Abstract

The purpose of this study was to find out the diversity index of molluscs (gastropods & bivalves) based on mangrove degradation level in Tobea Island of Muna Regency. The data were collected from May until July 2017, in Wangkolabu municipal area of Tobea sub-district, using observation method and direct observation in the research location to find out the index of ecology of molluscs such as index of diversity (H'), evenness (E), density (N), dominance (D), and dispersion pattern (Id) from both gastropod and bivalve classes based on mangrove degradation level. Direct identification in the field is applied to the type of mangrove vegetation and mollusc organisms that have been known by it’s scientific name. But if not, then the samples are taken and the documentation is then done further analysis at the Laboratory of Biology Unit of Ecology and Taxonomy of FMIPA UHO with reference to the guide book of Introduction of Mangrove in Indonesia and book Snail & Shell Indonesian. Then for water environment parameters in the form of water and substrate samples are further analyzed at Biology Laboratory of UHO Forensic and Biomolecular Unit using Miller triangle fraction method and APHA standard method. Statistical analysis is done using Excel Stat to determine the major components affecting the ecological index of molluscs in degraded mangrove areas. The results showed four species mangroves in Wangkolabu Village are Rhizophora mucronata, Rhizophora apiculata, Bruguiera gymnorrhiza, and Sonneratia alba and that mangrove degradation on the coast of Tobea Island, Wangkolabu village, Tobea sub-district has varied values for good mangrove criteria (Density/RDi = 1.600 individuals/hectare), medium mangrove (Density/RDi = 1.175 individuals/hectare), and mangrove broken/rare (Density/RDi = 375 individuals/hectare). Besides that, Cerithium spp., Nerita costata, Terebralia sulcata, and Clypeomorus sp., type gastropods is able to adapt to mangrove forests that have moderate density (RDi= 1.000–1.500 individuals/hectare), with mangrove substrate types in the form of sandy loam that has soil organic content (BO = 3,18%). While the gastropod type Telescopium telescopium prefers low density mangrove forests (RDi < 1.000 individuals/hectare) with the type of substrate in the form of clay containing organic matter (BO = 2,35%). Based on the results of the study it is expected that the need for mangrove forest management in the study area for the sustainability of aquatic biota, especially molluscs that can be developed as conservation areas by looking at the level of mangrove damage through non-fish aquaculture activities (shellfish) for the welfare of coastal communities and islands small in Southeast Sulawesi.

Abstract

Kata kunci: Vegetasi mangrove, zonasi, struktur, komposisi This research was aimed to describe the structure, dominance of mangrove vegetation, zonation, and natural regeneration at Kaledupa Island of Wakatobi National Park. Sampling of vegetation used line transect and plot methods. Data of mangrove vegetation was obtained through observation of each plot at all growth phases (trees, poles, sapling and seedling) covering species name, number of individual of each species, and size of the breast height diameter of stem. Data of water inundation at high tide was measured at each observation plot. Observation was conducted for 30 days. Dominance of mangrove vegetation was determined by importance value parameter, zonation by relative density parameter and natural regeneration by seedling density. Results of this research found that mangrove community at Kaledupa Island at trees stratum were dominated by species Bruguiera gymnorrhiza, poles stratum was dominated by species Rhizophora mucronata, and at sapling and seedling stratum were dominated by species Ceriops tagal. Mangrove zonation at Kaledupa Island consisted of four zones, that are R. mucronata, R. apiculata, C. taga, and C. decandra zone. Height of seawater inundation is controlling factor of formation of mangrove zonation at Kaledupa Island. Species C. tagal and C. decandra had high level of natural regeneration, while species R.mucronata, R. apiculata, B. gymnorrhiza, Xylocarpus granatum, Sonneratia alba, and Avicennia marina had low natural regeneration. Abstract

Several biotechnology industries are exploring the characteristics of lipase-dependent chaperones due to their distinctive biochemical traits. This study aimed to employ bioinformatics to analyze the molecular structure of Ralstonia picketii BK6's lipase-dependent chaperon (LipRM). The sequence mapping and amino acid distribution were examined using BioEdit (version 7.0.9.1). SignalP 5.0 and Interpro are employed for signal peptide detection, whereas Swiss-Model and VMD 1.9.2 are used for molecular dynamics modelling. The results showed that the Shine-Dalgarno sequence was discovered in the LipRM promoter, seven nucleotides upstream of the initiation codon (AUG) with the 5'-AGGAGA-3', and has a terminator region that facilitates the formation of a secondary structure. The protein's 3D structure prediction results indicate differences in the alpha helix chains (residues 166-174 and 254-271) between LipRM and the reference lipase. LipRM's molecular structure comprises a detachable signal peptide, and with variations in helix alpha chain conformation and ligand geometry.

Abstract

Abstract

Abstract. Hasidu LOAF, Jamili, Kharisma GN, Prasetya A, Maharani, Riska, Rudia LOAP, Ibrahim AF, Mubarak AA, Muhasafaat LO, Anzani L. 2020. Diversity of mollusks (bivalves and gastropods) in degraded mangrove ecosystems of Kolaka District, Southeast Sulawesi, Indonesia. Biodiversitas 21: 5884-5892. Mollusks are one of the mangrove organisms whose classes are bivalves and gastropods. It plays an important role in mangrove and marine ecosystems as filter feeders, predators, and herbivores. This study aims to knows the diversity and abundance of mollusks (bivalves and gastropods) in several mangrove ecosystems in the Kolaka coastline as well as the similarity of these locations. This study was conducted in mangrove ecosystems of Induha Village, Mangolo Village, Tahoa Village, and Towua Village of Kolaka District, Southeast Sulawesi, Indonesia, from July to August 2019. This is a transect method stretched along a 100 m line perpendicularly from the seaward. The size of the mollusks subplot was 1 m2 and placed along the line transect. Each line transect comprises 10 subplots. To analyze the diversity index, evenness index, and its abundance, Kaleida Graph 4.0 version was used. This research indicates that the mollusks consist of 4 families of bivalves with 6 species and 10 families of gastropods with 182 species. It also found out 23 species of mollusks scattered to each location. The molluscan species which spread in all four mangrove ecosystems were Terebralia sulcata, Nerita planospira, and Batillaria multiformis. In Induha, the mollusks species were Anadara notabilis and Drupella margariticola. Meanwhile, Saccostrea cucullata, Pirenella incisa, Clithon oualaniensis, and Clithon pulchellum were only found in Towua. The diversity index of bivalves in each location was categorized as low diversity index category, as well as gastropods were categorized as medium diversity index. The highest diversity index of gastropods was in Induha (H' = 1.96). It was supported by the good mangrove ecosystem for mollusks' habitat. The lowest diversity index of gastropods was in Towua (1.41). This research depicts that three kinds of species with high abundance rate whose rates were >1 ind/m2are located in two different locations namely; P. incisa (3.9 ind/m2) and S. cucullata (3.2 ind/m2) in Towua and followed by B. multiformis (2 ind/m2) and Isognomon ephippium (1.2 ind/m2) in Mangolo.
