Abstract

Penelitian ini bertujuan memberdayakan kader PAUD/PKK dalam membangun kebersamaan kelompok melalui tindakan pelatihan karawitan berbasis lagu dolanan tradisional Jawa agar terjadi penguatan nilai-nilai kehidupan pada kader PAUD di Kelurahan Caturtunggal Kecamatan Depok Sleman. Subjek penelitian adalah kader PAUD/PKK sebanyak 35 orang, dengan pilihan metode penelitian tindakan kelompok kader PAUD/PKK (participation action research). Hasil penelitian menunjukkan bahwa tindakan pelatihan karawitan dapat menanamkan nilai-nilai kehidupan seperti kerjasama, disiplin, tanggungjawab, percaya diri, toleransi, saling berbagi pengalaman, saling menolong, dan saling menghargai satu sama lain.

Abstract

 Monyet ekor panjang merupakan jenis primata yang memiliki kemampuan adaptasi yang tinggi untuk bertahan hidup pada berbagai tipe habitat yang berbeda. Kemampuan ini berkaitan dengan bagaimana jenis ini memanfaatkan sumberdaya yang terbatas di habitatnya selama waktu aktif untuk memenuhi kebutuhan hidupnya di alam. Tujuan penelitian ini adalah untuk menentukan pola aktivitas dan penggunaan strata vertikal oleh kelompok monyet ekor panjang yang sudah terhabituasi dengan baik di fasilitas penangkaran semi alami Pulau Tinjil, Propinsi Banten, Indonesia. Kelompok monyet ekor panjang diamati menggunakan metode Scan Sampling dengan interval lima menit antara bulan Maret dan September 2011. Berdasarkan hasil penelitian ini, kelompok monyet ekor panjang menggunakan waktu aktifnya paling banyak untuk berpindah (36,01%) dan beraktivitas paling banyak pada ketinggian 1-5 meter diatas tanah (31,98%).

Abstract

Development of human immunodeficiency virus type 1 (HIV-1) vaccines and anti-retroviral treatmentis currently hindered by the lack of models representing prominent symptoms of HIV-1 infections seen in humans. Simian-human immunodeficiency virus (SHIV) was constructed to resolve the limitations of SIVmac model and has been used in nonhuman primate model ofviral infections, particularly infections by the close relatives of HIV-1. Macaca fascicularis and M. nemestrina are being developed as model HIV/AIDS, by using chimeric virus SHIV produced by replacing the nucleotide structure of cyclophilin A binding region, vif gene and nef of HIV-1 with cyclophilin A binding region, vif gene and nef from SIV. The research aims to study the model of HIV/AIDS on nonhuman primates PBMC in vitro using SHIV. In particular, the study aims to obtain information about the capability of SHIV replication in PBMC of M. fascicularis and M. nemestrina. Results showed a cytopathic effect (CPE) in the form of multinucleated giant cells and expression of p24 protein in PHA-stimulated PBMC cultures of M. fascicularis and M. nemestrina after SHIV infection. The conclusion of this study is that SHIV can infect PBMC M. fascicularis and M. nemestrina in vitro based on CPE and expression of p24 protein.

Abstract

Bovine Herpesvirus (BoHV) is a member of Herpesviridae family that acts as pathogenic virus causing infectious bovine rhinotracheitis (IBR) among cattles, resulting in economic loss for cattle industry. BoHV-1 infection in cows is closely related to abortion, respiratory infection, reduced milk production, infertility, and low birth weight. The aim of this study was to identify and characterize the molecular of BoHV-1 and other virus types, as well as the possible presence of other Herpesviridae family using PCR to amplify DNA terminase gene. Four out of 210 nose swab samples were positive for herpes virus on DNA terminase gene. Further characterization of samples showed 99-100% similarity to BoHv-1 and BoHV-6 sequence. Genetic distance between genera BoHV-1 and BoHV-6 is 0.518 and within genera was 0.001 and 0.044. According to phylogenetic tree analysis of DNA terminase gene, the analyzed sequence clustered into 2 genera, namely Varicellovirus which is identical to BoHV-1 and Macavirus which is identical to BoHV-6. The study provides scientific information on molecular characteristics of Herpesviridae family, especially BoHV-1 which is prevalent in Indonesia with the highest density in the central ranches in Aceh province

