Abstract

Research was conducted to know the effect of salinity on the reduction of parasites in Gouramy (Osphronemus goramy). The Research was performed using completely Randomized design with 5 salinity treatments (water pond, water with salt concentration 0, 2, 4 dan 6 g/L) with 4 replications. The 5-7 cm of gouramy fingerling were reared as on treatments and the parasites were counted  using skin scrapping dan wet mount methods. Observation was performed every 3 day for 15 days on parasite intensity, prevalence, mortality, symptoms and water quality.The result showed that several parasites including Trichodina sp., Apiosoma sp., Ichthyophthirius sp., Oodinium sp., dan Henneguya sp were observed. The highest intensity and prevalence was found in Trichodina sp. The Salinity and lenght administration were significantly decrease number parasites.  Salinity treatments (2, 4 dan 6 g/L ) for 3 days could totally removing Trichodina sp. from fishes. Meanwhile the data of other parasite species were unsignificant due to the low level of its intensity and prevalence. Salinity administration in level 6 g/L could reduce mortality.

Abstract

Polymerase chain reaction (PCR) is a method that amplifies DNA which have been widely used in molecular biology technique. Based on the PCR, many methods have been developed on isothermal condition and the useful one is loop-mediated isothermal amplification of DNA (LAMP). LAMP reaction employs a Bst DNA polymerase and a set of four specific primers that recognizes a total of six distinct sequences of the target DNA and produces amount of different size of DNA. Many advantages have been achieved in LAMP such as the simple equipment for reaction and observation, short time, highly specific and sensitive procedure. LAMP has been used as a tools for detection many pathogens for human, animals and plants. Some fish pathogens as parasites, bacteria and viruses have been detected by LAMP. The principles and application of LAMP method are discussed in this paper.

Abstract

The purposes of this experiment were to establish loop-mediated isothermal amplification of DNA ( LAMP) method and its application to observe the presence and duration of koi herpes virus ( KHV) in freshwater fishes after infection. The infection was carried out using 4-6 cm length of java barb (Barbodes gonionotus), grass carp (Ctenoparingodon idella), gold fish (komet) (Carassius auratus), tambaqui (Colossoma macropomum) and koi (Cyprinus carpio koi) fishes. Fishes were infected intraperitoneally with KHV inoculums and 2 fishes were sampled everyday. DNA was extracted from gill and used for diagnosis with LAMP assays. The result of this experiment showed LAMP assay can be established and gave 100 times more sensitive than conventional PCR assay. Koi was confirmed as a host of KHV. Java barb, tambaqui, gold fish and grass carp can serve as vector for KHV and the presence of KHV was detected in java barb and tambaqui for 4 days, goldfish and grass carp for 5 days after infection.

Abstract

The purpose of this research was to observe the genetic gain value of 5 months white tilapia breeders of fi fth generation (F5) Singapore strain (ss) Janti compared with fourth generation (F4) and to revealed the differential selection value of 5 months white tilapia breeders of F5 ss Janti from average weight of top 100 compared to average weight of the population. Research was done for 5 months white tilapia (Oreochromis sp.) breeders of F5 ss Janti. The tilapia cultured in running water system for a month. The research performed for 5 months white tilapia breeders of ss Janti through individual selection method (selective breeding). The white tilapia breeders ss strain used in this research consisted of 400 male and 400 female. The selection of the white tilapia was done randomly. Parameter measured included total weight, length, survival rate, feed amount and water quality for culturing. The result showed that the F5 had increased growth weight of each individual with genetic gain value against the F4 was 30.12 % for the male and 27.92 % for female. The genetic gain value was better than several other selective breeding of tilapia before. The top 100 average weight of the 5 months white tilapia Janti breeders of F5 ss strain (184.03 ± 22.87 g to 224.88 ± 24.31 g) was  higher than average weight of the population (131.27 ± 38.98 g to 163.31 ± 47.17 g). The differential selection value of white tilapia breeders was 37.70 g for male and 40.19 g for female. The weight of male tilapia population was less various than female ones.

Abstract

Gouramy (Osphronemusgoramy) is an important fish comodity and preferred to be consumed by people. One of the limiting factor in freshwater aquaculture is bacterial infections. The objective of this research was to identify the species of pathogenic bacteria on gourami from Bantul Regency and to determine their pathogenicity. Eighteen diseased gouramy were collected from seven different fi sh farms. Bacteria were isolated from kidney and purifyon TSA medium. Characterization was conducted on the morphology of bacterial colonies and cells, biochemical tests. Koch Postulate Test was done to confirm pathogenic properties of the isolated bacteria. Pathogenicity test was performed by intraperitoneal injection of a serial doses of bacteria into gouramy and the Lethal Dosage 50 (LD50) was calculated. Twenty isolates of Aeromonas hydrophila, 5 isolates of Aeromonas salmonicida and one isolate of Vibrio cholerae,Vibrio anguillarum and Pseudomonas fluorescens respectively were isolated from and confi rmed as pathogen ingouramy. Pathogenic diversity was found among isolated bacteria, which a group of virulent bacteria as Aeromonas hydrophila GBNB, GGRB, and GCMA3 with LD50 values of 6.28 x 105, 2.09 x 105 and 5.37 x 105 CFU /fish; and a group of non virulent bacteria as A. hydrophila GKRA isolate and Vibrio cholera GPDA isolat with LD50 value of 7.14 x 107 and 3.45 x 107 CFU/fish respectively.

