Abstract

Enzim kitinase banyak digunakan dalam bidang medis, makanan, bioteknologi dan lingkungan. Banyaknya kebutuhan enzim kitinase menuntut penyediannya yang murah dan melimpah dengan teknologi produksi yang sederhana. Penelitian ini bertujuan untuk isolasi mikroba kitinolitik dari cairan fermentasi sampah organik, produksi dan uji aktivitas enzim kitinase serta mengetahui karakteristik dari enzim kitinase. Isolasi mikroba telah dilakukan dengan  metode spread plate. Aktivitas kitinase ditentukan secara kualitatif dengan pengukuran indeks kitinolitik dan secara kuantitatif dengan pengukuran absorbansi menggunakan spektrofotometer Uv-Vis pada panjang gelombang 660 nm berdasarkan banyaknya substrat kitin yang dihidrolisis oleh enzim kitinase. Satu dari beberapa isolat yang didapatkan, yaitu isolat A1 menunjukkan aktivitas kitinolitik tertinggi, yaitu sebesar 1,21. Hasil identifikasi mikrobiologi menunjukkan bahwa isolat A1dinyatakan sebagai Pseudomonas pseudomallei. Bakteri ini mampu menghasilkan kitinase secara optimum pada jam ke 18 waktu fermentasi, dengan penambahan molase 0,5% (b/v) dan 1% kitin (b/v) pada media produksinya. Kitinase yang dihasilkan P.  pseudomallei menunjukkan aktivitas optimum pada suhu 50 °C danpH sebesar 6.

Abstract

Natto and Soy Yogurt are fermented food products that are very beneficial for improving health. This community service activity aims to socialize the benefits and make them known to the wider community. However, community service activities during the Covid-19 pandemic cannot be carried out face-to-face and have activities with the community in large numbers. Therefore, this community service activity was carried out through webinar and practical activities. Online seminars was chosen so that the coverage was wider and more people could participate in the pandemic. Face-to-face and community activities are carried out with a limited number of participants, according to health protocols. The level of success, benefit and acceptance of the community from this community service activity is known from the responses of the participants during the activities. Based on the results of the assessment/ response of participants who were present virtually and those present at the location, In general it can be concluded that the PKM activity entitled "socializing the benefits and making natto and soy yogurt through webinars and practices" is going well, but better preparation is still needed.The material presented by the resource person is suitable for the current pandemic conditions, easy to understand, easy to practice and can be developed for home businesses. The Zoom Meeting application can be used as a medium for community service activities in pandemic conditions even though it still cannot reach the wider community.  Keywords: Socialization, natto, soy yogurt, webinar, practical

Abstract

Abstrak            Enzim lipase merupakan bagian dari enzim hidrolase yang bekerja pada ikatan ester. Enzim ini juga mengkatalisis beberapa jenis reaksi sehingga merupakan enzim yang potensial diaplikasikan ke berbagai bidang seperti industri tekstil, kulit, deterjen, makanan, dan lain-lain. Salah satu cara mendapatkan enzim novel ialah dengan pendekatan secara metagenomik dari sampel lingkungan tanpa melalui kultur di dalam laboratorium. Sampel lingkungan yang biasa diteliti ialah memiliki keadaan ekstrem atau memiliki sumber substrat enzim yang ingin dieksplorasi. Pada penelitian ini dilakukan eksplorasi gen enzim lipase pada tanah hasil pengolahan limbah kelapa sawit (POME) melalui pendekatan metagenomik menggunakan desain primer degenerate untuk gen enzim lipase untuk golongan HSL. Didapatkan fragmen gen lipase yang memiliki ukuran 250-300 pasang basa dan dilakukan kloning ke dalam plasmid pET-30a(+) dengan sel inang E. coli TOP10 menghasilkan pustaka fragmen lipase limbah sawit (PFL2S) sebanyak 26 klon.  Kata kunci: Lipase, metagenomik, primer degenerate