Abstract

Bats have been known as natural reservoirs for potential emerging infectious viruses, such as Lyssaviruses, Coronaviruses, Ebola viruses, Nipah virus, and many others. Because of their abudance in population, wide distribution and mobility, bats have a greater risk as source for zoonotic transmission than other animals. Despite the facts of their role as reservoirs for many pathogens, not until an epidemic of Severe Acute Respiratory Coronavirus (SARS-CoV) in 2003 and Middle-East Respiratory Syndrome Coronavirus (MERS-CoV) in 2012, that people pay much attention about coronavirus in bats. SARS-like virus also found in bats with a higher prevalence rate. This study aims to detect the coronavirus of bats in Gorontalo province Indonesia, characterization at the molecular level of the coronavirus genome and determining the level of kinship (through trees filogenetic). This study was conducted as part of bigger PREDICT Indonesia project, in particular to examine coronavirus in bats from Gorontalo province, Indonesia.  As many as  95 rectal swab samples collected from flying foxes (Pteropus alecto) were analyzed in the laboratory using Consensus Polymerase Chain Reaction (PCR) technique to amplify the target sequence from RNA-dependent RNA Polymerase (RdRp) gene with 434 basepair product, resulted 24 samples determined as presumptive positive. Eight out of 24 presumptive positive samples by PCR were analyzed further by nucleotide sequencing and confirmed coronavirus positive. Phylogenetic tree analyses to the eight coronavirus confirmed-sequences were constructed with MEGA-6.0 . The conclusion was 24 out of 95 samples suggested as presumptive positive to Bat CoV. Eight out of 24 samples were analyzed further by nucleotide sequencing and have similarities in the kinship. Three samples had the 98% nucleotide identity to BatCoV from Indonesia and five samples were 85-88% nucleotide identity to BatCoV from Thailand.

Abstract

Hepatitis B virus (HBV) is a DNA virus with liver as primary target organ.This virus caused chronic infection that can progress to cirrhosis, liver cancer and even death. In vitro model system of hepatocyte cultures is important and widely used to study a variety aspects of hepatitis B. Development of small animal Tupaia sp. for the in vitro model system is an alternative to the existinghepatocyte cultures. The specific purpose of the studyis to develop Tupaia javanica hepatocytes culture for HBV replication, and in a broader spectrum to answer the need for in vitro model of hepatocytes. Primary T. javanica hepatocytes (PTH) culture was successfully maintained for 14 days to reach 80% confluence, and infection of Javan gibbon HBV (GiHBV) and orangutan HBV (OuHBV) onto the culture on day 15 showed viral replication for up to eight days as measured by polymerase chain reaction (PCR). PCR quantification indicated that the highest copy number of DNA virus was detected onday two anddecreased until day 8 after infection. Cell receptor for HBV attachment, known as sodium taurocholate cotransporting polypeptide was expressed on the surface of PTH and shown as green luminenscent when observed by immunofluorescence assay. Sequence of partialS gene from the apes HBVs after the viruses have been infected to the PTH showed amino acid identity to their wildtype as high as 99.29% for GiHBV and 95.71% for OuHBV. This study suggested that the PTH can support the replication of GiHBV and  OuHBV.

Abstract

Melioidosis is an emerging zoonotic disease caused by the saprophytic Burkholderia pseudomallei, which infects human and a wide range of animal species. Melioidosis may lead to septicemia and pneumonia in human patients, which can be fatal if the patient is not treated accordingly. The disease is spread in tropical areas and is highly endemic to Southeast Asia and Northern Australia. However, melioidosis is poorly reported in Indonesia, especially in the veterinary field. This research provides serological evidence of antibodies to B. pseudomallei in both captive and wild nonhuman primates. Plasma samples were taken from a total of 390 monkeys in captivities and wild habitats in West Java and Bali, Indonesia. Enzyme-linked immunosorbent assay (ELISA) showed that the seroprevalence was 42.21% for Macaca fascicularis and 43.59% for Macaca nemestrina. Furthermore, the seroprevalence was 53.41% for captive macaques and 17.83% for wild macaques. The findings showed that exposure to B. pseudomallei happened in both captive and wild macaques. Based on this serosurveillance results, further studies such as comprehensive culture and clinical study are required to discover the clinical burden of the disease in nonhuman primates.