Abstract

African catfish (Clarias sp.) is one of important freshwater fish which prefer consumed by people due to many advantages. Aeromonas sp. bacteria are dangerous patogen for freshwater fishes. This objective of the research was to isolate, identify and determine pathogenicity of Aeromonas sp. bacteria from African catfish from District Ngawi. The samples of catfish with 20 - 25 cm length showing clinical signs disease were obtained from three sub districts. Bacteria were isolated from kidney and inoculated into GSP medium. Characterization and identification through morphology of bacterial colonies, cells and biochemical test. Postulate Koch was conducted to verify abaility to couse disease. Pathogenicity was analyzed by determination of value of Lethal Dosage-50 on catfish on 7 - 9 cm length. The result showed that the disease symptoms of fish infected by the bacteria were skin ulcer, abdominal swelling and kidney damage. Fifteen bacterial isolates were collected which five, six and four isolates were from Kecamatan Karang Jati, Geneng dan Paron.sub-district respectively. The result showed 12 isolates (GKJ1, GKJ3, GKJ4, GGN1, GGN2, GGN3, GGN4, GGN5, GGN6, GPR2, GPR3 and GPR4) were identified as Aeromonas hydrophila. Three isolates (GKJ2, GKJ5 and GPR1) were identified as A. salmonicida. Isolate of A. hydrophila GKJ1, GKJ4, GGN2, GGN5, GPR2 and GPR4 were virulent to African catfish with LD50 values of 1,55 x 105, 3,89 x 105, 7,24 x 105, 2,39 x 105, 6,61 x 104 and 1,95 x 104 cfu/fish.

Abstract

Edwarsiella tarda is a cosmopolitan bacterium and is a cause of Edwardsiellosis in various fish species. The bacterial infection causes large losses on aquaculture in Asia, especially Japan. This study was conducted to isolate and characterize E. tarda as causative disease in freshwater fishes, and to determine its pathogenicity to catfish (Pangasius sp.). Bacteria were isolated from kidney of diseased fishes on Tryptone Soya Agar medium. Identification was conducted based on morphological colonies, morphological cells and biochemical tests. Fulfillment of Koch Postulates was done by injecting bacteria intraperitoneally on 7-9 cm fishes at dose of 107 cfu/fish. Pathogenicity test was carried out by intraperitoneal injection at 104, 105, 106, and 107 cfu/fish to 7-9 cm-catfish (Pangasius sp.) and followed by observation of disease signs and mortality every six hours for 7 days. Pathogenicity was determined as Lethal Dosage (LD50) using Dragstedt Behrens method. In this research we have isolated three isolates E. tarda causing disease in fishes. The clinical signs of this disease were lose of pigmentation over the lession, swollen of stomach, haemorhage on fins , small cutaneous lesions, and necrotic on fins area. The LD50 of E. tarda isolate L2, L3, and N3 were 4.64 ± 0.35x105, 1.54 ± 0.07x105, and 1.13 ± 0.13x106 cfu/fish, respectively.

Abstract

Maggot Black Soldier Fly (BSF) is a nutritionally rich alternative food for fish. The slow digestibility requires the combination with other technologies to produce fish with excellent growth. This study aims to investigate the impact of probiotic application on the growth, microbiome, and gene expression in the intestines of tilapia-fed black soldier fly (BSF) larvae. Tilapia intestines were examined after two months of rearing with three replications and two treatments: A) 30% maggots and 70% commercial pellets with probiotics, and B) 30% maggots and 70% commercial pellets without probiotics. We discovered that probiotics have a significant impact on the gut microbiomes of fish and their absolute growth. The amplicon sequence variant in the probiotic treatment (A) was 25, with dominance by Cetobacterium, Acinetobacter, Enhydrobacter, and Gemmobacter, while the non-probiotic treatment (B) was 8, with dominance by Cetobacterium and Turicibacter. The probiotic treatment increased the expression of Ghrelin, Muc-2, IL-1β, and I-FABP genes, but not the CD36 gene. These findings suggest oral probiotics can help boost tilapia production when fed maggot black soldier fly (Hermetia illucens).
