Abstract

Propolis merupakan produk penting bagi lebah yang banyak digunakan sebagai salah satu alternatif pengobatan alami karena mengandung senyawa bioaktif yang dapat memberikan efek positif pada tubuh. Sebagai produk yang dihasilkan lebah untuk menutupi sarangnya dan mencegah predator maupun infeksi bakteri atau virus dari luar, propolis memiliki salah satu manfaat sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak propolis lebah Heterotrigona itama yang berasal dari 4 lokasi budidaya (perkebunan, persawahan, hutan sekunder, dan perkotaan) di Kalimantan Timur terhadap bakteri Propionibacterium acnes atau P.acnes dengan metode difusi lubang/sumuran pada media nutrient broth (NB) dengan kloramfenikol sebagai kontrol positif dan aseton sebagai kontrol negatif. Propolis diekstrak menggunakan metode maserasi dengan larutan etanol 96% yang kemudian dibuat dalam 3 (tiga) konsentrasi yaitu 500ppm, 250 ppm, dan 125 ppm. Hasil penelitian menunjukanfitokimia dari propolis di 4 lokasi budidaya dominan mengandung alkaloid, flavonoid dan kumarin. Uji bakteri pada konsentrasi 500 ppm,propolis H.itama dari lokasi perkebunan dan persawahan tergolong dalam kategori kuat dalam menghambat pertumbuhan bakteri, dari lokasi hutan sekunder tergolong sedang, dan dari lokasi perkotaan tergolong rendah.

Abstract

Propolis is a resinous material that bees gather from specific plant exudate streams and utilize to build hives, caulk or seal crevices, create funnels for bees to enter, store honey and pollen, maintain a warm environment, and function as a natural antibacterial. One type of propolis from the Meliponini Tribe, Propolis Homotrigona Apicalis, is made from resin of the damar tree. This research aims to present scientific data on the effects of propolis as an antibacterial in the human gastrointestinal system. The parameters examined included qualitative phytochemical activity, Porphyromonas sp, Steptococcus sp, and Eschericia sp activity as measured by the minimum inhibitory concentration (MIC) using the agar well method on Mueller Hiton Agar (MHA) media, as well as testing for antibacterial potency and comparing its activity to that of a positive chloramphenicol control. Using the cold maceration technique, 96% EtOH solvent was used to extract propolis over the course of three 24-hour periods. Alkaloids, flavonoids, triterpenoids, and carbohydrates were all active, according to the findings of phytochemical tests. According to the findings of the activity test, Porphyromonas sp, Steptococcus sp, and Eschericia sp all exhibited antibacterial activity. Propolis Homotrigona Apicalis has the potential to be utilized as a treatment that treats diarhea, gum inflammation, and bad breath since it has antibacterial action in these areas as well as a healthy digestive system.

Abstract

Ultisols in Indonesia have the potential for agricultural development, but the soils have low pH and nutrient contents that hinder plant growth and yield. Using animal manure can be an alternative to improve soil productivity and crop yields. This study aimed to examine the effects of animal manure on the chemical properties of Ultisol, yield, and secondary metabolic of Zingiber montanum. The treatments tested were combinations of types of manure (cow and chicken manure) and manure application levels, namely P0 (control), P1 (cow manure 20 t/ha), P2 (cow manure 40 t/ha), P3 (cow manure 60 t/ha), P4 (chicken manure 20 t/ha), P5 (chicken manure 40 t/ha), and P6 (chicken manure 60 t/ha). The results showed that the application of chicken manure of 60 t/ha increased N and P contents of the soil, and the application of cow manure of 60 t/ha increased soil cation exchange capacity. The application of cow manure of 60 t/ha gave the highest plant height, the number of leaves, and the number of at 18 weeks after planting, while the application of chicken manure dose of 60 t/ha produced the longest plant roots. The highest fresh and dry rhizome weight was observed for the 60 t/ha cow manure treatment. The highest secondary metabolic levels in each parameter were found in dry rhizomes (phenolic, flavonoid, and tannin) and fresh rhizomes (phenolic and flavonoid), with the highest tannin compound in the treatment of 40 t chicken manure/ha. The application of chicken manure at a dose of 60 t/ha resulted in a strong antioxidant yield in fresh and dry rhizomes.

Abstract

Green synthesis of Silver nanoparticles (AgNPs) using plant-based materials is an environmentally friendly path for the synthesis of silver nanoparticles instead of traditional chemical methods. This study presents the first biosynthesis of AgNPs using an ethanolic extract obtained from in vitro callus cultures of Myrmecodia tuberosa Jack, an endangered medicinal epiphyte and conservation‑friendly biotechnological platform. Cotyledon explants were successfully induced high-frequency friable, green callus and the ethanolic extract acted as reducing and stabilizing agent. UV‑Vis spectroscopy revealed surface plasmon resonance peaks at 420–450 nm with a visible color change from light brown to dark brown, confirming the AgNP formation. The most intense and well-defined SPR peak indicating optimal nanoparticle synthesis was observed after 6–8 days static incubation at 35°C using 2 mM AgNO₃. In the DPPH radical scavenging assay, it could moderate antioxidant activities, with IC₅₀ = 51.80±0.035 μg mL⁻¹) which was more than that of parent callus extract (IC₅₀ = 58.91±0.097μgmL⁻¹). This improvement is probably a result of synergistic effects between silver core and phytochemical capping layer. These results indicate that callus cultures of M. tuberosa is an environmentally friendly, sustainable and conservation-friendly source of reducing metabolites for the preparation of green nanomaterials applicable in biomedical practices.

Abstract

Bacterial infections remain a major problem for Indonesians. Bacterial resistance caused by inappropriate antibiotic use is a major factor in the prevalence of bacterial infections in the community. Therefore, the use of natural ingredients is needed as an alternative to natural antibiotics. One natural ingredient with antibacterial potential is bee bread, produced by the stingless bee species Heterotrigona itama.  The aim of this study was to determine the antibacterial activity contained in Heterotrigona itama bee bread extract in fighting pathogenic bacteria.  The test used the well method. The growth medium used was Mueller Hinton Agar (MHA) with a 24-hour incubation time. The concentrations used were 500, 250, and 125 µg/well.  The percentage and inhibition zone of Heterotrigona itama bee bread extract varied among bacterial species. Based on the diameter of the inhibition zone at a concentration of 500 µg/well, inhibition of gram-positive bacteria tended to be greater than that of gram-negative bacteria.  Bee bread produced by Heterotrigona itama bees has inhibitory activity against five types of pathogenic bacteria.  The results also provide opportunities for further research, including the formulation and production of applicable products.