Abstract

Aspek perkembangan bahasa merupakan salah satu aspek terpenting dalam tahap perkembangan anak. Kemampuan berbahasa bertujuan agar anak mampu mengungkapkan pikiran melalui bahasa yang sederhana secara tepat, mampu berkomunikasi secara efektif dan membangkitkan minat untuk dapat berbahasa Indonesia. Permainan yang dapat diterapkan untuk memperkaya kosakata anak salah satunya permainan ular tangga yang di desain khusus dan dibuat semenarik mungkin agar anak mudah dalam menambah kosakatanya. Penelitian ini menggunakan penelitian tindakan kelas dengan 19 subyek dari anak-anak TK kelompok B. Permainan ular tangga selain mampu meningkatkan kosakata juga mampu meningkatkan keaktifan anak dalam kegiatan. Kata kunci: peningkatan, kosakata benda, permainan ular tangga

Abstract

Pengelolaan Pendidikan Anak Usia Dini yang sesuai dengan kurikulum dan prinsip pembelajaran bagi anak usia dini diupayakan dapat mengoptimalkan perkembangan anak. Pelaksanaan pendidikan tidak serta merta dijalankan tanpa menganut pandangan tertentu atau hanya mengandalkan kemauan dan semangat mengembangkan PAUD saja. Pendidikan akan memiliki arah, tujuan dan pelaksanaan yang baik jika dari awal telah direncanakan secara matang. Filosofi merupakan salah satu hal yang harus dijadikan pijakan ketika akan menyelenggarakan pendidikan agar arah pelaksanaannya sistematis dan sesuai dengan tuntutan lingkungan. Pelatihan pengelolaan PAUD yang dilaksanakan di padukuhan sekitar UNY dihadiri sekitar 37 peserta kader PAUD. Metode kegiatandilaksanakan dalam bentuk ceramah, diskusi, dan praktek pengemasan pembelajaran. Kegiatan pelatihan dilaksanakan selama 2 hari, yaitu hari pertama pada tanggal 8 Juni 2013 dan hari kedua pada tanggal 22 Juni 2013.Kegiatan pelatihan ini terbagi ke dalam 3 (tiga) sesi. Kata Kunci: Pelatihan, Pengelolaan PAUD, Filosofi PAUD

Abstract

Pelatihan ini bertujuan untuk meningkatkan pengetahuan guru TK tentang kurikulum 2013. Tujuan yang kedua dari pelatihan ini adalah untuk meningkatkan pemahaman dan keterampilan guru TK dalam membuat rancangan Tema, RPPM berbasis budaya.Pelatihan ini dilaksanakan berdasarkan hasil PPM Joko Pamungkas pada tahun 2015 dimana belum semua guru mampu secara mandiri mengembangkan RPP sesuai kurikulum 2013. Tema budaya termasuk masih jarang digunakan dalam tema pembelajaran di PAUD karena dirasa sulit dikembangkan dalam kegiatan pembelajaran  anak usia dini.Metode dalam pelatihan ini berupa ceramah dan workshop. metode ceramah berisi tentang materi budaya, materi pendekatan saintifik dan materi pengembangan RPP kurikulum 2013. Khalayak  sasaran adalah guru-guru TK yang ada di wilayah DIY berjumlah 37 orang.Hasil dari PPM ini antara lain guru dapat mengembangkan RPP berbasis budaya. Tema budaya yang dapat dikembangkan yaitu tema budaya, ramadhan dan rekreasi. Lebih dari 75% peserta sudah memahami pengembangan kurikulum 2013 sesuai dengan materi dalam pelatihan ini. Kata kunci: kurikulum, PAUD, budaya.
