growing interest in fermented single bulb Allium sativum (FSBAS), a thermally processed form of garlic with enhanced bioactive properties. This study aimed to evaluate the antibacterial activity of ethanol extracts of FSBAS against ESBL-producing E. coli isolates. FSBAS was produced by fermentation at 70°C for 35 days. FSBAS was extracted using maceration with 96% ethanol. Antibacterial activity was assessed using the disc diffusion method. This experiment consisted of four treatment groups i.e. extract concentrations of 25%, 50%, 75%, and 100%; and two control groups, namely the negative control (1% DMSO) and the positive control (amoxicillin–clavulanate, 30 µg/disc). The ethanol extract of FSBAS showed measurable antibacterial activity against ESBL-producing E. coli. The diameter of the inhibition zone increased proportionally with the extract concentration, with the strongest effect observed at 100%. Statistical analysis confirmed treatment groups with concentrations of 50%, 75%, and 100% showing no significant differences with positive control (p > 0.05). The FSBAS extract shows promising antibacterial potential against multidrug-resistant E. coli.

Abstract

Abstract

Biofilm formation is a virulence factor contributing to the pathogenicity and antibiotic resistance of Klebsiella pneumoniae. This process occurs due to biofilm genes in clinical isolates, especially the mrkA, mrkD, and fimH genes. These three genes are related the quality of biofilms formed, which lead to chronic and prolonged infections. This study was conducted to determine the presence of mrkA, mrkD, and fimH genes as biofilm formers in Klebsiella pneumoniaeclinical isolates. The study used a descriptive in vitro laboratory study, with total of 24 Klebsiella pneumoniae clinical isolates from the collection of the Department of Microbiology, Faculty of Medicine, Universitas Tanjungpura. Isolates were subculture in MacConkey Agar, Gram staining, followed by DNA extraction and gene detection using PCR. The results showed the presence of the mrkA gene in 91.6% of the isolates, while the mrkD and fimH genes were each in 87.5% of the isolates. The high percentage of biofilm gene detected, should increase health workers and stakeholder awarness about the impact.
