Abstract

Abstract

Hydrilla verticillata is one of Allah SWT creations that potential as an antioxidant. This study’s objectives were to determine antioxidant activity and identify steroid isolates of n-hexane fraction of H. verticillata. The steroid compounds were extracted using methanol solvent. The methanol extract was hydrolyzed with HCl 2 N and partitioned using n-hexane. The n-hexane fraction was tested phytochemically and separated using thin-layer chromatography (TLC). The steroids compounds were identified using UV-Vis spectrophotometer, FTIR, and LC-MS/MS. The result showed that maceration extraction produced 5.14% yield, whereas n-hexane fraction produce 47.95% yield. Steroid separation through analytical TLC revealed that n-hexane: ethyl acetate (4:1) as the best eluant with 12 spots, while separation using preparative TLC yielded 19 spots. Steroid isolate from TLC preparative has antioxidant activity with EC50 of 5.109 ppm. Identification of steroid compounds using UV-Vis produced maximum wavelengths at 203.9 and 276 nm, while using FTIR indicated the presence of O-H group, geminal dimethyl, C=O, C=C, secondary C-OH, and =C-H (alkene) which might contain steroid compounds. The result of LC-MS/MS showed the presence of β-sitosterol. Abstract

Plastic has a fairly wide distribution in human life. The use of microplastics is very difficult to avoid because it has reduced various human activities. The use of plastics on a large and wide scale raises a problem for the environment and ecosytem through the degradation of plastics into microplastics. Microplastics have the potential to pose a threat to the environment and organisms, especially in water areas. Their small size allows microplastics to be ingested by zooplankton up to a higher trophic level. Damas Beach is one of the marine tourism destinations in the Prigi bay area with a potential fishery sector in East Java. The high activity of tourism and fishing causes Damas beach to be potentially polluted by microplastics. This study aims to determine the type of plastic polymer in the digestive tract of lisong tuna obtained in the Damas beach area, Prigi Bay, Treanggalek Regency, East Java. This study used a purposive sampling method with two repetitions. Types of microplastic polymers were analyzed using FTIR. The results of the analysis showed that the types of plastic polymers found in the digestive tract of lisong tuna include High Density Polyethylene (HDPE), Polyvinyl chloride (PVC), nylon (polyamide, Polyethylene Terephthalate (PET), Polypropylene (PP), and Polystyrene (PS).

Abstract

When the Covid19 pandemic occurred, Indonesia was the largest country in Southeast Asia with the most positive confirmed victims and the highest death rate. The consequences of the government's efforts to prevent the government from ordering vaccines reached 450 million doses for 187 million people in order to achieve individual immunity and herd immunity. As a country with the largest Muslim population in the world, the halal status of vaccines is very important. Of the six types of vaccines in Indonesia, namely; only the Sinovac vaccine received halal status, while other vaccines such as AstraZeneca, Pfizer, Moderna, Sinofarm were ultimately still used for emergency reasons due to insufficient halal vaccines. In the future, Muslim countries need to continue to develop alternative technologies and materials to ensure that the vaccines given are halal. Use of cell lines to produce vaccines such as Vero (Monkey), MDCK (Dog), BHK21 (Hamster), HEK (Human), PER (Human). The medium used for cell growth (upstream process) also contains Human Blood (HuS), Horse Blood (HS), Cow Blood (FBS). Likewise, the use of Pig and Animal Trypsin that is not slaughtered in a halal way, as well as the use of stabilizers in vaccine formulations such as Human or bovine (cow) Serum, Porcine or Cow Gelatin, Ethanol are critical points for fatwa institutions such as not passing the halal category. The use of alternative materials in vaccine production such as insect cells, yeast, fungi, non-animal medium and vegetable stabilizers has the opportunity to replace mainstream materials. The independence of halal vaccine production provides an opportunity to increase national resilience.

Abstract

Diabetes is closely related to immune response problems when it occurs chronically. Pegagan (Centella asiatica) is a medicinal plant with active compounds. Madecassoside is beneficial in treating diabetes, and nanoparticle technology is expected to enhance the medicinal potential and availability of pegagan compounds. The aim of this study was to determine the effect of chitosan-coated pegagan nanoparticles on the cytokine profile of chronic diabetic mice, which included CD4+TNF-α+, CD8+TNF-α+, CD4+IFN-γ+, CD8+IFN-γ+ and IL-6+. An experimental study with a randomized complete block design (CRD) consisting of six treatments with seven replicates was conducted. The groups were: healthy mice as negative control; diabetic mice treated with distilled water as positive control and diabetic mice treated with nanoparticle coated with chitosan (NPC) 20 mg/kg, 30 mg/kg, 40 mg/kg, and metformin 130 mg/kgBW. The data were tested using one-way analysis of variance (ANOVA) with a significance level of 5% and continued with the Duncan’s multiple range test. The results showed that pegagan NPC could significantly reduce the relative number of CD4+TNF-α+, CD8+TNF-α+, CD4+IFN-γ+ and CD8+IFN-γ+ and IL-6 in the dose of 20 mg/kg, 30 mg/kg and 40 mg/kg (p<0.05). The treatment dose of 20 mg/kg reduced CD4+TNF-α+, CD8+TNF-α+, CD4+IFN-γ+, CD8+IFN-γ+ to the levels of healthy mice and a dose of 30 mg/kg could reduce IL-6 as in healthy mice. These findings suggest that chitosan-coated pegagan nanoparticles are a promising therapy for diabetes, as they have the potential to modulate the immune response associated with chronic diabetes.

Abstract

Immunomodulators are substances acting as stimulants or suppressants against the immune system, either to specific or non-specific activity. The non-specific through the production of leukocytes as the first response in fighting against an attacking antigen. Meanwhile, the specific, hrough recognition of antigens by lymphocytes during reattack. This research aimed to determine the immunomodulatory activity of the extracts combination nanoparticles of Allium sativum, Curcuma mangga, and Acorus calamus on the leukocyte profile of mice. This study used 5 treatments and 6 repetitions. The treatment groups were K- (untreated group), P1 (extracts combination nanoparticle dose of 25 mg/kg), P2 (extracts combination nanoparticle dose of 50 mg / kg), P3 (subur kandungan herbal medicine dose of 75 mg / kg), and P4 (Clomiphene citrate dose of 0.9 mg / kgBW). The parameters used included the total number of leukocytes and their differential value. The data that met the parametric assumptions, such as normally distributed and homogeneous were examined using the One Way ANOVA test, and when there was a significant difference, it was processed with the Duncan assessment. While those that did not meet the assumptions were evaluated using a non-parametric analysis. The statistical results showed that administration of extracts combination nanoparticles of A. sativum, C. mangga, and A. calamus at doses of 25 mg/kg and 50 mg/kg were able to suppress the inflammatory reaction by decreasing the total number of leukocytes. However, the differential leukocyte count was able to maintained or modulate immune system, indicating by the percentage of neutrophil, basophil, and eosinophil in the normal range. At a dose of 50 mg/kg, decreased the percentage of lymphocytes, while for monocytes, all dosage ranges were able to increase their number.