Abstract

Introduction: in general, residents of flats are a low-income group of people. Economic improvement for Rusunawa residents with concrete efforts is needed to be able to alleviate poverty. Purpose: the community service program's touch is aimed at initiating entrepreneurship among the residents of the flats through providing training on making a variety of microbial bioproducts with economic value, targeting PKK Rusunawa Penjaringan Sari, Rungkut-Surabaya mothers. Method: Community service activities are carried out in 3 stages, namely (1) survey and coordination, (2) workshops and training on making bioproducts from microbes, and (3) Evaluation of activities. Results: Community service participants who took part in the workshop on microbial bioproducts experienced an increase in knowledge about the meaning of microbial bioproducts, types and manufacturing technology. The number of participants who scored 60 and above increased by 95% in the post-test compared to the pre-test. The workshop was able to build participants' skills in making three types of microbial bioproducts, namely yogurt, biosurfactant and liquid organic fertilizer. Positive responses in the form of sentences were very useful for the workshop material, many were focused on the results of the feedback. Especially for yogurt bioproducts, almost all participants considered it very tasty, not inferior to commercial products already on the market. This shows an indication of a very high level of satisfaction. Conclusions: the understanding of community service participants on microbial bioproduct material increased through the workshop, indicated by an increasing of the participants who scored above of 60 in the post-test by 95% compared to the pre-test. Community service members' feedback through testimonials showed that 100% of the participants responded that it was very useful, indicating that the participants were very satisfied with the activity. The workshop can equip participants with skills in making yogurt bioproducts, biosurfactants and liquid organic fertilizers, which in the future are expected to be able to initiate entrepreneurship for the participants.

Abstract

Estrogen is a hormone that influences the growth of uterus. Ovariectomized rats lose their ovaries as the main source of estrogen so their uterus tends to shrink. The objective of thise study was to evaluate the potency of cocoa as an estrogen substitute, which was done through an experiment that tested the influence of cocoa products on the uterus weight of ovariectomized rats. There were five treatments and in each of the treatments, six wistar rats were given the substance for three days. Four groups of treatment consisted of ovariectomized rats given cocoa extract (875.2 ppm of polyphenols, 1 g/kg of body weight, BW), cocoa powder (369.8 ppm of polyphenols, 1 g/kg BW), olive oil (10 mL/kg BW), or estradiol valerate (1 mg/kg BW). The fifth group consisted of intact (non-ovariectomized) rats given 10 mL/kg BW of water. The finding showed that the ovariectomized rats given olive oil had lower uterus weight than that of intact rats, while the ovariectomized rats given estradiol valerate had higher uterus weight compared to the intact and olive oil groups. The ovariectomized rats given cocoa powder and extract had higher uterus weight compared to those given only olive oil; although they were not significantly different. Correlation between the body weight and uterus weight varied across treatment groups. The rats given olive oil showed significant, positive correlation, while the intact rats showed moderate, positive correlation. The rats given estradiol valerate and cocoa powder showed non-significant correlation. Since the polyphenol content in cocoa powder was at lower concentration than that in the cocoa extract, it was predicted that cocoa polyphenols are more potential in the lower concentration. This study concludes that even though consumption of cocoa powder and extract did not significantly induced uterus growth, cocoa is still considered having estrogenic activity by lowering the correlation between the body weight and uterus weight in ovariectomized rats.

Abstract

Estrogen depletion increases the risk of dyslipidemia by triggering higher levels of total cholesterol (TC), triglyceride (TG), and low density lipoprotein (LDL) besides depressed level of high density lipoprotein (HDL). This study was conducted to investigate the potential of cocoa products to affect serum lipid profile in the estrogen-deficient rats. Thirty adult female wistar rats were divided into five groups i.e. four groups contained ovariectomized rats, and one group consisted of intact rats. The test articles were all dissolved in olive oil and administered orally, comprising of 1 g/kg body weight (BW) cocoa powder, 1 g/kg BW cocoa extract, 1 mg/kg BW estradiol valerate, 10 ml/kg BW olive oil as carrier. Intact group was given drinking water. After three-day administration, the rats were terminated and serum lipid profile was observed. The trial obtained ethical approval from the Animal Care and Use Committee, Veterinary Faculty, Airlangga University (Certificate No. 620-KE). The result showed that carrier group developed higher LDL and lower HDL levels, as well as greater LDL/HDL ratio compared to that of intact group. Estradiol valerate group had significantly elevated TG level. Cocoa powder and cocoa extract groups showed small and non-significant changes in TC, TG and HDL. Surprisingly, consumption of cocoa extract that is rich in polyphenols had resulted highest mean of LDL levels among other groups. It was hypothesized that polyphenol in cocoa extract had affected expression of LDL receptors (LDLR) due to an antagonistic activity against estrogen receptor alpha (ERá). To conclude, neither cocoa powder nor cocoa extract exhibits significant estrogenic effect on the serum lipid profile of estrogen-deficient rats.

