Abstract

The research on isolation and characterization of amylothermophilic bacteria has been done in Research Laboratory of Microbiology, Biology Department, Mathematics and Natural Sciences Faculty, Andalas University, Padang. The aimed to obtain amylothermophilic isolate and describe their characteristic. The study used purposive sampling method and descriptive analysis. The study results obtained four amylothermophilic isolates with amylolitic index ranged from 2.23 until 3.51 were different macroscopic characters, rod shape, motile and produce catalase enzyme, three isolates  were  Gram positive and spore forming, one isolate was  Gram negative.

Abstract

The study about “characterization of amilo-thermophilic obligat bacteria from Semurup Hot Spring” has been conducted from November 2013 to March 2014 in Laboratory of Microbiology, Biology Department, Faculty of Mathematic and Natural Science, Andalas University, Padang. The purpose of this study was to find out and analyze isolate amilo-thermophilic obligat bacteria. The result of this study showed that isolate SII-7 from Semurup Hot Spring survived at 70 ºC, that produced amylase with unique macroscopic characters: random, cream and smooth surface. The isolate SII-7 was bacille, Gram positive, has endospore as a product, motility and catalase positive.Keyword: Characterization, thermophilic obligat, bacteria, amylase.

Abstract

Research on the antibacterial potency of fresh extract from leaves of Jamaican cherry (Muntingia calabura L.) in inhibiting the growth of Shigella dysenteriae had been conducted in the Microbiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Andalas University. The study aimed to determine the potential of fresh extract from  Jamaican cherry leaves in inhibiting the growth of S. dysenteriae and to determine its Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) against S. dysenteriae. The results showed that the fresh extract of Jamaican cherry leaves was potent as an antimicrobial agent against pathogenic bacteria S. dysenteriae. It was shown through 12 - 14 mm diameter of inhibition zone which was classified as strong inhibition. The MIC was measured at 3.125% while MBC was undetermined. According to these findings, it can be concluded that the fresh extract from Jamaican cherry leaves was potent to inhibit the growth of S.dysentriae at 3.125% concentration, yet unable to kill it.

Abstract

A study on screening and characterization odf cellulolytic thermophylic bacteria from Sungai Medang hot spring was conducted on May to June 2012 using purposive sampling methods. This study aimed to find a cellulolytic thermophylic bacterium and to analyze its cellulolyties. Twenty eight cellulolytic thermophylic bacteria were isolated from Sungai Medang hot springs. The highest enzymatic activity (amylolytic and proteolytic activity) was shown by isolate MII2.1 from a location at which the temperature was 78oC. This isolate was characterized as an aerobic bacterium, gram negative and non motile. The form of colony was a circular margin, smooth shiny surface and elevation flat.Keywords: amylolytic, bacterium, cellulolytic, proteolytic, thermophylic

Abstract

This study aimed to observe the ability of isolated bacteria from Sungai Medang hot spring in producing amylase and to determine the activity and characteristic of isolated amylolytic bacteria which gave the highest amylolytic index. The water samples were collectied by using purposive sampling based on different level of temperatures. This study has successfully collected 48 bacterial isolated from the hot spring. To find the highest amylolytic activity, we tested ten isolated bacteria which have high amylolytic indices. We found that MV2.7 isolated from 740C of hot spring gave the highest amylolytic activity. The isolated bacteria was positive proteolytic, cellulolytic and negative lipolytic.
