Abstract

Carica papaya is known to reduce the quality of spermatozoa in morphology and viability of spermatozoa. Biochemical compound in Carica papaya is unstable and can easily degraded in disgetive track. Formulation of papaya seeds in the form of self-nano emulsion (SNE) is an alternative drug delivery system to increase bioavailability by lipophilic means. The sample consisted of 18 rats which were divided into 3 groups, namely the control group which was not given any treatment; the treatment group which was given papaya seed ethanol extract as much as 100 mg/kgBW and the treatment group which was given papaya seed extract-SNE as much as 100 mg/kgBW, the administration was carried out orally for 28 days. Examination of the quality of rat spermatozoa includes morphology, motility, and viability. Data analysis using One Way ANOVA and Chi-Square test. The results of the analysis showed that the decrease in morphology and viability of spermatozoa was significantly different with a result (p < 0.05) between treatment groups. As for motility, it does not have a significant difference with a result (p > 0.05) between treatment groups.. Based on the results of the study, it can be concluded that the administration of papaya seed ethanol extract and papaya seed SNE extract significantly reduces the quality of spermatozoa in the form of morphology and viability, and does not significantly reduce the motility of Wistar strain white rat spermatozoa.

Abstract

The kipahit plant (Tithonia diversifolia) is commonly used in traditional medicine to treat various diseases, particularly diabetes. Investigating the bioactive compounds of T. diversifolia as α-glucosidase enzyme inhibitors is very promising to be carried out for antidiabetic drug development. A combination of in vitro and in silico studies was conducted to determine the inhibitory interaction of these compounds. In vitro assay was performed using the spectrophotometry method on methanol extracts and revealed that the stems (IC50 = 105.0 ppm) exhibited higher bioactivity than the leaves (IC50 > 500 ppm). Metabolite profiling of the methanol extract of the T. diversifolia stems revealed 94 compounds, which continued for in silico methods (pharmacokinetics and toxicology, followed by molecular docking with flexible-rigid method) for antidiabetic activity and drug-likeness parameters. Among the identified compounds, eight showed promise as drug candidates for inhibiting the α-glucosidase enzyme. The compound 1-(7-hydroxy-2-(hydroxymethyl)-2-methyl-2H-chromen-6-yl)ethanone was the most effective candidate among the eight candidates, based on its high similarity liked positive control, the lowest binding affinity value (−7.739 kcal/mol), and the inhibition constant (97.0 μM). The research findings suggested that the compounds in T. diversifolia had the potential to inhibit the α-glucosidase enzyme and could be developed into antidiabetic drugs.

Abstract

The co-grinding method has been used to produce solid dispersions that increase the solubility of drug substances by utilizing hydrophilic polymers. The purpose of this study was to evaluate the effect of chitosan and variations of sodium alginate as polymers on the dissolution rate of cefixime solid dispersion using the co-grinding technique. The cefixime solid dispersion formulation was made in three variations of sodium alginate formulas, namely 200 mg, 250 mg, and 300 mg. Sample characterization was carried out using XRD, FTIR, SEM, solubility testing, dissolution rate, and flow properties. The results showed that Formula 2 (F2) cefixime solid dispersion was the best formula because the degree of crystallinity decreased to 21.71%, and FTIR analysis showed the functional group interaction. Evaluation of cefixime solid dispersion showed changes in particle morphology. In addition, there was an increase in the transmittance percentage in SIF of 98.587 ± 0.019 and an increase in the dissolution rate of cefixime of 83.61%, an increase in the flow rate of 6.3 ± 0.14 grams/second, an angle of repose of 26.4 ± 0.4◦, a compressibility index of 16.3 ± 0.29%, and a Hausner ratio of 1.19 ± 0.35.

Abstract

ABSTRACT The butterfly pea flower (Clitoria ternatea) contains antioxidants that help combat premature skin aging. Peel-off masks are popular with consumers as topical formulations because they are unique and practical. An essential component of peel-off masks is the gelling agent Polyvinyl Alcohol (PVA), which acts as a film-forming agent. However, PVA at the wrong concentration can result poor mask physical properties. Combining PVA with other gelling agents can obtain peel-off masks with better physical properties. This study aims to identify the effect of PVA and iota carrageenan on the optimal formula of peel-off masks regarding physical properties, stability, and antioxidant activity. Butterfly pea flower extract was obtained by maceration in 70% ethanol. Optimization of PVA and iota carrageenan was conducted using a factorial design method, observing responses such as viscosity, spreadability, and drying time. Peel-off masks with butterfly pea flower extract were prepared with concentration variations of PVA (6%, 10%) and iota carrageenan (0.5%, 1%). The optimal formula was then tested for stability using cycling test method and for antioxidant activity using DPPH method. Increasing concentrations of PVA and iota carrageenan significantly affects viscosity, spreadability, and drying time, with a p-value < 0.05. The optimal peel-off mask formula contained 16% PVA and 0.5% iota carrageenan, with a viscosity of 16.473 cPs, spreadability of 5.6 cm, and drying time of 23 minutes and 11 seconds. The optimal formula demonstrated good stability during storage, with a significance p-value > 0.05, and was categorized as having weak antioxidant activity with an IC50 value 171.738 µg/mL.