Abstract

Modification of electrode using polymelamine (PM) and gold nanoparticles (AuNPs) has been successfully developed via electropolymerization and electrodeposition onto carbon paste electrode (CPE) using cyclic voltammetry (CV) technique. The modified electrode (AuNPs/PM/CPE) was applied as voltammetry sensors in a simultaneous of dopamine (DA) and ascorbic acid (AA). AuNPs/PM/CPE presented an effective surface area 5 times wider than CPE and demonstrated good electrocatalytic performance in the oxidation of DA and AA in 0.1 M phosphate buffer solution (pH 3) with a scan rate of 100 mV s−1. The differential pulse voltammetry (DPV) technique was chosen as the best method for separating potential peaks of DA and AA. The linear response for determining DA and AA using the DPV technique produced a concentration range of 0.1–13 and 0.4–12 µM with coefficient linearity of 0.9999 and 0.9997, the limit of detection of 0.1405 and 0.2187 µM, the accuracy of 89.62–109.16%, and 83.63–105.08%, and the precision of 0.017–0.701% and 0.066–0.626%, respectively. In addition, this electrode was applied in a real sample of infant urine with a concentration of 1 µM by spike method and found 98.86 and 98.28% as percent recovery of DA and AA, respectively.

Abstract

Community empowerment and household waste processing are two issues that need to be resolved by the government. An independent and educated society is an important pillar to support the development of country. Meanwhile, waste that continues to accumulate without processing has a negative impact on health and the environment. Public education about household waste processing can be a solution to the two problems above. Education was carried out at Junior High School YBPK Surabaya and residents of Dukuh Kupang Surabaya, and other participants who joined in a hibrid meeting with a total of 116 participants. Activities at YBPK Surabaya took place on 25 March and 5 August 2023 and activities at Dukuh Kupang Surabaya took place on 27 May and 26 August 2023. The types of waste processed are organic waste and used cooking oil, which are then converted into eco-enzymes and used cooking oil Activities are divided into 2 stages. there are community education activities and program sustainability monitoring activities. The community education activities includes presentation and workshop were hold in March 2023 at YBPK Surabaya Middle School and May 2023 at Dukuh Kupang Surabaya. The evaluation of sustainability program including individual practice and monitoring of activities was carried out in August 2023 at Dukuh Kupang Surabaya. Methods for implementing activities include presentation, practice accompanied by a mentor, individual practice, and monitoring program sustainability. Based on the analysis of participants' ability, the participant scores before and after the activity was increase. It means that this training succeeded in increasing participants' knowledge and skills in processing waste, especially organic waste and used cooking oil waste. A total of 72 out of 116 participants (62%) had processed household waste either individually or in groups. Activity evaluation data shows that these activities are necessary and on target to create an independent and creative society.

Abstract

Pectinase is an enzyme that can hydrolyze substances of pectin. It has several applications in daily life, for example in juice factories, wine factories, pulp and paper factories, coffee and tea factories that use fermentation. Pectinase consists of pectate lyase, pectin lyase, and polygalacturonase. The aim of this research is to partially purify pectinase protein by ammonium sulfate precipitation and dialysis. A preliminary test of pectinase activity used halo zone analysis. The partial purification was needed to optimize ammonium sulfate and get the optimal condition in 40–90% of the ammonium sulfate composition. Pectinase activity was measured using DNS method, and the concentration of protein was measured using Bradford method. The result of this partial purification of Anoxybacillus flavithermus TP-01 was 70%. Decreasing od pectinase activity was resulted in this research due to the loss of cofactor. Based on the result, it can be concluded that the pectinase from Anoxybacillus flavithermus TP-01 was polygalacturonase.

Abstract

Background: Candida albicans grows in the gastrointestinal tract as a normal microflora that may cause intestinal candidiasis, characterized by formation of biofilm and inflammation. Candida is recognized by toll-like receptors (TLRs). This study aimed to explore the relationship between intestinal TLR 2 and TLR 4 expression levels in candidiasis at each phase of inflammation.Methods: An experimental study was performed using a simple randomized sampling on 30 male Wistar rats divided into a control and a treatment group. Each group was inoculated with Candida albicans. Dysbiosis conditions were designed in the treatment group using multiple antibiotics and on day 5 the rats were injected with subcutaneous cortisone acetate. The groups were terminated in five different times (days 7, 14, 21, 28 and 35). On the termination day, intestinal tissue was isolated and the TLR 2 and TLR 4 expression were analyzed by immunohistochemistry. The data were analyzed by parametric test with SPSS (p<0.05) and completed by post-hoc test Least Significance Difference (LSD) to compare pairs of groups.Results: The expression of TLR 2 and TLR 4 between control and treatment groups showed significant differences (p=0.005). In the treatment group, there was a gradual increase in the TLR2 and TLR 4 expressions.  Positive expression of TLR appeared more in the submucosal or basal area than apical surface. The treatment group showed the highest expression of both TLR2 (82.37%) and TLR4 (87.40%) on termination day 35.Conclusions: Inflammation caused by intestinal candidiasis can result in increased expression of intestinal TLR2 and 4 contributing to an increased risk of biofilm formation.
